The genetic parameters used in National Hanwoo Genetic Evaluation(NHGE) were needed to be monitored and updated periodically for accounting any possible changes in population parameters due to selection and environmental changes. Genetic parameters were estimated with single and two-trait models with MTDFREML package using 2,791 carcass records of steers collected from Hanwoo Progeny Test Program(HPTP). Single and two-trait models gave similar parameter estimates for all traits. The heritability estimates from single and two-trait models for carcass weight(CW), dressing percentage(DP), eye muscle area(EMA), back fat thickness(BFT) and marbling score(MS) were 0.30, 0.30, 0.37, 0.44 and 0.44, respectively. The heritability estimates for all the traits except BFT were slightly lower than those used in NHGE but seemed to be within the acceptable ranges. However, further monitoring is needed because the data might not have fully reflected the changes such as carcass grading standards in performance testing program. In order to shift statistical model of NHGE from single trait model to multiple-trait model, the genetic correlations between carcass traits were estimated with pairwise two-trait models. The genetic correlation coefficients between CW and DP, between CW and EMA, between CW and BFT and between CW and MS were 0.44, 0.63, 0.17 and 0.06, respectively. Those between DP and EMA, between DP and BFT and between DP and MS were 0.29, 0.40 and 0.20. Those between EMA and BFT and between EMA and MS were -0.24 and 0.15, respectively. The genetic correlation coefficient between BFT and MS was 0.03.

The study was carried to identify the impact on nation-wide genetic evaluation and to obtain basic materials for the development of strategies in Swine Improvement Network Project (SINP). Data consisted of pedigree records of 235,511 and performance records of 70,747 for Duroc from 1987 to 2010 were collected by Korea Animal Improvement Association. Performance traits included three point back fat thickness (Shoulder, Belly, Waist), loin area, days to 90 kg and average daily gain. Exchange of genetic resources cross the breeding farms was not high, and furthermore the sizable farms which can accommodate genetic evaluation within the farm were scarce. Three data sets (individual farm evaluation: I, two sub-group evaluation: S, and whole eight farm evaluation: P) were used for genetic analysis. Genetic variances were larger in subordinate farms than in joiners farms for connectedness, and consequently the heritabilities were generally higher in subordinate farms than in joiner farms with I. The standard errors of heritability were small in the order of I, S and P. Estimated average inbreeding coefficients were 1.12%, 0.95% and 1.53% for joiner and subordinate group with S and population with P, respectively. The estimated correlations of breeding values with I and P were lowest. The correlations of breeding values with I and P for traits ranged 0.22 to 0.45 for moved parent animals and 0.24 to 0.72 for all animals. The results in the study suggest that nation-wide evaluation uses more pedigree information and improves accuracy. Furthermore SINP for connectedness could help to improve the accuracy of evaluation.
Germplasms and breeding lines of malting barley at National Yeongnam Agricultural Experiment Station (NYAES) were evaluated for malting barley quality improvement. Among the numerous malting barley quality parameters, the mean of protein content was 12.3% between 10.7% to 14.0% and range of the $\beta$-glucan content was from 3.5% to 5.8%, the mean was 4.6 percent. The length of acrospire, non-germination rate, friability, the speed of filtration, extract yield, kolbach index, diastatic power were significantly different between the individual varieties, however the other traits were not significantly different. The results of correlation analysis among 15 quality parameters showed significant positive correlation between crude protein content and malt protein content. However, other quality parameters such as sugar content, fiablity, extract yield, and kolbach index had negative correlation with crude protein content. Therefore, crude protein content could be one of the major factors that deteriorate quality. The varieties of Viva, Nishino-chikara, Kinukei 9, Kinukei 12, Sacheon 6 and Jinyangbori that showed over the 80% in extract yield and the higher diastatic power, will be used by crossing parents for improve the quality of malting barley.
This study was conducted to develop an SNP set that can be useful for marker-assisted breeding (MAB) in watermelon (Citrullus. lanatus L) using Genotyping-by-sequencing (GBS) analysis of 20 commercial elite watermelon inbreds. The result of GBS showed that 77% of approximately 1.1 billion raw reads were mapped on the watermelon genome with an average mapping region of about 4,000 Kb, which indicated genome coverage of 2.3%. After the filtering process, a total of 2,670 SNPs with an average depth of 31.57 and the PIC (Polymorphic Information Content) value of 0.1~0.38 for 20 elite inbreds were obtained. Among those SNPs, 55 SNPs (5 SNPs per chromosome that are equally distributed on each chromosome) were selected. For the understanding genetic relationship of 20 elite inbreds, PCA (Principal Component Analysis) was carried out with 55 SNPs, which resulted in the classification of inbreds into 4 groups based on PC1 (52%) and PC2 (11%), thus causing differentiation between the inbreds. A similar classification pattern for PCA was observed from hierarchical clustering analysis. The SNP set developed in this study has the potential for application to cultivar identification, F1 seed purity test, and marker-assisted backcross (MABC) not only for 20 elite inbreds but also for diverse resources for watermelon breeding.

Precise, fast, and cost-effective identification of crop cultivars is essential for plant breeder's rights. Traditional methods for identification of persimmon cultivars are based on the evaluation of sets of morphological characteristics. However, it is difficult to distinguish closely related cultivars using only morphological traits. This study was conducted to develop DNA markers for identification of the 32 persimmon cultivars in Korea and Japan. A total of 309 randomly amplified polymorphic DNA (RAPD) markers were identified using 40 different random primers. Various number of polymorphic bands ranged from 4 (OPP-08) to 14 (UBC159) were detected with an average of 7.7. Resulting 57 RAPD fragments were selected, and their sequences were determined for developing sequence characterized amplified region (SCAR) markers. As a result, 15 of 57 RAPD fragments were successfully converted to SCAR markers. Single polymorphic bands of the same size as or smaller than the RAPD fragments were amplified depending on SCAR markers. Among these markers, a combination of eight SCAR markers (PS225_200, PSN05_420, PSF13_523, PSN11_540, PS372_567, PS485_569, PSP08_635, and PS631_735) provided sufficient polymorphisms to identify 32 persimmon cultivars. These newly developed markers will be a fast and reliable tool to identify persimmon cultivars.
The data were collected in the dairy herd improvement program from January 2000 to July 2005. Test data included 825,157 records of first parity and animals with both parents known were included. This study aimed to describe the effect of incomplete lactation records of latest generation to the change in sire's breeding value using Random Regression model (RRM) in genetic evaluation. Estimation of genetic parameter and breeding value for sire used REMLF90 and BLUPF90 program. The phenotypic value on the number of test day records between group TD11, TD8, TD5, TD2 showed no large differences. For all the group heritability of test day milk yield range from 0.30 to 0.36. However TD2 group showed low heritability the least test day recode on the latest generation. The correlation of above 50% between test day and TD11(0.610), TD8(0.616), TD5(0.661) and TD2(0.682) with different records in latest generation. Sire's rank of breeding value varied widely depending on the records on the number of lactation from start to the latest generation. Study showed that change in breeding value ranked if daughter's test recode more so it should have at least 5 test day records. The use of RRM in dairy cattle genetic evaluation would be desirable if complete lactation records for latest generation daughters of young bulls when selection for proven bulls. Random Regression model (RRM) require at least 5 test-day lactation recode.
Heritability and genetic correlation for reproductive traits in Yorkshire pig breed were estimated using Bayesian method via Gibbs sampling. The data set consisted of 9,609 reproductive records at pig breeding farm in Korea. For estimating those parameters using Gibbs sampling, 5,000 cycles of ‘burn-in’ period were discarded among a total of 55,000 samples. Out of the remaining 50,000 samples, 5,000 estimates by each parameter were retained and used for analyses to avoid any correlation among adjacent samples. The reproductive trait considered in this study were total number of born piglets(TNB) and estimated by two different models. The estimated heritability and permanent environmental effect using Gibbs sampler were 0.12±0.020 and 10.9±1.63, respectively. Estimated genetic correlations considered parities as different traits were distributed from 0.99 to -0.13. Such results indicated that reproductive traits for sows should be considered as different traits.
To improve yeast strains for bioethanol production, yeasts with ethanol tolerance, thermotolerance, and ${\beta}$-1,3-glucanase activity were bred using yeast genome shuffling. Saccharomyces cerevisiae $BY4742{\Delta}exg1$/pAInu-exgA, which has extracellular ${\beta}$-1,3-glucanase activity, and the Aspergillus oryzae and S. cerevisiae YKY020 strains, which exhibit ethanol tolerance and thermotolerance, were fused by yeast protoplast fusion. Following cell fusion, four candidate cells (No. 3, 9, 11, and 12 strains) showing thermotolerance at $40^{\circ}C$ were selected, and their ethanol tolerance (7% ethanol concentration) and ${\beta}$-1,3-glucanase activity were subsequently analyzed. All the phenotypes of the two parent cells were simultaneously expressed in one (No. 11) of the four candidate cells, and this strain was called BYK-F11. The BYK-F11 fused cell showed enhanced cell growth, ethanol tolerance, ${\beta}$-1,3-glucanase activity, and ethanol productivity compared with the $BY4742{\Delta}exg1$/pAInu-exgA and YKY020 strains. The results prove that a new yeast strain with different characters and the same mating type can be easily bred by protoplast fusion of yeasts.

A total of 424 plants was regenerated from the seed-culture of a rice cultivar, 'Ilpum'. The regenerated plants were grown in a greenhouse. The 297 plants with high fertility were selected among 424 plants. The harvested seeds from each plant were planted to each line at experiment field in 2008 and 2009. The each line was evaluated for the agronomic and morphological traits, also. The 64 lines (21.5%) showed significant differences in agronomic and morphological traits from donor cultivar 'Ilpum' among 297 lines. The heading date different from donor cultivar 'Ilpum' showed highest frequency in 297 lines, and accounts for 9.1% (29 lines). The phenotype of opaque endosperm and rolling leaf account for 1.7% and 1.3% in 297 lines, respectively. The genetic segregation was observed in dwarf/semi-dwarf, rolling leaf and opaque endosperm at $S_1$ generation, but not in $S_2$ generation. These results suggest that the mutant derived from a tissue-culture will be one of the promising genetic resources, due to its wide variation and high frequency of mutation, comparatively.
The 1,874 rice lines were selected from 3,000 Ds insertional mutant pool by Basta herbicide treatment and were surveyed for trait variation and molecular characteristics of genes knocked out by Ds insertion. Compared with "Donjin", an original japonica cultivar used for transformation, Ds insertion mutant pool showed large variation in major agronomic traits including tiller, panicle, and heading etc. Southern blot analysis demonstrated that these lines on the average had two Ds copies in Donjin genome, resulting in 38.4% of one copy, 32.5% of two copies, 16.7% of three copies, and 11.3% of over four copies. GUS analysis showed that 3.9% of lines (73/1,860) had tissue-specific expression in leaves, nodal parts, floral organs such as stigma and pollen, and roots. Data set obtained from agricultural trait variation and molecular characteristics for individual Ds insertional lines would provide researchers with more information for understanding the function of unknown rice genes controlling economically important traits.
