Comparison of Active Ingredients between Field Grown and In Vitro Cultured Rhizome of Korean Native Ginger (Zingiber officinale Roscoe) (조직배양생강과 한국재래종 생강의 유효성분 비교)

  • This study was carried out to compare and analyze the active ingredients of Korean native ginger and rhizome derived from in vitro shoot-tip culture of Korean native ginger. Proximate compositions, mineral nutrients, free sugars, fatty acids, volatile components, 6-gingerol, and 6-shogaol were analysed and evaluated. Korean native ginger was proved to have a little more contents than in vitro rhizome in proximate compositions (crude ash, crude lipid, crude protein, carbohydrate). Mineral nutrient contents (Cu, Fe, Mn, Zn) of in vitro rhizome were higher than those of Korean native ginger. Among the mineral nutrients, the quantity of K was the highest, followed by P, Mg, Na, and Ca. Free sugar contents (fructose, glucose, sucrose) of in vitro rhizome were higher than those of Korean native ginger. Fatty acids containing less than C14 was the major among the fatty acids in ginger. Citral ingredient of the unique aromatic compound of Korean native ginger was stronger than that of the rhizome derived from in vitro shoot-tip culture. Gingerol concentration was increased by shoot-tip culture.

Effects of Cultural Condition on the Yield and Contents of Effective Components in Alisma orientale(Samuels.) Juzepcz. (택사(澤瀉)의 재배조건(栽培條件)이 수량(收量) 및 유효성분(有效成分)의 함량(含量)에 미치는 영향(影響))

  • This research was carried out to clarify effects of cultural condition on the content of Alisol B-monoacetate, whose function is antihypercolesterol in blood, and yield in Alisma Rhizomes. When the corm part of Alisma Rhizomes was extracted by shaking extraction at $25^{\circ}C$ and reflux extraction at $40^{\circ}C$ to $100^{\circ}C$ four times, the total content of Alisol B-monoacetate was 0. 402%, 0. 425% and 0. 402% at $25^{\circ}C$, $40^{\circ}C$ and $60^{\circ}C$ respectively. The recovery rate was 97% by three times extraction at $25^{\circ}C$, and 96% and 97% by three times extraction at $40^{\circ}C$ and $60^{\circ}C$ respectively. When the corm was harvested on Oct. 30, the content of Alisol B-monoacetate was 0. 46% but it was increased to 0. 71% on Nov. 30. In the case of Oct. 30, the corms of $S(4{\sim}14g\; FW)$ size were determined to contain the highest level of Alisol B-monoacetate (0. 53%), and the content was decreased as the corn size was increased. This tendency was also adopted on Nov. 30. On the other hand, the content of crude protein and starch was changed rarely by the period of harvest. When the planting depth was $0{\sim}1cm$, the yield was the highest as 206kg/10a, and the yield was decreased as the planting depth was deep. The variation of the content of Alisol B-monoacetate was small as 0. 33% to 0. 39% by planting depth. From the above results it could be concluded that the optimum time for harvest of Alisma Rhizomes is Nov. 30 and the optimum planting depth is $0{\sim}1cm$.

Optimization of Extraction Process for Antioxidant from Persimmon Leaf and Thistle Using Response Surface Methodology (반응표면분석법을 이용한 감잎과 엉겅퀴로부터 항산화성분의 추출공정 최적화)

  • In this study, we extracted an antioxidant from natural products which are known to have a high antioxidant content and also optimized the extraction process by applying a response surface methodology (RSM). In addition, by measuring the total flavonoids and total polyphenols of the active ingredient extracted persimmon leaf and thistle, the functionality of the active ingredient was evaluated. Both ultrapure water and alcohol were used as extraction solvents and the ratio of ultrapure and alcohol, amount of sample, extraction time, extraction temperature were set as independence variables. Also, the yield, total polyphenols, and flavonoids was set as the response. Optimal extraction conditions were as follows; for persimmon leaf, the extraction time = 3.1 h, ratio of alcohol/ultrapure = 63.4 vol%, and temperature = $54.6^{\circ}C$ while for thistle the extraction time = 2.9 h, ratio of alcohol/ultrapure = 40.7 vol%, and temperature = $68.4^{\circ}C$. Also, the response were as follows; for persimmon leaf, the yield = 27.7%, total polyphenols = 33.2 mg GAE/g, and total flavonoids = 47.8 mg QE/mg dw, whereas for thistle the yield = 27.0%, total polyphenols = 17.9 mg GAE/g, and total flavonoids = 28.8 mg QE/mg dw at the optimal conditions. The overall satisfaction level was 71.7%.

Anti-diabetic Effects of Banaba Leaf Extracts (Lagerstroemia speciosa Pers.) through Solvents (추출용매 변화에 따른 바나바(Lagerstroemia speciosa Pers.) 잎 추출물의 항당뇨 효과)

  • This study investigated that the antidiabetic effects of banaba extracts with variety solvents selectivity in vitro and in vivo. Banaba extracts were prepared with water, 70% ethanol, 90% ethanol, 100% ethanol and water-ethanol that of extract twice times sequentially water and ethanol. Cell toxicity and insulin secretion of banaba extracts was tested by MTT (3-[4,5-dimetylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay on hamster insulinoma cell line, HIT-T15. Also we tested that insulin, body weight, blood glucose, total cholesterol, HDL-cholesterol, triglyceride and free fatty acid in streptozotocin diabetic rats. Water-ethanol extract has remarkable antidiabetic effect compare with the other banaba extracts. For water-ethanol extract has both of hydrophilic and hydrophobic antidiabetic materials from banaba. Expecially, corosolic acid, as known as unique polyphenol, has antidiabetic effect studied by many researchers till nowadays. But corosolic acid does not solve in water. Otherwise, we suggest that banaba extract of hydrophilic and hydrophobic materials (polyphenol and antioxidants) mixture more increased antidiabetic effects.

Differences of Major Compounds in Valerian fauriei var. dasycarpa HARA and Valerian officinalis L. Grown at Different Places in Korea (광릉쥐오줌풀과 넓은잎쥐오줌풀의 산지별 유효성분의 변화)

  • This study was conducted to know the effect of cultivation area with different sea levels on essential oil content and its composition and content of major active compounds in Valerian fauriei var. dasycarpa HARA and Valerian officinalis L. The total contents of essential oil were not different according to the locality in both valerian, however, its compositions were varied. The major components in V. fauriei were detected bornyl acetate and sesquiterpene alcohol in three cultivation areas. The major compounds were borneol, borney acetate, ${\beta}\;-sesquiohellandrene$ in V. officinalis other. Among the valepotrate components known as major pharaceutically active components in Europe and Nepal valerian, valtrate was detected in a small amount in V. fauriei, but its quantity was not changed according to the cultivation area.

Chemical Component of Mugwort (Artemisia princeps Pampanini) leaves Collected in Korea (수집약쑥의 유효성분 함량변이)

  • Mugwort, Artemisia princeps Pampanini, has been used as natural medicine traditionally in the far east countries including Korea. This study was performed with the object of provoding basic data an leaf quality improvement in mugwort. The 210 mugwort were collected in 48 locations from 2004 to 2006, leaves of 210 genotypes collected were analyzed for eupatilin, jaceosidin and sesamin content. The eupatilin content was averaged to 43.8mg (100 g leaves) and ranged from 0 to 228.0 mg. The average jaceosidin and sesamin contents were 12.8 mg and 3.58 mg and the ranges of those were 0 to 73.8 mg and 0 to 6.56 mg, respectively. The eupatilin content of mugwort harvested in the western region was higher compared with those harvested in the resemble altitude of the southern and eastern of Korea.

Anti-aging Effect of Astradalus membranaceus Root Extract (황기추출물의 항노화 효과)

  • Astradalus membranaceus root extract (ARE) contains saponins, flavonoids, and their glycosides. Two flavonoids, calycosin and formononetin, were predominant constituents with the different amount by diverse extraction conditions. ARE showed the anti-oxidative activities, inhibited tyrosinase activity, and increased type I procollagen expression in proportion to calycosin and formononetin contents. The clinical study also showed that skin wrinkles were significantly reduced in the test group treated with a cream formulation containging 0.1% ARE compared with a group treated with a cream formulation without 0.1% ARE (p < 0.05). These results suggest that ARE may have potential as an anti-aging ingredient in cosmetics.

  • Four heat conduction models were examined for defatted soy-protein curds in order to get the 'intrinsic' thermal conductivity of soy-protein. As the result of examination, the 'intrinsic', thermal conductivities of soy-protein, frozen and unfrozen states, were determined on the basis of series model to be 0.488 W/m.K and 0.300 W/m.K, respectively. By using the 'intrinsic' thermal conductivity values of soybean protein and the series model, the effective thermal conductivity of soybean curds, with and without fat, at frozen and unfrozen states, was predicted satisfactorily, The temperature dependency of the effective thermal conductivity of soybean curd was mostly observed to correlate with the thermal conductivity of water and ice.

Development of Neutron Skyshine Evaluation Method for High Energy Electron Accelerator Using Monte Carlo Code (몬테카를로 코드를 이용한 고에너지 전자가속기의 중성자 skyshine 평가방법 개발)

  • The skyshine effect is an essential and important phenomenon in the shielding design of the high energy accelerator. In this study, a new estimation method of neutron skyshine was proposed and was verified by comparison with existing methods. The effective dose of secondary neutrons and photons at the locations that was far away from high-energy electron accelerator was calculated using FLUKA and PHITS Monte Carlo code. The transport paths of secondary radiations to reach a long distance were classified as skyshine, direct, groundshine and multiple-shine. The contribution of each classified component to the total effective dose was evaluated. The neutrons produced from the thick copper target irradiated by 10 GeV electron beam was applied as a source term of this transport. In order to evaluate a groundshine effect, the composition of soil on the PAL-XFEL site was considered. At a relatively short distance less than 50 m from the accelerator tunnel, the direct and groundshine components mostly contributed to the total effective dose. The skyshine component was important at a long distance. The evaluated dose of neutron skyshine agreed better with the results using Rindi's formula, which was based on the experimental results at high energy electron accelerator. That also agreed with the estimated dose using the simple evaluation code, SHINE3, within about 20%. The total effective dose, including all components, was 10 times larger than the estimated doses using other methods for this comparison. The influence of multiple-shine path in this evaluation of the estimation method was investigated to be bigger than one of pure skyshine path.
