Development and Application of SITES (부지환경종합관리시스템 개발과 적용)

  • SITES(Site Information and Total Environmental Data Management System) has been developed for the purpose of systematically managing site characteristics and environmental data produced during the pre-operational, operational, and post-closure phases of a radioactive waste disposal facility. SITES is an integration system, which consists of 4 modules, to be available for maintenance of site characteristics data, for safety assessment, and for site/environment monitoring; site environmental data management module(SECURE), integrated safety assessment module(SAINT), site/environment monitoring module(SUDAL) and geological information module for geological data management(SITES-GIS). Each module has its database with the functions of browsing, storing, and reporting data and information. Data from SECURE and SUDAL are interconnected to be utilized as inputs to SAINT. SAINT has the functions that multi-user can access simultaneously via client-server system, and the safety assessment results can be managed with its embedded Quality Assurance feature. Comparison between assessment results and environmental monitoring data can be made and visualized in SUDAL and SITES-GIS. Also, SUDAL is designed that the periodic monitoring data and information could be opened to the public via internet homepage. SITES has applied to the Wolsong low- and intermediate-level radioactive waste disposal center in Korea, and is expected to enhance the function of site/environment monitoring in other nuclear-related facilities and also in industrial facilities handling hazardous materials.

Biodegradation of Biphenyl by Sphingbium yanoikuyae BK-10 (Biphenyl의 Sphingobium yanoikuyae BK-10에 의한 분해 특성)

  • Bacterium capable of using biphenyl as a sole source of carbon and energy were isolated from soil, and based on the results of 16S rDNA sequence, strain BK10 identified as a Sphingobium yanoiktiyae. The optimum cultural conditions were as follows; $NH_4NO_3$ 1g, $K_2HPO_4$ 1g, $MgSO_4{\cdot}7H_2O$ 0.5g, $CaCO_3$ 0.2 g per 1 liter of distilled water. The Sphingobium yanoikuyae BK10 strain was completely utilized biphenyl in mineral salt media containing biphenyl at concentration 500 $\mu$g/ml of biphenyl as a sole carbon and energy source within 48 hours. Optimumal pH and temperature for biphenyl degradation and cell growth of strains were 6.0$\sim$8.0 and 20$\sim$50$^{\circ}C$, respectively. Especially, at 30$^{\circ}C$, cell-growth were higher than other temperature. Cell grown on biphenyl has been shown to have a higher removal rate for biphenyl than grown on sucrose. This study shows that Sphingobium yanoikuyae BK10 strain had a high biodegradation capability of biphenyl and can be simulate a candidate compounds the bioremediation of PCBs (Polychlorinated biphenyl) contaminant soil and water.

Effectiveness of an Environment-friendly Fumigator for Microorganisms that Can Cause Biological Damage to Records in Japan, Malaysia, and Oman (일본, 말레이시아, 오만의 기록물에서 분리한 유해미생물에 대한 친환경 기록물 소독장비 효과검증)

  • Archives have historical and academic values. That is why the study of preservation conditions and the environment for the effective management of various records progress. However, materials such as paper and adhesives will inevitably cause biological damages. These damages spread into the inner side of archives, which would make it impossible to recover. For example, archival records from Malaysia and Oman are easily exposed to biological damage because of these countries' hot and humid climate. As such, once records are damaged, disinfection operations are customary in these areas. Methyl bromide (MeBr) and ethylene oxide (EtO) were used in the disinfection process as they showed good insecticidal effect. However their antibacterial effect are negligible. As such, the need for an environmental-friendly fumigator was evident. An environmental-friendly fumigator is designed to improve storage stability. Such equipment is expected to contribute to prevention of damages to cultural heritages through its use of vaporization technology, maintenance of a disinfection space (chamber), and self-generation of nitrogen.

Analysis on the Legal Control Levels and GHS Classification Information Status for Strongly Acidic Hazardous Materials (강산성 유해화학물질의 법적관리 수준 및 GHS 분류정보 제공 실태분석 연구)

  • Objective: This study inspected incident cases, legal control levels, and GHS(Globally Harmonized System of Classification and Labeling of Chemicals) classification results of strong acids such as hydrogen fluoride, hydrogen chloride, nitric acid, and sulfuric acid, which have been responsible for many recent chemical accidents. As a result, it is deemed necessary for legal control levels of these strong acids to be revised and GHS classification be managed nation-wide. Methods: This study inspected incident cases and legal control levels for strong acids such as hydrogen fluoride, hydrogen chloride, nitric acid, and sulfuric acid. The study analyzed and compared chemical information status and GHS classification results. Results: There were 76 domestic incidents involving strongly acidic hazardous materials over the five years between 2007 and 2011. They include 37 leakage incidents(46.7%) within a workplace, 30 leakage incidents(39.5%) during transportation, and nine leakage incidents(13.8%) following an explosion. The strongly acidic materials in question are defined and controlled as toxic chemicals according to the classes of Substances Requiring Preparation for Accidents, Managed Hazardous Substance, Hazardous Chemical(corrosive) as set forth under the Enforcement Decree of the Toxic Chemicals Control Act and Rules on Occupational Safety and Health Standards of Occupational Safety and Health Act. Among them, nitric acid is solely controlled as a class 6 hazardous material, oxidizing liquid, under the Hazardous Chemicals Control Act. The classification results of the EU ECHA(European Chemicals Agency) CLP(Commission Regulation(EC) No. 790/2009 of 10 August 2009, for the purposes of its adaptation to technical and scientific progress, Regulation(EC) No 1272/2008 of the European Parliament and of the Council on classification, labeling and packaging of substances and mixtures) and NIER (National Institute of Environmental Research) are almost identical for the three chemicals, with the exception of sulfuric acid. Much of the classification information of NITE (National Institute of Technology and Evaluation) and KOSHA(Korea Occupational Safety and Health Agency, KOSHA) is the same. NIER provides 12(41.4%) out of 29 classifications, as does KOSHA.

Isolation and Characterization of a Bacteriocin-Producing Lactobacillus sakei B16 from Kimchi (김치에서 박테리오신을 생산하는 Lactobacillus sakei B16의 분리 및 특성 분석)

  • Lactic acid bacteria (LAB) are able to secrete antimicrobial peptides called bacteriocins, which inhibit other bacteria such as pathogenic microorganisms. Therefore, bacteriocin-producing starters can be used as natural biopreservatives for various foods. The objective of this study was to screen and characterize bacteriocin-producing LAB from Kimchi and to investigate their applicability as a starter in Kimchi fermentation. To screen bacteriocin-producing LAB, gram-positive and gram-negative bacteria were used as indicators. To measure the antimicrobial activities of isolates, agar well diffusion assay method was used. According to the results, bacteriocin produced by $Lb.$ $sakei$ B16 showed antimicrobial activity against $Listeria$ $monocytogenes$ ATCC 19115, $Escherichia$ $coli$ KCTC 1467, and$Lactobacillus$ $plantarum$ KTCT 3104. Furthermore, bacteriocin was very stable after treatment with high temperature and high and low pH, but its effects were inhibited by treatment with proteolytic enzymes such as trypsin, proteinase K, and ${\alpha}$-chymotrypsin, revealing their bacteriocin-like protein- based structure. These results suggest that $Lb.$ $sakei$ B16 and its bacteriocin are good candidates as a functional probiotic and natural biopreservative, respectively, in fermented foods.

A Study on the Introduction of Green IT Based on the Cases of Implementing Green Internet Data Center (그린 데이터센터 구축 사례에 기반한 그린 IT 도입 방안에 관한 연구)

  • As global climate changes, the interest in environmental crisis is increasing and a number of international agreements and regulations against this crisis are being established. Global information technology(IT) corporations are building their own pro-environmental green IT strategies to cope with the regulatory measures. Green IT broadly refers to pro-environmental technologies designed to replace hazardous materials, maximize energy effectivity, and find alternative energies. In the current stage of the IT industry development, Green IT specifically refers to the technologies that deal with the server heat generation and the energy reduction in data center. This study defines the concept of Green IT and reviews its origin and necessity. Then, it examines the issues regarding Green IT industry in Korea as well as other countries and compares the Green IT strategies developed in each country. Reviewing the recent development of IT and data center market enables us to see that overall Green IT strategies focus on the establishment of Green Internet Data Centers. Therefore, this study analyzes the cases in which some domestic and foreign corporations introduced Green Data Centers in order to examine the protocol and legal requirements for building Green IT, the aspects of environmental evaluation and design, and specific strategies for launching Green IT strategies and its future assignments. The conclusions of this study are as follows. First, to introduce Green Data Center as a strategy to build Green IT, the government and corporations should cooperate with each other. Partial introduction at the initial stage is desirable because, through the process, mutual trust between the two parties can be built more smoothly. Second, CEO's determination to build Green IT and continue its operation is indispensable. CEO's are required to have clear understanding as to why Green IT needs to be built and how it should be constructed. Those who initiate the construction of Green Data Center for Green IT need to know the definition and necessity of Green IT while at the same time understanding the implicit meanings of Green IT. They also need to be aware of future-oriented values of Green Data Center and readjust their corporate business activities in the pro-environmental direction. Finally, not only the CEOs' pro-environmental activities but also the change of mind on the part of all corporate employees is required to realize Green IT. It should be remembered that pro-environmental Green IT starts with minor activities.

  • Aquatic ecosystems are receiving various harmful effects due to anthropogenic chemical pollutions. To protect wildlife, risk assessments of the chemicals are conducted using reference indexes of toxicity estimated by species-level laboratory tests and/or micro-/mesocosm community-level studies. However, the existing micro-/mesocosm communities are structurally too complicated, and it is also difficult to compare the experimental results directly with those from species-level tests. Here, we developed a procedure of a simple bi-trophic microcosm experiment which contains the common species (a green algae, Pseudokirchneriella subcapitata and a cladoceran, Daphnia magna) for testing chemical toxicities. For the proper operation of bitrophic microcosm experiment, the minimum required concentration of primary producer (P. subcapitata) is $5{\times}10^5cells\;mL^{-1}$. The microcosm system showed higher stability when the initially introduced D. magna population was composed of neonates (<24-h old) than adults and those mixture. This simple microcosm system would be an applicable tool to estimate the disturbing impacts of pollutants on plant-herbivore interactions, and linking the species- and population-/community level risk assessments in the future studies.

Stress Evaluation to Heavy Metal Exposure using Molecular Marker in Chironomus riparius (분자지표 유전자 발현을 통한 Chironomus riparius 중금속 노출 스트레스 평가)

  • Heavy metals are common pollutants in the freshwater environment and have toxicological effect in habitat organisms. The heavy metals highly accumulated in sediment and organism, and observed various physiological responses. In this study, we investigated the molecular response to heavy metal toxicity (Al, Aluminum; Cr, Chromium; Cu, copper; Mn, Manganese; Zn, Zinc) through expression of heat shock protein 40, 70, 90 (HSP40, 70, 90), cytochrome 450 (CYP450), Glutathione S-transferase (GST) and Serine-type endopeptidase (SP). HSPs showed up-regulation in Cu and Zn exposures. Furthermore, HSPs expression in treated groups tended to be higher than the control group. The tendency of CYP450 and GST mRNA expression was higher for Cr and Cu than for other exposure group. The expression of SP gene was low at Al exposure and other group were measured to be similar to control. These results suggest that heavy metal toxicity in freshwater ecosystem may affect physiological and molecular process. Also, the comprehensive gene expression in the aquatic midge Chironomus riparius give useful information to potential molecular biomarkers for assessing heavy metal toxicity.

Proteome in Toxicological Assessment of Endocrine Disrupting Chemicals (프로테오믹스를 이용한 내분비계 교란물질 환경독성 연구)

  • It is important to understand the potential human health implications of exposure to environmental chemicals that may act as hormonally active agents. It is necessary to have an understanding of how pharmaceutical and personal care products and other chemicals affect the ecosystem of our planet as well as human health. Endocrine disruption is defined as the ability of a chemical contaminating the workplace or the environment to interfere with homeostasis, development, reproduction, and/or behavior in a living organism or it's offspring. Certain classes of environmentally persistent chemicals such as polychlorinated biphenyls (PCBs), dioxins, furans, and some pesticides can adversely effect the endocrine systems of aquatic life and terrestrial wildlife. Research continues to support the theory of endocrine disruption. However, endocrine disruption researches have been applied to proteomics poorly. Proteomics can be defined as the systematic analysis of proteins for their identity, quantity and function. It could increase the predictability of early drug development and identify non-invasive biomarkers of tonicity or efficacy. Proteome analysis is most commonly accomplished by the combination of two-dimensional gel electrophoresis (2D/E) and MALDI-TOF mass spectrometry (MS) sr protein chip array and SELDI-TOF MS. Proteomics have an opportunity to play an important role in resolving the question of what role endocrine disruptors play in initiating human disease. Proteomics can also play an imfortant role in the evaluation of the risk assessment and use of risk management and risk communication tools required to address public health concerns related to notions of endocrine disruptors. Understanding the need for the proteomics and possessing knowledge of the developing biomakers used to abbess endocrine activity potential will he essential components relevant to the topic of endocrine disruptors.

A Survey of Consumer's Consumption Characteristics of Environmental-Friendly Agricultural Products (EFAP) -Focused on Consumers in Discount Stores- (친환경농산물 소비행태 조사 -대형할인마트 고객을 중심으로-)

  • We investigated the consumption inclinations of consumers for environmentally-friendly agricultural products (EFAP) for better production and distribution of EFAP. Consumer inclinations in large supermarkets from April 7th to 30th, 2008 were surveyed. Data were analyzed using the SPSSWIN 12.0 program. Results showed that 90.4% of consumers recognized EFAP and 76.4% of those who recognized the products had actually bought the products. Members of the "have purchased group" bought EFAP once a week because they believed these products were safe from hazards, such as pesticide. The amount spent on EFAP was less than 20% of the total food budget and the major consumed items were green vegetables and fruits. The sale location was mainly large supermarkets and agricultural cooperation outlets. Although many people were satisfied with EFAP from the viewpoint of nutritional and sensory qualities and safety compared with general agricultural products, they were notsatisfied with the price. Many consumers trusted that EFAP were safe and nutritional. Members of the "not purchased group" also trusted the nutritional and sensory qualities of EFAP and recognized the safety issue, but they were not satisfied with the price, compared with general agricultural products. Consequently, to succeed in the market, EFAP should maintain the confidence of the consumer while decreasing in price.
