Evaluation of Biohydrogen Production Using Various Inoculum Sources (다양한 접종원을 이용한 바이오수소 생산 평가)

  • In this study we evaluated biohydrogen production potential as operational parameters (substrate, salt concentration, and temperature) using eight inoculum sources. While the volumetric biohydrogen production rate was significantly affected by temperature and inoculum sources, substrate and salt concentration did not have a significant effect on the biohydrogen production. Mesophilic temperature (37℃) was also found more appropriate for the hydrogen production than thermophilic temperature (50℃). Rate, while the eight inoculum sources, anaerobic digestion sludge exhibited the fastest biohydrogen production. The maximum production rate from anaerobic digestion sludge was 2,729 and 1,385 ml-H2·l-1·d-1 at mesophilic and thermophilic temperature, respectively.

The Effects of Feeding Feed Additives Containing Microorganisms on Characteristics of Excreta in Growing Pigs (육성돈에 미생물제제 급여시 분뇨 특성에 미치는 효과 연구)

  • The effects of microbial feedstuff additives on feed conversion rate and physical and chemical characteristics of excreta in growing pigs were investigated. Three different products (A, B and C) were compared. Microbial population tests showed B contained higher numbers of total bacteria, Lactobacillus spp. and yeasts. The amylase activity of B was also higher than that of A and C. The daily feed intake rates fer control, A, B and C were 2.06, 2.13, 2.17 and 2.34 kg, respectively. Pigs feed product C had the highest liveweight gain(2.89 kg). However, the results of feed conversion rate were not significantly different between treatments. Amount of faces excreted for control, A, B and C was 1.18, 1,19, 1.23 and 1.32 kg, respectively. Urine volume for control, A, B, and C was 1.91, 1.80, 2.19 and 2.31 kg respectively. Moisture content, T-N, $P_2O_5$ and $K_2O$ in pig manure were not significantly different between treatments. The range of BOD values was 63,453 to $73,758mg/\ell$ for faeces, and 5,678 to $7,428mg/\ell$, for urine. SS values of solid and liquid excreta ranged from 142,200 to 176,000 and from 710 to $1,025mg/\ell$, respectively.

Inhibitory Effects of Natural Additives on Pathogenic Microorganisms Growth during Storage of Commercial Chicken (닭고기 저장 동안 천연첨가물에 의한 병원성 미생물 억제 효과)

  • The purpose of this study was to examine the antimicrobial effects of natural additives on pathogenic microorganisms in commercial chicken during storage. Chicken skin ($20cm^2$) inoculated with Salmonella Typhimurium or Pseudomonas aeruginosa was treated with 2% natural additives, including chitosan, epigallocatechin gallate (EGCG) and garlic, during storage at $4^{\circ}C$ for 12 days. A combination of the three natural additives significantly inhibited growth of S. Typhimurium compared to treatment with chitosan, EGCG or garlic alone (p<0.05). This combination also inhibited growth of P. aeruginosa compared to treatment with one or two of the natural additives (p<0.05). These results suggest that combined chitosan, EGCG, and garlic may be used as an antimicrobial agent in commercial chicken during refrigerated storage.

Characteristics of Autographa californica Nuclear Polyhedrosis Virus in Spodoptera exigua Cell Line. (파밤나방 세포주에서 Autographa californica 핵다각체병 바이러스의 감염 특성)

  • To study the usefulness of Se301 cells, which is originated from Spodoptera exigua and has susceptibility to the Autographa californica NPV (AcNPV), as a host for the AcNPV-based expression vector system, we compared the characteristics of AcNPV in Se301 and Sf-21 cells. The symptom by viral infection was similar in both of cells, but the ratio of polyhedra released from the cell was higher in Se301 cells than in Sf-21 cells. The overall PIB productivity of AcNPV was similar in both cells but the size of polyhedra was larger in Se301 cells. While the polyhedrin expression efficiency was about 2.4 times higher in Se301 cells than in Sf-21 cells, the viral growth was higher in Sf-21 cells. These results suggested that Se301 cell is very useful in the AcNPV-based expression system as a host.

Increase of Epigallocatechin in Green Tea Extract by Lactic Acid Bacteria Fermentation (젖산균 발효를 통한 녹차 추출물의 Epigallocatechin 함량의 증대)

  • Hydrolytic enzyme activities, including those of ${\beta}$-glucosidase, ${\beta}$-glucuronidase, ${\beta}$-xylosidase, ${\beta}$-galactosidase, ${\beta}$-arabinofuranosidase, ${\beta}$-arabinosidase, and ${\beta}$-arabinopyranosidase, which are useful for bioconversion, were explored in lactic acid bacteria isolated from Korean traditional fermented foods. Nine bacterial strains were selected for the fermentation of green tea extract prepared by supercritical fluid extraction. Changes in the concentrations of catechin, epicatechin, epicatechin gallate, epigallocatechin, and epigallocatechin-3-gallate in green tea extract were investigated after fermentation by the selected lactic acid bacteria strains. The strain Leuconostoc mesenteroides MBE1424, which showed the highest ${\beta}$-glucuronidase enzyme activity among the tested bacterial strains, increased the epigallocatechin content of the green tea extract by 60%. In addition, L. mesenteroides MBE1424 was more resistant than the control strain at high temperature and showed a maximum specific growth rate at $40^{\circ}C$. L. mesenteroides MBE1424 was presumed to have an enzyme system containing ${\beta}$-glucuronidase with utility in the bioconversion of green tea extract.

Effects Of the Inoculation Of Candida rugosa CY-10 OH the Reducing Odours in Pig Slurry Medium (Candida rugosa CY-10의 접종에 따른 돈분배양액내 악취저감 효과에 관한 연구)

  • This study was carried out to isolate and identify the yeasts from the the composts, which were effective to reduce odor of the pig feces, and to investigate their physiological properties. In yeasts, one of 30 isolates was obtained on 10% pig feces extract medium. Judging from the morphological and biochemical characteristics, the CY-10 isolated from the compost were identified as Candida rugosa. This isolated strain showed the deodorizing activity by reducing the concentration of $NH_3$ and R-$NH_2$ than that of the control. The CY-10 had completely utilizing butyric acid and iso-butyric acid including 10% pig slurry of the volatile fatty acids, which are the specific malodorous agents of pig feces. Compared to control, this yeast was found effective for decrease in $NH_4$-N, Soluble-N and BOD, 20%, 12.6%, and 9.82% respectively.

Applications of Agro-Based Materials for Water Dropwort (Oenanthe stolonifera DC) Organic Farming (미나리 유기재배를 위한 활용자재 시용효과)

  • Organic farming is a type of agricultural practices based on naturally occurring processes excluding or strictly limiting the use of synthetic fertilizers, pesticides, and other chemicals. This study was conducted to investigate the influences of agro-based materials, effective microorganisms (EM), liquid silicate (LS), and organic liquid fertilizer (OLF) for water dropwort (Oenanthe stolonifera DC.) cultivation. Soil pH, soil organic matter, and plant available phosphorous decreased with LS application. Exchangeable Ca and Mg decreased with EM application, and electrical conductivity and exchangeable Ca and K decreased with OLF application. Most of essential nutrient contents in water dropwort were reduced with the treatments of LS, EM, and OLF as compared with those in control plot, except nitrogen and phosphorus. However, diseases and insect pests were almost not observed in the water dropwort in the agro-based material application plots, except cluster caterpillar (Spodoptera litura). Productivity of water dropwort tended to be reduced: its higher productivity in the OLF and EM+LS plots and lower in the LS and control plots.

Natural Purification Treatment using Soil Brick with Combined Effective Microorganisms and Emergent Plants (복합유용미생물 및 수생식물을 활착시킨 흙블록을 이용한 자연정화 처리방법 연구)

  • In this study, using soil brick with combined effective microorganisms and emergent plants was identified which it can increase the effect of conservation and improvement of water. Lab-test was consist of four kind of reactors and each of reactors were A(rawwater), B(soil brick), C(emergent plant) and D(soil brick+emergent plant). Iris pseudoacorus, Phargmites australis, Typha angustifolia and Zizania latifolia were used for emergent plant. Evaluation of application on various environment were performed on agricultural waterway and pond. The pH measurement test of soil brick was performed due to evaluate whether a strong alkaline water flows out of the soil brick. Result of lab-test, removal efficiency of D was better than removal efficiency of A presenting 20.9%, 27.9% 21.5%, 33.8% and 58.4% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. Removal efficiency of soil brick on agricultural waterway was revealed to be 49.5%, 45.0%, 43.7%, 37.3% and 28.6% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. And removal efficiency of soil brick on the pond was revealed to be 12.7%, 10.5%, 9.32%, 10.4% and 36.3% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. Result of pH measurement test of soil brick was neutral which was about 6 to 8.

Inhibition of HIV-1 Replication in CD4+ Peripheral Blood Lymphocytes by Intracellular Expression of RNA Aptamer (RNA aptamer 발현을 통한 CD4+ peripheral blood lymphocytes에서의 인간 면역결핍 바이러스의 증식 억제)

  • We have previously demonstrated that intracellular expression of an RNA aptamer termed RRE40, which was selected in vitro to bind HIV Rev 10-fold much tighter than wild-type RRE, efficiently protected human CD4+ T cell line, CEM, from HIV-1. In this study, to evaluate the efficacy of the RRE40 RNA in clinical settings, polyclonal CD4+ peripheral blood lymphocytes (PBLs) were transduced with retroviral vectors expressing RRE40 decoy RNA and then challenged with clinical isolates of HIV-1. In contrast to the control cells transduced with vectors expressing control tRNA, intracellular expression of RRE40 RNA more effectively inhibited HIV-1 replication in CD4+ PBLs. However, transient and diminished inhibition, rather than complete inhibition, of HIV-1 replication in PBLs expressing RRE40 decoys have been observed. These results suggest that RRE40 decoy RNA would be useful to inhibit HIV-1 replication in cells. However, development of more efficient gene transfer protocols and/or more effective decoy RNAs would be needed to apply RNA decoy to modulate HIV-1 patient.

Mannitol Production by Aureobasidium pullulans (Aureobasidium pullulans에 의한 Mannitol의 생산)

  • Aureobasidium pullulans produced high concentration of polyols extracellularly in the media of sucrose, glucose and mannose as sole carbon source. Mannitol was the main polyol produced during the late exponential and stationary phases of growth together with small quantities of glycerol. Sucrose and glucose were rather rapidly metabolized to mannitol among carbon sources examined where the initial glucose concentration showed no difference in the amount of mannitol. In contrast 20%(w/v) of sucrose was the most appropriate concentration tested. However, the yield of mannitol based on substrate used($Y_{p/s}$) was independent on the initial concentration, and the mean value of mannitol yield in 10% glucose and sucrose media was 0.144 and 0.188, respectively. Mannitol production was reduced in response to an elevated water stress imposed by salts within the range from 0.25 to IM of NaCl or KCl as stress solutes. However, glycerol contents and its ratio to mannitol were increased at the conditions of high salinity. Based on the results, extracellular mannitol produced by A. pullulans probably resulted partly from osmoregulation(in case of glycerol) and mainly from, as known to occur in most of fungi, enzymatic reduction of the corresponding hexoses through phosphate pathway.
