Analysis of Bacterials Community Structure in Leadchate-Contaminated Groundwater using Denaturing Gradient Gel Electrophoresis (Denaturing Gradient Gel Electrophoresis를 이용한 매립지 침출수로 오염된 지하수의 세균 군집 분석)

  • This research has been performed to clarify the relationship between hydrogeochemistry and bacterial community structure in groundwater contaminated with landfill leachate. We collected and analyzed samples from 5 sites such as leachate (KSG1-12), treated leachate (KSG1-16), two contaminated groundwaters (KSG1-07 and KSG1-08) and non-contaminated groundwater (KSG1-13). pH was 8.83, 8.04, 6.87, 6.87 and 6.53 in order; redox potential (Eh) 108, 202, 47, 200 and 154 mV; electric conductivity (EC) 3710, 894, 1223, 559 and 169.9 $\mu$S/cm; suspended solids (SS) 86.45, 13.74, 4.18, 0.24 and 11.91 mg/L. In KSG01-12, the ion concentrations were higher especially in $Cl^-$ and $HCO_3^-$ than other sites. The concentrations of Fe, Mn and $SO_4^{2-}$ were higher In KSG1-07 than in KSG1-08, and vise versa in $NO_3^{2-}$. In the comparison of DGGE fingerprint patterns, the similarity was highest between KSG1-13 and KSG1-16 (57.2%), probably due to common properties like low or none contaminant concentrations. Otherwise KSG1-08 showed lowest similarities with KSG1-13 (25.8%) and KSG1-12 (27.6%), maybe because of the degree of contamination. The most dominant bacterial species in each site were involved in $\alpha$-Proteobacteria (55.6%) in KSG1-12, $\gamma$-Proteobacteria (50.0%) in KSG1-16, $\beta$-Proteobacteria (66.7%) in KSG1-07, $\gamma$-Proteobacteria (54.5%) in KSG1-08 and $\beta$-Proteobacteria (36.4%) in KSG1-13. These results indicate that the microbial community structure might be changed according to the flow of leachate in grounderwater, implying changes in concentrations of pollutants, available electron accepters and/or other environmental conditions.

Analysis of the Concentration of Metal Elements in Freshwater Fish Otolith Using LA-ICP/MS (LA-ICP/MS를 이용한 담수 어류 이석 내 금속 원소 농도 분석)

  • In this study, the possibility of a follow-up study on environmental pollution in domestic freshwater was identified by analyzing fish otoliths using Laser ablation-inductively coupled mass spectrometry (LA-ICP/MS). Fish otolith are known to be affected by the environment in which fish live. As a result, research on this subject is active in many countries; however, this is not the case in Korea. Therefore, in this study, the possibility of tracing environmental pollution using fish otoliths was identified by analyzing the components of metal elements used as indicators for environmental pollution. For the component analysis of metallic elements LA-ICP/MS, which can shorten analysis time by reducing the pretreatment process, was used. Sampling was conducted by dividing the research and the background area and carp, a freshwater species, was selected as the experimental fish species subject. Based on the established LA-ICP/MS conditions, the concentration of the metallic elements in the fish otoliths collected in the research area was 2202.9 mg kg-1, 2.03 times higher than the 1,086.3 mg kg-1 in the background area. All elements except for Li and U, were found to be higher in the research area than in the background area. Compared with the sediment measuring net analysis data, the distribution tendency of Zn, Pb, and Cu in sediment metal element concentrations in the two regions and distribution of metal element concentration in fish otoliths were similarly shown. These results confirm that fish otoliths can be used to track environmental pollutants, such as in sediments.

Analysis of Sensitivity to Prediction of Particulate Matters and Related Meteorological Fields Using the WRF-Chem Model during Asian Dust Episode Days (황사 발생 기간 동안 WRF-Chem 모델을 이용한 미세먼지 예측과 관련 기상장에 대한 민감도 분석)

  • The purpose of this study was to analyze the sensitivity of meteorological fields and the variation of concentration of particulate matters (PMs) due to aerosol schemes and dust options within the WRF-Chem model to estimate Asian dusts affected on 29 May 2008 in the Korean peninsula. The anthropogenic emissions within the model were adopted by the $0.5^{\circ}{\pm}0.5^{\circ}$ RETRO of the global emissions, and the photolysis option was by Fast-J photolysis. Also, three scenarios such as the RADM2 chemical mechanism and MADE/SORGAM aerosol, the MOSAIC 8 section aerosol, and the GOCART dust erosion were simulated for calculating Asian dust emissions. As a result, the scenario of the RADM2 chemical mechanism & MADE/SORGAM aerosol depicted higher concentration than the others' in both Asian dusts and the background concentration of PMs. By comparing of the daily mean of PM10 measured at each air quality monitoring site in Seoul with the scenario results, the correlation coefficient was 0.67, and the root mean square error was $44{\mu}gm^{-3}$. In addition, the air temperature, the wind speed, the planetary boundary layer height, and the outgoing long-wave radiation were simulated under conditions of no chemical option with these three scenarios within the WRF or WRF-Chem model. Both the spatial distributions of the PBL height and the wind speed of u component among the meteorological factors were similar to those of the Asia dusts in range of 1,800-3,000 m and $2-16ms^{-1}$, respectively. And, it was shown that both scenarios of the RADM2 chemical mechanism and MADE/SORGAM aerosol and the GOCART dust erosion were interacted on-line between meteorological factors and Asian dusts or aerosols within the model because the outgoing long-wave radiation was changed to lower than the others.

Distribution Characteristics of Heavy Metals for Tidal Flat Sediments in the Saemankeum Area (새만금 갯벌의 중금속 분포 특성)

  • This study was conducted to evaluate distribution and behavior characteristics of heavy metals for tidal flat sediments in the Saemankeum area. The value of heavy metal contents in the tidal flat sediments were higher than that surveyed in 1994, but showing that it's lower when compared with the value of Shiwha lake. Enrichment factors in the tidal flat sediments showed below 1 except for Pb and Zn. Metal excesses was appeared that Cu, Cr, Fe, Mn lacked and Pb, Zn were high. The correlation analysis between heavy metals and organic matters was found high positive relationship (r=0.424$\~$0.839), especially correlation coefficient between ignition loss and oxide Al, Fe, Mn showed high positive relationship above 0.7. The correlation analysis between Brain size and hear metals was found that as grain size was small, heavy metals contents were increased. Correlation coefficient between silt and heavy metals except for Cu, Pb showed high positive relationship as 0.552$\~$0.732.

Trace Metals in Surface Sediments of Garolim Bay, Korea (가로림만 표층 퇴적물 내 미량금속 분포 특성)

  • In 2010 and 2015, total 77 surface sediment samples were collected to assess the anthropogenic effects of trace metals in surface sediments of Garolim Bay, and the physical characteristics (particle size and specific surface area) and geochemical components (major (Al, Ca, Fe, K, Ba) and trace metals (Mn, Cs, Cr, Co, Ni, Cu, Zn, Pb), organic carbon and calcium carbonate) were analyzed. Mean grain size of Garolim Bay surface sediments ranged from 0.51-5.58 Ø (mean 3.98 Ø) and increased from the inlet of bay to the inner bay, and from the waterway to the land. Most of the metal concentrations except for some elements showed the similar distribution to those of mean grain size and specific surface area. As the particle size decreased and the specific surface area increased, the metal concentration increased. In order to estimate the factors controlling the concentration of trace metals, factor analysis was performed, and three factors were extracted (92.7% of the total variation). Factor 1 accounted for 71.3% of the total variation, which was a grain size factor. Factor 2 accounted for 14.2% of the total variation, Factor 3 accounted for 7.2% of the total variance. Enrichment factor was calculated using the particle size corrected background concentration. Metals with a enrichment factor of 1.5 or higher and the number of samples were 4 for Cr (St. 1, 16, 27, 39) and 1 for Pb (St. 39), but there were little differences in the concentrations of 1M HCl leached metals for these metals. The percentage of 1M HCl leached fraction to total metal concentration decreased in the order of Pb~Co>Cu>Zn~Mn>Ni>Cr. Comparing this value with contaminated and clean sediments in other coastal areas, the percentages for each metal were similar regardless of the trace metal levels in all regions. This fact might be resulted from the reaction between the 1M HCl solution and the different sediment constituents, indicating that there is a limit to apply this percentage of leached metal to the estimation of the contamination extent.

Characterization of Concentrations of Fine Particulate Matter in the Atmosphere of Pohang Area (포항지역 대기 중 초미세먼지(PM$_{2.5}$)의 오염특성평가)

  • The purposes of this study are to investigate the concentration levels of fine particles, so called PM$_{2.5}$, to identify the affecting sources, and to estimate quantitatively the source contributions of PM$_{2.5}$. Ambient air sampling was seasonally carried out at two sites in Pohang(a residential and an industrial area) during the period of March to December 2003. PM$_{2.5}$ samples were collected by high volume air samplers with a PM$_{10}$ Inlet and an impactor for particle size segregation, and then determined by gravimetric method. The chemical species associated with PM$_{2.5}$ were analyzed by inductively coupled plasma spectrophotometery(ICP) and ion chromatography(IC). The results showed that the most significant season for PM$_{2.5}$ mass concentrations appeared to be spring, followed by winter, fall, and summer. The annual mean concentrations of PM$_{2.5}$ were 36.6 $\mu$g/m$^3$ in the industrial and 30.6 $\mu$g/m$^3$ in the residential area, respectively. The major components associated with PM$_{2.5}$ were the secondary aerosols such as nitrates and sulfates, which were respectively 4.2 and 8.6 $\mu$g/m$^3$ in the industrial area and 3.7 and 6.9 $\mu$g/m$^3$ in the residential area. The concentrations of chemical component in relation to natural emission sources such as Al, Ca, Mg, K were generally higher at both sampling sites than other sources. However, the concentrations of Fe, Mn, Cr in the industrial area were higher than those in the residential area. Based on the principal component analysis and stepwise multiple linear regression analysis for both areas, it was found that soil/road dust and secondary aerosols are the most significant factors affecting the variations of PM$_{2.5}$ in the ambient air of Pohang. The source apportionments of PM$_{2.5}$ were conducted by chemical mass balance(CMB) modeling. The contributions of PM$_{2.5}$ emission sources were estimated using the CMB8.0 receptor model, resulting that soil/road dust was the major contributor to PM$_{2.5}$, followed by secondary aerosols, vehicle emissions, marine aerosols, metallurgy industry. Finally, the application and its limitations of chemical mass balance modeling for PM$_{2.5}$ was discussed.

A Numerical Study of Smoke Movement in Atrium Fires with Ceiling Hea Flux (천장에 열 유속을 갖는 대형 공간에서 화재 발생시 연기거동에 대한 수치해석적 연구)

  • This paper describes the smoke filling process of a fire field model based on a self-developed SMEP(Smoke Movement Estimating Program) code to the simulation of fire induced flows in the two types of atrium space containing a ceiling heat flux. The SMEP using PISO algorithm solves conservation equations for mass, momentum, energy and species, together with those for the modified k- epsilon turbulence model with buoyancy term. Compressibility is assumed and the perfect gas law is used. The results of the calculated upper-layer average temperature and smoke layer interface height has shown reasonable agreement compared with the zone models. The zone models used are the CFAST developed at the Building and Fire Research Laboratory NIST U.S.A. and the NBTC one-room of FIRECALC developed at CSIRO, Australia. The smoke layer interface heights that are important in fire safety were not as sensitive as the smoke layer temperature to the nature of ceiling heat flux condition.

The Effect of Stream Anion and River-Bed Materialson Aquatic Insects (계류수의 음이온과 하상재료가 수서곤충에 미치는 영향)

  • This study was carried out to obtain basic data on the kinds of aquatic insects and their living conditions in the mountain stream. The investigation was done in Bongmyung stream. Experimental Forest, Kangwon National University on aquatic insects, anions and river-bed materials. The results are as follows. 1. At every plot surveyed, diversity index, richness index and evenness index of aquatic insects appeared higher at upper stream than at lower stream in erosion control dam. 2. Anion concentrations were almost the same in plots A, B, C, D and E, but plot F at the lower stream showed 1.5 to 89 times higher concentration than the others. 3. In river-bed materials analysed, particle diameter was bigger at the upper stream than at the lower stream. At the down stream of erosion control dam showed high pebble composition ratio. 4. The number of aquatic insects showed the negative relation with the anion concentration and the positive one with the size of river-bed materials. Especially, they were affected much by the distribution chart of boulder.

Tolerance of Juvenile gobiidae, Tridentiger trigonocephalus Exposed to Various Salinity (염분농도에 따른 두줄망둑, Tridentiger trigonocephalus 치어의 내성)

  • The juvenile gobiidae, Tridentiger trigonocephatus were reared for 40 days under the salinity ranges between 0 and 33.6$\textperthousand$ to examine the effects of salinity on the survival, growth and oxygen consumption rates of the fish. Survival rates were significantly declined for the fish reared under 3.4$\textperthousand$, and daily growth rates were also reduced below 6.7$\textperthousand$ Body length and weight of gobiids reared below 6.7$\textperthousand$ were smaller than those reared above 13.4$\textperthousand$. Oxygen consumption rates in the salinity conditions $\leq$ 10.1$\textperthousand$ were significantly reduced with decreasing salinity. This study reveled that low salinity reduced survival, growth and oxygen consumption rates of the juvenile gobiids suggesting potential influence on the natural mortality of Tridentiger trigonocephalus in the estuarine areas where experience the extreme salinity fluctuations in Korean waters.

Leaching Characteristics on Clay Ground induced by Artesian Pressure (피압에 의한 점토 지반의 용탈 특성)

  • This paper performed consolidation tests on soft ground with and without artesian pressure conditions to find out characteristics of leaching effects using two types of one-dimensional column equipment(height : 1,100mm, outer diameter : 250mm). Artesian pressure of 5.5kPa was applied to the bottom of soft ground inside column equipment. Distribution of salinity and shear strength with soil depth were measured after the consolidation test. From the results, it was found that distribution of undrained shear strength and salt concentrations were similar at the top of clay ground irrespective of artesian pressure condition. However, at the bottom of clay ground, the values of undrained shear strength and salt concentration under artesian pressure were lower than those without artesian pressure. This result indicates that structure of soft soil with artesian pressure was weakened by salt leaching. Electronic resistance results showed that void ratio under artesian pressure condition was more reduced than that without artesian pressure condition.
