Protective Effects of HEAC and its Primary Active Compound on HCl-ethanol Induced Gastritis in Rats (흰쥐에서 염산-에탄올로 유발된 위염에 대한 HEAC 추출물과 그 주성분의 억제효과)

  • The purpose of this study is to evaluate the gastric mucosal damage effect by Hericium erinaceus cultibated with Artemisia capillaris (HEAC) with 0.15 M HCl in ethanol in rats. Among them, 80% ethanol extract showed potent inhibitory effect (67.7%) on gastritis. Using solvent fractionation, 80% ethanol extract of HEAC was separated into five fractions with $n$-hexane, methylene chloride, ethyl acetate, $n$-butanol and water. As a result, fractions obtained with methylene chloride exhibited strong inhibitory effects on gastritis. With this result, the methylene chloride extract was analyzed by GC/MS, HPLC, IR and NMR($^1H$, $^{13}C$) to identify primary active components and the main component in the extract was identified as ethyl linoleate. The effective does ($ED_{50}$) values of 80% ethanol extract of HEAC and ethyl linoleate showed 22.6 and 6.4 mg/kg, respectively and these values were higher than those of stillen (44.2 mg/kg) and selbex (46.5 mg/kg). Therefore, the administration of 80% ethanol extract of HEAC and ethyl linoleate have a strong protective effect agasint the gastritis induced by HCl-ethanol and may be promising new drug for treating gastritis and gastric ulcer.

Protective effects of red ginseng according to steaming time on HCl/ethanol-induced acute gastritis (염산/에탄올로 유도된 급성 위염 동물모델에서 증숙시간에 따른 홍삼의 보호 효과)

  • The objective of the present study was to evaluate the protective effect of red ginseng (RG) according to steaming time on 150 mM HCl/60 % ethanol induced gastric ulcer models in mice. The sample was divided into 3 groups-G (dried ginseng), RG 4 (steamed 4 h and dried ginseng), RG 6 (steamed 6 h and dried ginseng), and determined through in vitro experiments, such as 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid radical scavenging activity, HPLC analysis, total polyphenol, and flavonoid contents. In vitro experiment results were depended on steaming hours. Based on the results, we chose two samples (G, RG 6) and conducted in vivo experiments. Mice were divided into 5 groups: Nor (normal group), Con (acute gastritis mice treated with distilled water), G (gastris induced by HCl/Ethanol treated with 100 mg/kg G), RG 6 (gastris induced by HCl/ethanol treated with 100 mg/kg RG 6), and SC (gastris induced by HCl/Ethanol treated with 10 mg/kg sucralfate). In our results revealed that RG 6 suppressed elevated reactive oxygen species, and inflammatory related makers, such as cyclooxygenase-2, inducible nitric oxide synthase, tumor necrosis factor alpha, and interleukin-1 beta. In addition, gastric lesion area was improved. These results suggest that RG 6 protects the stomach by attenuating oxidative stress and inflammatory response under gastric ulcer conditions. Therefore, RG 6 should be a potential therapeutic agent for the treatment of acute gastric ulcer.

  • 호산구는 염증 반응에 의해 활성화되며, 주로 기생충 감염이나 알러지 질환 등에 대한 면역 작용을 담당한다고 알려져 있다. 호산구 증가증은 약물 반응, 알러지, 국소적인 기생충 감염 등에 의한 경우가 많지만, 자가면역성 질환이나 종양에 의한 경우도 있다. 최근 연구를 통해 위염의 대표적인 원인균 중 하나인 유문나선균 역시 위점막에서 나타나는 조직 호산구 증가증의 원인이 될 수 있다고 밝혀지고 있으나, 유문나선균에 의한 호산구 증가증 발생 기전이나 빈도는 아직까지 확립되지 않고 있다. 위점막 내 호산구 침윤과 동반되는 위염은 복통, 오심, 구토, 설사, 장폐색 등을 일으킬 뿐만 아니라, 아토피성 피부염, 천식, 위식도 역류, 염증성 장질환 등의 발생과 관련이 있다고 보고되고 있다. 위염 및 다양한 관련 질환에 의한 증상은 공중 근무자들의 임무 수행 능력을 저하시켜 항공기 사고를 발생시킬 수 있는 가능성을 가지고 있다는 점에서 항공의학적으로 매우 중요하며, 실제로 호산구성 위염이나 유문나선균 감염의 치료 여부가 공중 근무자에게 일시적 또는 영구적 비행임무정지를 부과할 수 있는 기준이 되기도 한다. 본 연구에서는 대한민국 공군 장병 환자를 대상으로 내시경을 통해 얻은 위점막 조직 내 호산구수를 측정하고, 이를 위점막 표면의 유문나선균 존재 유무와 관련지어 보았다. 111명 중 20명의 환자에서 한 고배율 시야 당 30개 이상의 호산구가 관찰되었고, 63명의 환자의 위점막 표면에서 유문나선균을 확인하였다. 또한 위점막 내 호산구의 밀도와 유문나선균의 존재 간의 관계는 통계학적으로 유의하였다. 본 연구의 결과는 대한민국 공군 장병을 대상으로 하여 조직 호산구 증가증과 유문나선균의 빈도 및 상호 관계를 최초로 분석했다는 점에서 의의가 있으며, 추후 유문나선균이 어떤 기전으로 위점막 조직 내 호산구의 증가에 관여하는지에 대한 연구를 진행하기 위한 기초 자료로서 활용될 수 있을 것이다.

Classification method of chronic gastritis by modeling of pulse signal (맥파 모델링을 통한 만성위염 분류 기법)

  • Chronic gastritis is the disease that is occuring in one in every 10 persons in Korea. In western medicine, endoscopy is needed to diagnose chronic gastritis, but it causes patients a pain and budget of expense. According to the TEM (Traditional Eastern Medicine), on the other hand, the 'Guan' position of the right wrist is related to a stomach. Thus we can diagnosis chronic gastritis by analyzing of pulse signal. However, pulse signal diagnosis is depended on oriental doctor's knowledge and experience. In this study, a systematic approach is proposed to analyze the computerized pulse signal. The pulse signals are firstly pre-processed, Gaussian model is adopted to fit the pulse signal, and then some related parameters are extracted from the model. Consequently, disease-sensitive parameters are selected by T-test and statistical difference. Finally, the selected parameters are entered into a Fuzzy C-Means (FCM) algorithm for classification. Classification results show that healthy persons and chronic gastritis patients are 95% and 87%, respectively.

Analysis of Gene Expression in Helicobacter pylori-associated Nodular Gastritis in Children Using Microarray (소아의 Helicobacter pylori 감염에 의한 결절성 위염의 유전자 발현 양상 분석)

  • Purpose: Nodular gastritis is a characteristic finding of Helicobacter pylori infection in children. The aim of this study was to evaluate the difference in gene expression in the gastric mucosa of H. pylori-infected and non-infected children, and to analyze the difference in gene expression using cDNA microarray analysis of nodular gastritis caused by H. pylori infection. Methods: Twelve children (6 boys and 6 girls; mean age 9.8 years) who underwent upper gastrointestinal endoscopy and biopsy at Seoul National University Bundang Hospital were included in the study. The subjects were divided into three groups according to the presence of H. pylori infection and nodular gastritis on endoscopic examination. Gastric mucosa tissue was kept at $-70^{\circ}C$ and RNA was extracted to perform cDNA microarray analysis in each patient. Results: cDNA microarray analysis in children revealed a clear distinction between H. pylori-infected and non-infected gastric mucosa. Specifically, 182 over-expressed genes and 29 under-expressed genes were identified in H. pylori-infected gastric mucosa compared to non-infected mucosa. H. pylori-infected nodular gastritis revealed different gene expression patterns from H. pylori-infected normal gastric mucosa; five genes were over-expressed and five genes were under-expressed. Conclusion: In the presence of H. pylori infection, gastric mucosa shows distinct differences in gene expression, and nodular gastritis with H. pylori infection in children may be associated with over- or under-expression of some genes. Further studies are required to clarify the host response and the pathogenesis of nodular gastritis in children.

Protective Effect of Canavalia gladiata on Gastric Inflammation Induced by Alcohol Treatment in Rats (알코올성 위염 동물 모델에서 작두콩 추출물의 보호 효과)

  • The objective of this study was to investigate the protective effect of extracts from Canavalia gladiata (CGE) on gastric inflammation induced by alcohol treatment in SD rats. Rats were divided into four groups: G1 (normal group), G2 (gastric inflammation induced by alcohol), G3 (gastric inflammation induced by alcohol with lansoprazole pretreatment), G4 (gastric inflammation induced by alcohol with 250 mg/kg b.w. CGE pretreatment), G5 (gastric inflammation induced by alcohol with 500 mg/kg b.w. CGE pretreatment). After the oral administration of 40% alcohol and samples for seven days, acute gastritis was induced with 70% alcohol and 0.15 M HCl. After 1 h of alcohol administration, the animals were sacrificed. Groups pretreated with lansoprazole or CGE showed an attenuation of gastric mucosal injury, including decreases in sub-epithelial loss, hemorrhages, and gastric juice secretion induced by administration of alcohol. The oral administration of CGE (500 mg/kg b.w.) significantly decreased the levels of TBARS. To examine molecular factors that regulate inflammation, the protein expression of NF-${\kappa}B$ and COX-2 were measured through immuno-histochemistry. Compared with the normal group (G1), the expression of NF-${\kappa}B$ and COX-2 were clearly increased in G2. COX-2 and NF-${\kappa}B$ were expressed even higher in groups pretreated with CGE compared to G2. In conclusion, our data show that Canavalia gladiata has inhibitory and protective effects on gastric inflammation induced by alcohol treatment in SD rats.

노두 수침엑스의 위염 및 위궤양에 대한 효과

  • 위염(gastritis), 위궤양(gastric ulcer)은 소화기 질환중에서 가장 빈도수가 높은 질병으로 위장관 점막이 위산에 의해 소화되어 버리므로서 궤양을 형성하는 상태를 말한다. 위염 및 위궤양의 발생빈도가 높은 것에 비해 발생원인은 정확히 밝혀져 있지 않으며 대체로 공격인자와 방어인자의 불균형 즉, 공격인자의 증가나 방어인자의 약화 또는 세균에 의한 감염에 의해 발생되는 것으로 알려져 있다. 일련의 식물엑스에 대하여 항위얌 및 항궤양효능에 관한 검색을 실시하여 노두의 M?H엑스가 현저한 효과가 있음을 예지하였으므로 그에 대하여 보다 구체적인 실험을 실시하였다. 즉 MeOH 엑스를 Hexane, CHCl$_3$, BuOH로 계통적으로 추출하여 상기의 분획 및 잔사인 물분획을 제조하여 이에 대한 실험을 실시하였다.

Protective Effect of Water Extract of Leonurus japonicus Houttuyn against HCl/EtOH-induced Gastric Mucosal Damage and Genotoxicity Evaluation using Micronucleus Test (익모초 물 추출물의 위염 예방효과 및 소핵시험을 이용한 유전독성 평가)

  • Gastritis is an inflammatory disease involving the stomach and is caused by several factors, including stress, non-steroidal anti-inflammatory drugs such as aspirin, liquor, and Helicobacter pylori. In Korea, Leonurus japonicus Houttuyn (LJW) has been used as traditional medicine for vaginal bleeding, hematuria, and bruise. Previous studies have reported that LJW exhibited hepatoprotective, cardioprotective, and anti-hyperlipidemic effect. However, the effect of the water extract of LJW on gastritis was not elucidated. Thus, we evaluated the anti-gastric effect and genotoxicity of LJW. LJW effectively prevented the degeneration of surface mucous cells and glandular epithelial cells and vascular congestion induced by HCl/EtOH. Micronucleus assay indicated that the rate of micronucleated polychromatic erythrocytes/polychromatic erythrocytes was not significantly different compared that of the control. Further experiments are required to determine the role of LJW in the gastric injury process such as cyclooxygenase signaling pathway and the secretion of mucus in the stomach.

The protective effect of Citrus unshiu Peel water extract through PI3K/Akt/NF-κB signaling pathway in mice with HCl/ethanol-induced acute gastritis (HCl/ethanol로 유발한 급성 위염 마우스에서 PI3K/Akt/NF-κB 신호전달경로를 통한 진피 열수 추출물의 보호 효과)

  • This study aimed to verify the effect of Citrus unshiu peel water extract (CUP) on a mouse model of acute gastritis (AG) induced by HCl/ethanol. Several studies have found that CUP has anti-inflammatory effects. The AG model was induced by oral administration of 150 mM HCl/60% ethanol (550 µL) to all groups except the control group. Also, for drug treatment, sucralfate (10 mg/kg) and CUP (100 or 200 mg/kg) were orally administered for 90 minutes before induction. The effect of CUP treatment was confirmed by gross gastric mucosal damage measurement, and the levels of Glutamic Oxaloacetic Transaminase (GOT), Glutamic Pyruvic Transaminase (GPT), and myeloperoxidase were reduced as well as the levels of oxidative stress biomarkers and their related proteins. In addition, the levels of inflammatory proteins, mediators, and cytokines were significantly downregulated byPI3K/Akt signaling. Taken together, these results show that CUP treatment alleviates AG by regulating PI3K/Akt signaling.
