Decomposition and Leaching of Bastnasite by Sulfation and Recovery of Cerium Hydroxide from Leached Solution (황산화반응에 의한 불탄산염 희토류광(Bastnasite)의 분해, 침출 및 세륨수산화물의 회수)

  • This study was carried out to investigate the optimum leaching conditions for the sulfation and water leaching, and separation of cerium from rare earth elements in leached solution by acid-adjusting method. The optimum conditions for the sulfation and water leaching from bastnasite concentrates are that the equivalent ration of sulfuric acid to concentrates is 2.5, calcination temperature and time are $600^{\circ}C$ and 2 hrs respectively, and the pulp density in the water leaching is 9.1%. The yield of rare earth oxide is about 93% at the above condition. The process of recovery of cerium hydroxide from leached solution by acid-adjusting method was carried out as following steps. The first step is the oxidation of the solution at pH 5 by using twice the equivalent of $H_2O_2$ solution as an oxidant. The second step is the precipitation to obtain cerium complex salt and cerium hydroxide after lowering the solution to pH 2. The last step is the oxidation-precipitation by using equivalent of $H_2O_2$ solution. From these results, it was possible to prepare cerium hydroxide with the yield of 60% and the quality of 80%.

Preparation and Characterization of Polysaccharide-based Nanofiber Using Electrospinning Method (전기방사 방법을 이용한 천연 다당류 나노섬유 제조 및 특성 연구)

  • In this study, alginate/poly(ethylene oxide) (PEO), and chitosan/PEO solution are prepared by dissolving alginate and chitosan into specific solvent for electrospinning. Solutions are poured into 10 mL plastic syringes with a metal nozzle supplied a high voltage power. The solution of alginate and chitosan is controlled by polymer concentration, temperature, relative humidity, applied voltage, distance from nozzle and flow rate of solution. Morphologies of fabricated nanofiber are observed by scanning electron microscopy (SEM). Optimal conditions for electrospinning of alginate nanofiber membrane are 2 wt% of alginate, 2 wt% of PEO at $60^{\circ}C$, 15 cm from the nozzle, $8{\mu}m/min$ flow rate and 20~24 kV. The conditions for elctrospinning of chitosan nanofiber membrane are 2 wt% of chitosan, 2 wt% PEO at $25^{\circ}C$, 15 cm from the nozzle, $8{\mu}m/min$ flow rate and 24 kV. The fabrication conditions of complex nanofiber prepared with chitosan and alginate are 20 cm from the nozzle, $8{\mu}m/min$ flow rate and 26 kV.

Quenching Curves for VOCs in the Analysis of Groundwater $^{14}C$ using Liquid Scintillation Counter (액체섬광계수기를 이용한 지하수의 $^{14}C$ 측정에서 휘발성유기화합물(VOC)에 대한 소광곡선)

  • Optimal quenching curves have been studied for the accurate analysis of $^{14}C$ in groundwater polluted by reducing efficiency of volatile organic compounds (VOCs) in liquid scintillation counter (LSC). The quenching parameters (SQP(E)) were measured for ten VOCs such as benzene, toluene, ethylbenzene, o-(m-,p-)xylene, trichloroethylene (TCE), tetrachloroethylene(PCE), carbon tetrachloride and chloroform. The quenching curves were plotted using $^{14}C$ standard solution and chloroform as a quenching agent. Optimal plotting conditions were determined for standard solution, LSC measuring time and the concentration of chloroform. The quenching effects of chlorinated organic compounds such as TCE, PCE, carbon tetrachloride and chloroform were greater than those of BTEX (benzene, toluene, ethylbenzene and xylene). Optimum measuring time was 100 minutes far 7,000 dpm/mL standard solution. A few mL of chloroform should be added for good quenching curves. These quenching curves have good correlation coefficients (> 0.99) and the curves could be applied to accurate analysis of $^{14}C$ in groundwater and tap water.

Investigation on Formation of Nanotube Titanium Oxide Film by Anodizing on Titanium in NaF Electrolytes (NaF 전해용액을 이용한 양극산화에 의한 타이타늄 표면의 나노튜브구조의 형성에 관한 연구)

  • The aim of this study is to find the condition of forming the favorable nanotubes by anodizing with NaF and $H_3PO_4$. Machined Ti discs were used for anode, and Platinum net was used for cathode. For electrolyte, $H_3PO_4$ and NaF solution were mixed. We controlled voltage, electrolyte concentration, anodizing time and formed nanotubes on Ti discs. After that, these were washed with distilled water for 24 hours and dried in the $40^{\circ}C$ oven for 24 hours. The surface structure of specimens were analyzed. The results were as follows : At 0.5 wt % NaF, according as increasing voltage and anodizing time, early state of nucleating pores were generated. At 1.0 wt % NaF, 20 V, 20 & 25 min, well-formed nanotubes were observed. At 1.0 wt % NaF, 30 V, structure of nanotube became bigger and interconnected. At 2.0 wt % NaF, no nanotubes were formed and it was unrelated with voltage and time. At 1.0 wt % NaF, 20 V, 20 - 25 min, well-ordered nanotubes were generated on Ti discs. For the formation of favorable nanotubes, it is considered that proper parameters such as electrolyte concentration, voltage, anodizing time are necessary according to the kind of electrolytes.

Incubation conditions affecting biogenic amines degradation of probiotic lactic acid bacteria (프로바이오틱 유산균의 바이오제닉 아민 분해능에 영향을 미치는 배양 조건)

  • The purpose of this study was to investigate the inhibitory effect of antibacterial substances produced by probiotic lactic acid bacteria (LAB) against biogenic amines-producing bacteria and the influence of culture conditions on the antibacterial activity of bacteriocin and organic acid. The bacteriocin solutions of Lactobacillus plantarum FIL20 (64 AU/ml) and Lactobacillus paracasei FIL31 (128 AU/ml) showed strong antibacterial activity against Serratia marcescens CIH09 and Aeromonas hydrophilia RIH28, respectively. And the lactic acid contents in the cell-free culture supernatants (CFCS) obtained from FIL20 and FIL31 strains were $107.3{\pm}2.7mM$ and $129.5{\pm}4.6mM$, respectively. Therefore, the bacteriocin solution (200 AU/ml) and the CFCS ($200{\mu}l/ml$) produced by L. plantarum FIL20 and L. paracasei FIL31 significantly (P < 0.05) decreased the bacterial numbers and histamine and tyramine production ability of S. marcescens CIH09 and A. hydrophilia RIH28. The amounts of histamine and tyramine produced by the CIH09 strain under conditions of low initial pH (5.0) and incubation temperature ($15^{\circ}C$) was significantly reduced by treatment with bacteriocin solution and CFCS obtained from L. plantarum FIL20. In addition, the bacterial counts and biogenic amines contents of CIH09 strain were significantly decreased (P < 0.05) when sodium chloride (5%) or potassium nitrite (200 mg/g) were mixed with the antibacterial substances of L. plantarum FIL20. Consequently, the bacteriocin and organic acid solution of L. plantarum FIL20 and L. paracasei FIL31 can be used as a biological preservation to effectively control the production of biogenic amines by the application of hurdle technology.

A Study on an Effective Decellularization Technique for a Xenograft Cardiac Valve: the Effect of Osmotic Treatment with Hypotonic Solution (이종 심장 판막 이식편에서 효과적인 탈세포화 방법에 관한 연구; 저장성 용액(hypotonic solution)의 삼투압 처치법 효과)

  • Background: Cellular remnants in the bioprosthetic heart valve are known to be related to a host's immunologic response and they can form the nidus for calcification. The extracellular matrix of the decellularized valve tissue can also be used as a biological scaffold for cell attachment, endothelialization and tissue reconstitution. Thus, decellularization is the most important part in making a bioprosthetic valve and biological caffold. Many protocols and agents have been suggested for decellularization, yet there ave been few reports about the effect of a treatment with hypotonic solution prior to chemical or enzymatic treatment. This study investigated the effect of a treatment with hypotonic solution and the appropriate environments such as temperature, the treatment duration and the concentration of sodium dodecylsulfate (SDS) for achieving proper decellularization. Material and Method: Porcine aortic valves were decellularized with odium dodecylsulfate at various concentrations (0.25%, 0.5%), time durations (6, 12, 24 hours) and temperatures ($4^{\circ}C$, $20^{\circ}C$)(Group B). Same the number of porcine aortic valves (group A) was treated with hypotonic solution prior to SDS treatment at the same conditions. The duration of exposure to the hypotonic solution was 4, 7 and 14 hours and he temperature was $4^{\circ}C$ and $20^{\circ}C$, respectively. The degree of decellularization was analyzed by performing hematoxylin and eosin staining. Result: There were no differences in the degree of decellularization between the two concentrations (0.25% 0.5%) of SDS. Twenty four hours treatment with SDS revealed the best decellularization effect for both roups A and B at the temperature of $4^{\circ}C$, but there was no differences between the roups at $20^{\circ}C$. Treatment with hypotonic solution (group A) showed a better ecellularization effect at all the matched conditions. Fourteen hours treatment at $4^{\circ}C$ ith ypotonic solution prior to 80S treatment revealed the best decellularization effect. The treatment with hypotonic solution at $20^{\circ}C$ revealed a good decellularization effect, but his showed significant extracellular matrix destruction. Conclusion: The exposure of porcine heart valves to hypotonic solution prior to SDS treatment is highly effective for achieving decellularization. Osmotic treatment with hypotonic solution should be considered or achieving decellularization of porcine aortic valves. Further study should be carried out to see whether the treatment with hypotonic solution could reduce the exposure duration and concentration of chemical detergents, and also to evaluate how the structure of the extracellular matrix of the porcine valve is affected by the exposure to hypotonic solution.

Synthesis of High Purity Tungsten Oxide with Tungsten Chloride from the Chlorination of Scheelite (회중석의 염소화 생성물로부터 고순도 WO3의 합성)

  • In this study we developed the synthesis of high purity tungsten oxide from tungsten chlorides obtained by the chlorination of scheelite in a fluidized bed reactor. Within a minute of dissolution time, tungsten chlorides were almost dissolved in $H_2O_2$ solution. The proper dissolution conditions for the tungsten chlorides were as follows : $H_2O_2$ concentration 0.5%, dissolution temperature $15^{\circ}C$ and $H_2O_2$ amount to 0.5g tungsten chlorides 30ml. Under above conditions, the tungsten oxide prepared from dissolved product was identified as $WO_3$ and the purity of $WO_3$ was 99.53%.

  • Optimum counting condition of a low level liquid scintillation counter for radiocarbon dating has been determined to minimize the contribution of tritium activity and get the highest FOM by the variation of scintillation solutions, scintillation solution to benzene ratio, and ${\beta}$-ray spectrum counting ranges. Under the condition, background count rate has been reduced to $2.26{\pm}0.03cpm$, and the maximum measurable age reached to about 40,000 years. And the practicality of counting system has been verified by the analysis of shell and peat samples.

  • The wet etching is a process using chemical solution and occurring chemical reaction on substrate surface. when we do wet etching process, we have to consider stoichiometry, etching time and temperature of etchant for good resolution. In this experiment, we used Cr, Al andIndium-tin-oxide (ITO) metal and we deposited them with DC sputtering machine. The Cr thin film metal thickness is about $1300{\AA}$, ITO films show a low electrical resistance and high transmittance in the visible range of an optical spectrum and Ai film is used for signal line. We measured and analysed wet etching properties on the metal thin films.

  • Using biodegradable polycaprolactone, the microcapsules were prepared by solvent evaporation method. Retinol was selected as a core material, which was used as an important ingredient material in cosmetic fields. Poly(vinyl alcohol) was used as a stabilizer. The shape and property of the microcapsules were characterized by scanning electron microscope and differential scanning calorimeter, and the release rate of the microcapsule was measured by UV spectrophotometer. The microcapsules were prepared, changing the concentration of wall material, the stirring rate, and the concentration of stabilizer. Under the optimum condition, the microcapsules were formed, which showd 5~6 um in diameter and got the homogeneous sphere shape.
