The Mechanisms for Xylose Transport into Yeasts (효모내로의 Xylose 운반 기작)

  • The biochemical study of sugar uptake in yeasts started five decades ago and led to the early production of abundant kinetic and mechanistic data. However, the first accurate overview of the underlying sugar transporter genes was obtained relatively late, due mainly to the genetic complexity of hexose uptake in the model yeast, Saccharomyces cerevisiae. The genomic era generated in turn a massive amount of information, allowing the identification of a multitude of putative sugar transporter and sensor-encoding genes in yeast genomes, many of which are phylogenetically related. This review aims to briefly summarize our current knowledges on the biochemical and molecular features of the transporters of pentoses in yeasts, when possible establishing links between previous kinetic studies and genomic data currently available. Emphasis is given to recent developments concerning the identification of D-xylose transporter genes, which are thought to be key players in the optimization of S. cerevisiae for bioethanol production from lignocellulose hydrolysates.

  • Acetylation of triazinylated monosaccharides was carried out. The products were identified by elementary analysis. IR and NMR with Methyl $[T]-{\beta}-L-acetyl-arabopyranoside$ and Methyl $[T]-\alpha-D-acetyl-lyxopyranoside$ respectively. The triazine derivatives of uronic acids had no remarkable biological activities against dish and rice, and also showed not to be effected on antifungal and antibacterial activities.

Optimization of Microwave-Assisted Pretreatment Conditions for Enzyme-free Hydrolysis of Lipid Extracted Microalgae (탈지미세조류의 무효소 당화를 위한 마이크로파 전처리 조건 최적화)

  • The purpose of this study was to effectively produce the biosugar from cell wall of lipid extracted microalgae (LEA) by using microwave-assisted pretreatment without enzymatic hydrolysis process. Response surface methodology (RSM) was applied to optimization of microwave-assisted pretreatment conditions for the production of biosugar based on enzyme-free process from LEA. Microwave power (198~702 W), extraction time (39~241 sec), and sulfuric acid (0~1.0 mol) were used as independent variables for central composite design (CCD) in order to predict optimum pretreatment conditions. It was noted that the pretreatment variables that affect the production of glucose (C6) and xylose (C5) significantly have been identified as the microwave power and extraction time. Additionally, the increase in microwave power and time had led to an increase in biosugar production. The superimposed contour plot for maximizing dependent variables showed the maximum C6 (hexose) and C5 (pentose) yields of 92.7 and 74.5% were estimated by the predicted model under pretreatment condition of 700 w, 185.7 sec, and 0.48 mol, and the yields of C6 and C5 were confirmed as 94.2 and 71.8% by experimental validation, respectively. This study showed that microwave-assisted pretreatment under low temperature below $100^{\circ}C$ with short pretreatment time was verified to be an effective enzyme free pretreatment process for the production of biosugar from LEA compared to conventional pretreatment methods.

  • 세계적인 자원고갈과 지구온난화와 같은 환경문제가 발생됨에 따라 대체에너지 개발에 대한 연구가 활발히 이루어지고 있다. 섬유소 기질을 이용한 바이오에탄올 생산은 세계적으로 막대한 자원이 있으며 광합성에 의해 재생산되는 무한한 재원으로서 환경적으로도 대기오염물질을 적게 배출하여 유용한 에너지원으로 각광받고 있다. 하지만 섬유소 기질은 cellulose, hemicellulose, lignin과 같은 고분자 화합물이 유기적으로 결합된 단단한 결정구조로 이루어져 있어 이를 분해하여 원하는 물질을 얻기 위해서는 전처리 과정이 필요하다. 전처리 공정은 바이오에탄올을 생산하는 당화 및 발효공정의 효율 및 반응시간 단축에 기여하며, 특히 섬유소 기질일 경우에는 필수불가결한 공정이다. 전처리 공정은 물리적, 화학적, 생물학적 방법으로 나누어지며, 이러한 방법들 중 기질의 특성과 처리효율에 따라 기술들을 병합하여 사용하기도 한다. 이에 본 연구에서는 산 처리, 암모니아 처리, 과산화수소 처리 및 효소를 이용한 생물학적 처리를 단독 또는 병행하여, 전환된 당 성분 및 함량을 조사함으로서 섬유소계 기질인 채소 음식물류 쓰레기를 대상으로 바이오에탄올을 경제적으로 생산하기 위한 적합한 전처리법을 검토하였다. 전처리 방법별 당화율을 살펴보면, 산 처리와 암모니아-과산화수소-계면활성제 처리가 각각 65.3 % 및 65.7 %로 가장 높았으며, 과산화수소 처리는 16.2 %로 가장 낮았다. 반면 전처리 공정 없이 효소를 이용한 당화만을 실시한 경우에는 4.3 %의 낮은 당화율을 나타내었다. 섬유소계 기질의 전처리 효율을 향상시키기 위해 첨가하는 계면활성제의 효과는 암모니아-과산화수소 및 암모니아-과산화수소-계면활성제 처리의 당화율을 비교한 결과, 뚜렷한 효과를 확인할 수 없었다. 전처리 방법별 당의 성분 및 함량을 비교한 결과 육탄당은 암모니아-과산화수소-계면활성제 전처리에서 가장 많이 검출되었다. 오탄당은 산 처리 후 그 함량이 현저히 높았으며, 오탄당 중 xylose의 함량이 60.49 mg/g로 가장 많이 차지하고 있었다. 이 결과로부터 전처리 방법에 관계없이 당화율은 유사한 수준을 보이지만, 당화된 당의 성분 및 함량에는 큰 차이가 있음을 알 수 있었다. 이당류의 경우 과산화수소 및 암모니아-과산화수소 처리를 제외한 나머지 전처리 방법에서 유사한 수준을 나타내었다. 암모니아 처리 및 과산화수소 처리를 순차적으로 병행한 암모니아-과산화수소 처리에서는 각각의 처리시보다 육탄당의 함량은 증가하였으나 암모니아 처리시보다 이당류의 함량은 감소한 것으로 나타났다.

  • It is indispensable to increase the conversion rate of a reducing sugars such as pentose and hexose derived from cellulosic wastes for a highly efficient bioethanol fermentation from food wastes. The saccharification liquid from cellulosic substrates such as vegetable food wastes contained lots of hexose like glucose and pentose like xylose. Since Saccharomyces-based yeasts could not convert xylose to bioethanol, Pichia stipitis which could directly ferment xylose to ethanol was chosen. After selecting Saccharomyces coreanus and P. stipitis, fermentation characteristics by mixture of two yeasts were investigated. As a result, it was verified the production of ethanol was enhanced by the co-fermentation, although there were somewhat differences between the fermentation characteristics by the aeration methods. Moreover, the consumption of pentose, hexose and disaccharide was obviously observed, and aeration in the process of fermentation seemed to stimulate the activity of P. stipitis.

  • Bacillus stearotheymophilus, a potent xylanolytic bacterium isolated from soil, was tested for the strain's strategies of pentose utilization and the evidence of substrate preferences. The strain metabolized glucose, xylose, ribose, maltose, cellobiose, sucrose, arabinose and xylitol. The efficacy of the sugars as a carbon and energy source in this strain was of the order named above. The organism, however, could not grow on glycerol as a sole growth substrate. During cultivation on a mixture of glucose and xylose or arabinose, the major hydrolytic products of xylan, B. stearothermophilus displayed classical diauxic growth in which glucose was utilized during the first phase. On the other hand, the pentose utilization was prevented immediately upon addition of glucose. Cellobiose was preferred over xylose or arabinose. In contrast, maltose and pentose were co-utilized, and also no preference on between xylose and arabinose. Enzymatic studies indicated that B. stearothermophilus possessed constitutive hexokinase, a key enzyme of the glucose metabolic system. While, the production of $^{D}$-xylose isomerase, $^{D}$-xylulokinase and $^{D}$-arabinose isomerase essential for pentose phosphate pathway were induced by xylose, xylan, and xylitol but repressed by glucose. Taken together, the results suggested that the sequential utilization of B. stearothermophilus would be mediated by catabolite regulatory mechanisms such as catabolite inhibition or inducer exclusion.

  • Effects of nitrogen sources on free amino acids, and on photosynthsis-respiration equilibrium point of various positional leaf in soybean were investigated in relation to phosphorus sensitivity. The content of free amino acids was highest in ammonium and lowest in urea treated leaves. Glycine, serine, alanine and especially histidine were high in the ammonium treated leaves. Aspartic acid was high in the nitrate treated leaves. Photosynthesis respiration equilibrium point was higher in the sensitive cultivars, and higher with ammonium than with nitrate. The excess ammonium in plant appears to draw out an intermediate metabolite from carbon fixation pathway resulting in photosynthetic inhibition and activate pentoses phosphate pathway and photorespiration. Such phenomena were likely accentuated in phosphorus sensitive variety.

  • A piscicidal substance was isolated from the culture medium of Streptomyces umbrosus by avicel column chromatography and avicel thin layer chromatography after extration with chroloform. Bluegreen fluorescence was emitted under UV irradiation. Factors which govern toxin production and nutrition requirement for high toxin titres were observed. Nutritional uptake for toxin production was not curresponded with that for cell growth. Alanine, valine, serine asparagine, arginine, histidine, urea and sodium nitrate as a carbon source and glucose, mannose, rhamnose, xylose, arabitol and starch as a carbon source were recognized as a favorable nutrient for high toxin production. Magnesium was essential factor whereas vitamins were not of effective. Most of toxin was formed simultaneously with cell growth in esponential phase. Maximal production was observed for six day culture at 3$0^{\circ}C$. Tissues of gill, kidney and pnacreas in Cyprinus carpio were denatured extreamly after treating with the substance. Atrophied nucleous, indented membrane and degradated cytoplasm with necrotic affectness were noted on each tissue. The chemical formula of the substance was designated as $C_{38}$ $H_{66}$ $NO_4$.

Ethanol Fermentation by Pichia Stipitis in a Mixture of Pentoses and Hexoses (오탄당과 육탄당의 혼합용액에서 Pichia stipitis에 의한 에탄올 발효)

  • P. stipitis CBS5776 was cultivated to examine the characteristics of ethanol fermentation for hexoses (mannose, g1ucose, and galactose) and pentoses(xylose and arabinose). Glucose was the best carbon source among the sugars used in terms of ethanol yield. Glucose was used to produce ethanol with an yield coefficient 0.376g ethanol/g glucose, whereas mannose was converted to produce ethanol with an yield coefficient 0.326g ethanol/g mannose. P. stipitis CBS5776 was also grown in a mixture of sugars to study the pattern of carbon utilization. The yeast utilized glucose and mannose firsts and then galastose and xylose as carbon sources. Arabinose was partially used for biomass when it was present as a sole carbon source, but it was not metabolized at all in a mixture of carbon sources. P. stipitis produced $12.2g/\ell$ ethanol with a yield coefficient 0.332 g ethano1/g sugar in a mixture of sugars.

  • As a basic studies for the laboratory scale production of alfalfa inoculum, Rhizobium meliloti M14 was characterized for its carbon and nitrogen sources, and some parameters for broth cultivation in a chemostat were studied by semi-continuous operation. The result s obtained were as follows. 1. Growth rate of the strain was increased by disaccharides than by monosaccharides tested, and pentoses resulted in poor growth than hexoses. Sugar alcohols including inositol supported the best growth among sugars. 2. Mannitol in the yeast-mannitol-broth was substituted by natural carbon sources such as malt extract or molasses. 3. Ten per cent of fresh yeast water appeared to supply enough amount of growth factor s for the strain, and the effect was equivalent to 0.24 percent of the commercial yeast extract powder. 4. Batch growth of the stain in a chemostat, New Brunswick Micro Ferm 28L, reached in the early stationary growth phase of $5{\sim}7{\times}10^9cells/ml$ after 36 hours of incubation. The culture at this stage was switched to semi-continuous cultivation, and the culture broth of four-fifth of the working volume was recovered every 24 hours when the maximal count was obtained.
