Effects of Korean Food-based Dietary Inflammatory Index Potential on the incidence of diabetes and HbA1c level in Korean adults aged 40 years and older (40세 이상 성인 한국인에서 한국형 식사염증지표 수준에 따른 당뇨병 발생률 및 당화혈색소 수준 변화 연구)

  • Purpose: The present study examined the associations of Korean Food-based Index of Dietary Inflammatory Potential (FBDI) scores with the prevalence of diabetes and hemoglobin A1c (HbA1c) level of diabetes patients in Korean adults. Methods: The Korean Genome and Epidemiology Study (KoGES) Health Examinee baseline data, collected between 2004 and 2013 and followed up between 2012 and 2016, were used in our study. A total 56,391 participants including diabetes (n = 5,733) and non-diabetes (n = 50,658) were analyzed. The subjects were classified into quartiles of FBDI scores using the semi-quantitative food-frequency questionnaire developed for KoGES. The prevalence rate of diabetes under FBDI scores was assessed by Cox proportional risk models and the severity of the diabetes was analyzed by multiple regression analysis. Results: There were 775 incident cases of diabetes after a mean follow-up of 3.97 years. There was no statistically significant association between FBDI scores and incidence of diabetes. Among diabetes patients at baseline, FBDI scores were related to the risk of progression of diabetes which was represented by greater than 9% HbA1c (Q1 vs. Q4; odds ratio, 1.562 [95% confidence intervals, 1.13-2.15]; p for trend = 0.007). The stratified analysis showed a stronger association in females, irregular exercise group, and higher body mass index group. Conclusion: These results suggest that a pro-inflammatory diet is not associated with the incidence of diabetes but is related to the HbA1c level of diabetes patients. Thus, further longitudinal studies with longer periods are required to determine a relationship between dietary inflammatory index and diabetes in Korea.

Effects of Resistance Training on Serum Inflammatory Markers and CatSper 1-4 Protein Expression in Testis of OLETF Rats (저항성 운동이 OLETF 쥐의 혈청 염증반응지표 및 고환조직의 CatSper 1-4 단백질 발현에 미치는 영향)

  • PURPOSE: This study was to investigate the effects of resistance exercise on serum inflammatory markers and CatSper 1-4 expression in testis of OLETF rats. METHODS: Male OLETF rats were divided into two groups; control group (n=12), resistance exercise group (n=12). The exercise group performed a total of 8 weeks of moderate intensity resistance exercise on a 1.35 m vertical ladder with weights secured to their tails. RESULTS: The results of this study was following; The exercise group showed a significant decreased in the inflammatory ($IL-1{\beta}$, IL-6, $TNF-{\alpha}$) levels as compared to the control group. But CRP was no significant difference between control and exercise groups. Also, CatSper 1 and 2 were significantly increased in the exercise group compared to the control group, whereas CatSper 3, 4 were no significant difference between both groups. CONCLUSIONS: This study suggests that resistance exercise training can contribute to reduce pro-inflammatory responses in whole body and it affects male reproductive function by improve sperm quality and CatSper protein expression.

Anti-inflammatory Effects of Aroma Oil Complex on DNCB-Induced Allergic Contact Dermatitis in Dogs (개에서 DNCB에 의해 유발된 알레르기성 접촉피부염에 대한 아로마 오일 합제의 항염증 효과)

  • Allergic contact dermatitis (ACD) is an inflammatory skin disease and regarded as a prototype of T-cell mediated delayed-type hypersensitivity reactions. Aroma Oil Complex (AOC) is composed of lavender true oil, chamomile roman oil and tea tree oil. This study was performed to assess the effects of AOC in a canine model of ACD. ACD was induced on the back of dogs induced by sensitization and repeated application by 2,4-dinitro-1-chlorobenzene (DNCB). Topical treatment of AOC was applied once a day for 8 days and skin biophysical parameters including transepidermal water loss (TEWL), skin hydration, skin thickness and erythema index, were measured every two days during experimental periods. Histopathology and immunohistochemistry were performed to evaluate the anti-inflammatory effect. In skin biophysical parameters, TEWL, skin hydration, skin thickness and erythema index were significantly increased, with a maximum increase appeared on day 2 (p<0.05). After the completion of AOC treatment, skin biophysical parameters were significantly reached those of baseline in a time-dependent manner (p<0.05). In histopathology, marked increases of epidermal thicknesses were induced after DNCB challenge with numerous inflammatory cell infiltrations and edematous changes, decreases of connective tissue occupied regions in dermis. In addition, marked increases of cytokine - tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interferon-${\gamma}$ (IFN-${\gamma}$)-immunoreactivities in the dermis and of apoptotic markers - caspase-3 and PARP-immunoreactivities in the epidermis were observed in DNCB control as compared with intact control, respectively (p<0.01). The decrease of infiltrated inflammatory cells and related decreases of pro-inflammatory cytokine immunoreactivities were observed in AOC treated skin (p<0.01). Based on these findings, AOC may have anti-inflammatory and alleviatory effects in the allergic contact dermatitis.

Protective Effects of Akebia quinata Fruit Extract on Acute Alcohol-induced Hepatotoxicity in Mice (급성 알코올 간독성을 유발한 생쥐에 있어서 으름 열매 추출물의 간 기능 보호효과)

  • We studied the effects of Akebia quinata fruit extract (AQ) on acute alcohol-induced hepatotoxicity in mice. AQ (30-1,000 mg/kg body weight (BW) per day) was orally administered to the study group, once daily for 1 week. On the last day of AQ treatment, ethanol (6 mg/kg BW) was orally administered to induce acute liver injury. The AQ-treated group showed significantly lower levels of alanine aminotransferase and aspartate aminotransferase, compared to the only ethanol-treated group (ETG). The glutathione level in the AQ-treated group elevated up to 20.6%, compared to that observed in the ETG. The mRNA expression of glutathione synthetic enzymes was also higher in the AQ-treated group, compared to the ETG. The AQ-treated group also exhibited lower levels of expression of NADPH oxidase 4 and tumor necrosis factor alpha mRNA. Thus, these results show that AQ treatment can be a potential method to reduce oxidative stress and inflammation in ethanol-treated mouse liver and also that AQ can be a useful therapeutic agent for acute alcohol-induced hepatotoxicity.

Effect of 4 Weeks' Walking Exercise with Blood Flow Restriction on Inflammatory index, Isokinetic Muscle Function, and Thigh Circumference in Obese Women (4주간의 혈류를 제한한 걷기운동이 비만여성의 혈중 염증지표와 등속성근기능, 대퇴둘레에 미치는 영향)

  • Blood flow restriction(BFR) exercise is defined as low intensity and short term exercise using pneumatic pressure belts at the top of limbs, which affects the physiological functions of the body. The purpose of this study was to investigate the effects of walking exercise with BFR on inflammatory index, isokinetic muscle function, and thigh circumference in obese women. Eleven obese women(> BMI $25kg/m^2$ & > body fat 30%) wore pneumatic pressure belts on both femurs and performed walking exercise twice per day, 3 days/wk for 4 weeks (walking 2 min; resting 1 min). Data analysis was carried out using paired t-test. Body weight, BMI, and body fat significantly decreased after exercise(p<.05), and right thigh circumference significantly decreased(p<.05). The concentration of plasma IL-6 significantly increased(p<.05) after exercise. TNF-${\alpha}$ level was not statistically different but tended to slightly increase. CRP slightly decreased, although it did not reach statistical significance after exercise. Muscle strength significantly increased in the $60^{\circ}/sec$ of right/left side extension, left side flexion, and $180^{\circ}/sec$ of left side extension after training(p<.05). These results suggest that 4 weeks of blood flow restriction walking exercise has positive effects on inflammatory index and isokinetic muscle function. Therefore, we consider that blood flow restriction exercise can be used for treatment of obesity, related chronic diseases, and metabolic syndrome. Further, blood flow restriction exercise for a short time has similar effects as a high intensity resistance program.

The Effect of Polycan-Calcium Gluconate Complex on Inflammatory Mediators from Periodontitis Patients (Polycan-Calcium Gluconate 복합제에 의한 염증인자의 감소 효과)

  • The aim of this study was to examine the effect of polycan-calcium gluconate complex on the levels of immune and inflammatory mediators in gingival crevicular fluid and serum from patients with periodontitis. A total of 39 patients with periodontitis took placebo (placebo group) or polycan-calcium gluconate complex (treatment group) twice a day for 4 weeks. At baseline and 4 weeks, gingival crevicular fluid (GCF) and blood was collected from each subjects. The secretion level of interleukin $(IL)-1{\beta}$ in GCF was measured by enzyme-linked immunosorbent assay (ELISA). Western blot analysis was conducted to determine the expression of matrix metalloproteinase (MMP)-8 and tumor necrosis factor $(TNF)-{\alpha}$ in GCF. Serum samples were analysed for MMP-8 by ELISA and C-reactive protein (CRP) by turbidimetric immuno assay. MMP-8 and $TNF-{\alpha}$ were significantly decreased in the treatment group at 4 weeks. The level of $IL-1{\beta}$ in the treatment group was significantly lower than those of the placebo group. No differences were observed in CRP levels. Taken together, these data indicate that taking of polycan-calcium gluconate complex led to reduction of inflammatory biomarkers in serum and GCF of periodontitis patients.

  • Chrysanthemum coronarium L. contains various antioxidants such as beta-carotene, vitamins A and C, and polyphenols, and is known to have anti-inflammatory effects. Under the assumption that the PDRN contained in the extract can mediate the anti-inflammatory response, the mouse macrophages, RAW264.7 cells, were stimulated with LPS to induce the conversion to inflammatory cells, and then the addition of PDRN extracted from the extract was effective in inhibiting inflammation. It was analyzed whether there was The gene expression of IL-1β and TNF-α was used as an anti-inflammatory index, and the relative expression levels of each gene were confirmed by RT-PCR. As a result, RT-PCR confirmed the effect of PDRN-induced inhibition of inflammation in both IL-1β and TNF-α genes. Therefore, based on this study, it is considered to be a precedent data that can be developed as a treatment for inflammatory diseases, and it will be helpful in research and development of a treatment that can improve the anti-inflammatory mechanism.

  • Arabinoxylan (Ara) rice bran has been shown to be a potent biological response modifier as manifested by stimulation of different arms of the immune system. We examined the effects of Ara rice bran and exercise on the immune function and cytokines in lipopolysaccharide (LPS)-stimulated rats. As the results, tumor necrosis factor-${\alpha}$ as representative inflammatory cytokines showed a significantly lower in Ara supplement group, thus the Ara rice bran had a higher inhibitory activity than the both exercise and control group. However, 4 weeks of exercise training significantly increased inflammatory reactions rather than treatment with Ara in LPS-treated rats. The Ara rice bran acted to decrease the inflammatory reaction. These results suggest that the supplement of Ara rice bran is likely contribute to inflammation response and the Ara rice bran can be used as a possible safe alternative to the immunotherapeutic modalities.

A Pilot Study to Develop Gamrosu, a Modified Fasting Therapy Beverage: Case Series (절식보조음료 감로수(甘露水) 개발을 위한 선행연구: 증례보고)

  • Gamrosu is a modified fasting therapy beverage (431 kcal/d). A retrospective observational study was conducted to investigate the detoxification feasibility of Gamrosu. Three cases were reviewed which participants have finished the fasting with Garmrosu for 10 consecutive days. Detoxification profiles of Gamrosu were reviewed at pre- and post-fasting sessions. Post-Gamrosu session, -6.3% of average weight, -6.5% of average body fat mass and -6.6% of average muscle mass were reduced with the nutrition indices being improved. The inflammation indices showed the significant diminished profiles. Liver/kidney functions and the standard of electrolytes were maintained within normal range in stable manners, however, marginal elevation of total bilirubin and mild ketoacidosis were observed. The indices of oxidative stress decreased and those of antioxidative activity increased. The fatigue scale scores indicated lower scores except insomnia symptom. Taken together, detoxification profiles of Gamrosu were sufficiently feasible and the observed findings should be considered for further clinical studies.

Anti-inflammatory effects of Cudrania tricuspidata twig sawdust fermented with Ganoderma lucidum mycelium (영지버섯균 발효 꾸지뽕나무 가지 톱밥 추출물의 항염증 활성)

  • In this study, we evaluated the anti-inflammatory effect of extract from Cudrania tricuspidata twig sawdust fermented with Ganoderma lucidum mycelium. Fermented Cudrania tricuspidata twig sawdust extracted with 70% ethanol and elucidated the potential signaling pathway in lipopolysaccharide (LPS)-induced RAW264.7 cells. Fermented Cudrania tricuspidata twig sawdust inhibits LPS-stimulated nitric oxide (NO) production without affecting cell viability in a dose-dependent manner and production of LPS-induced pro-inflammatory cytokines such as interleukin (IL)-1β, tumor necrosis factor (TNF)-α and prostaglandin2 (PGE2). Fermented Cudrania tricuspidata twig sawdust also suppressed the expression of the pro-inflammatory mediators such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in LPS-stimulated RAW264.7 cells. Moreover, Fermented Cudrania tricuspidata twig sawdust significantly attenuated LPS-induced IkappaB (IκB) degradation and suppressed nuclear factor kappa B (NF-κB) nuclear translocation. These results suggest that fermented Cudrania tricuspidata twig sawdust may have great potential for the development of anti-inflammatory agent.
