Solvent Leaching Characteristics of Dark Brownish Pigment from Activated Charcoal used in Decolorization of Crude Polysaccharide from Auricularia auricula (흑목이 버섯 다당류의 탈색에 사용된 활성탄으로부터 흑갈색 색소의 용매 침출 특성)

  • A dark brownish pigment in the crude polysaccharide from Auricularia auricula was adsorbed by activated charcoal. The leaching of the pigment adsorbed on activated charcoal and regeneration of activated charcoal used was investigated with eight kinds of solvents. The highest leaching capacity was obtained with the alkaline solution (KOH). The optimum volume of 1 M KOH solution per activated carbon was 45 mL/g, and the treatment for 10 min during single stage leaching was sufficient to achieve the leaching equilibrium. Second-order kinetic model provided the best fitting for the pigment leaching. The pigment leaching capacity of 88.9% was obtained by seven times of treatment with 1 M KOH solution at 25$^{\circ}C$, while at 95$^{\circ}C$, leaching capacity of 82.6% was achieved with single stage alone showing the significant increase of leaching capacity with increasing temperature. The regenerated activated charcoal was nearly as effective as fresh activated charcoal in pigment adsorption of crude polysaccharide from Auricularia auricula.

Study on the Antimicrobial Effects of Citrus Peel by Different Extract Methods (추출방법에 따른 감귤과피 추출물의 항균효과)

  • The antimicrobial activity of the extract of citrus peel prepared by the method of hot water, ethanol and sugar was examined. The results showed that the extract of citrus peel prepared by hot water or ethanol did not have antimicrobial activity, but the extract by 10$\%$(w/v) sugar revealed the high antimicrobial activity. Extracted in 10%(w/v) sugar solution for 9 days, showed the highest antimicrobial activity against 8 strains of bacteria. The minimum inhibition concentration was found to be 0.5$\%$(v/v) against S. aureus, 1.5$\%$(v/v) against B. subtilis, M. luteus and E. coli, and 2.0$\%$(v/v) against S. mutans. The antimicrobial activity of the citrus peel extract was stable regardless of the treatment at 40 $\~$ 100 $^{\circ}C$C for 20 min and unstable response to the change of pH. The results suggested the development of citrus peel as heat-stable antimicrobial agents.

  • The extraction and separation methods of protein-bound polysaccharides from the mycelium and culture broth of Coriolus versicolor (Fr) Quel were investigated. The use of 2% solution of surface active agent Triton X-100, was effective for extraction of the protein-bound polysaccharides from the mycelium. For the separation and partial purification of the protein-bound polysaccharides, the column chromatography using DEAE-Cellulose and DEAE-Sephadex proved to be effective.

Bioethanol Production from Rice straw by Irpex consors (송곳니구름버섯을 이용한 볏짚에서의 에탄올 생산)

  • This study was initiated to evaluate ethanol production by a Korean isolate of white rot fungus Irpex consors. It was found that the fungus could produce ethanol by converting glucose, mannose, xylose, and cellobiose under semi-aerobic condition with yields of 0.23, 0.19, 0.21, and 0.17 g ethanol per g sugars, respectively. Furthermore, the strain produced ethanol by simultaneous saccharification and fermentation of rice straw treated with steam pressured boiling water, 3% NaOH, and 3% $H_2SO_4$ with maximum yields of 0.12, 0.15, and 0.19 g ethanol per g rice straw, respectively. These results suggested that I. consors could produce ethanol from the components of cellulose and hemicellulose including glucose, mannose, xylose, cellobiose as well as rice straw treated with steam pressured boiling water, dilute sodium hydroxide, and dilute sulfuric acid. This is the first report that I. consors mycelia produce ethanol from various sugars and lignocellulosic substance including rice straw.

  • The Buncheon manganese ore deposits occur in vein along the fault of $N20^{\circ}E$, cutting the foliation of Yulri Series. The deposits consist of primary manganese silicate ores in the deeper part and superficial manganese oxide ores near the surface. The spatial distribution of manganese oxide ores with respect to the manganese silicate ores suggests that the manganese oxide ores are the supergene oxidation product of the manganese silicate ores. Manganese silicate ores consist mainly of fine-to coarse-grained pyroxmangite with minor rhodochrosite, quartz, sulfides and chlorite. Manganese oxide ores are composed of supergene manganese oxides such as nsutite, birnessite, manganite and todorokite, and other associated minerals. Paragenetic sequence of formation of the manganese minerals are as follows: $\array{{rhodochrosite{_{\rightarrow}^o}todorokite{_{\searro}^o}}\\pyroxmangite{_{\line(10){90}}^o}{\nearro}}birnessite{_{\rightarrow}^o}nsutite{_{\rightarrow}^s}manganite$ In order to elucidate the mineralogy of the manganese minerals, microscopic, X-ray, IR spectroscopic, and thermal studies were made for manganese and associated minerals.

  • There are many typical epithermal deposits scattering in the Taran of central Kalimantan. Indonesia. To get the shallow geological information, we carried out the geophysical exploration: spontaneous potential survey, in-suite magnetic susceptibility measurement and relistivity survey method in this area for a few weeks from September 30th to October 27th in 1997. SP survey (Fluke 27 multimeter)/magnetic susceptibility (EK -7 meter) measurement was carried out with a 250m$\times$10m lattice net in N45$^{\circ}$W direction. The dipole-dipole array resistivity survey was conducted with an electrode spacing of ${\alpha}$=30 m and electrode separation index n=7 at the line 5. The result shows that was the gold bearing quartz vein area can be divided into two type lone: low sulfidation type and high sulfidation type zone. The low resistivity value in the survey line 5 indicated a fracture Bone associated with the high sulfidation zone.

  • Exfoliated graphite oxide (EGO) was prepared by graphite oxide in an aqueous solution of TMAOH. The hybrid graphene/Zn-Al LDH material was fabricated by the hydrothermal reduction of the solution of EGO, $Zn(NO_3)_2{\cdot}6H_2O$, $Al(NO_3)_3{\cdot}9H_2O$, urea, and trisodium citrate. That is, metal ions were absorbed on the surface of EGO, and Zn-Al LDH material was randomly dispersed on the surface of graphene along with a reduction process of EGO to graphene by hydrothermal treatment. The composition, morphology, and thermal property of the obtained graphene-based hybrid material were studied by FE-SEM, EDX, TEM, FT-IR, XRD, TGA, and DSC.

Mineral Chemistry of Talc from Different Origins in the Dongyang Talc Deposit (동양활석광상에서 산출되는 서로 다른 기원의 활석에 대한 광물화학)

  • Mineral chemistry of talc from the Dongyang talc deposits was studied to characterize the differences between dolomite-origin talc (talc I) and tremolite-origin talc (talc II). Average iron and aluminum contents are higher in talc II, 2.18 wt% FeO and 0.31 wt% $Al_2$O$_3$), than in talc I, 1.48 wt% FeO and 0.08 wt% $Al_2$O$_3$). Consistently lower Mg/(Mg+Fe+Mn) ratios and higher AI concentrations with uniform values of talc II compared to talc I seem to reflect the compositional differences of the original materials, tremolite and dolomite, respectively. Relative enrichment of Al as well as Fe in talc II compared to tremolite can be attributed to their immobile behaviors during alteration process and the rapid diffusion of hydrothermal fluids, which can accelerate instantaneous nucleation with immaturity growth of talc II. Increase in the concentrations of talc II can lower the ore grade by increasing concentrations of impure components such as AI and Fe, and by abundant presence of tremolite as well.

Browning and Pungent Taste Reduction Techniques in Onion Extract (양파추출물의 갈변 및 매운맛 억제기술)

  • The onion extractions of MIX treatment (5% $\beta$ cyclodextrin+1% calcium chloride+1% soluble starch mixture solution) using hot water (100${^{\circ}C}$ and 80${^{\circ}C}$) and ultrasonic treatment (25${^{\circ}C}$) incresed L values and decreased a and b values apart from the extraction methods. Extent of the browning reaction as caused by the MIX treatment (0.093) following 100${^{\circ}C}$ water extraction resulted in as low as 31% O.D. level, as compared to the control(0.296). Analysis of the pyruvic acid showed that the control had higher content of pyruric acid than MIX-treated samples. The MIX treatment had lower intensities and higher preferences of browning color and pungency taste compared to the control. The total and coliform microbial counts increased continuously during storage period, while the MIX treatment reduced the number of viable cells. Finally, it was concluded that the MIX treatment was highly effective in suppressing the undesirable browning color and pungent taste after processing, and the microbes increment during storage.

Processings of Intermediate Flavoring Substance from Low-Utilized Longfinned Squid (저활용 소형 창오징어를 이용한 풍미소재의 가공)

  • To develop natural intermediate flavoring substances, optimal processing conditions and qualities for two stage enzyme hydrolysate (TSEH) from low-utilized small longfinned squid were investigated. The optimal conditions for TSEH method were found as digestion with Alcalase (0.2% w/w-sample, pH 8.0) at 55$^{\circ}C$ 3 hours at the 1st stage and with Neutrase (exo-peptidase, 0.2% w/w-sample, pH 6.0) at 45$^{\circ}C$ for 2~3 hours at the 2nd stage. Among the method of water extract, autolytic extract and various kinds yields, transparency and organoleptic taste. From the results of chemical experiments and sensory evaluation, longfinned squid TSEH is flavorable as the natural intermediate taste-active substances for fisheries products such as soup base, squid-taste pasty and snacks.
