Biological Activity of Phenol Compound from a Cactus Cheonnyuncho (Opuntia humifusa) in Korea (천년초선인장으로부터 분리한 페놀성 화합물의 생리활성 효과)

  • After Cheonnyuncho (Opuntia humifusa) was extracted by 70% ethanol and partitioned with hexane, chloroform, ethyl acetate, butanol, and water in order, ethyl acetate fraction with excellent antioxidant activities was acquired. Ethyl acetate fraction was conducted by TLC, and the third spot of 10 spots was selected due to its best antioxidant activities. When the third fraction further isolated on silica gel chromatography, the fourth fraction of 10 fractions had the best antioxidant activities and purified by HPLC. The molecular weight on EI-MS, and $^1H$- and $^{13}C$-NMR spectrum showed that the purified compound was taxifolin. In addition, antioxidant activities were analyzed, and the purified compound from Cheonnyuncho was found to be effective as much as $\alpha$-tocopherol, BHA. According to the analysis on antibacterial activities, the purified compound also showed more activity than benzoic acid against all pathogenic bacteria.

Effect of Dandelion (Taraxacum Officinale) Extracts on the Intestinal Microorganisms of Streptozotocin-Induced Diabetic Rats (민들레추출물이 당뇨유발 흰쥐의 장내 미생물 균총에 미치는 영향)

  • This study was designed to test the effect of dandelion (Taraxacum officinale) extracts on the intestinal microorganisms of streptozotocin-induced diabetic rats. Male Wistar rats were divided into normal, diabetic control and dandelion extract groups. The extracts were prepared by water, ethylacetate and ether from leaf and root, respectively. Diabetes was induced by injecting streptozotocin (55 mg/kg BW, i.p.) in citrate buffer. The extract was supplemented in 11.45g of raw dandelion / kg diet for 50 days. The growth of Lactobacillus was more enhanced in dandelion leaf-water and leaf-ethylacetate extract group than that of the diabetic control group, whereas the growth of E. coli decreased. Results indicate that the dandelion extracts would be effective to improve the intestinal microorganisms.

Extractives from the Allium victorials (산마늘(Allium victorialis)의 추출성분)

  • Allium victorials var. platyphyllum were ground, extracted with 95% EtOH, concentrated, and fractionated with a series of n-hexane, methylene chloride, ethyl acetate, butanol and water on a separatory funnel. A portion of ethyl acetate soluble fraction was chromatographed on a Sephadex LH-20 and silica gel column using various solvent system as eluents. The isolated compounds were identified by TLC, $^1H-$, $^{13}C$-NMR, HMBC and EI-MS. Astragalin (compound 1), kaempferol (compound 2), quercetin (compound 3), ferulic acid (compound 4) were isolated from the ethyl acetate soluble fraction of A. victorials.

Induction of Apoptosis by Scolopendra subspinipes mutilans in Human Leukemia HL-60 Cells through Bcl-xL Regulation (왕지네(Scolopendra subspinipes mutilans)의 Bcl-xL 조절에 의한 HL-60 세포의 아폽토시스(Apoptosis) 유도)

  • The anticancer properties of Scolopendra subspinipes mutilans extract were investigated. The extract from S. subspinipes mutilans by 80% EtOH was fractionated with n-hexane, dichloromethan ($CH_2Cl_2$), ethylacetate (EtOAc), and butanol (BuOH) in order. The EtOAc fraction showed the highest inhibitory activity (about 80%) against human leukemia (HL-60) cell growth at $50\;{\mu}g/mL$. To explore the mechanism of cytotoxicity, we used several measures of apoptosis to determine whether these processes were involved in EtOAc fraction-induced HL-60 cell death. Our results showed EtOAc fraction induced cell shrinkage, cell membrane blebbing, apoptotic body, and DNA fragmentation. The EtOAc fraction gradually decreased the expression of anti-apoptotic Bcl-xL and led to the activation of caspase-3, -9 and cleavage of PARP. These findings suggest that S. subspinipes mutilans exhibits potential anticancer properties.

Antioxidant and Genotoxic Inhibition Activity of Ethanol Extract from the Inonotus obliquus (차가버섯 분획물의 항산화활성 및 유전독성 억제효과)

  • This study was performed to observe the antioxidative and genotoxic effect of the fractions from Inonotus obliquus using DPPH test and micronucleus assay. Stepwise fractionation of the ethanol extract from Inonotus obliquus was done by using hexane, chloroform, ethyl acetate, butanol and water to obtain effective fraction. Each fraction was tested in 1.5 ${\times}$ 10$^{-4}$ DPPH. Among six fractions, the ethyl acetate fraction showed the highest electron donating activities (46.5 $\mu\textrm{g}$/mL). The results on genotoxic effects on insoluble fractions and most of fractions showed cytotoxic effects more than 90% activity. These results suggest that some components contained in the Inonotus obliquus showed such activities and much more studies have to perform.

Inhibitory Effects of the Solvent Fractions from Persimmon Leaves on Xanthine Oxidise Activity (용매별 감잎 추출물의 Xanthine Oxidase 저해효과)

  • The inhibitory effects of xanthine oxidase by the methanol extract and the solvent fractions obtained from persimmon leaves were investigated. The inhibition ratio of xanthine oxidase was 78% by addition of 2.0mg/ml of the methanol extract. Among the solvent fractions, the ethylacetate fraction showed the strongest inhibitory effect against the xanthine oxidase, followed by the hexane fraction. The effect increased with addition of the ethylacetate fraction. At a concentration of 2.0mg/ml of the ethylacetate fraction, 65% of the enzyme activity decreased within 1.0 min of incubation with xanthine oxidase. But the activity of xanthine oxidise did not decrease significantly by the length of the incubation time.

Antibacterial Activities of Galla Rhois Extracts against Fish Pathogenic Bacteria (오배자 추출물의 어병 세균에 대한 항균 효능)

  • Galla Rhois was extracted with various solvents such as ethyl alcohol, ethyl acetate, n-hexane, and n-butyl alcohol, and antibacterial activity of the extracts were also tested. The ethyl acetate extracted Galla Rhois showed high antibacterial activities and was the most effective extract, was further fractionated into 8 subfractions with silica gel column chromatography. The subfractions 4 and 5 exhibited an excellent antibacterial activity against Streptococcus iniac KCTC3657.

Phenolic Compounds from the Fruits of Paulownia coreana Uyeki (오동나무 열매 추출물의 페놀성 화합물)

  • Paulownia coreana Uyeki fruits were collected, extracted with acetone-$H_2O$ (7:3, v /v), fractionated with n-hexane, methylene chloride, and ethyl acetate, then freeze dried to give some dark brown powder. The ethyl acetate soluble mixture was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. Spectrometric analysis such as NMR and MS including TLC were performed to establish the structures of the isolated compounds. From the ethyl acetate fraction, one phenolic acid (I) and five flavonoids (II~VI) and were isolated and elucidated. The antioxidative activities were tested on the isolated compounds, crude and fractionated extractives by DPPH radical scavenging method. The result showed that caffeic acid, kaempferol, luteolin, quercetin and the EtOAc soluble fraction exhibited higher activities than those of ${\alpha}$-tocopherol and BHT.

Antioxidative and Antimicrobial Activities of Castanopsis cuspidata var. sieboldii Extracts (구실잣밤나무 추출물의 항산화 및 항균 활성)

  • This study was designed to investigate the possible utilization of Castanopsis cuspidata as a source of antiseptic agents. The leaves of C. cuspidata, extracted by 80% ethanol, were sequentially fractionated with n-hexane, dichloromethane, ethylacetate, and n-butanol. In order to effectively screen for a natural preservative agent, we first investigated the antioxidant activities such as DPPH radical scavenging capacity, superoxide radical scavenging capacity, and xanthine oxidase inhibitory activity of the C. cuspidata ethanol extracts and fraction. Using a screening system, we found that the ethylacetate fraction had the strongest antioxidant activity, which followed a dose-dependent manner. The antimicrobial activities were shown in the ethylacetate fraction of C. cuspidata. Among the five fractions, the ethylacetate fraction showed the highest antimicrobial activities against microorganisms tested, which were Bacillus sublitis, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella enteritidis and Salmonella typhimurium. In addition, leaf extracts of C. cuspidata could be suitable for the development of food preservatives.

Physiological Activities of Water Extract and Solvent Fractions of an Edible Mushroom, Pholiota adiposa (검은비늘버섯 물 추출물 및 유기용매 분획물의 생리활성 효과)

  • This study was conducted in order to investigate the physiological activities, including antioxidative, fibrinolytic, thrombin inhibitory, and ${\alpha}$-glucosidase inhibitory activities of the water extract and solvent fractions isolated from Pholiota adiposa. The antioxidative activities of the water extract and water fraction were 57.57% and 48.27%, respectively. The fibrinolytic activity was strong only in the ethyl acetate fraction at 0.70 plasmin units/mL. The ethyl acetate fraction showed high thrombin inhibitory activity, and a-glucosidase inhibitory activity at 77.67% and 89.32%, respectively. The ethyl acetate fraction hydrolyzed both $A{\alpha}$ and $B{\beta}$ subunits of human fibrinogen, but did not show reactivity for the ${\gamma}$ form of human fibrinogen. Fibrinolytic activity of the ethyl acetate fraction was not decreased by heating for 10 min at $100^{\circ}C$.
