매생이 에탄올 추출물의 생리기능성과 아질산염 분해능

  • 매생이의 에탄올 추출물의 기능적 특성과 추출액의 플라보노이드와 페놀함량과의 상관성을 검토하고, 아질산염 분해능과 효소의 영향 등을 측정하였다. 매생이 에탄올 추출액의 클로로필 함량은 물추출보다 에탄올 추출의 경우가 높았으나, 플라보노이드 함량은 물추출액에서 약간 높게 나타났다. 추출액의 TPC함량은 TFC함량이 높을수록 높게 나타났으나 직접 비례하지는 않았다. 그리고 매생이에 함유된 아질산염 함량 측정결과와 아질산염 분해능 결파 비교로부터 에탄올 추출의 경우에는 아질산염 분해능이 시료자체의 아질산염 함량과 직접적인 영향이 없는 것으로 보였으며, 매생이 에탄올 추출액이 나타내는 아질산염 분해작용은 낮은 pH에서 높게 나타났으나, 매생이 자체에 존재하는 효소의 관여는 극히 미약하였다.

The Effects of Extraction Conditions on the Antioxidative Effects of Extracts from Campbell Early and Muscat Bailey A Grapevine Leaves (추출조건에 따른 캠벨얼리와 Muscat Bailey A 포도잎 추출물의 항산화 효과)

  • We investigated the physiological activities of extracts from Campbell Early and Muscat Bailey A (MBA) grapevine leaves. Total phenol and flavonoid contents were highest in ethanol extracts from MBA grapevine leaves compared to extracts from Campbell Early grapevine leaves. Specific polyphenols higher in ethanol extracts from MBA grapevine leaves include gallic acid, epicatechin, caffeic acid, naringin, and resveratrol. Resveratrol content from MBA grapevine leaves increased when extracted for more than two hours in ethanol and water. The hydroxyl radical scavenging ability of ethanol extracts was higher than the water extract from both strains of grapevine leaves. DPPH and total antioxidants were highest in ethanol extracts from MBA grapevine leaves among the other extracts. Therefore, these results suggest that ethanol extracts from MBA grapevine leaves are a highly valuable resource for the development of natural functional foods.

A Study on the Skin Improvement of Ethanol and Hot Water Extracts from Scutellaria baicalensis (황금 에탄올 및 열수 추출물의 피부 개선 연구)

  • In this study, we investigated the antioxidant activity, anti-inflammatory activity, whitening, moisturizing, wound-healing, cell proliferation and cell protective effects of 70% EtOH and hot water extract from Scutellaria baicalensis. For the anti-oxidative test, the 70% EtOH and hot water extract showed DPPH radical scavenging activities. In the anti-inflammatory tests, 70% EtOH and hot water extract inhibited the production of NO, pro-inflammatory cytokine (IL-6) and prostaglandin (PGE2). In addition, it was confirmed that the 70% EtOH and hot water extract inhibited the melanin production, and increased production of hyaluronic acid (HA), a moisturizing factor. As a result of cell migration and proliferation assay, 70% EtOH extract promoted the cell growth in HaCaT cell. Additionally, 70% EtOH and hot water extract showed cell protective effects against UVB, and 70% EtOH extract also showed cell protective effects agianst blue light. Based on these results, it is concluded that the 70% EtOH and hot water extract from Scutellaria baicalensis could be potentially applicable as anti-oxdiative, anti-inflammation, whitening, moisturizing, wound-healing, cell proliferation and cell protective effects in cosmetic natural materials.

Functional Properties of Medicinal Plant Extracts (한방식재료 추출물의 기능성)

  • The purpose of this study was to investigate the functional properties of medicinal plant extracts. Four kinds of medicinal plants, Dioscorea batatas(DB), Armeniacae Semen(AS) Crataegus pinnatifida Bunge(CP) and Ponciri Fractus(PF), were extracted with water and 70% ethanol and the extracts were tested for their electron donating ability(EDA), nitrite scavenging ability(NSA) and inhibitory effects on cancer cells(MDA cell and A549 cell)growth. EDA at 100-1,000 ppm of water extract ranged from 3% to 14%, 14% to 36%, 29% to 72% and 14% to 43%, and that of ethanol extract ranged from 9% to 62%, 27% to 59%, 33% to 89% and 14% to 44%, in DB, AS, CP and PF, respectively. NSA of extracts measured at various pH(1.2, 3.0, 4.2, 6.0) showed the highest ability in all extracts at pH 1.2 and decreased with increasing pH. The highest NSA of water extracts of 1,000 ppm at pH 1.2 was 6%, 31%, 55% and 44% and that of ethanol extract was15%, 32%, 69% and 52%, in DB, AS, CP and PF, respectively Inhibition ratio of water and ethanol extracts on MDA cell growth was 24% and 17%, 51% and 93%, 46% and 69%, and 48% and 47%, while that on A549 cell was 18% and 9%, 6% and 3%, 7% and 3%, and 43% and 11%, at 1,000 ppm, in DB, AS, CP and PF, respectively.

Antioxidant activities of ethanol extracts from barley sprouts (새싹보리 에탄올 농도별 추출물의 산화방지 활성)

  • Phenolic compounds and antioxidant activities of ethanol extracts from barley sprouts were evaluated in this study. Barley sprouts were extracted using water and ethanol in various concentration (25, 50, and 75%) using reflux extraction methods. Ultra performance liquid chromatography (UPLC) analysis showed that barley sprouts are mainly composed of rutin, gallic acid, ferulic acid, and ${\rho}$-coumaric acid. The 75% ethanol extracts had higher total polyphenol contents ($44.01{\pm}1.32mg/g$) and total flavonoid contents ($102.96{\pm}2.49mg/g$). 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity ($EC_{50}$ value: $1.65{\pm}0.02mg/mL$) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging activity ($EC_{50}$ value: $1.67{\pm}0.02mg/mL$) of the 75% ethanol extracts of barley sprouts were found to be the most effective. The 75% ethanol extracts of barley sprouts exhibited a strong reducing activity and ferric reducing antioxidant activity. As a result, the 75% ethanol extracts of barley sprouts showed stronger antioxidant activity than other extracts.

Inhibitory Effects of Maesaengi (Capsosiphon fulvescens) Extracts on Angiotensin Converting Enzyme and α-Glucosidase (매생이 추출물의 angiotensin converting enzyme 및 α-glucosidase 활성 저해 효과)

  • Physiological activities of hot water (MHW) and 80% ethanol (MEH) extracts from Maesaengi (Capsosiphon fulvescens) were investigated in this study. For the evaluation of antioxidant activities for MHW and MEH, 2,2-diphenyl-1-pic-rylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity were measured. DPPH radical scavenging activity and SOD-like activity of MHW, and MEH were increased weekly in a dose-dependent manner, and were about 10.8, 13.8, 62.4, and 27.1% at 10 mg/ml, respectively. The angiotensin converting enzyme (ACE) inhibitory activities of MHW and MEH were about 5.9% and 49.7% at 1 mg/ml, respectively. The ${\alpha}$-glucosidase inhibitory effect of MHW and MEH were about 1.4% and 67.3% at 1 mg/ml, respectively. To determine the influence of MHW and MEH on alcohol metabolizing activity, the generating activities of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were measured. Facilitating rates of ADH activity by MHW and MEH were increased weekly in a dose-dependent manner and ALDH activities were not detected. Elastase inhibitory activities of MHW and MEH were 75.9% and 51.2% at 10 mg/ml, respectively.

Antioxidant and Antimicrobial Activities of Smilax china Leaf Extracts (청미래 덩굴 잎 추출물의 항산화 및 항균 활성)

  • Hot-water and 70%-ethanol Smilax china leaf extracts were prepared, and their total polyphenol and flavonoid contents, DPPH-radical-scavenging ability, nitrite-scavenging ability, and antimicrobial activity were determined. The total polyphenol contents of the hot-water and ethanol extract were $5.433{\pm}0.171$ and $13.060{\pm}0.110mg/g$, respectively; their flavonoid contents were $1.599{\pm}0.017$ and $3.005{\pm}0.084mg/g$; their DPPH-radical-scavenging abilities, assayed at 1.0 mg/mL, were 33.6 and 92.3%; and their nitrite-scavenging abilities, assayed at 0.1-2.0 mg/mL, were 37.9-61.6 and 38.4-77.8%. The 70%-ethanol extract showed higher antimicrobial activity than the hot-water extract. The antimicrobial activities were high in Bacillus cereus, Vibrio parahaemolyticus, Salmonella typhymurium, and Staphylococcus aureus, in that order. The antimicrobial substances in the two extracts were maintained after heating at $65-125^{\circ}C$ for 30 min.

Physiological activities of Lespedeza cuneata extracts (야관문 추출물의 생리 활성)

  • This study was conducted to examine the physiological activities of Lespedeza cundata extracts. The extraction yield of 50% ethanol extract (17.60%) was higher than that of hot water extract (12.60%). The total phenolic and total flavonoid contents of the 50% ethanol extract were 242.26 mg/g and 160.73 mg/g, respectively. The DPPH radical scavenging activities of the hot water and 50% ethanol extracts were 92.07% and 96.38%, respectively. The superoxide radical scavenging activities of hot water and 50% ethanol extracts on $250{\sim}1,000{\mu}g/mL$ were 54.89~85.68% and 44.50~94.46%, respectively. The tyrosinase inhibition activity of the 50% ethanol extract at $1,000{\mu}g/mL$ (63.31%) was the highest. The nitrite scavenging activity of the 50% ethanol extract was higher than that of the hot water extract. The nitric oxide production of 50% ethanol extract ($7.15{\sim}20.61{\mu}M$) improved with an increase in the treatment concentration. The hot water and 50% ethanol extracts at $1,000{\mu}g/mL$ inhibited the proliferation of the cancer cell lines A549, HeLa, Hep3B, and Sarcoma180. There results suggest that the 50% ethanol Lespedeza cuneata extracts may be useful as a functional food material in the food industry.

Quality Characteristics of Fried Fish Paste Added with Ethanol Extract of Onion (양파 에탄올 추출물을 첨가한 튀김어묵의 품질 특성)

  • The effects of ethanol extract of onion (EEO) on the qualify of fried fish paste were investigated. EEO was added at 0, 1, 3 and 5% level. Quality attributes including moisture content, pH, acid value, TBA value, volatile basic nitrogen, color value, viable cell count and sensory evaluation were analyzed. Moisture content was not changed by addition of EEO. Increasing the amount of EEO, pH, acid value and TBA value of fried fish paste tended to decreased but volatile basic nitrogen increased. L-value was decreased and a-value and b-value were increased by addition of EEO. Viable cell count was decreased by addition of EEO. In sensory evaluation, the higher amount of EEO obtained higher favorite score in flavor and taste, and 3% EEO had the best score in overall acceptance. These results suggest that EEO can be applied to fried fish paste for the purpose of high quality and functionality.

Hepatoprotective Effect of Curdrania tricuspidata Extracts against Oxidative Damage (산화적 손상에 대한 꾸지뽕나무 잎, 열매 및 줄기 추출물의 간세포 보호효과)

  • We investigated the antioxidant and hepatoprotective effects of extracts from the leaves, stems, and fruit of Cudrania tricuspidata (CT) against $H_2O_2$ or ethanol-induced oxidative damage. The total polyphenol and flavonoid content was the highest in the 80% ethanol extracts from the leaves of the plant (CTL80). Also, the radical scavenging activity of DPPH and ABTS in the CTL80 was significantly higher than that of the non-treated control. To determine the hepatoprotective effects of CT in $H_2O_2$ and ethanol-induced oxidative damage, cell viability was measured using an XTT assay. Pre-treatment of CTL80 significantly increased cell viability compared with the non-treated control cells by 71.21% and 80.40%, respectively. The data suggests that CTL80 exhibits hepatoprotective antioxidant effects. Therefore, CTL80 may be considered a potential agent to control $H_2O_2$ or ethanol-induced liver damage.
