Applications of Focused Ion Beam for Biomedical Research (의생물 연구 분야에서 집속이온빔장치의 응용)

  • A focused ion beam (FIB) system produces a beam of positive ions (usually gallium) which are heavier than electrons and can be focused by electrostatic lenses into a spot on the specimen. With its ability milling of the specimen material by 10 to 100 nm with each pass of the beam, FIB is widely adopted in materials science, semiconductor industry, and ceramics research. Recently, FIB has been increasingly employed in the field of biomedical sciences. Here we provide a brief introduction to FIB and its applications for a wide variety of biomedical research. The surface of specimen can be in situ processed and quasi-real time visualized by two beam combination of FIB and field emission scanning electron microscope (FESEM). Due to its milling process, internal structures can be exposed and analyzed: yeast cells, fungus-inoculated wheat leaf, mannitol particles in inhalation aerosols, and oyster shell. Serial blockface tomography with the system kindles 3-dimensional reconstruction researches in the realm of nervous system and life sciences. Two-beam system of FIB/FESEM is a versatile tool to be utilized in the biomedical sciences, especially in 3-dimensional reconstruction studies.

Antibiotic Susceptibility of Vibrio parahaemolyticus Isolated from Commercial Marine Products (시판 어패류에서 분리한 장염 비브리오균의 항생제 감수성 특성)

  • Ninety three strains (4.2%) of Vibrio parahaemolyticus were isolated from 2,239 samples of commercial marine products during 2006-2008 in Seoul, Korea. We examined 16 antimicrobial susceptibilities of 93 V. parahaemolyticus isolates. Antibiotic resistance of V. parahaemolyticus was most frequently observed to ampicillin (93.5%), followed by cephalothin (90.3%), streptomycin (87.1%), ticarcillin (55.9%), and amikacin (40.9%). Antibiotic susceptibility was most frequently observed to nalidixic acid, chloramphenicol and ampicillin/sulbactam (100%), followed by trimethoprim/sulfamethoxazole (98.9%), gentamicin and tetracycline (82.8%), and ceftriaxone (63.4%). In addition, the isolates also displayed intermediate resistance to kanamycin (79.6%), ciprofloxacin (64.5%), amikacin (47.3%) and cefoxitin (43.0%). All isolates were resistant to more than two drugs. The most prominent multiple drug resistance was 3 drug resistance (37.6%), followed by 4 drug (24.7%), 5 drug (17.2%), and 6 drug resistance (11.8%). The most prominent multiple drug resistance pattern was the cephalothin-streptomycin-ampicillin resistance pattern (22.6%), followed by cephalothinticarcillin-streptomycin-ampicillin (18.3%) and cephalothin-ticarcillin-streptomycin-ampicillin-amikacin (9.7%). Multiple drug resistance patterns of V. parahaemolyticus from marine products require continuous monitoring.

Changes in Enzymatic Activities during Eoyukjang Fermentation (어육장 발효 시 생성되는 효소의 활성 변화)

  • Eoyukjang is a traditional sauce-type of Korean food that is similar to a soybean sauce made from fermented soybeans, and it is produced from a fermented mixture of sea foods, meats, and meju (soybean paste). This study examined periodical changes in the enzymatic activities of ${\alpha}$-amylase, esterase, ${\beta}$-glucosidase, protease, lipase, and lipoxygenase within the culture broth and solids of eoyukjang during 1 year of fermentation. The eoyukjang solids had 234-532% higher protein content than the culture broth. The specific activities of ${\alpha}$-amylase, esterase, ${\beta}$-glucosidase, and protease increased in both the culture broth and solids. Particularly, in the culture broth, ${\alpha}$-amylase, esterase, ${\beta}$-glucosidase, and protease activities rapidly increased (3- to 8-fold) until 10 months of fermentation, and then drastically decreased. However, the activities of lipase and lipoxygenase in both the culture broth and solids were less than 0.05 unit/mg of protein, respectively, throughout fermentation; thus, their activity levels were low and changed little over the 12 months. Overall, while the solids had higher protein content than the culture broth, the broth had greater enzyme activity levels during eoyukjang preparation.

Associations of Cognitive Function and Dietary Factors in Elderly Patients with Alzheimer's Disease (알쯔하이머병 노인들의 인지기능과 관련된 식이 요인)

  • The purpose of this study was to investigate nutrients or food factors related to cognitive function of elderly having Alzheimer's disease. In this study 38 subjects who were over 65 years old have participated in dementia clinic at A medical center. After they were diagnosed to Alzheimer's Disease (AD) through blood analysis, neuropsychological test, brain image and interview by medical specialist, we examined for their general information, anthropometry, blood pressure and dietary intakes. Dietary intakes were investigated using the 24-hour recall record. Energy intake was adequate and the energy composition of carbohydrate, protein and fat was 60.8 : 16.2 : 23.0, but dietary intakes of calcium, vitamin A and folate were less than 75% of the recommended intake levels for Koreans. The multiple regression analysis adjusted with age, sex and educational level showed that cognitive function was positively related to intakes of zinc, fishes and shellfishes, beans & nuts, sugars and fats, and negatively related to intakes of plant calcium and eggs. These results indicate that intakes of specific nutrients or food groups are associated with the specific domains of cognitive function in elderly with AD.

Evaluation of dietary intake in Korean adults according to energy intake from eating-out: Based on 2013~2014 Korea National Health and Nutrition Examination Survey (성인 남녀에서 외식을 통한 열량 섭취 정도에 따른 영양소 및 식품군별 섭취 상태 평가 : 2013~2014 국민건강영양조사 자료를 이용하여)

  • Purpose: The objective of the present study was to evaluate nutrient and food group intakes in Korean adults according to energy intake from eating-out. Methods: This study was a cross-sectional study based on the 2013~2014 Korea National Health and Nutritional Examination Survey, and study subjects were 5,186 males (2,151) and females (3,035) aged 19~64 years. Energy intake from eating-out was analyzed using the 24-hour dietary recall method, and groups were classified by quartile according to energy intake from eating-out. Results: Subjects who had more energy intake from eating-out tended to be younger, highly educated, have a higher income, and have higher alcohol consumption. Total energy intake and fat intake density of the highest quartile group of energy intake from eating-out were higher than those of the other groups. However, fiber, vitamin $B_1$, calcium, phosphorus, potassium, and iron intake density levels were significantly lower in the highest quartile group of energy intake from eating-out. The highest quartile group of energy intake from eating-out consumed significantly more meat and beverages compared to the other groups. In addition, regarding diet quality (Index of nutritional quality), significantly lower vitamin $B_1$, calcium, phosphorus, and iron levels were observed in the highest quartile group of energy intake from eating-out compared to those in the other groups. Conclusion: In conclusion, in Korean adults, consumption of eating-out decreased dietary quality, including vitamin $B_1$, calcium, phosphorus, and iron levels. Further studies are needed to confirm these findings.

Quantitative Cell Count of Vibrio vulnificus Cells Based on MPN-PCR Method (MPN-PCR 방법을 이용한 Vibrio vulnificus 균수 정량분석)

  • The objective of this study was to establish a quantitative count method of Vibrio vulnificus cells. Plate count method is often used to count the number of V. vulnificus cells using thiosulfate citrate bile salts sucrose (TCBS) agar plate. However, this method is unsuitable for counting V. vulnificus cells due to growth inhibition and cell injuries in TCBS medium. In this study, we suggested a most probable number-polymerase chain reaction (MPN-PCR) method using alkaline peptone water medium for the quantification of V. vulnificus. This MPN-PCR method showed 2 log higher cell number than TCBS agar plate method. Similar results were also found in the control using, Luria-Bertani agar containing 2% NaCl. Thus, this MPN-PCR method can be used a sensitive method for quantitative count of viable V. vulnificus cells in fish and shellfish samples.

Prevalence of Toxin Genes and Profiles of Antibitoc Resistance in Vibrio vulnificus Isolates from Fish, Fish Tanks, and Patients (어류, 수족관수 및 환자에서 분리된 Vibrio vulnificus의 독소유전자 분포 및 항생제 내성)

  • Prevalence of toxin genes and profiles of antibiotic resistance in Vibrio vulnificus were investigated for prevention of Vibrio sepsis and selection of effective antibiotics. A total of 23 V. vulnificus strains were isolated from Vibrio sepsis patients, fish, and water samples collected from fish tanks in restaurants in Jeonnam province during 2015-2017 period. Prevalence of toxin genes including, RtxA, viuB and vvhA were assessed and susceptibilities to 15 different antibiotics were determined. As a result of the toxin gene profile, the RtxA toxin gene was detected in 19 (82.6%) out of 23 strains, and vvhA and viuB toxin genes were positive in all strains. These results showed that V. vulnificus tested in this study possessed at least one more toxin gene, and the toxin gene detection rate was higher than in previous reports. Therefore, there is always a risk of Vibrio sepsis through eating fish or having contact with aquarium water at seafood restaurants. Especially, it was deemed necessary to provide preventive education about Vibrio sepsis for workers in such restaurants. The results of antibiotic susceptibility tests presented 94.4% resistance to cepoxitin antibiotics but all strains showed susceptibility to 14 kinds of antibiotics including chloramphenicol and tetracycline. The currents antibiotic therapy using chloramphenicol and teteracycline against Vibrio sepsis was judged to be useful.

Effects of Nutritional Education on Food Behavior of Unbalanced Diet Children : An Investigation of Children at Elementary Schools in Incheon (영양교육이 편식 아동의 식행동에 미치는 효과 - 인천지역 초등학생을 중심으로 -)

  • This study investigated the effects of a 7 week nutritional educational program on the unbalanced dietary habits, tastes, dietary attitudes, and nutritional knowledge of 127 fourth grade students. The subjects were selected from seven elementary schools that were under the supervision of the Dongbu Office of Education in Incheon. Results showed that most of the subjects were fully aware of the fact that they were on an unbalanced diet. It was also revealed that the main reason for their unbalanced diet was that they tended not to eat foods that did not fit their tastes. After implementation of the 7 week nutritional education, the subjects'unbalanced dietary habits significantly decreased, but their desires to fix their unbalanced diet failed to increase. The subjects'tastes showed a positive change in their perception towards vegetables, seafood, and seaweed to a significant degree; however, their appetite far meat, eggs, beans, frozen floods, and processed goods did not show any noticeable change. A significant increase was also found on the level of the subjects'nutritional knowledge but not in their dietary habits and dietary attitudes.

Analysis of General Composition and Harmful Material of Protaetia brevitarsis (흰점박이꽃무지(Protaetia brevitarsis)의 일반성분 및 유해물질 분석)

  • To evaluate Protaetia brevitarsis as a food material, we investigated its composition of nutritional and harmful components. Analysis of nutritional composition (moisture, crude protein, crude fat, crude ash, crude fiber, and carbohydrates) showed that the content of crude protein, fat, fiber, and ash were $57.86{\pm}0.01$%, $16.57{\pm}1.81$%, $5.31{\pm}0.10$%, and $8.36{\pm}0.10$% in Protaetia brevitarsis powder, respectively. Amino acids were composed of 17.68% essential and 33.97% non-essential in Protaetia brevitarsis powder. Protaetia brevitarsis powder contained 61.10% unsaturated fatty acid with oleic acids. Additionally, Protaetia brevitarsis powder had a large quantity of minerals related to body organization, such as K (1597 mg/100 g), P (724.1 mg/100 g), Mg (366.3 mg/100 g), and so on. We also confirmed that all bacteria and all heavy metals analyzed in this study, except for very small amount of Hg ($0.1{\pm}0.042$ mg/kg), were not detected in the lysophilized Protaetia brevitarsis powder.

STUDIES ON THE VARIATION IN CHEMICAL CONSTITUENTS OF THE SEA MUSSEL, MYTILUS EDULIS (진주담치 Mytilus edulis의 성분에 관한 연구)

  • This paper deals with the proximate composition of the cultured sea mussel (Mytilus edulis) which are distributed along the whole coastline of Korea, particularly abundant in the southern coast Studies on seasonal variation in chemical constituents of the mussel at Northern Bay of Choongmu were carried out from March to December 1968 and the results obtained are as follows: 1. Moisture content in mussel meat was $78.5\%$ on the average; the maximum amounted to $81.3\%$ during May-June, while the minimum was $77.8\%$ in September. 2. Crude protein was in the range of $10.9-13.7\%$; the maximum was In September-October, the minimum appeared in March, and the average value was $12.8\%$. 3. Lipids on the average was $2.5\%$ and there was no markable difference .between the high and low contents. 4. Total sugar was $5\%$ during September-October in its highest, while there appeared minor contents during winter season. 5. Crude ash on the average was $1.5\%$ the and maximum was in November-December. As for minerals in the ash, $963mg\%$ of phosphate, $82mg\%$ of calcium, and $188mg\%$ of iron were found respectively. 6. pH was in the range of 6.02-6.55, but it generally declined to acidity in the summer season. 7. In amino acid contents, there found 16 kinds; $710mg\%$ of glutamic acid, $696mg\%$ of aspartic acid, $383mg\%$ of Iysine, $225.4mg\%$ of valine, $225.1mg\%$ of proline, etc. 8. The amounts of Protein, lipids and total sugar are tend to increase from August to October, particularly in September.
