Anti-diabetic Activity of Polysaccharide from Salicornia herbacea (함초 다당체의 항당뇨 활성)

  • The present study investigated the effect of physiologically active polysaccharide (SP1) isolated from Salicornia herbacea on streptozotocin-induced diabetic rats. Male Spraque-Dawley rats were divided into four groups which were normal control group (NC), diabetic control group (DC), diabetic CSP group (DCSP), and diabetic SP1 group (DSP1). Animals were administrated with 2% experimental drinks for 6 weeks. The levels of glucose, triglyceride, total cholesterol, and high density lipoprotein (HDL)-cholesterol in the serum were measured before and after intake of test compounds. The levels of glucose and triglyceride in the DSP1 were significantly lower than those in the DC by 25% and 20%, respectively. The levels of total cholesterol and high density lipoprotein (HDL)-cholesterol in the DSP1 were similar to those in the DC. These results suggest that SP1 substantially exhibit anti-hyperglycemic and anti-hypertriglyceridemic activity in diabetic rats. Therefore SP1 is believed to show remarkable anti-diabetic effect on streptozotocin-induced diabetic rats.

Comparison of Anti-Oxidative Activities of Perilla frutescens Extracts by Extraction Methods (추출 방법에 따른 자소엽 추출물의 항산화 효과 비교)

  • Perilla frutescens (P. frutescens) is one of evergreen shrubs belonging to the Labiatae and is grown wildly in Korea. This study was carried out to evaluate the anti-oxidative effects of Perilla frutescens Extracts by Extraction Methods (water, heating and sonication). Anti-oxidative effects were measured using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and total phenol content. Cell viability and hepatoprotective effects were identified by 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) assay. Among various extracts, P. rutescens extracts by sonication showed the highest DPPH radical scavenging activity at $5000{\mu}g/mL$. Total phenolic content in P. frutescens extracts by sonication was $51.60{\pm}1.06mg\;GAE/g$ extract. However, P. frutescens extracts did not show hepatoprotective effects. This study identified anti-oxidative effects of P. frutescens extracts by sonication, and it would be necessary to perform further studies of P. frutescens extracts by sonication.

Antifungal Activity of Methylene Chloride Fraction of Pimpinella brachycarpa Against Aspergillus niger (참나물 Methylene Chloride 분획의 Aspergillus niger에 대한 항진균 활성)

  • In order to develop safe and economic novel antifungal agents, we prepared 73 methanol extracts from medicinal and edible herbs and examined their 365 solvent fractions using n-hexane, methylene chloride, ethylacetate, butanol and water residue based on the sequential organic solvent fraction method. When using the various fractions in the screening step for antifungal activity, we discovered ethylacetate fraction of Morus alba L., methylene chloride fraction of Pimpinella brachycarpa (MCPB), and n-hexane fraction of Salvia miltiorrhiza Bunge, which all have activities in methanol extracts, as potential sources of antifungal agents. Amongst these, the antifungal activity of P. brachycarpa has not to date been reported on. In addition, the mycelial growth inhibition and spore germination inhibition activities of MCPB against A. niger were confirmed by disc-diffusion assay in a 10 day culture. The MIC and MFC of MCPB were determined as 0.25 and 0.5 mg/ml, respectively. The MCPB has no hemolytic activity against human RBC at 0.5 mg/ml and glycoside-flavonoids are theorized to be active constituents. These results suggest that MCPB has a prominent antifungal activity and that the application of sequential organic solvent fractions, instead of simple natural product extracts, is useful in the screening process of novel bioactive substances.

Effects of Vitis coignetiae on the Quality and Antihypertension of Vitis hybrid Red Wine (쉐리단 포도주의 항고혈압성과 품질에 머루첨가의 영향)

  • The goal of this study was to develop a high value Korean red wine possessing antihypertension activity. The effects of some medicinal plants and grapes on the alcohol fermentation process and the angiotensin I-converting enzyme (ACE) inhibitory activity of Vitis hybrid red wine were investigated. Various Vitis hybrid red wines were vinified by the fermentation of a mixture of Vitis hybrid must and some medicinal plants and grapes at $25^{\circ}C$ for 10 days. Of these red wines, the Vitis hybrid-Vitis coignetiae red wine exhibited a high ethanol content of 12.0% and had a good level of acceptability. It also showed a high antihypertensive ACE inhibitory activity of 68.5%. After post-fermentation of 60 days, the ACE inhibitory activities of the Vitis hybrid-Vitis coignetiae red wine exhibited the highest ACE inhibitory activity of 80.7% ($IC_{50}$: 28 mg/mL) and also had the best acceptability. The $C_{18}$ solid phase extracts of the Vitis hybrid-Vitis coignetiae red wine, after 60 days post-fermentation, showed clear antihypertensive effects on spontaneously hypertensive rats. Our results reveal that the Vitis hybrid-Vitis coignetiae red wine has the potential to become a new functional red wine due to its good acceptability and high antihypertensive activity.

Immunomodulating Activity of Salicornia herbacea Extract (함초 추출물의 마우스 면역 증강 활성)

  • Immunomodulating effect of Salicornia herbacea extract on the mouse splenocytes was investigated. Crude S. herbacea polysaccharide extract (CSP) and other kinds of fine S. herbacea polysaccharides (SPI and SPII) were prepared from S. herbacea by hot water extraction and further ultrafiltration and gel filtration chromatography. In vitro experiment, the mouse splenocytes and separated T cells were treated with CSP, SPI or SPII (0.5, 1, 2, 4 mg/ml). In vivo experiment, three different S. herbacea extracts were orally administrated everyday for one week. For the basic data, body weight and physiological parameters such as organ weight and spleen index were observed. The proliferation of the cells was used as an index for immunemodulating activity and the effect of proliferation was evaluated using MTS assay. The CSP, SPI and SPII directly induced the proliferation of splenocytes and separated T cells in a dose-dependent manner. In results, the proliferation was more increased in the SPI and SPII treated cells than in the CSP treated cells. The best proliferation was shown in the splenocytes cultured with SPI at the concentration of 4 mg/ml for 24 hr. The proliferation of splenocytes and separated T-cells was higher (3.2 and 3.5 times, respectively) than the control. Moreover, when the mouse splenocytes were treated with mitogen, the efficient proliferation was shown in the splenocytes cultured with SPI. In conclusion, polysaccharides from S. herbacea showed a substantial immunomodulating activity in the mouse immune cells.

Biological Activities of Essential Oil from Chamaecyparis obtusa (편백(Chamaecyparis obtusa) 정유의 항균, 항염, 항산화 효과)

  • The essential oil from Chamaecyparis obtusa was investigated for biological activities in anti-oxidative, anti-inflammation and antibacterial method, respectively. The Growth inhibitory effect of C. obtusa oil on the bacteria was evaluated with MIC (minimum inhibitory concentration), $IC_{50}\;(50\%$ inhibitory concentration), and paper disc method. Two kinds of gram positive strains and two kinds of gram negative strains were used in this study. Gram positive strains were B. subtilis and S. aureus. and Gram negative strains were E. coli and P. aeruginosa. Gram positive strains showed much more intensive effect than gram negative strains. Anti-oxidative effect was investigated with DPPH (1,1-diphenyl-2-picrylhidrazyl) in methanol based and $IC_{50}\;was\;0.78\%.$ Our results suggest that the essential oil from Chamaecyparis obtusa has effects on anti-bacterial, anti- oxidative and anti-inflammation in in vitro and in uiuo. Then this material could be expect synergic effect with other candidated extracts and oils.

Exploration of Preservatives that Inhibit Wood Feeding by Inhibiting Termite Intestinal Enzyme Activity (흰개미 장내 효소 활성 억제로 목재의 섭식을 저해하는 보존 처리제의 탐색)

  • In Korea, damages to wooden cultural properties by termites have been reported all over the country, including Ulleungdo Island and Jeju Island. In order to reduce the damage caused by termites, most cultural properties damaged by them are treated with fumigation or drug treatment on the soil to kill or repel them. The number of treatments is decreasing due to cost and safety problems, and new methods needed to cope with this situation. After extracting an enzyme by selecting only worker termites from the termites collected in Shinan, Jeonnam, as a result of measuring the enzyme activity using xylan of cellulose and hemicellulose that are the components of wood, the activity of termite intestinal enzymes in xylan was higher than that of cellulose having a high molecular weight. Therefore, in this study, as a result of exploring over 600 species of medicinal plant extracts that inhibit the activity of termite intestinal enzymes using xylan as a substrate, the inhibitory effect was significant in Borneolum Syntheticum, Ephedra sinica, and Menthol. Selected Borneolum Syntheticum, Ephedra sinica, and Menthol's extracts not only inhibited the activity of termite intestinal enzymes, but also confirmed that they have insecticidal activation and inhibitory effects on feeding in the result of the direct treatment.

The Physiological Activities of Bark Extract of Albizia julibrissin (자귀나무(A. julibrissin) 수피 추출물의 생리활성)

  • Three bark extracts of Albizia julibrissin were prepared using water (AW), 70% (v/v) ethanol (AE), and hot water (AHW). Organic solvent fractions were analyzed for total flavonoids and polyphenols, antioxidant activities, and inhibitory activities against xanthine oxidase. Total flavonoid and polyphenol contents of the AHW extract were 8.57 mg/g and 108.67 mg/g, respectively. The SOD-like activities of all extracts, assayed at 1.0 mg/mL, were 10.46-16.73%. The nitrite-scavenging ability of the AHW extract, assayed at pH 1.2, was 60.82%, and the $IC_{50}$ value was $770.18\;{\mu}g$/mL. The electron-donating ability of the AHW extract, at 0.3 mg/mL, was 92.30%; the $IC_{50}$ values of the AW and AHW extracts were $31.31\;{\mu}g$/mL and $36.22\;{\mu}g$/mL, respectively; thus higher than that of ascorbic acid ($39.06\;{\mu}g$/mL). Xanthine oxidase inhibition by the AHW extract, at 1.0 mg/mL, was 94.05%. These results indicate that the AHW of A. julibrissin has potential as a natural antioxidant, for addition to foods and nutraceuticals.

Variation of Phenolics Contents and Antioxidant Activity of Vaccinium oldhamii Miq. (국내 자생 정금나무의 페놀류 함량과 항산화 활성의 다양성)

  • Vaccinium oldhamii Miq. is a Korean native tree, which is deciduous and shrub tree with broad leaf. Fruits were used primarily for edible or medicinal purposes for bladder infection in Korea and China. However, there is a lack of information on the degree of variation of phytochemical contents and antioxidant activity of these native clonal. We investigated morphological characteristics, phytochemical contents (polyphenol, flavonoid, anthocyanin) and antioxidant activity for the fruits of 24 individual variation from 4 different regions in Korea. The results in terms of horticultural traits and biochemical characteristics showed significant differences among populations and among individuals within population. The total phenolic contents ranged from 4.40 to 10.58 mg GAE/g and the total flavonoid contents ranged from 2.02 to 8.09 mg NE/g. The total anthocyanin contents ranged from 232.5 to 684.3 mg CGE/100g. Reducing power fruits ranged from 0.15 to 1.43 and DPPH free radical scavenging activity ranged from 49.1 to 97.5% at 400 ppm. Results revealed that the region of origin greatly influenced the phytochemical contents and antioxidant activity of V. oldhamii, which might help for the selection and validation of the most productive cultivars in the food and pharmaceutical industries.

Investment Beneficial Analysis of Culture of Mountain Medical Plant Resources - Open field Culture of Schizandra - (산지약용식물자원의 재배 투자 수익성 분석 - 오미자 노지재배 -)

  • This study is to give informations about management Situations and investment returns to someone who will newly cultivate schizandra or expand area of culture. This study used IRR, B/C Ratio and NPV for beneficial analysis. We surveyed fifty one among schizandra cultivation farmhouses in chief producing districts Gangwon Inje, Gyeongbuk Munkyung, Kyungnam Sancheong and Jeonnam Whasoon in Korea from May till September in 2009. So this study surveyed about inputted labors and materials, land price and etc. by working process to calculate producing and operating cost. Fruits of schizandra are sold from 5,000 won to 6,000 won per kg at locals. And IRR(Internal Rate of Returns), B/C Ratio and incomes ratio is calculated by three senarios 5,000 won, 5,500 won and 6,000 won per kg at locals. Discount rate is 3.00%. As the result of this study IRRs are -16.00%, -6.91%, 0.40% one after another and B/C Ratios are 0.81, 0.89, 0.97 one after another. And schizandra cultivation isn't profitable. When price of schizandra fruits is 6,200 won per kg, IRR is 3.00% and B/C Ratio is 1.00. Therefore, if price of schizandra fruits is over 6,200 won per kg, we judge that it is profitable. And income ratio is 23.9% on 6,200 won per kg.
