The effects of tetracycline gel with and without citric acid on periodontally diseased root surface - in vitro study (테트라싸이클린 젤 및 구연산함유 테트라싸이클린 젤의 도포가 치주염에 이환된 치근표면에 미치는 영향에 관한 실험 연구)

  • The purpose of the present study was to evaluate the effects of 5% tetracycline(Tc) with or without citric acid on periodontally diseased root surfaces. Six single-rooted teeth extracted from one patient was selected and received thorough scaling and root planning, followed by saline irrigation. Each one tooth was divided into eight fragments with a thin separating disc. Total 48 fragments were prepared and setted into 4 groups for this study. Group I (control group)were treated with saline. Group II were treated with 5% Tc gel. Group III were treated with 33% citric acid-5% Tc gel. Group IV were treated with Tc solution. All the specimens are evaluated under Scanning Electron Microscope(SEM). Group I showed large amount of debris in spite of thorough scaling and root planing, but Group II, III & IV showed clean and soft root surface texture. In higher magnification(x3, 000), Group II, III & IV showed nunmerous dentinal tubules, especially Group IV showed collagen fibrils. In the present study, Tc gel and Tc gel with citric showed clinically successful result when treated on periodontally diseased root surface, in vitro.

Roxithromycin Treatment of Tsutsugamushi Disease (Scrub Typhus) in Children (소아 쯔쯔가무시병에서 록시쓰로마이신 치료)

  • Purpose : Although chloramphenicol and doxycycline have been used for the treatment of tsutsugamuchi disease, a difficulty exists in determining which drugs to use in treating children because of potential complications such as aplastic anemia or teeth discoloration. We evaluated the effect of roxithromycin, a macrolide antibiotic, on tsutsugamushi disease in children. Methods : A retrospective analysis was conducted on 39 children with tsutsugamuchi disease(scrub typhus) who were treated with doxycycline(DC), chloramphenicol(CM), or roxythromycin(RM) between 1991 and 2000. We divided the patients into a DC-treated group(DC group; 16 children), a CM-treated group(CM group; 14 children), and RM-treated group(RM group; 9 children) and compared these groups. Results : Most cases(97%) developed in October and November. Fever and rash were observed in all 39 cases and an eschar was noted in 36 cases(92%). No statistical differences could be found between the three groups in mean age, duration of fever before admission, white blood cell(WBC) count, and complications including abnormal liver enzymes. In most cases defervescence after treatment was within 24 hours(34 cases, 87%), and during 24-48 hours in two cases in the DC group, one in the CM group, and two in the RM group(no statistical difference). Conclusion : Roxythromycin was as effective as conventional doxycycline or chloramphenicol, in children with scrub typhus and may be safer to use.

Antibiotic Uptake by Plants from Soil Applied with Antibiotic-Treated Animal Manure (가축분뇨 시용 토양에서 식물체의 축산용 항생물질 흡수양상)

  • Food contamination through antibiotic uptake by plants has been one of the major concerns regarding animal manure amendment to agricultural land. Antibiotic uptake by plants was tested with three veterinary antibiotics such as chlortetracycline (CTC), tylosin (TYL), and sulfamethazine (SMT) and three plants such as lettuce, tomato, and hairy vetch. Antibiotic-treated swine slurry was applied to a greenhouse soil before transplanting or sowing the plants. The treated antibiotic concentrations to the swine slurry were 22.9 mg CTC $L^{-1}$, 27.8 mg TYL $L^{-1}$, and 32.4 mg SMT $L^{-1}$. Lettuce was harvested on 64 days after transplanting and 94 days for tomato and hairy vetch. Concentrations of CTC were 3.4 ng $g^{-1}$ for lettuce and 0.7 ng $g^{-1}$ for tomato on a fresh weight basis. For TYL, 20.1 ng $g^{-1}$ from lettuce leaves and 3.0 ng $g^{-1}$ from whole hairy vetch were detected. Sulfamethazine showed greatest concentration among the three antibiotics, 63.3 ng $g^{-1}$ in lettuce leaves, 30.2 ng $g^{-1}$ in tomato fruits, and 20.9 ng $g^{-1}$ in hairy vetch. The results imply that antibiotic uptake by plants may be dependent on antibiotic type and plant type.

  • The purpose of this case report was to present an example of an esthetic and functional rehabilitation of anterior teeth with tetracycline discoloration and minor morphological abnormality of a 39-year old female. A chairside computer-aided design/computer-aided manufacturing (CAD/CAM) system with CEREC AC was applied for the prosthetic procedure and all ceramic crowns made with lithum disilicate (IPS e.max CAD) restored the esthetic and functional features of sixteen anterior teeth successfully.

  • The purpose of this study was to evaluate the substantivity of experimentally developed gel type tetracycline HCl and a mixture of tetracycline-citric acid gel, and compare to those of solution type tetracycline HCl. 11 extracted anterior teeth were subjected to this study. After scaling and root planing teeth were randomly divided into 3 treatments groups : group 1; 3 teeth were irrigated with tetracycline HCl(50mg/ml) solution , group 2; tetracycline gel (5%) was inserted in the periodontal pockets of 3 teeth, group 3; a mixture of tetracycline and citric acid gel was inserted in the pockets of 3 teeth. And 2 teeth treated in 0.9 % sterile saline served as controls. After 5-minute exposure, each tooth immediately extracted and incubated at room temperature for 22 days in tris-buffered saline as a desorption media. The total volume of TBS was removed and replaced with fresh TBS, at 24-h intervals. Removed desorption media transferred to a sterile vial and stored at -70 oC. This procedure was repeated every 24 h throughout the 22-day desorption period. Using Porphyromonas gingivalis as an indicator organism, a microtiter assay was used to evaluate antimicrobial activity desorbed from the teeth. 1. 50mg/ml tetracycline HCl solution exhibited the longest antimicrobial activity. Compared to saline treated group, it showed significant difference on the day 1 and day 2 desorption period. 2. The ODs of 5% tetracycline gel and a mixture of tetracycline-citric acid gel were significantly different during the first 24 hour only. 3. There was no statistically significant difference after the day 3 between the groups.(p<0.05). Despite our expectation a mixture of tetracycline-citric acid gel did not show longer antimicrobial activities than those of tetracycline gel, and the solution type exhibited the longest activities. Because the gel type agents may stay in the subgingival environment longer than the solution, if the teeth were not extracted immediately after the delivery of the agent, the result could be different. hus this result suggests the possibilities of practical use of these kind of gel type agents.

Occurrence of Jujube Witches' Broom Disease and Control Efficacy of Trunk Injection with Oxytetracycline in Boeun Area from 2018 to 2019 (2018-2019년 보은지역 대추나무 빗자루병 발생 및 옥시테트라사이클린 수간주사 방제 효과)

  • Jujube witches' broom (JWB) disease is a high-risk plant disease that occurred on and devastated jujube orchards throughout South Korea in the 1950s. Recently, as the jujube cultivation area has been significantly increased nationwide, we investigated the disease in jujube orchards in the Boeun region to obtain basic information for proactively evaluating the risk of JWB outbreaks from 2017 to 2019. A field trial was conducted to evaluate the effect of the trunk injection of Sungbocycline (a. i. oxytetracycline calcium alkyl trimethyl ammonium 17%, OTC). The incidence of JWB disease was very low in Boeun, but it was gradually increased mainly in the area and on the farms where it had occurred previously. The trunk injection with OTC was most effective in the early stages of growth. However, even though the trees were treated with OTC, disease symptoms appeared in other healthy branches in severely affected trees. When the tree was first injected with OTC in June, the control efficacy of second injection was higher one month after the first injection than five months. These results suggest that early diagnosis based on various symptoms and early control based on them can be very effective in preventing JWB disease in advance and increasing control efficacy.

Effect of Cimetidine and Antacid on Pharmacokinetics of Minocycline (시메티딘 및 제산제가 미노싸이클린의 약물동태에 미치는 영향)

  • Effects of aluminum magnesium hydroxide (A) and cimetidine (C) on the pharmacokinetics of minocycline (M) were investigated in female rats. Blood samples were collected at various time intervals until 36 hrs following oral dosing of drugs. Plasma minocycline concentrations were determined by HPLC. Control group (M), $T_1$ group (M+A), $T_2$ group (A+M after 2 hrs), $T_3$ group (M+A after 2 hrs), $T_4$ group (M+C) and $T_5$ group (C+M after 2 hrs) were divided to examine interaction of the drugs with minocycline. Plasma minocyline level-time curves were well described by two-compartment open model with first-order absorption in rats. Antacid treatment was associated with reduced of 71.0, 45.9, 35.7% in minocycline absorption rate $constant(K_{\alpha})$, maximum plasma $concentration(C_{max})$, and relative $bioavailability(F_{rel})$, respectively. Cimetidine treatment group exhibited no significant changes in plasma level-time curve when compared with control group and did not affect minocycline absorption as by any of these three parameters.

  • Solid phase extraction(SPE) and matrix solid phase dispersion(MSPD) have been studied as preparation procedures for tetracyclines analysis by high-performance liquid chromatography(HPLC) in meat. The recovery range was 74${\sim}$98% for SPE, and 72${\sim}$93% for MSPD at spiked levels of 100ng/g for oxytetracycline(OTC), tetracycline(TC), chlortetracycline(CTC), and doxycycline(DC). The detection limits were 15${\sim}$78ng/g for SPE and 25${\sim}$84ng/g for MSPD, respectively. Analytical method was HPLC with UV detector. The purpose of this study was developing a practical, accurate and precise method for rapid extraction and quantitation of tetracycline residues in meat.
