Cultural Characters, Nutrition of the Colletotrichum spp. Isolated from Anthracnose of Lycium chinense and Effect of Fungicides on Disease Incidence (구기자(枸杞子) 탄저병균(炭疽病菌)의 배양적(培養的) 성질(性質), 영양(營養) 및 탄저병(炭疽病) 발병(發病)에 미치는 약제(藥劑)의 영향(影響))

  • Mycelial growth of Colletotrichun dematium and C. gloeosporioides isolated from anthracnose of Lycium chinense was favorable on V-8 Juice Agar media and Oatmeal Agar media of the four different media tested. Sporulation of C. dematium was favorable on PDA media and V-8 Juice Agar media and that of C. gloeosporioides was favorable on all media tested. The optimum temperature for mycelial growth of C. gloeosporioides and C. dematium was $25^{\circ}C$ and $28^{\circ}C$, respectively and sporulation of the fungi was stimulated under alternating NUV light and darkness. Of the seven nitrogen sources tested, gelatin was the most favorable for mycelial growth of C. gloeosporioides and glycin and gelatin were favorable for that of C. dematium. Of the eleven carbon sources tested, dextrin and sorbitol were favorable for mycelial growth of C. gloeosporioides and dextrin was the most favorable for that of C. dematium. Among the eleven fugicides tested, Benomyl, Folpet, Dithianon, Carbendazim and S-3308L were found superior for the inhibition of mycelial growth and spore germination of the fungi. They were, also, found superior for the control of anthracnose disease of L. chinense when they were applied to the plants in the field.

Genetic Diversity of Korean Cylindrocarpon destructans Based on Virulence Aassay and RAPD Analysis (병원성 검정 및 RAPD 분석에 의한 국내 인삼뿌리썩음병균(Cylindrocarpon destructans)의 유전적 다양성)

  • Ginseng root rot caused by Cylindrocarpon destructans is one of the most destructive diseases of ginseng(Panax ginseng). We analyzed the features of the species through pathogenicity test and genetic diversity analysis of C. destructans in Korea, for its application as basic data to attempt for effective control. C. destructans isolated from rotted ginseng roots exhibited a variety of colonial colors on media. It was assumed that there may exist genetic diversity in the population by the diversity of pathogenicity among isolates observed when artificially inoculated into ginseng roots. Pathogenicity tests using ex vivo wound inoculation with agar mixture inoculation on ginseng roots were performed similar results as were observed appear to be useful for rapid pathogen inspection. According to RAPD analysis results, Korean C. destructans isolates formed a single genetic group which can be distinguished readily from closely related other fungi. C. destructans group was divided into two small groups. Therefore, we were able to confirm pathogenicity and genetic difference between the isolates in each of the groups of the pathogen.

Effects of algae on chronic assessment endpoints observed with Korean freshwater Cladocerans (한국산 물벼룩의 먹이조건별 번식영향)

  • The influence of different feeding rates of three green algae on growth and reproduction in four Korean cladocera, Daphnia sp., Daphnia obtusa, Moina macrocopa, and Simocephalus vetulus was investigated. The growth and reproduction rates of Daphnia sp. fed Chlorella vulgaris cells were significantly increased than those of daphnid fed Senedesmus subspicatus and Selenastrum carpricornutum. The total offsprings of Daphnia sp. fed $25\times10^4$ and $50\times10^4$ cells Chlorella vulgaris for 21days were $124{\pm}2.8$ and $168{\pm}22.1$, respectively. The Daphnia obtusa fed $25\times10^4$ and $50\times10^4$ cells Selenastrum carpricornutum was shown good reproduction rates, $72.2{\pm}7.9$ and $79.6{\pm}22.9$, respectively. The Moina macrocopa fed $25\times10^4$ and $50\times10^4$ cells Senedesmus subspicatus as well as Simocephalus vetulus and $25\times10^4$ and $50\times10^4$ cells Chlorella vulgaris were shown good reproduction rates. All of algae tested were sufficient as a good diet for Korean Cladocerans in this study. However, the preferred alga was Chlorella vulgaris and the optimal feeding concentrations were $2.5\times10^5\sim5\times10^5$ cells/mL, because the alga was shown most stable and generally high production rates in all cladocerans tested.

  • The inhibitory effect of epigallocatechin gallate (EGCG), one of the antioxidants in green tea (Camellia sinensis), against Salmonella Enteritidis was evaluated in commercial braised quail eggs at two temperatures (4 and $25^{\circ}C$). Although S. Enteritidis was dose-dependently suppressed by EGCG in pure culture at $25^{\circ}C$, it was not inhibited in the sauce or eggs at this temperature. At low temperature ($4^{\circ}C$), S. Enteritidis was inhibited in both the sauce and eggs by $400{\mu}g/mL$ EGCG. Thus, EGCG at an appropriate concentration could be a useful food additive for inhibiting S. Enteritidis in braised quail eggs at low temperatures.

Inhibitory Effects of an Eicosanoid Biosynthesis Inhibitor, Benzylideneacetone, Against Two Spotted Spider Mite, Tetranychus urticae, and a Bacterial Wilt-causing Pathogen, Ralstonia solanacearum (아이코사노이드 생합성 저해제인 벤질리덴아세톤의 점박이응애(Tetranychus urticae)와 세균성풋마름병 세균(Ralstonia solanacearum)에 대한 억제효과)

  • A monoterpenoid compound, benzylideneacetone (BZA), is a metabolite of an entomopathogenic bacterium, Xenorhabdus nematophila. Its primary biological activity is an inhibitor of phospholipase $A_2$, which catalyzes the committed step of biosynthesis of various eicosanoids that are critically important to mediate insect immune responses. When BZA was applied to two-spotted spider mite, Tetranychus urticae, it exhibited a dose-dependent mortality in leaf-disc assay. Subsequently BZA was tested against T. urticae infesting apples in a field orchard, in which it showed a significant control efficacy, which was not statistically different with that of a commercial acaricide. BZA also had significant antibacterial activities against three species of plant pathogenic bacteria when it was added to the bacterial cultures, in which it showed the highest inhibitory activity against a bacterial wilt-causing pathogen, Ralstonia solanacearum. The bacterial pathogen caused significant disease symptom to young potato plants. However, BZA significantly suppressed the disease occurrence. This study suggests that BZA can be used to develop a novel crop protectant to control mite and bacterial pathogen.

The Influence of Water Temperature and Salinity on the Filtration Rates of the Short-necked clam, Ruditapes philippinarum (수온과 염분 변화에 따른 바지락의 여과율 변동)

  • The present study was performed to describe the influence of water temperature and salinity on the filtration rates of the short-necked clam, Ruditapes philippinarum. The clams were collected at tidal flat near Yeosu city, Cheollanamdo, Korea, from July 2001 to August 2001. Diatoms, Phaeodactylum tricornutum (KMCC B-128), were indoor-cultured by f/2 medium, and were used to measure the filtration rate of the clams. Filtration rates of the clams were measured by indirect method. Cell concentrations of food organisms were determined by direct counting cells using the hemacytometer under the light microscope. The filtration rate of the clams increased with temperatures up to the optimum temperature, circa 25$^{\circ}C$. Above this optimum temperature, the filtration rate decreased drastically. Also the filtration rate of the clams increased with salinity up to 35 psu. The maximal filtration rates of the clams were recorded at 20-25$^{\circ}C$, similar to be known as the optimal temperature for their growth, and 25-35 psu, respectively. The minimal filtration rates of the clams were recorded at 5$^{\circ}C$ and 15 psu. At the similar temperature and salinity, the filtration rate of the younger clams was higher than that of the older ones. Thermal coefficient, Q$_{10}$ values at low temperature range were much higher than those at high temperature range. These results indicate the short-necked clam is more sensitive in cold water. As they grow up, they become more stronger against their ambient environmental changes, such as thermal-shock, salinity changes.

Pseudoepidemic of Mycobacteria Other Than Tuberculosis (MOTT) Due to Contaminated Bronchoscope (기관지경 오염에 의한 비결핵항산균증의 위발생)

  • Background: The development of the flexible fiberoptic broncoscope by Ikeda was an important technologic advance in the diagnosis and management of patients with pulmonary disease. But, cross contamination related to fiberoptic bronchoscope was reported in cases involving tubercle bacilli, MOTT and other agents. Therefore, cleaning and disinfecting of fiberoptic bronchoscope requires careful attention. Methods: From September 1991 to May 1992, medical records of all patients with positive culture for MOTT in bronchial washing specimens were reviewed. Also to evaluate bactericidal effect of 2% glutaraldehyde, culture was performed after inoculum of MOTT, Serratia marsescens and Pseudomonas aeruginosa to the disinfectant solution. Results: In 2% alkaline glutaraldehyde, MOTT was not survived only after 30 minute exposure, but P. aeruginosa and S. marsescens were rapidly inactivated with no survivors after exposure to 2% glutaraldehyde. Since vigorous mechanical cleansing and more than 30 minute of contact time within washing machine, no more outbreak was observed. Conclusions: It is also very important that bronchoscopes must be meticulously cleaned after each procedure and more than 30 minute exposure would be required for eradication of MOTT with 2% glutaraldehyde. However even the most strictly applied infection control measures cannot exclude contamination completly and clinicians have to stay alert to this possibility. Prompt detection of pseudoepidemics is possible if abrupt increase in isolation rates, especially if they involve unusual or generally nonpathogenic organisms, are readily recognized.

  • The influence of starvation on morphological change of the red sea bream larvae was examined at Song-ji fish hatchery, Tongyong-Gun, Kyongnam Provice in July 1988. The results obtained are as follows: 1) The larvae of red sea bream began to feed on rotifers in 2 days after hatching. In case of non-feeding, all of the larvae died in 5 days after hatching and the larvae which feeding delayed 1 and 2 days from normal first feeding schedule also died 100 in 6 days after hatching. 2) With the exhaustion of the yolk, the total length, body length, myotome height and gut height of unfed larvae decreased. 3) The ratio of height to myotome height in unfed larvae has declined most rapidly compare to other demensions while starving. At 5 days after hatching, the ratios of these of starving larvae and fed larvae were 0.306 and 0.010, respectively. 4) The morphology of starving larvae at 6 days after hatching are characterized as sharpened jaw, projected edge of lower part of clavicle and slender gut.

Quantitative Analysis of Bacillus amyloliquefaciens GR4-5 in Soil (Bacillus amyloliquefaciens GR4-5 균주의 토양 내 정량 분석)

  • Bacillus amyloliquefaciens GR4-5 was isolated from the rhizosphere soil of Korean ginseng and displayed broad-spectrum suppression of ginseng root rot pathogens. The survivability of B. amyloliquefaciens GR4-5 in soil was investigated under three different conditions; indoor, outdoor - of which soil was put in 14 mL tube after treatment - and field environments. Soil samples were collected over a four-week period from three experimental designs, and assessed for 16S rRNA gene copy number by quantitative polymerase chain reaction (qPCR). In outdoor condition, the 16S rRNA gene copy number of Bacillus spp. was 8.35 log copies g $soil^{-1}$ immediately after the GR4-5 treatment. Two weeks later, the 16S rRNA gene copy number of Bacillus spp. (6.70 log copies g $soil^{-1}$) was similar to that of the control (6.38 log copies g $soil^{-1}$). In indoor condition, the 16S rRNA gene copy number of Bacillus spp. maintained in a certain level for a longer period than those in outdoor and field. The 16S rRNA gene copy number of Bacillus spp. in field experiment was reduced faster than that of outdoor condition. Our results show that B. amyloliquefaciens GR4-5 can survive in bulk soil for 1 week, indicating its potential use as a biocontrol agent following 7 day application intervals. This study presents that outdoor microcosm system design could be a useful method to assess easily the survivability of beneficial microorganisms.

Overview of UV-B Effects on Marine Algae (자외선이 해조류에 미치는 영향에 관한 고찰)

  • Numerous observations revealed strong evidence of increased middle ultraviolet radiation or UV-B (280 ~ 320 nm) at the earth's surface resulting from stratospheric ozone depletion. UV is the waveband of electromagnetic radiation which is strongly absorbed by nucleic acids and proteins, thus causing damage to living systems. It has been recorded in the East Sea, Korea that solar UV-B impinging on the ocean surface penetrates seawater to significant depths. Recent researches showed that exposure to UV-B for as short as 2h at the ambient level (2.0 Wm$^{-2}$) decreased macroalgal growth and photosynthesis and destroyed photosynthetic pigments. These may suggest that UV-B could be an important environmental factor to determine algal survival and distribution. Some adaptive mechanisms to protect macroalgae from UV-damage have been found, which include photoreactivation and formation of UV-absorbing pigments. Post-illumination of visible light mitigated UV-induced damage in laminarian young sporophytes with blue the most effective waveband. The existence of UV-B absorbing pigments has been recognized in the green alga, Ulva pertusa and the red alga, Pachymeniopsis sp., which is likely to exert protective function for photosynthetic pigments inside the thalli from UV-damage. Further studies are however needed to confirm that these mechanisms are of general occurrence in seaweeds. Macroalgae together with phytoplankton are the primary producers to incorporate about 100 Gt of carbons per year, and provide half of the total biomass on the earth. UV-driven reduction in macroalgal biomass, if any, would therefore cause deleterious effects on marine ecosystem. The ultimate impacts of increasing UV-B flux due to ozone destruction are still unknown, but the impression from UV studies made so far seems to highlight the importance of setting up long-term monitoring system for us to be able to predict and detect the onset of large -scale deterioration in aquatic ecosystem.
