Changes in Organic and Inorganic Nutrients in Terminal Shoots of 'Fuyu' Persimmon during Spring Growth (감나무 정단신초의 봄 생장 동안 유기 및 무기 양분의 변화)

  • To understand changes in composition and distribution of nutrients during early shoot growth of persimmon, organic compounds and inorganic elements of terminal shoots were analyzed for about 40 days from the time of foliation. Sample shoots were collected from mature 'Fuyu' trees for this three-year experiment and they were divided to stem, leaves, and the fruits including flower buds at the earliest stage. During shoot growth, concentration of soluble sugars increased in both leaves and fruits, but that of starch increased only in leaves. Those of amino acids tended to decrease in all the parts but there was no consistent change in proteins. As shoots grew, contents of all the organic compounds in a shoot increased, and they were especially higher in May leaves accounting for more than 60% of the shoot total for each nutrient. Along with shoot growth, concentrations of N and P gradually decreased in all three parts, while K decreased only in stem. However, those of Ca and Mg did not show notable changes in all the parts with wide variations depending on the year. Due to the quantitative increase in growth, contents of inorganic elements in a shoot increased in all the parts and the leaves accounted for 54-82% of the shoot total. At the cessation time of extension growth, a shoot contained 526-768 mg of soluble sugars, 245-844 mg of starch, 26-31 mg of amino acids, and 66-103 mg of proteins for three years. On the other hand, a shoot contained 203-388 mg of K, the greatest among the inorganic elements, followed by 132-159 mg of N. Changes of the nutrients in a shoot were much greater during the earlier stage of growth after foliation than during the later stage toward growth cessation, suggesting the importance of mobilizing reserve nutrients for the early growth of the shoots. The results of this study also suggested that the rate of nutrient changes, especially during the earlier stage of shoot growth, could be affected by environmental and cultural conditions.

In Vitro Propagation of Guzmania cv. Cherry by Axillary Shoot Culture (측지배양에 의한 Guzmania cv. Cherry의 기내 대량번식)

  • Guzmania was propagated through in vitro culture of lateral shoots. When new shoots grown in greenhouse were cut and cultured in vitro, contamination rate was very high at about 80% in the first stage of in vitro culture. Among cytokinin treatments for agar medium, 2.0 mg/L BA was most effective for shoot multiplication, and those with 0.5 mg/L kinetin and 0.5~1.0 mg/L BA were favorable for shoot multiplication. BA was more effective for shoot multiplication than kinetin, and shoot multiplication was more enhanced when 2.0 mg/L BA was combined with 0.1~0.5 mg/L IAA than 2.0 mg/L BA alone. The medium with 2.0 mg/L BA and 0.1 mg/L IAA showed the highest rate of shoot multiplication with about 8.7 in shoot number, and those with 2.0 mg/L BA and 0.5~1.0 mg/L IAA also resulted in high multiplication of shoots. Shoots were multiplicated more in liquid rotation culture(80 rpm) with the medium containing 0.5 mg/L BA and 0.1 mg/L IAA than liquid stagnating and solid cultures. Regenerated shoots formed roots very favorably in the medium supplemented with 2.0 mg/L IBA.

  • Daphne pseudomezereum var. koreana is native to Korea and is distributedin Kangwon-do, Jeollabuk do, and Gyeongsang-do. This economically valuable species has experienced a dramatic decrease in natural habitat due to climate change and is difficult to cultivate. In this study, we investigate a mass propagation method for D. pseudomezereum through in vitro culture and genetic resource preservation.WPM medium was better than the MS medium for shoot growth. As a result, we compared the shoot number and length of apical (W/AP) and non-apical shoots (W0/AP) with BA and GA3 treatments in WPM medium. Their shoots and length grew well in both BA 8ìM + GA38ìM-treated apical shoot and without-apical shoot. NAA did not effectively induce rooting of the in vitro plantlet.

Radioresistance of Acorus calamus to Gamma Ray Irradiation

  • Acorus calamus is an indigenous hydrophyte used as phytoremediation and water purification as well as medicinal herb. Irradiation of gamma ray was performed to investigate the effect on radioresistance of A. calamus on in vitro. Various doses (20-500 Gy) of gamma rays were irradiated to in vitro plantlets. The effect of irradiation was studied on survival rate, shoot formation and growth rate. The survival and multiplication rate was remarkably decreased with the increase of radiation dose. The growth of plantlets was significantly increased in comparison to the control at low doses. The hermetic effect of radiation was appeared in A. calamus. It was significantly decreased or interrupted above 250 Gy. The mean 50% lethal dose ($LD_{50}$) was extremely high as 240 Gy. We conclude that A. calamus had remarkably high radioresistance compared with other plant species.

Effect of Plant Growth Regulators and Medium Salt Strength on In Vitro Propagation of Belamcanda chinensis DC (범부채의 기내증식에 미치는 식물생장조절물질 및 무기염류농도의 효과)

  • The effects of plant growth regulators on callus formation and organogenesis in shoot tip explant of Belancanda chinensis were examined. Shoot tip explants cultured in full salt strength of MT(Murashige and Tucker) medium containing 2,4-D 1.0 or 2.0mg/l were vigorously formed callus. Full salt strength of MT medium and 1/2 MT medium supplemented with zeatin 1.0mg/l were more effective than that with combination treatments of 2,4-D on the formation of shoots from calli. When shoots regenerated from shoot tips were transplanted into 1/2 MT medium added with 1.0mg/l, 41% of shoots formed roots.

  • This study was conducted to observe the effects of urea foliar spray to replace soil fertilization on defoliation ratio, shoot growth and fruit setting in satsuma mandarin (Citrus unshiu Marc.). Twice foliar sprays of 1% urea after harvest increased N and chlorophyll contents in leaves, and decreased defoliation during the winter season. The growth of the ovary and spring shoot was stimulated by the twice 0.8% urea foliar spray around the time of bud emergence. In addition, the treatments increased the number of flowers and fruit setting, but didn't influence fruit quality.

Effect of Culture Method and Medium Composition on Shoot Regeneration from Sporophytes of Cyrtomium caryotideum var. coreanum Nakai. (참쇠고비(Cyrtomium caryotideum) 포자체로부터의 식물체 재생에 미치는 배양방법 및 배지구성물질의 영향)

  • This study was conducted to investigate the efficient propagation method of Cyrtomium caryoptideum var. coreanum by sporophyte culture. The influence of origin of explant sources (rhizome, blade, or stipe) and homogenization of culture materials on shoot regeneration were investigated. As a result, only rhizome explant exhibited the organogenic capacity and the shoot regeneration was promoted by homogenization of culture material. Vigorous and excellent growth of multiple shoots was induced on the half-strength of inorganic salts containing MS medium. It was appeared that optimum nitrogen content of shoot regeneration was half-strength of nitrogen containing MS medium (30mM) and optimum sucrose concentration was 1%. Addition of $NaH_2PO_4$ to culture medium generally enhanced shoot multiplication and promoted growth of the regenerants. The organogenic capacity of homogenized rhizomes was especially promoted on medium supplemented with $5{\mu}M$ kinetin plus $5{\mu}M$ IBA. The incorporation of $0.1\sim0.2%$ activated charcoal on medium supplemented with growth regulators prevented the formation of multiple bud primordia - nodule-like bud clusters and improved the normal morphogenesis of sporophytes.

A Selection of Korean Native Sedum spp. for Evergreen Ground Covers of the Central Districts of Korea (중부지방 상록지피용 기린초 속의 조경용 소재 선발)

  • Sedum takesimenses among the Korean Native Genus Sedum was selected as evergreen ground covers during winter in the central districts of Korea. In order to obtain basic information for production as a pot flower with good quality, the effects of pinching frequencies on growth and flowering of S. takesimense were investigated. S. takesimense plant height became much shorter and numbers of lateral shoots were increased with increased pinching. Shoots of S. takesimense treated with 30% pinching during 2004 and 2005 showed greater length and less number of lateral shoots than those with 60% pinching. Leaves of S. takesimense treated with 70% shading showed a dark green color and had a longer shoot length and a lower number of shoots than cultivation under full sunlight. Therefore, Korean native S. takesimense will be utilized as ground cover treated with 70% shading or under a tree. Therefore, S. takesimense will be promising as ground cover materials in the central districts of Korea.

Effect of complex fertilizer treatment on a recovery of reduced turfgrass caused by competing with trees in golf course (복합비료 시비가 골프코스 수목근부 잔디고사 회복에 미치는 영향)

  • This study was conducted to effect a complex fertilizer treatment on a recovery of damaged turfgrass caused by competing with tree at area under trees, and to seek for effective management system on damaged areas under trees in golf courses. Available phosphorous and potassium were enough to grow up turfgrass in plots of East valley and Ji San Golf Club. But these plots were acid soil ranged from pH 5.3 to pH 5.5, so that lime fertilizer was required for improving the chemical of soil. The effect on complex fertilizer showed significant f3r the recovery of damaged turfgrass. Turfgrass recovery ratio by complex fertilizer was better in low-density(LD) section of fertilizer than in high-density(HD) section of control. As the result of surveying turfgrass characters according to dates, dry matters in HD sections of control plots were higher than these in LD sections of fertilizer plots in 6 Aug. before sprinkling a complex fertilizer. But dry matters in LD section of fertilizer were, on the contrary, higher than in 6 Sep. after sprinkling complex fertilizer. In view of the result so far conducted, a turfgrass recovery to LD sections of complex fertilizer was batter than that of HD sections of control. Sprinkling complex fertilizer on turfgrass damaged by competing with trees will maintain the turfgrass growth, even though happen to compete between trees and turfgrass.
