Growth Characteristics of in Vitro Mass Propagated Plantlets of Ever-bearing Strawberry 'Goha' according to Aeration Rate in Bioreactor (사계성 딸기 '고하' 조직배양묘의 대량증식 시 생물반응기 내 공기주입량에 따른 생육 특성)

  • This study was conducted to determine the optimal aeration rate for mass propagation of ever-bearing strawberry by bioreactor culture. The aeration rate was treated in four levels: 0.1 vvm (air volume/medium volume/min), 0.2 vvm, 0.3 vvm, and 0.4 vvm. In 0.2 vvm conditions, shoot length was the longest at 9.03 cm in bioreactor culture, leave numbers were 40.4 ea and fresh weight was 6,106 mg. Plant growth rate at 0.2 vvm condition was faster than other treatments. In the aeration condition, 0.2 vvm was most effective to increase aerial part growth and to decrease medium consumption. As the culture periods increased, the fresh weight also increased rapidly. After six weeks of cultivation, shoots were emerged with 10.4 ea per plantlet, resulting in developing a complete plant. As a result, the bioreactor culture system for mass propagation of strawberry is required to continuously supply the air by 0.2 vvm speed and cultivate at least for six weeks.

Growth and Yield Response of Highbush Blueberry 'Duke' to Hydroponic Cultivation (블루베리 '듀크' 품종의 양액재배 시 수체 생장과 수량 반응)

  • The hydroponic culture for growing 'Duke' blueberry was evaluated in a protective greenhouse provided with similar environmental conditions to the conventional blueberry cultivations. One year old 'Duke' blueberry bushes planted in 180 L containers filled with 130 L peat moss and 40 L pearlite (v/v) were selected for the experiment. A nutrient solution consisted with NO3-N 4.6, NH4-N 3.4, PO4-P 3.3, K 3, Ca 4.6, and Mg 2.2 mmol-1 was supplied to the plants, comparing non-hydroponic treatment (provided with only underground water). Hydroponic culture increased number of shoot per bush by 18% and total shoot length by 24% compared with non-hydroponic culture. Total dry weight of a bush increased in the hydroponic with vigorous root growth 1.4-fold more than the non-hydroponic. Higher concentrations of inorganic elements and organic compounds were found in the hydroponic, indicating active nutrient absorption of the bush. The hydroponic produced high yield similar to adult bush from 4 years old age, maintaining the yield until 8 years old age. The findings of this study indicated that hydroponic cultivation systems will be useful for growing blueberry crop.

Berry Production Using Secondary Shoots in 'Campbell Early' Grapevines (포도 캠벨얼리 품종의 2차지를 이용한 과실생산)

  • For the production of second crop in 'Campbell Early' grape, the primary shoots were pruned at 3rd, 6th or 9th nodes from the shoot bases on 13 days, 23 days and 33 days after full bloom date on 7th June. Secondary shoots were sprouted 7~8 days after the pruning, and it took 19~25 days for the flowering on the secondary shoots. The flower cluster number on secondary shoots were 2.8 for 13 days after full bloom, and 3.2 for 23 days and 33 days after full bloom, meaning little effect by pruning time. The 3rd node pruning produced 2~2.4 flower clusters with flower cluster length of 9.3~10.4 cm, while the 6th or 9th node pruning produced 3.1~3.8 flower clusters with flower cluster length of 12~14.9 cm, showing superior flower cluster length for the 6th or 9th node pruning. The secondary shoots developed from the buds pruned 13 days after full bloom with pruning bud positions of 6th nodes demonstrated superior fruits with higher soluble solids and lower acidity than the rest of the pruning times and positions.

Plant Regeneration through Direct Somatic Embryogenesis from Immature Zygotic Embryo of Prunus yedoensis in Mt.Halla (한라산 자생 왕벚나무의 미성숙 접합자배로부터 직접 체세포배 발생을 통한 식물체 재생)

  • Two types of somatic embryos were directly induced from the immature zygotic embryos of the wild prunus yedoensis in Mt. Halla after 16 weeks of culture on MS medium supplemented with 0.1mg/L $GA_3$ and 0.1mg/L BAPor 0.5mg/L $GA_3$ and 0.1mg/L BAP. One was normal single embryo with a single basal part. Normal somatic embryos germinated successfully on 1/2 MS medium. However, abnormal nulticotyledonary somatic embryos, formed shoots only on hormone free MS medium and about 80% of shoots rooted on MS medium with 0.5mg/L IBA. The mximum frequency (62.5%) of normal somatic embryos was directly obtained from the zygotic embryo 30 days after full blooming but it was decreased with further maturation.

  • This experiment was conducted to investigate the effects of length of storage period under low temperature, $CO_2$ enrichment and addition of plant growth regulators in Murashige and Skoog medium on the plant regeneration of Korean ginseng (Panax ginseng C. A. Meyer). Seeds were treated for 60 and 80 days respectively under $5^{\circ}C$ environment. 2500ppm of $CO_2$ was enriched by ventilation. Three plant growth regulators added to the medium were Indolbutyric acid, Benzyladenin and Gibberellic acid (GA3). The result indicated that : The capacity of differentiation was higher in the aged cotyledons from the seeds treated for 80 days under low temperature condition than in those treated for 60 days. $CO_2$ enrichment had stimulating effects on the growth and development of shoot primordium significantly but less effects on the formation of adventitious buds. From one zygotic embryo hundreds of plantlets were differentiated. $CO_2$ enrichment had effects on the formation of both indirect somatic embryo and direct somatic embryo. Indirect somatic embryo showed little growth and differentiation, being undifferentiated vascular stele and epicotyl. Direct somatic embryos were formed on the epidermis of backside basal part of cotyledon. Those embryos developed to whole plant having latent bud.

Effects of Culture Condition on Embryogenesis in Microspore Culture of Brassica napus L. Domestic Cultivar 'Tammiyuchae' (국내 육성 품종 '탐미유채'의 소포자 배양 시 배양조건이 배발생에 미치는 영향)

  • For the establishment of an efficient embryogenesis from microspore culture in Brassica napus L. domestic cultivar 'Tammiyuchae', four different factors affecting microspore embryogenesis and plantlet regeneration were investigated. The highest embryogenesis rate was achieved when microspores at late uninucleate to early binucleate stage were isolated from flower buds with a length of 3.0~3.5 mm. On average, 388 embryos generated from 1 ml of microspores media. The highest number of embryos was obtained when microspores were subjected to $32.5^{\circ}C$ for 2 days. Embryogenesis of 'Tammiyuchae' was increased with increasing microspore culture density up to about $5{\times}10^4ea/mL$. Gradually higher culture density repressed embryogenesis of microspores. Regeneration rate of shoots from microspore-derived embryos was observed in MS solid medium supplemented with $0.5mg{\cdot}L^{-1}$ NAA and $1.0mg{\cdot}L^{-1}$ BA, and grew well in MS solid medium without plant growth regulators.

Establishment of optimal conditions for micropropagation by node culture and multiple shoots formation from sucker explants of thornless Blackberry (Rubus fruticosus L. cv. BB21) (가시없는 블랙베리(Rubus fruticosus L. cv. BB21)의 근맹아를 이용한 다경유도와 절간배양을 통한 식물체 증식조건의 확립)

  • This study was conducted to develop a simple, rapid, and reliable method for in vitro propagation of disease-free and true-to-type clones from sucker explants of thornless blackberry (Rubus fruticosus L. ${\times}$ R. parvifolius L.). To induce multiple shoots, the sucker explants were sterilized in 1% NaOCl solution, and then were aseptically cultured on the full and 1/2 MS solid medium supplemented with BAP (0.1, 0.5, 1.0, 2.0 mg/L). After six weeks of culture, the highest frequency (85.4%) of shoot formation from sucker explants was obtained on the full-strength MS medium with 1.0 mg/L BAP. Node explants obtained from multiple shoots were cultured on the various media of full- or half-strength of AD, B5, MS, SH, QL, WPM media, respectively. After 30 days of culture, plant growth was good on the half-AD, half-QL medium. After 90 days of culture, plant growth was good on the full MS and full SH medium. The survival rate of the plantlets after transfer to plastic pots containing soil mixture (sand: soil: vermiculite was 1:1:1, vol.) in the greenhouse was 98%. The results indicate that a multiple-shoot procedure can be applied for an efficient mass propagation of Rubus fruticosus L. ${\times}$ R. parvifolius L.

  • The flowering locus T (FT) gene, of which expression will be controlled at high temperature by heat shock promoter (it printed as to HSproFT), was introduced into spray-type chrysanthemum (Dendranthema grandiflorum (Ramat.) Kitamura) 2 cultivars ('Pink PangPang' and 'Pink Pride' by co-cultivation with Agrobacterium tumefaciens strain C58C1 harboring pCAMBIA2300 containing the HSproFT gene. After leaf segments of the 2 cultivars were infected with the A. tumafaciens with C58C1 as explants, shoots were regenerated from the explants cultured on the $1^{st}$ selection medium (MS basal salts + 1.0 mg/L BA, 0.5 mg/L IAA + 10 mg/L kanamycin + 0.7% plant agar, pH 5.8). The shoots were transferred into the $2^{nd}$ selection medium (MS basal salts + 1.0 mg/L BA, 0.5 mg/L IAA + 20 mg/L kanamycin + 0.7% plant agar, pH 5.8). One hundred seventeen plantlets from 'Pink PangPang' and 5 ones from 'Pink Pride' were confirmed as transformants by PCR analysis. Twenty six of the transformants and non-transformants were acclimatized and established well in a green house. Eights of 26 transgenic lines showed flower bud 1.7~10 days earlier than nontransgenic plants, and 24 of them flowered 1~6 days earlier than non- transgenic plants. The shape and color of flower of all HSproFT-transgenic lines were not different with those of non- transgenic plants.

The Effect of Rubber Banding Material on Root Development after Transplanting of Landscape Trees - For Pine Trees - (고무밴드 결속재가 조경수목 이식 후 뿌리발달에 미치는 영향 - 소나무류를 대상으로 -)

  • This study conducted an experiment to clarify the effect of rubber bands used as a root connector during the process of transplanting landscape trees on the development of the root system and the rooting process. The research period was four years, from April 2007 to April 2011, and the test conducted for this study was performed at the experimental field located at 398-2 Bangdong-ri, Sacheon-myeon, Gangneung-si, Gangwon-do. Twenty 15-year-old Pinus densiflora Siebold & Zucc. with good growth conditions were harvested and transplanted from the forest in Jebi-ri, Gujeong-myeon, Gangneung-si, Gangwon-do for the field experiment. A completely randomized design was applied for plot design, with 10 pines without rubber bands and 10 pines with rubber bands. Pinus densiflora for. multicaulis Uyeki was selected as the official tree of the pot test and was planted in a transparent pot to observe the development of the root system. The results of this research on the effect of rubber bands used as a root connector on root system development and the rooting process are as follows. 1. The rate of height growth in the field test was 4.1% lower in the trees with rubber bands when compared to trees without rubber bands. Trees with rubber bands were 4.2% wider than those without rubber bands in root diameter. The chlorophyll content was 6.8% higher in trees without rubber bands, but the rate of height growth, root diameter, and chlorophyll content were not significantly correlated. 2. In the comparison of fresh root weight in the field test, trees with rubber banding had roots weighing 1,740.0kg and those without rubber bands had roots weighing 1,433.3kg. Root dry weight was 522.3g in trees with rubber bands and 450.0g in those without rubber bands, but showed no significant difference depending on whether the rubber band was attached. 3. In a comparison of root number between surfaces touching and not touching the rubber band in trees with rubber banding, the surface touching the rubber band was observed to have more roots growing, the difference of which was deemed significant. 4. The shoot growth rate in the pot test was 1.1% higher in trees without rubber bands when compared with trees with rubber bands. The chlorophyll content was 0.02 higher in trees with rubber bands but the difference was not significant. 5. In the pot test, no significance was found in comparison of root number, root length, and root dry weight in trees with and without rubber bands. These test results imply that removing rubber bands as a connector does not present any significant effects on the ground growth or root development of transplanted pine trees. As it is shown that surface touching rubber bands grow more roots in trees with rubber bands, more active related research must be undertaken.

Somatic Embryogenesis and Organogenesis via Callus Culture from Hydrocotyl maritima Honda (선피막이의 캘러스 배양을 통한 체세포 배발생과 기관분화)

  • Hydrocotyle maritima Honda used as medicinal plants for a hemostatic agent was investigated for in vitro regeneration. The petiole explants of H. maritima were cultured on callus induction medium containing growth regulators ($0{\sim}5\;mg/l$, NAA and $0{\sim}5\;mg/l$ 2,4-D) either in single or in combination with $0.1{\sim}2\;mg/l$ BA for 6 weeks. Although single treatments of 2,4-D or NAA resulted in callus formation, the best results were combination of $0.5\;mg/l$, 2,4-D and $0.5\;mg/l$, BA. $5\;mg/l$, NAA and $0.5\;mg/l$, BA, respectively. The highest number of shoot (12 shoots per callus) was achieved with $3\;mg/l$, Kinetin. Also, when pieces of embryogenic callus induced on the medium supplemented with $0.5\;mg/l$, 2,4-D and $0.5\;mg/l$, BA were subcultured on hormone-free medium, somatic embryos were differentiated and developed further into welldeveloped plants.
