To seek for restoration method of Abies koreana forest with planting seedlings, Abies koreana seedlings(2, 3 and 5 year old) were planted by date(spring and summer) and surface treatment (coir-mat, coir-net and control) at Janggumok area in Hallasan. Survival rate and shoot growth of the seedlings were measured and compared by planting date, seedling age and surface treatments. For summer planting, survival rates of the seedlings are increased with increasing seedling ages, and those are not differed between surface treatments. For spring planting, survival rates of the seedlings are increased with decreasing seedling ages, and those are not differed between surface treatments. For summer planting, shoot growth of the seedlings are significantly differed between surface treatments. Shoot growth of the seedlings was the highest value 2.11mm at coir-matted plot, and the lowest value 1.49mm at control B)lot. Shoot growth of the seedlings are not differed between the seedling ages, but the highest shoot growth was 2.26mm at coir-matted and 3-year old plot. For spring planting, survival rates and shoot growth of the seedlings are not differed between seedling ages and surface treatments.
This study was conducted to provide useful information for improvement on the efficency of transformation mediated by Agrobacterium tumefaciens. The result from the sensitivity test of cotyledon explants of tomato to kanamycin suggested that 50mg/L could be a proper concentration for selection media. Two hundred mg/L of cefotaxime was selected as a proper concentration to remove Agrobacteria from media without any negative effect on explants. Both callus formation and shoot regeneration from cotyledon explants of tomato were significantly suppressed by the cocultivation with Agrobacterium. Three days of cocultivation was effective on callus formation and shoot regeneration in all of tomato cultivars tested. Confirmation of transformation for regenerated shoots was carried out by histochemical GUS assay and PCR analysis using NPTII primer, and transgenic shoots were obtained from all of 3 tomato cultivars tested.

This study aimed to evaluate the anti-obesity effects of Cudrania tricuspidata leaf extract in the order of leaf development on the shoot (L0, L1, L2, L3, L4, and L5). The leaves at the apex of a Cudrania tricuspidata shoot were classified as L0; the next leaves of the apex were classified as L1, L2, L3, and L4 from highest to lowest; and the lowest leaf was classified as L5. A series of 70% ethyl alcohol leaf extracts were screened for the inhibitory effects of adipogenesis in 3T3-L1 preadipocytes. We found that the apical leaf extract of Cudrania tricuspidata (CTL0) was the most effective. Next, a study was conducted on the inhibitory action mechanism of CTL0. Treatment with CTL0 significantly suppressed the differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by the decrease in lipid droplet content observed with Oil Red O staining. Treatment with 12.5 ㎍/ml, 25 ㎍/ml, and 50 ㎍/ml of CTL0 significantly reduced the lipid droplet content. Glucose and cellular triglyceride concentrations were reduced in the 3T3-L1 cells on the CTL0-treated medium compared to the differentiation medium (DM control, DMEM + insulin + dexamethasone + rosiglitazone). Compared with DM, CTL0 significantly inhibited the expression of key pro-adipogenic transcription factors, including peroxisome proliferator-activated receptor γ (PPARγ), LPL, A-FABP, and Glut4. These findings show that CTL0 extract has potent anti-obesity effects.

In order to compare salt tolerance among potato cultivars (Solanum tuberosum L.), in vitro plants of each cultivar were cultured on the Murashige and Skoog's medium containing different levels of NaCl (0, 75, 150, and 225 mM). The suitable level of NaCl for a comparison of the shoot and root growth between potato cultivars was 75mM. Shoot length and weight were better in 'Dejima', 'Superior', 'Jayoung', and 'Haryeong' than in other cultivars such as 'Goun' and 'Atlantic'. Normal root growth was observed in 'Seohong', 'Superior', and 'Haryeong', while 'Goun', 'Atlantic', 'Dejima', 'Jowon', and 'Chuback' showed no root. Proline contents in all the cultivars increased as affected by NaCl levels, then the increase amount was fewer in the cultivars showed a better growth in both shoot and root than in others showed a poor growth. As a result of this study, 'Superior' and 'Haryeong' were determined as salt tolerant cultivars and therefore using these cultivars it would be possible to potato production at salt arable land such as reclaimed tidal.
In order to establish a micropropagation system for buffalo gourd (Cucurbita foetidissima ) and common milkweed (Asclepias syriaca), the effects of several plant growth regulators and culture temperature on shoot multiplication and rooting were investigated. In buffalo gourd, the greatest number of shoot from shoot tip culture and well growth of formed shoot were obtained on the MIS medium supplemented with 1.0 mg/L BA and 0.3 or 0.6 mg/L IAA. Whereas kinetin and 2iP were not effective for shoot multiplication in vitro. It was found that 22$^{\circ}C$ and $25^{\circ}C$ were suitable for shoot multiplication. Roots were easily formed by the addition of auxins, especially 1.0 or 2.0 mg/L IBA and 2.0 mg/L IAA. Over 90% of plants survived successfully after being transferred into the field. In common milkweed, BA was more effective than kinetin or 2iP for its micropropagation in vitro. The increased shoot weight and number of nodes per shoot were obtained on the medium containing 3.0 mg/L BA and 0.3 or 0.6 mg/L IAA. But 2iP promoted the shoot elongation. In addition. common milkweed was sensitive to culture temperature in vitro. Temperature around 22$^{\circ}C$ was favorable for shoot multiplication and growth, whereas temperature higher than $25^{\circ}C$ usually reduced the rate of shoot survival rate.
This study was carried out to induce plant regeneration via shoot formation from sucker explants of Rubus fruticosus L. To induce adventitious shoots, sucker explants were sterilized in $1.2\%$ NaOCl solution, and cultured on the MS solid medium supplemented with kinetin (0.5, 1.0, 3.0 mg/L) and BA (0.5, 1.0, 3.0 mg/L), respectively. As above, to induce adventitious shoots, sucker explants were cultured on the MS solid medium supplemented with IBA (0, 0.1, 1.0 mg/L) and BA (0, 0.1, 1.0, 2.0 mg/L). After 4 weeks of culture, the highest frquency $(100\%)$ of shoot formation from sucker explants was obtained from the medium with 1.0 mg/L BA. The highest shoot number per explant from in vitro shoot explants was 5.3. After 10 weeks of culture, the number of shoot per explant was increased. The highest frequency $(85\%)$ of root formation was obtained at 0.5 mg/L glycine medium, when the explant with shoot were cultured on the MS medium containing glycine at various concentrations from 0 to 2.0 mg/L. The survival rate of the plantlets after transfer to plastic pots containing sand, soil, and vermiculite (1:1:1, vol.) was $95\%$. The results indicate that micropropagation procedure can be applied for an efficient mass propagation of Rubus fruticosus.

In order to establish an efficient adventitious shoot regeneration conditions from leaf explants for Asian pear 'Whangkeumbae', the effect of concentration and kinds of plant growth regulator and carbon source was investigated. Leaf explants of cultures grown on Murashige and Skoog (MS) medium containing 8 g/L plant agar were used. When the medium contained 0.25 mg/L thidiazuron (TDZ) and 0.3 mg/L indolebutyric acid (IBA), the adventitious shoot regeneration rate (ASRR) was greater as 61.1% than others treated and higher TDZ concentrations (2.5 and 5 mg/L) treatment significantly reduced the ASRR. As the effect of IBA and indoleacetic acid (IAA) concentration on the ASRR, 0.5 mg/L TDZ plus different concentration of IAA exhibited relatively high ASRR and 0.5 mg/L TDZ plus 0.3 mg/L IAA showed the highest ASRR of 76.7%. Also the effect of sucrose and sorbitol as carbon source on regeneration was examined. The highest ASRR and the most shoots per explants averaged 94.4% and 3.49 by treatment of 30 mg/L sorbitol, respectably. Sorbitol is considered better carbon source than sucrose for shoot regeneration of 'Whangkeumbae' pear.
This study was carried out to establish the micropropagation system of Bupleurum latissimum Nakai that is a Korean native endangered species. Callus were induced from the leaf, petiole and floral bud and the percentage of callus formation was highest in the floral bud on the MS medium containing 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D. Especially, callus induced from floral bud was formed 77.8% and the percentage of shoot formation was 42.6% on the MS medium containing 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$$L^{-1}$ TDZ. For simultaneously callus formation and shoot regeneration, 1/2 MS medium was more effective than MS medium. The percentage callus formation, shoot regeneration and rooting were 46.3%, 13.0%, 13.0% in 1/2 MS medium, respectively. Soot regeneration from callus was good in 1/2 MS medium supplemented with 2.0 mg ${\cdot}$$L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$$L^{-1}$ BA where percentage of shoot regeneration was 74.1 %, and the number of shoot per explant was 2.4. The percentage of rooting was lowest (57.8%) in control while it was highest (97.8%) in 1.5 mg ${\cdot}$$L^{-1}$ NAA. In acclimatization of regenerated plantlets, the percentage of survived plantlets was highest (86.1%), and plant height, root length and fresh weight were good in the soil for horticulture.

Veronica rotunda var. subintegra (Nakai) T. Yamaz. is perennial herb belonging to the family Scrophulariaceae, native to Korea. Plant growth regulators, cutting mediums, cutting position, and leaf size were examined to investigate the requirements for cutting propagation of V. rotunda var. subintegra. This study was conducted over a period of 6 weeks from March to May 2017 in the Division of Plant Resources, Korea National Arboretum. For all treatments, survival and rooting percentage was 100%. In PGRs experiment, the number of roots and shoot length treated with 100 mg/L IBA treatment was significantly higher than that of NAA treatment. The shooting percentage was decreased with an increase of the PGRs concentration. In growing substrates experiment, the number of roots on commercial soil (61.2) was the highest, followed commercial + kanuma soil (45.7), and the lowest was kanuma soil (41.8). The shoot length treated with commercial soil (8.9 cm) was significantly greater than that of kanuma soil (4.1 cm). In cutting positions experiment, the number of roots treated with apical (56.6) was significantly higher than that of basal (38.8). In leaf-cut experiment, the number of roots and of shoot length treated with full leaf was significantly higher than that of half leaf. As a result, we recommend treating cuttings with 100 mg/L IBA treatment, commercial soil, apical position, and full leaf for successful cutting propagation of V. rotunda var. subintegra.

We analyzed the correlation among fruit bearing branch (FBB) and shoot and fruit characteristics in order to select the length of FBB suitable for producing high-quality fruits of 'Soomee', a peach tree developed in Korea. The length and diameter of FBB were 26.1 cm and 6.1 mm, respectively, shoot and leaf number per FBB were 3.2 and 38.6, respectively. Of these, the coefficient of variation was very high in the shoots and leaf number. The average weight and soluble solid content (SSC) of fruit were 298.6 g and 12.2 Brix, respectively, and coefficient of variation of the fruit weight was 18.0 %, which was higher than that of SSC. As the FBB of 10-20 cm and 20-30 cm length per tree were 27.1 % and 25.4 %, respectively, the sum of short and middle FBB frequency per tree was more than 50 %. Fruits of 250-350 g and 11.0-13.0 Brix per tree were distributed in 68.6 % and 74.0 %, respectively. As a result of correlation analysis, fruit weight and shoot number were affected by the length of FBB. In particular, length of FBB showed the relation of fruit weight with $y=-0.0482x^2+2.4512x+277.36$. As a result, the length of FBB that can maximize fruit weight was analyzed as 25.4 cm. Therefore, in the filed, the suitable FBB for producing 'Soomee' peach is estimated to be about 20-30 cm.
