Effects of Aluminum Solution Treatment on the Growth of Forsythia koreana and Platanus occidentalis Cuttings(2) (알루미늄용액 처리가 개나리와 플라타너스삽수의 생장에 미치는 영향(2))

  • To examine aluminum toxicity on woody plants, Forsythia koreans and Platanus occidentalis cuttings were grown in the pot(48$\times$33$\times$9cm) filled with sand, and treated aluminum solution and ground water (pH 6.75) 3times per week from April 28, 1993 to June 16. Aluminum solution were prepared 1.0, 2.5 and 5.0mM aluminum potassium sulfate, dilulted with ground water. Growth-related characters (Shoot growth, leaf number and leaf chlorophyll content ) and root growth were measured and compared among the treatments. In all growth-related characters(Shoot growth, leaf number and leaf chlorophyll content), differences among the treatments were highly significant. In root growth, differences among the treatments were highly significant for Forsythia koreana cuttings, but not for Platanus occidentalis cuttings.

  • Girdling and girdling+pinching treatments on 'Sekaiichi' apple remarkably reduced June drop and accelerated fruit growth in early stage of fruit development. Girdling+pinching and pinching reduced bourse shoot growth compared with non-treated control. There were no significant differences in flower bud formation in the following year among treatments. It was assumed that the optimum period for girdling is between 5 days before full bloom and full bloom period.

In vitro Growth of Shoot Derived from Shoot Tip in Asparagus cochinchinensis (Lour.) Merr. (천문동 경정 유래 신초의 기내생장)

  • This study was carried out to investigate the optimal conditions for efficient in vitro growth of Asparagus cochinchinensis (Lour.) Merr. shoot derived from shoot tip. Shoots were successfully cultured in MS medium. It was found that $23^{\circ}C{\sim}25^{\circ}C$ were suitable for shoot growth. The growth of shoot was greatly influenced by cytokinins. The shoots derived from shoot tip were well elongated on MS medium supplemented with BA and zeatin. In vitro shoots were very poor or no growing. Especially, 3.0 mg/l BA on MS medium was very effective in elongation of shoot. Root formation from in vitro growth of plant let was achieved on MS medium supplemented with 0.5 mg/l IBA. The results suggest that selection of plant growth regulator could be important factor to achieve an efficient in vitro growth.

Change of Rooting Potential as Affected by Cutting Time in Corylopsis coreana (히어리(Corylopsis coreana)에 있어서 삽목시기에 따른 발근력의 변화)

  • This study was conducted to find out a proper cutting time as related with a growth pattern for the mass propagation of Corylopsis coreana, which is one of the Korean endemic plant. New shoots began to grow in early April soon after end of blooming time and grew rapidly until the end of June. Shoot growth became dull after June and stopped in early September, showing a typical primary growth curve of temperate deciduous trees which show only one flush of growth throughout an annual growth cycle. The moisture content of shoot was the highest in an early growing season of May, and decreased with shoot maturity. Moisture content of cuttings taken on June 20 was 71.2%. Rooting was better in softwood cuttings than in hardwood cuttings. The highest value of rooting percentage was obtained in cuttings taken on June 20.

Improvement of Catechin Productivity in Callus Cultures of Camellia sinensis Leaves (차나무 잎의 캘러스 배양을 통한 카테킨류의 생산성 개선)

  • The effects of thiamine-HCl or polyvinylpyrrolidone (PVP) on growth and catechin contents of calli from Camellia sinensis leaves were investigated to improve catechin productivity in callus cultures on the selective medium. The growth of calli was great on the proliferation medium (the MS medium with 0.2 mg/L 2,4-D and 1.0 mg/L TDZ) supplemented with $20{\sim}30\;mg/L$ thiamine-HCl. Although the caffeine content was low in calli compared to young leaves, the total content of catechins was high in calli. Particularly,(-)-epicatechin (EC) which not detected in young loaves was also detected in calli on the propagation medium containing thiamine-HCl or PVP. In addition, the (-)-epicatechingallate (ECG) content was significantly higher in calli than in young leaves. In conclusion, the proliferation medium supplemented with 30 mg/L thiamine-HCl as a vitamin source seemed to be optimal condition for the growth and catechin production in callus culture.

Flowering and Growth of Dendrobium phalaenopsis as influenced by Photoperiod, Temperature, and Plant Growth Regulators (온도, 일장 및 식물생장조절제에 의한 덴파레의 생육 및 개화 반응)

  • This study was conducted to investigate effect of temperature, photoperiod, and plant growth regulators on the growth and flowering of Dendrobium phalaenopsis. Photoperiod treatments of 10 or 16 day length were from Mar. 24 to May 23. After treatments, all plants were exposed under natural photoperiod. Temperature treatments promoted development of new shoots (leads) of Dendrobium phalaenopsis 'Semi Alba' at $30/25^{\circ}C$(day/night) for 12 weeks, and 16 hr-photoperiod treatment promoted development of new shoots and flowering. The 16 hr photoperiod Dendrobium phalaenopsis 'Candistraipe ${\times}$ Tedtakiguz' no difference in the numbers of flower, flower width, and stalk length compared to the 10 hr photoperiod. Days to first flowering from appearance of new shoot were 241 days under natural photoperiod, 243 days under 10 hr, and 216 days under 16 hr, so that 16 hr-photoperiod treatment was faster about 25 days than that of natural or 10 hr treatment. There was not significant difference in growing and flowering of Dendrobium phalaenopsis among plant growth regulators.

Effects of Soil Moisture Control and Dormancy Breaking Agents on Bud Burst and Fruiting for Double Cropping System in a Year in 'Kyoho' Grapes (포도 '거봉' 2기작재배를 위한 하계휴면타파에서 토양수분 조절과 휴면타파제 처리가 발아에 미치는 영향)

  • Double cropping system in a year in Kyoho grapes (Vitis labruscana L.) has currently been attempted in the plastic greenhouse. One of the problems in double cropping system is the promotion of bud break in summer season and shoot fertility. Effects of the control of soil moisture tension near the root zone and treatments of bud dormancy breaking agents on bud breaking in summer were examined to promote the bud break for the second fruiting. The lignification of shoots was induced in July or August by the control of soil moisture tension in root zone environment. The first shoot growth was almost the same as that in common plastic greenhouses. The highest bud break value appeared in the plot of cyanamide chemicals mixed with merit blue as over 75% bud break rate. The bud break rate in the discontinuing plot in irrigation showed significantly higher in bud break than that in the continuing plot in irrigation. Despite of the final high bud break rate, the time of bud break was irregular.

Induction of Mutants by Irradiation of $\gamma$-Ray on In vitro Shoots of Persimmon (기내$\gamma$-선 처리에 의한 감(Diospyros kaki Thunb.) 돌연변이 유기)

  • This study was carried out to know the optimum dose of gamma-ray for the induction of mutation in vitro and the characteristics of the mutants induced by gamma-ray in persimmon (Diospyros kaki Thunb.). The LD50 (50% lethal dose) for in vitro shoots of the cultivar, Nishimurawase was between 1 krad and 2 krad and about 1 krad for the cultivar, Ichikikeijiro. As the dose of gamma-ray increased, the length of shoots decreased and necrosis of buds increased. For the cultivar, Nishmurawase, 37.5∼58.3% shoots rooted and the rooting rate and the number of roots per shoot was low in high gamma-ray. The irradiated young plants which were grown in the growth cabinet for 6 weeks were shorter in shoot length and had more branches than non-irradiated plants. The survival rate of irradiated plants grown in the green house for 3 months was 33%, while 77% for control plants.

Plant Regeneration from Cell Suspension Culture Using Leaf Callus in Actinidia deliciosa X A. arguta Clone 118 (양다래X다래 클론 118의 엽조직 캘러스를 이용한 세포 현탁배양으로부터 식물체 유도)

  • Calli were induced by culturing the leaf segment of Actinidia deliciosa ${\times}$ A. arguta clone 118 on MS medium supplemented with 0.5 mg/L 2,4-D, 0.1 mg/L NAA and 0.05 mg/L BA for 8 weeks in light condition. The induced calli were inoculated in liquid MS medium containing 0.5 mg/L 2,4-D, 0.1 mg/L NAA, 0.05 mg/L BA and 3% sucrose to establish cell suspension culture. The cells at the exponential stage and the stationary stage could be observed between 5-11 days and after that 12 days in culture, respectively. The fresh weight of callus induced from the suspended cells did not vary much among the media containing eight different combinations of plant growth regulators tested. The highest frequency of shoot induction (88.3%) was observed in MS medium containing 2.0 mg/L zeatin. Either BA or zeatin mixed with thidiazuron (TDZ) seemed to be effective in shoot induction. The induced shoots were transferred to MS medium containing 0.2 mg/L zeatin for further shoot growth. And then the shoots were transferred to Standardi (ST) medium containing 1.0 mg/L indolebutyric acid (IBA) for rooting. Plantlets could be obtained through cell suspension culture of Actinidia deliciosa ${\times}$ A. arguta clone 118.

Multiple Shoot Induction on the New Cltivar, Saxifraga fortunei 'Greenstar' by Different Media and Plant Growth Rregulators (배지 및 생장조절제 종류를 이용한 바위떡풀 신품종 '그린스타' 의 다신초 유도)

  • This study was conducted to develop in vitro propagation techniques of new cultivar, 'Greenstar', bred by Highland Agriculture Research Center. The multiple shoot induction and plant growth of in vitro plant were analyzed by MS media concentration (1/2 MS, 1 MS and 2 MS medium), plant growth regulators and its proper concentration; CPPU [Forchlorfenuron(N-(2-chloro-4-pridyl)-3-phenylurea) (0, 0.5, 1.0 and $2.0mg{\cdot}L^{-1}$), thidiazuron [(TDZ), (0.01, 0.1, 0.5 and $1.0mg{\cdot}L^{-1}$), zeatin (0, 0.5, 1.0 and $2.0mg{\cdot}L^{-1}$), and BA [6-benzylaminiopurine(BA), (0, 0.5, 1.0 and $2.0mg{\cdot}L^{-1}$)] in MS (3% sugar and 0.8% agar with pH 5.7) media. The highest number of induced shoots, leaves and roots were shown in 1/2 MS medium concentration. On the 1/2 MS medium, shoot numbers, shoot length, leaf numbers and root numbers were 11.0, 1.9 cm, 24.7, and 8.0, respectively. On the absence of CPPU in the 1/2 MS medium, shoot length and root numbers was greater than CPPU treatment, but the highest number of shoots was induced by the $2.0mg{\cdot}L^{-1}$ of CPPU concentration in 1/2MS medium. TDZ, zeatin, and BA treatments were not effective on the induction of multiple shoot in vitro culture. As a result, in vitro culture of new Saxifraga fortunei, 'Greenstar' with $2.0mg{\cdot}L^{-1}$ of CPPU in 1/2 MS medium was most effective for the rapid multiplication.
