Cytotoxicity of Hydrogen Peroxide and Effects of Rhizoma Gastrodiae Against Hydrogen Peroxide in Mouse Cerebral Neurons (생쥐의 배양 대뇌신경세포에 대한 Hydrogen Peroxide의 세포독성 및 천마의 영향)

  • To elucidate the toxic effect of oxygen free radicals on cultured mouse cerebral neurons damaged by hydrogen peroxide(H₂O₂)-induced neurotoxicity, we examined the neurotoxicity induced by oxygen radicals by NR assay when cultured cerebral neurons were grown in the media containing various concentrations of H202 for 6 hours. In addition, neuroprotective effects of herb extracts such Rhizoma Gastrodiae(RG) on H202-induced neurotoxicity in cultured cerebral neurons were evaluated after cultured cerebral neurons were preincubated with various concentrations of herb extract, RG for 2 hours before 50uM H₂O₂ for 6 hours. H₂O₂ decreased remarkably cell viability in dose-and time-dependent manner in these cultures, and also herb exract, RG decreased LDH activity of cerebral neurons damaged by H₂O₂. From the above results, it is suggested that H₂O₂ was toxic in cultured cerebral neurons from mouse, and RG was effective in blocking the neurotoxicity induced by oxygen radicals in these cultures.

Nitric Oxide-Mediated Cytotoxicity of Manganese in Basal Ganglia Neuronal Cells (대뇌 기저핵 신경세포에서 Nitric Oxide를 매개로 한 망간의 세포독성)

  • Objectives:eurotoxicity is mediated by nitric oxide(NO) in the rat primary neuronal cultures and assess the effect of $Mn^{2+}$ on the N-methyl-D aspartate(NMDA) receptors. Methods: We have used 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT)assay to examine the effect of cytotoxicity of $MnCl_2$ in neuronal cells , NO production was determined by measuring nirites, a stable oxidation product of NO. The neurons in the rat that contains neuronal nitric oxide synthase(nNOS) were examined by immunofluorescence and confocal microscopy. The effects of $Mn^{2+}$ on the NMDA receptors was assesed by the whole cell voltage clamp technique. Results: We showed that the NO release and NOS expression was increased with 500uM $MnCl_2$ treatment and an NOS inhibitors, $N^G-nitro-L-arginine$, prevented neurotoxicity elicited by manganese. In the electrophysiological study, $Mn^{2+}$ does not block or activate the NMDA receptors and not pass through the NMDA receptors in a neurons of basal ganglia. Conclusions: It is concluded that manganese neurotoxicity in basal ganglia was partially mediated by nitric oxide in the cell culture model.

Induction of Glutathione S-transferase Activity by the Extracts of Glycyrrhiza uralensis Fischer (감초 추출물이 glutathione S-transferase의 유도 활성에 미치는 영향)

  • In the present study, we investigated the glutathione S-transferase (GST) induction of medicinal plants using a cultured PC12 cells. The methanol extracts of Dendrobium nobile Lindley, Schizonepeta tenuifolia Briquet, Glycyrrhiza uralensis Fischer, Paenoia lactiflora Pall were tested. As the result, exposure of PC12 cells to G. uralensis resulted in the significant induction of GST. On the continuous experiment, G. uralensis was extracted with methanol, ethanol, and acetone. Among these extracts, methanol extracts showed the highest GST induction. The methanol extracts were further fractionated with hexane, diethyl ether, ethyl acetate, and water layer according to the degree of polarity. The diethyl ether layer showed the highest exhibited GST induction on PC12 cells. Based on these results suggest that the extracts of G. uralensis can be applicable for the development of a new antioxidant agent.

Fine Structural Study on Compensation Effect of Red Ginseng against Methylmercury Injury to Brain and Gill of Fightingfish (메틸수은으로 손상된 버들붕어 뇌와 아가미에 대한 홍삼의 보상효능에 관한 미세구조적 연구)

  • The toxic effects of methylmercury on the ultrastructures of the brain and gill tissues of fightingfish and compensative effects of red ginseng were investigated by means of electron microscopy. The brain neuron of methylmercury exposure group showed dilatation of dendrite and axon, numerical decrease of ribosomes, partial loss of nucleoplasm and cytoplasm and considerable swelling of mitochondria as compared with the normal neuron. And necrotic cell with ruptured nucleus and vacuolated mitochondria was noticeable. While, slight swelling of mitochondria, some dilation of dendrite and axon and numerical increase of ribosome occurred in the neuron of methylmercury-red ginseng treatment group as compared with the methylmercury exposure group. In the gill lamella of methylmercury exposure group, collapse of pillar cells and arms, dilated epithelial cell and thickened membrane were observed. While, in the gill lamella of methylmercury-red ginseng treatment group, arms were slightly disintegrated and basement membrane was some thickend as compared with the methylmercury exposure group. From the above results, it is concluded that red ginseng has detoxication effect on methylmercury toxicity and so takes compensative effect on injured tissues caused by methylmercury intoxication.

An Analysis about Building-related Health Symptoms of Workers at Dental College Hospitals in Seoul (치과대학병원 종사자의 실내자각증상 관련요인 분석 -서울시내 소재 치과대학병원을 중심으로-)

  • In this study, recognized building-related health symptoms of various workers in dental college hospitals are surveyed and analyzed in order to improve indoor environment at dental college hospitals. We conducted a questionnaire survey for 682 workers in three dental college hospitals located in Seoul from Sep. 20 to Oct. 8, 2004. Most workers at dental hospitals complained about noise, poor ventilation, dusty environment, and air dryness. SBS symptoms score for subjects was 1.43${\pm}$0.74 (often feel) and major symptoms was throat, CNS, eye, and nose symptoms. Recognized Building-related health symptoms are significantly related to tasks, working experience, job-satisfaction, indoor environment recognition index, medical history, used to special material, and number of dental unit chair. Also working places and ventilation are significantly related to the symptom. Based on the results, it can be said that special dental treatment causes undesirable environment such as noise, dust, and bad odor. It is also related to recognized building related health symptoms reported by workers at dental hospitals. So managemental efforts are needed to improve indoor environment which is related by dental treatment.

  • In recent years, we have been working on the improvement project of mercury 3-lecture type at the Geummundo lighthouse for more than 100 years. We have installed toxic mercury removal, LED light source, and dualization system for nonstop. It can be seen as an example. Circular restoration of the entire frame is performed, and the real-time status can be grasped through the installation of the double-ball bearing and the remote control system. After installation, optical measurement through Hanvit secured the optical performance of 2,000,000 cd of floating brightness of 300cd, proving the efficient possibility of introducing the domestic rotary type light source using LED light source.

  • This study was carried out to develop the antitoxic compound about cytotoxicity of cadmium on cultured rat neuroglial cells. These cells divided into 3 groups; control group (medium only) or $MTT_{50}$ group (neuroglial cell, $61{\mu}M$ cadmium) and experimental group ($61{\mu}M$ caffeic acid). Neutral red (NR) and tetrazolium MTT of the colorimetric assay were performed to evaluate the cytotoxicity of cell organelles. The light microscopic study was carried out to morphological changes of cultured rat neuroglical cells. The results indicated that caffeic acid showed detoxification effect on cytotoxicity of cadmium in $61{\mu}M$ concentration. According to the spectroscopic study of 1:1 complex of cadmium and caffeic acid, it showed that this formation of complex eliminated cadmium from cultured rat neurogllal cells.

  • Brodie abscess is a localized form of subacute or chronic osteomyelitis which is common in children but may also occur in adulthood. When Brodie abscess is located in the posterior metaphysis of the proximal tibia, open biopsy and curettage have a difficulty in approach to the lesion and can cause neurovascular injury or soft tissue contamination. We report a case wherein a novel surgical technique was used to treat a Brodie abscess in the posterior proximal tibial metaphysis in 48 year-old-male with endoscopic-assisted curettage by commercial anterior cruciate ligament targeting device(Rigid Fix; Mitek, Johnson & Johnson, Norwood, MA). Two portals were created toward the abscess site and, through each portal interchangeably, the granulation tissue and sclerotic bone could be excised. We believe that endoscopic-assisted curettage presents safe technique, decreased morbidity, accurate assessment of the extent of the abscess and possible improvement in long-term outcomes.

Effect on Wnt/β-catenin Pathway of Methanol Extracts from Native Plants in Korea (국내자생식물 메탄올추출물의 Wnt/β-catenin 신호전달체계에 대한 효과)

  • The Wnt/β-catenin signaling pathway is mandatory in adipogenesis. In this study, we investigated the applicability of functional materials for the treatment of obesity by examining Wnt/β-catenin pathway reporter activity related to adipocyte differentiation inhibiting with korean native plant extracts. The luciferase activity of HEK 293-TOP cells increased the reporter activity approximately 152% and 130% by treatment with Sanguisorba officinalis and Thuja orientalis, respectively. Ricinus communis were represented about 90% higher activity, two samples(Rosa rugosa and Sophorae Flos) showed 80% higher activity than the control. Three samples of plant extracts (Zanthoxylum piperitum, Pueraria thunbergiana, Solanum nigrum) were about 70% higher activity compared with the non-treated control. Cytotoxicity of plant extracts was not detected in the rat neural stem cells. These results suggest that the selected eight plant extracts are safe compounds. Our findings indicate that Wnt/β-catenin pathway reporter activity could be used for high throughput screening system. In addition, the plant extracts selected as candidates for adipocyte differentiation inhibiting may be potential therapeutic agents for obesity, it will be exploring the possibility of developing an anti-obesity materials through further experiments with selected plant extracts.

Neurotropism and Expression Pattern of lacZ Inserted PRV-Bartha in Geniculohypothalamic Tract Tracing (lacZ가 삽입된 PRV-Bartha 종의 신경친화성 및 무릎시상하부로 추적시 발현양상 분석)

  • To localize the connection between intergeniculate nucleus and suprachisasmatic nucleus through geniculohypothalamic tract in postnatal mongolian gerbil, we injected lacZ inserted PRV-Bartha strain into suprachiasrnatic nucleus and tried to immunostain against it with Rb134 and mouse $anti-{\beta}-galactosidase$. The numbers of immunoreactive neurons in intergeniculate leaflet were $8{\pm}3.2$ in P1 Period, $10{\pm}4.1$ in P3 Period and $13{\pm}6.2$ in P7 Period, and was statistically significant (p<0.05) and had tendency to increase with time consuming. The results showed that intergeniculate leaflet had projected some axons into suprachiasrnatic nucleus through geniculohyptothalamic tract in postnatal mongolian gerbil. But we could not exclude the possibility of direct projections from dorsal and ventral geniculate nuclei into suprachisamatic nucleus completely.
