Effects of Scutellaria baicalensis Water Extract on Lipid Metabolism and Antioxidant Defense System in Rats Fed High Fat Diet (황금추출물이 고지방 식이를 급여한 흰쥐의 지질대사 및 항산화방어계에 미치는 영향)

  • The objective of this study was to determine the effects of a Scutellaria baicalensis water extract (SDWE) on lipid levels, lipid peroxidation and antioxidant enzyme activities on rats fed a high fat diet for 6 weeks. Thirty-two male Sprague-Dawley rats (4-weeks-old) were randomly divided into four groups: normal diet and deionized water (ND), normal diet and Scutellaria baicalensis water extract (NDS), high fat diet and deionized water (HFD), high fat diet and Scutellaria baicalensis water extract (HFDS). The food intakes were significantly lower, but the food efficiency ratios were significantly higher in the high fat diet groups than those in other groups. The level of HDL-cholesterol and HDL-cholesterol/total cholesterol ratio in plasma were significantly higher and AI (atherogenic index) in HFDS group was significantly lower than that in HFD group. The level of triglyceride in plasma was significantly decreased in SDWE groups. The triglyceride of liver was significantly increased in the high fat diet groups and the total cholesterol of liver in the HFDS group was significantly lower than that in the HFD group. The plasma and liver concentrations of thiobarbituric acid reactive substances (TBARS) in the NDS group were significantly lower than those in the ND group. The total antioxidant status (TAS) in plasma was significantly increased in the HFDS group compared to the HFD group. The activities of SOD, catalase and GST were significantly increased in SDWE groups compared to ionized water groups. The activity of GSH-Px and the concentration of GSH in liver in the HFDS group were significantly higher than those in the HFD group. These results suggest that a supplement of SDWE on rats fed high fat diet reduce levels of lipid and lipid peroxidation in plasma and liver and improve the antioxidant defense systems.

Effect of Dandelion Leaf Extracts on Lipid Metabolism in Rats Fed High Cholesterol Diet (민들레잎추출물의 흰쥐 체내 지질대사 개선 효과)

  • The effect of dandelion leaf extracts on lipid metabolism was investigated in high cholesterol-fed rats. Four groups of rats were given a high cholesterol diet containing 10g cholesterol/kg and 2.5 g sodium cholate/kg for 6 weeks. The control group received without dandelion leaf extracts and the other three groups received with one of dandelion leaf extracts (water, ethyl acetate, ether), respectively. Weight gain and feed intake were significantly higher in water group than control group. Serum triglyceride, total cholesterol, cholesteryl ester, chylomicron and VLDL-cholesterol concentratins were lower, whereas HDL-cholesterol and phospholipid concentrations were significantly higher in dandelion leaf extract supplemented-groups than control group. Dandelion extract increased HDL-C/total-C ratio and lowered atherogenic index. Hepatic triglyceride, total cholesterol, free cholesterol and cholesteryl ester concentrations lowered in dandelion leaf extract groups compared to control group. Especially, hepatic triglyceride and free cholesterol levels were lower in ethyl acetate extract group than in water and ether extract groups. Dandelion leaf extract supplements did not affect hepatic phospholipid cencentration. Hepatic HMG-CoA reductase activity was significantly lower in water extract group than the others. ACAT activity was significantly decreased by dandelion extract supplements.

Physicochemical Properties of Dextran Produced by Leuconostoc mesenteroides SM according to Concentration of Yeast Extract and its Modulation of Rheological Properties (효모 추출물 농도에 따른 Leuconostoc mesenteroides SM에 의해 생산된 dextran의 물리화학적 특성 및 물성개량)

  • Dextran was produced by $Leuconostoc$ $mesenteroides$ SM with various contents of yeast extract and its rheological properties were modulated, via an addition of hydroxypropyl methylcellulose (HPMC) of different viscosities. The conversion yield reached 90% after adding 3% yeast extract, which remained constant, thereafter. The acidity of the cultures was approximately 1.4 and 0.9% after fermentation for 24 h at 25 and $30^{\circ}C$, respectively. The total dextran content (107.3 g/kg) was the highest in the presence of the 3% yeast extract. Under the same conditions, the consistency, viscous modulus (G"), and elastic modulus (G') of the cultures were $37.6\;Pa{\cdot}s^n$, 38 Pa, and 50 Pa, respectively. The rheological properties of the culture were changed drastically by the fortification with HPMC of higher concentration and viscosity. The addition of 10% HPMC (4,000 cp) resulted in a significant increase in G" to 1,950 Pa. Furthermore, adding HPMC to a viscous culture resulted in a remarkable increase in both hardness and firmness.

Effects of Barley Noodles Contained Mulberry Leave Extracts on Blood Glucose Regulation in Diabetic Mice (당뇨 쥐에서 상엽추출물을 첨가한 보리면의 혈당조절효과)

  • This study aimed to investigate the blood glucose regulatory effect of barley noodles contained mulberry leaves water extract (MLWE) in diabetic mice in order to provide fundamental data as a functional food. ICR mice were divided into six groups as follows: normal control, positive control, barley control, mulberry control, barley+MLWE 2.5% and barley+MLWE 5% groups. High fat/high carbohydrate diet was fed for 4 weeks and diabetes mellitus was induced by i.p. injection of streptozotocin and nicotinamide. After diabetes induction, experimental diet was supplemented for 2 weeks and simultaneous measurement of body weight, blood glucose (including glucose tolerance test), liver function and blood lipid profile was conducted. As a result, there was less decrease in body weight compared to the positive control group in the groups that supplemented barley and MLWE together than the group ingesting barley and MLWE alone. In addition, regulatory effects on blood glucose, liver function and blood lipid concentration were also stronger in barley and MLWE concurrently supplemented groups. Consequently, combination of barley and MLWE was effective in the regulation of body weight, blood glucose, liver function, and blood lipid in diabetic mice. Therefore, this recipe could be a promising strategy for beneficial effects in the patients of diabetes mellitus.

Estimation of the Efficiency of Dietary Protein Utilization Based on the Urinary Excretion of Acid-Soluble Peptides in Rats (뇨중의 산가용성 펩타이드에 의한 식이 단백질 이용 효율의 추정)

  • Nutritional factors affecting the urinary excretion of acid-soluble peptides(ASP) in rats were studied using protein-free diet, gluten diet, casein diet, and gluten supplemented with lysine and threonine(GLT) diet. The content of urinary ASP was lowest in protein-free diet group among the four kinds of diets above. But the amino acid pattern of urinary ASP in the four dietary groups were similar each other, suggesting that urinary ASP is mainly from endogenous origin under these nutritional conditions. The efficiency of dietary protein utilization was significantly lower in gluten diet than that of casein diet or GLT diet. Those findings suggest that the rate of urinary excretion of ASP-form amino acids can be employed as an index of protein metabolism, particularly as a simple index in the assessing the status of protein nutrition.

Antimicrobial Activity of Extract (삼백초(Saururus chinensis Baill) 추출물의 항균활성)

  • In order to develop natural food preservatives, the ethanol and water extracts of the Saururus chinensis Baill were prepared. Antimicrobial activity was examined against 10 kinds of harmful microorganisms. The ethanol and water extracts showed the most active antimicrobial activity against B. subtilis and E. coli. The ethanol extract showed stronger antimicrobial activity than that of the water extract. However, the extracts did not show any antimicrobial activity against lactic acid bacteria and yeast. The minimum inhibitory concentrations (MIC) of ethanol extracts against B. subtilis and E. coli were 5 to 10 mg/mL, respectively. Antimicrobial activity of the ethanol extract was not destroyed at 40-12$0^{\circ}C$ and pH 3∼11. The ethanol extract was fractionated in the order of hexane, diethyl ether, ethyl acetate, and water fractions. The highest antimicrobial activity was found in the diethyl ether fraction.

Effects of Plantago asiatica L. on antioxidative activities and lipid levels in hyperlipidemic Sprague-Dawley(SD) rats (질경이(Plantago asiatica L.) 추출물이 고지혈 유발 흰쥐의 항산화활성 및 지질농도에 미치는 영향)

  • This study was carried out to investigate the effects of Plantago asiatica L. extract on anti oxidative potential, free radical generation and the lipid levels in rats. Sprague-Dawley(SD) rats were divided into two groups based on their diet, as follows: the AIN-76 diet (control group), and the modified AIN-76 diet(cholesterol 0.5%) with 0.5% P. asiatica extract, for 7 weeks. The body weight and teed efficiency ratios of the two groups did not significantly differ. The antioxidative potentials more significantly increased in the group that was fed P. asiatica extract than in the control group(p<0.05). There was no difference in the rate of free radical generation, though. The weights of the organs, such as heart, kidney, liver, and spleen, of the rats in the two groups did not differ, though. The ratio of the HDL cholesterol to the total cholesterol in the P. asiatica group was significantly higher than that in the control group, and the other serum lipid parameters (total cholesterol, HDL cholesterol, triglyceride, and phospholipids) did not differ between the two groups. These results imply that supplementation with P. asiatica extract may improve the antioxidant potential and decrease the lipid levels in the blood.

Effects of Cucurbita moschata, Adlay Seed, and Cudrania tricuspidata Leaf Mixed-powder Diet Supplements on the Visceral Fat, Fecal Amount, and Serum Lipid Levels of the Rats on a High-Fat Diet (늙은 호박, 율무, 꾸지뽕잎 혼합조성물이 고지방식이를 급여한 흰쥐의 내장지방, 배변량 및 혈청지질농도에 미치는 영향)

  • This study investigated the effects of the diet with Cucurbita moschata (CM), adlay seed (AS), and Cudrania tricuspidata leaf (CTL) mixed-powder on the visceral fat, fecal amount, and serum lipid profiles in rats fed a high-fat diet (HFD). The mixed ratio of CM, AS, and CTL as a functional food ingredient was 1:1:1 on a weight basis. Male Sprague-Dawley rats were fed a high-fat diet (D12492; 45 kcal fat) for 5 weeks during the first phase. In the second phase, which lasted 5 weeks, the rats were divided into six experimental groups. The groups were: a high-fat diet group as a control during the 10 weeks of experimental period; and a high-fat diet with 5, 10, 15, 20, and 30% mixed-powder supplement group. The adipose tissue (RFP, EFP) weights were significantly decreased in the 20% and 30% mixed-powder supplement groups than in the HFD group (p<0.05). The fecal weight produced by the 15%, 20%, and 30% mixed-powder supplement groups was significantly increased during the second phase compared to the first phase (p<0.05). In terms of serum lipids, TG (triglyceride) content was decreased in the 10% and 15% mixed-powder supplement groups. The blood glucose levels were significantly decreased in all experimental diet supplement groups than in the HFD group (p<0.05). The findings show that the high supplement groups could improve the visceral fat accumulation, fecal amount, and blood glucose content compared to the HFD group.
