Characterization of 65 kD Protein in Latex Excreted from Euphorbia lathyris (Euphorbia lathyris에서 분비되는 Latex 65kD 단백질의 특성규명)

  • Soluble latex protein fraction excreted from Euporbia lathyris laticifer was resolved by 10% SDS-polyacrylamide gel electrophoresis to identify distinctively displayed latex major protein bands including ELp65, ELp55, ELp43, ELp32 and ELp23. Among them, ELp65 was purified by ammonium sulfate precipitation, gel permeation chromatography and ion exchange chromatography. Its N-terminal amino acid sequencing revealed its homology to the leading region of mature peptide of tomato p69a subtilisin-like protease, suggesting a certain role involved in plant defense system. In the analysis of Southern blot hybridization using PCR-amplified tomato p69a probe DNA, E. lathyris genome was suggested to have a gene family consisting of 3-5 gene members putatively encoding subtilisin-like proteases.

Effects of Plant Extract containing Creams on UVB Radiation-induced Inflammatory Responses in Mice (쥐에서 자외선 B가 유도한 염증 반응에 대한 식물 추출물 함유 크림의 영향)

  • Ultraviolet radiation (UV) is a major cause of photodamages to human skin and the immediate responses of the skin to UV include the erythema and edema. In an attempt to find effective UV-protecting agents to be used in cosmetics, a number of plant extracts were screened in the cell-based assays. Among the total of 38 plant extracts tested, 3 plant extracts derived from Sasa quelpaertensis, Althaea rosea, and Dryopteris crassirhizoma attenuated the UVB-induced cytotoxicity as well as melanin synthesis in cultured human epidermal melanocytes. The anti-inflammatory effects of these plant extracts were further examined in animal models. A control or test cream containing 1% of a plant extract was topically applied to ears of a C57BL/6 mouse or the dorsal skin of a SKH-1 hafirless mouse before and after the exposure to UVB. The change in ear thickness or dorsal skin redness due to UVB exposure was determined to monitor edema and erythema, respectively. All three test creams exhibited anti-inflammatory effects in both experiments. The creams containing Sasa quelpaertensis, Althaea rosea or Dryopteris crassirhizoma extract alleviated the UVB-induced edema response on day 4 by 53.8 %, 56.4 % and 31.1 %, respectively. They also inhibited the erythema formation on day 2 by 45.7 %, 34.1 % and 20.5 %, respectively. This study suggests that the selected plant extracts formulated in cosmetics may attenuate skin inflammation caused by overexposure to UV.

  • The environmental pollutions were a serious problem in Korea recently. So many researcher have studied the effect of environmental pollution on plants and agro-ecosystem, but the basic mechanisms of environmental stresses were various. One of the important mechanisms was oxidative stress caused by active toxic oxygen. The toxic oxygen was generated by several stresses, abnormal temperature, many xenobiotics, air pollutants, water stress, fugal toxin, etc. In the species of toxic oxygen which is primary inducer of oxidative stresses, superoxide, hydrogen peroxide, hydroxyl radical and singlet oxygen were representative species. The scavenging systems were divided into two groups. One was nonenzymatic system and the other enzymatic system. Antioxidants such as glutathione, ascorbic acid, and carotenoid, have the primary function in defense mechanisms. Enzymatic system divided into two groups; First, direct interaction with toxic oxygen(eg. superoxide dismutase). Second, participation in redox reaction to maintain the active antioxidant levels(eg. glutathione reductase, ascorbate peroxidase, etc.).

  • Parameter uncertainty and sensitivity of KFOOD model for calculating the ingestion dose via terrestrial food-chain pathway was analyzed with using Monte-Carlo approach. For the rice ingestion pathway, estimated values from KFOOD code were very conservative. Most sensitive input parameters in model were deposition velocities and soil-to-plant transfer coefficient of radionuclides.

  • Gibberellic Acid-Stimulated Arabidopsis (GASA) genes are involved in plant hormone signaling, cell division and elongation, as well as in responses to stress conditions in plants. In this study, we isolated a GASA gene from hybrid poplar (Populus alba ${\times}$ P. glandulosa) and analyzed its physiological phenotype and molecular functions in poplar. PagGASA cDNA encodes a putative protein composed of 95 amino acids containing an N-terminal signal peptide and a conservative cysteine-rich C-terminal domain. Southern blot analysis revealed that one or two copies of the PagGASA are present in the poplar genome. The PagGASA transcripts were highly detected in flowers and roots. Moreover, the expression of PagGASA was induced by growth hormone (gibberellic acid) and stress hormones (abscisic acid, jasmonic acid, and salicylic acid). By using transgenic analysis, we showed that the upregulation of PagGASA in poplar provides high tolerance to drought stress. Therefore, our results suggest that PagGASA plays an important role in drought stress tolerance via stress-related plant hormone signaling in poplar.

  • Quantitative reverse transcription PCR is used for gene expression analysis as the accurate and sensitive method. To analyze quantification of gene expression changes in apple plants, 10 housekeeping genes (ACT, CKL, EF-1α, GAPDH, MDH, PDI, THFs, UBC, UBC10, and WD40) were evaluated for their stability of expression during infection by Apple stem grooving virus (ASGV) or in cold-stress apple plant buds. Five reference-gene validation programs were used to establish the order of the most stable genes for ASGV as CKL>THFs>GAPDH>ACT, and the least stable genes WD40CKL>UBC10, and the least stable genes were ACT

  • In order to investigate the relation between the phytoplankton and the catch fluctuation of set net fishing grounds located in the coastal waters of Yeosu, phytoplankton observations on the fishing ground were carried out by the training ship of Yeosu, Fisheries University from April to November in 1990, and the data obtained were compared with the catch data from the joint market of yeosu fisheries cooperative society in 1990. The results obtained are summarized as follows: 1) The phytoplanktons were more appeared in summer than in spring or autumn and their quantity was much in the shore of Dolsan-do, and little in the offshore waters distributed in the shores of Sori-do and Sejon-do, Thus, the quantity of the planktons could be used for estimating the distribution of watermass. 2) The fishes caught by the set net were arranged in the order of catch amounts as follows: Spanish mackerel > Hair tail > Common mackerel > Sardine > Anchovy > Horse mackerel > Yellow tail. The catches of anchovy and Sardine were high in April to May and Hair tail. Horse mackerel and Common mackerel were caught from June to October. But Spanish mackerel were caught during the whole period of fishing. 3) The catches by set nets showed a correlation with the quantity of phytoplanktons. The planktons appeared most in the inner waters. influenced largely by the inflow of land waters in summer. and the catches were high in summer when the offshore water was distributed least. Therefore, the most important factor influencing the catches were regarded to be the productivity of food organism in inner water into which abundant nutrients were supplied by the inflow of land waters. That is, the fluctuation of catches by set nets seemed to be influenced mainly by the productivity of food organism.

Disease Resistance-Based Management of Alternaria Black Spot in Cruciferous Crops (병 저항성 기반 십자화과 작물의 검은무늬병 관리)

  • Alternaria black spots or blights in cruciferous crops have been devastating diseases worldwide and led to economic losses in broccoli, Chinese cabbage, kale, radish, rapeseed, etc. These diseases are caused by different Alternaria spp., including A. brassicae, A. brassicicola and A. raphani transmitted from infected seeds or insect vectors. Efforts to excavate disease resistance traits of cruciferous crops against Alternaria black spots or blights have been demonstrated. Genetic resource of disease resistance was investigated in the wild relatives of cruciferous crops, and different cultivars were screened under different inoculation conditions. Development of the disease-resistant lines against Alternaria black spots or blights was also tried via genetic transformation of the cruciferous crops using diverse plant defence-associated genes. Plant immunity activated by pre-treatment with chemicals, i. e. β-amino-n-butyric acid and melatonin, was suggested for reducing Alternaria black spots or blights in cruciferous crops. The disease resistance traits have also been evaluated in model plant Arabidopsis originating from different habitats. Various plant immunity-related mutants showing different disease responses from wild-type Arabidopsis provided valuable information for managing Alternaria black spots or blights in cruciferous crops. In particular, redox regulation and antioxidant responses altered in the Alternaria-infected mutants were discussed in this review.

  • 사람의 흡충류감염(吸蟲類感染)은 대부분(大部分) 열대(熟帶) 및 동부아세아지역(東部亞細亞地域) 여러 국가(國家)에서 발생(發生)한다. 주혈흡충증(住血吸蟲症), 간흡충증(肝吸蟲症), 태국간흡충증(泰國肝吸蟲症), 간흡충증(肝吸蟲症), 요꼬가와흡충증(吸蟲症), 이형이형흡충증(異型異型吸蟲症) 및 폐대흡충증(肺大吸蟲症)등은 어떤 일부(一部) 아세아국가(亞細亞國家)에 있어서 공중보건상(公衆保健上) 중요(重要)한 문제(間題)가 되어 있다. 이와 같은 패류매개성기생충감염(貝類媒介性寄生蟲感染)은 풍토병(風土病)을 일으키고 있다. 사람들의 생활습관은 풍토병을 일으키게될 원인이 될 뿐만 아니라 때로는 그 질병(疾病)을 유행(流行) 전파(傳播)시키는 기본적 요소가 되기도 한다. 주혈흡충을 제외한 흡충류감염은 우리가 중간숙주(中間宿主)을 먹었을때 그곳에 있었던 피낭유충(被囊幼蟲)에 의하여 감염이 이루어 진다. 식물(植物)의 생식(生殖)(간질(肝蛭), 폐대흡충(肺大吸蟲)), 게 및 가재 생식(폐흡충), 민물고기 생식(간흡충, 요꼬가와흡충, 이형이형흡충), 또는 우렁이 생식(극구흡충(棘口吸蟲)), 기타(몇가지 우연적감염(偶然的感染))등은 사람의 흡충류감염의 주 원인이 되는 것이다. 한편 주혈흡충 감염은 요염수원(汚染水源)에 접촉한 피부을 뚫고 들어온 쎌카리아 유충(幼蟲)에 노출되기 때문에 일어난다. 흡충류감염에 대하여 예방과 방어에 관한 공중보건학적 조치는 많다. 이들의 조치중에는 화학요법에 의한 감염원을 감소시키는 것, 패류숙주의 박멸, 만족할만한 위생시설의 비치, 급수시설, 보건교육, 진단기술의 보급, 환경관리 및 개선 등이 포함된다. 그러나 감염자들에 대한 화학요법은 가장 빠르고 효과적인 억제방법인 것으로 나타나고 있다. 흡충류감염을 예방하는데 있어서 가장 중요한 변화는 프라지콴텔과 같은 신약의 개발이었다. 프라지콴텔은 모든 종류의 주혈흡충, 간흡충, 폐흡충 및 여러가지 장내기생 흡충류등 사람의 병원성흡충류에 대한 치료제로서 가장 좋은 특효약이다. 흡충류감염이 중요한 보건문제가 되어 있는 곳에서는 그 예방과 억제책에 있어서 장단기계획(長短期計劃)을 수립하는 것이 좋을 것 같다. 단기는 집단화학요법을 조기에 실시하여야 하고 자기는 "비특이성 기구"로써, 예를들면 위생시설 및 급수시설의 설치, 보건교육등으로 감염을 감소시켜 감염유지 수준이하로 보지(保持)시키는 것이다. 이와같은 억제조치를 착실히 수행하기 위하여 많은일이 있다. 예를 들면 집단참여, 역학적 조사, 약물의 공급, 환경관리, 보건교육, 지역사회관련, 일차보건 진료와의 협조 및 재정후원등에 대하여 검토하고 연구하여야 할 것이다.

Exploring on the Defense Strategies against Hervivory of Several Broad-leaved Tree Species in Solomon Islands (솔로몬제도에 생육중인 몇 활엽수종의 초식에 대한 방어전략 탐색)

  • To explore on the defense strategies against hervivory of broadleaved tree species growing in tropical rain forest, Solomon Islands, morphological characteristics of the leaf, leaf domatia structure and the number, herbivores insects and mites on the leaves of 26 tree species, collected from the trees growing in Guadalcanal, New Georgia, and Tetepare islands, were investigated from Feb. 25 to March 5, 2010. The results are summarized as follows. Six tree species including Terminalia catappa(23.08%) among total 26 species have domatia structure. Especially, domatia of T. catappa reveals two types; pouch type and pit+tuft type. Predatory mites are observed on the leaves of only three species including T. catappa. Predatory mites' number per leaf or leaflet proved a few, 0.33~0.40. Insect pests destroying the leaf of tree species are such as aphids, Diaspididae sp., larva of Lepidoptera, and Eriosoma sp.(Pemphlogidae). Ants are co-works with aphids and Diaspididae sp., and supports herbivores. These results indicate that protective mutualisms between tree and predatory mites may be less frequent in woody species of tropical rain forest in Solomon Islands.
