Ecophysiological Studies on the Water Relations of Economic Tree Species - Temporal Changes of Stomatal Responses to Soil Moisture Regimes and Exogenous Abscisic Acid in Oaks and Ash - (주요 경제 수종의 수분 특성에 관한 생리생태학적 연구 - 토양수분 조건 및 ABA 처리에 따른 참나무류와 물푸레나무 기공의 시계열적인 변화 반응 -)

  • Seasonal and diurnal changes of stomatal diffusive resistance(S.D.R.) and transpiration rate(T.R.) were investigated for determining the ecophysiological water relations of economic tree species subjected to chronic water stress or exogenous abscisic acid treatment. Four species of oaks including Quercus serrata, Q. mongolica. Q. acutissima, and Q. variabilis were used as the experimental materials and also Fraxinus rhynchophylla was studied together with oaks. Stomatal diffusive resistances were repeatedly measured on the containerized 1-0 year seedlings subjected to two kinds of soil moisture regime (wet and dry) in June, August, and September by LI-1600 Steady State Porometer of LI-Cor, Inc.. Exogenous abscisic acid (ABA) solutions of 0.5 mM and 0.05 mM in July and August, respectively, were absorbed into shoots cut from the containerized seedlings for determining their effects on stomatal behavior and transpiration. Most of measurements in stomatal diffusive resistance maintained about 5 s/cm in the morning after sunrise despite of different treatments. But the values fluctuated frequently to high level above 20 s/cm through the afternoon until sunset in the seedlings subjected to dry soil moisture regime. Despite of various treatments and environmental conditions, stomatal diffusive resistances of Q. variabilis were more stable than those of Q. serrata or Q. acutissima. Their values of F. rhynchophylla changed more irregularly in comparison with those of oak species. Exogenous abscisic acid absorbed into shoots cut from seedlings increased stomatal diffusive resistance obviously in most of the species studied. The stomatal responses to abscisic acid treatment were more sensitive in July especially in Q. serrata than in Q. variabilis and Q. acutissima. But the effects of ABA treatment were presented more remarkably in Q. acutissima in August. The responses to abscisic acid were not certain in F. rhynchophylla because of their various fluctuation patterns.

Studies on. the Green-Soybean Cultivation as preceding Crop of the Rice in the Paddy-Field in the middle Parts of Korea (중부지방에 있어서 풋콩의 답전작재배에 관한 연구)

  • The followings are the results obtained in a series of experiments concerning of the varieties, the short-day-treatment, the fertilizer application, and the planting space of soybean, which were carried out to investigate the methods best suited for cultivating green soybean as the preceding crop of the rice. in the paddy-field in the middle parts of Korea at the practice farm attached to Agriculture College of Kon Kuk University in 1972. 1. Though the varieties of soybean was planted on the hot-bed on March 15 and then set in the main plot, none of them did flower within May 15, which is the limit time of flowering in growing soybean as the preceding crop of the rice in the paddy-field without the short-day-treatment being applied during raising seedlings. 2. The earliest-maturing variety groups such as HOKKAl#l, WASEMIDORI, YAEHUSANARI, MIT AKARAHAKUCHO, and VERDE flowered within May 15 by the short-day-treatment during raising seedlings. 3. The optimum hours of the day length was known to be 7 to 9 in the medium-maturing and late-maturing variety groups and 7 to 11 in the early-maturing and the earliest-maturing variety groups in the case of appling the short-day-treatment for 10 days from the beginning of the primary regular compound leaf development. 4 The optimum days in appling the short-day-treatment for 11 hours a day was recognized to be about 10 days regardless of the maturity of varieties. 5. Reduction of days required to flower by the short-clay-treatment, that is, light-sensitivity was remarkably higher in the medium-maturing and the late-maturing variety groups than in the earliest-maturing and the early-maturing variety groups. 6. The yield showed an increase of about 17 per cent in the case of appling the standard amount of nitrogen(4.0 kg/10 a), but it tended to reduce on the contrary in appling the increased amount of nitrogen. 7. The application of increased amount of phosphate had less significant effect on the yield increase than in the case of application of its standard amount( 4.0 kg/l0 a). 8. When the number of transplantation plant was changed from 54 to 130 per 3.3 $m^2$, the yield in 130 plant plot was about two times so higher than in 54 plant plot that the effect of close planting cultivation on the yield was proved to be remarkable. 9. Conditions possible for cultivating green soybean as the preceding crop of the rice in the paddy-field in the middle parts of Korea are turned to be as follows: (a) to plant the earliest-maturing-variety groups on the hot-bed on March 15. (b) to apply the short-day-treatment by 11 hours a day for 16 days from April 16, which is about the time when the primary regular leaf begin to develop. And it was, found to be a most remarkable in the increased yield that apply nitrogen 4.0 kg, phosphate 4.0 kg, and potassium 6.0 kg per 10 a as basic manuring totally and apply the close planting by 130 plants per $3.3{m^2}$(50 cm $\times$ 5 cm).

Effect of Microbial Additives on Metabolic Characteristics in Sheep and Milking Performance of Lactating Dairy Cows (미생물제제의 첨가가 면양의 반추대사 및 젖소의 유생산성에 미치는 영향)

  • Two experiments were conducted to observe the effects of direct fed microbials on metabolic characteristics in sheep and milking performance in dairy cows. A metabolic trial with four ruminally cannulated sheep(60±6kg) was conducted in a 4×4 Latin square design to investigate the supplementation effects of Saccharomyces cerevisiae, Clostridium butyricum or mixed microbes of S. cerevisiae and C. butyricum on ruminal fermentation characteristics and whole tract digestibility. Sheep were fed 1.25 kg of total mixed ration(TMR, DM basis) supplemented with S. cerevisiae (2.5g/day), C. butyricum (1.0g/day) or its mixture(S. cerevisiae 1.25g/day+C. butyricum 1g/day), twice daily in an equal volume. But control sheep were fed only TMR. A feeding trial with 28 lactating Holstein cattle was also conducted for 12 weeks to investigate the effects of the same microbial supplements as for the metabolic trial on milking performance. The cows were fed the TMR(control), and fed S. cerevisiae(50g/day), C. butyricum(15g/day) or its mixture (S. cerevisiae 25g/day + C. butyricum 7.5g/day) with upper layer dressing method. Total VFA concentration and the digestibility of whole digestive tract in the sheep increased by supplementation of S. cerevisiae, C. butyricum or their combined microbials compare to control group. The proportion of propionic acid at 1h(P<0.039) and 3h(P<0.022) decreased by supplementation of S. cerevisiae while tended to increase acetic acid proportion at the same times. Daily dry matter intake(DMI) was not influenced by the microbial treatments, but milk yield(P<0.031) and feed efficiency(milk yield/DMI, P<0.043) were higher for the cow received C. butyricum than those for other treatments. The milk fat content was higher (P<0.085) when cows fed S. cerevisiae(4.11%) than that fed the control (4.08%), the diets with C. butyricum (3.85%) and the microbial mixture. Based on the results obtained from the current experiments, supplementation of C. butyricum or mixture with S. cerevisiae might be increased milk fat content and milk productivity of lactating daily cows. Surfactant Enhanced In-Situ Soil Flushing Pilot Test for the Soil and Groundwater Remediation in an Oil Contaminated Site (계면활성제 원위치 토양 세정법을 이용한 유류 오염 지역 토양.지하수 정화 실증 시험)

  • Surfactant enhanced in-situ soil flushing was performed to remediate the soil and groundwater at an oil contaminated site, where had been used as a military vehicle repair area for 40 years. A section from the contaminated site (4.5 m $\times$ 4.5 m $\times$ 6.0 m) was selected for the research, which was composed of heterogeneous sandy and silt-sandy soils with average $K_d$ of 2.0$\times$$10^{-4}$cm/sec. Two percent of sorbitan monooleate (POE 20) and 0.07% of iso-prophyl alcohol were mixed for the surfactant solution and 3 pore volumes of surfactant solution were injected to remove oil from the contaminated section. Four injection wells and two extraction wells were built in the section to flush surfactant solution. Water samples taken from extraction wells and the storage tank were analyzed on a gas-chromatography (GC) for TPH concentration in the effluent with different time. Five pore volumes of solution were extracted while TPH concentration in soil and groundwater at the section were below the Waste Water Discharge Limit (WWDL). The effluent TPH concentration from wells with only water flushing was below 10 ppm. However, the effluent concentration using surfactant solution flushing increased to 1751 ppm, which was more than 170 times compared with the concentration with only water flushing. Total 18.5 kg of oil (TPH) was removed from the soil and groundwater at the section. The concentration of heavy metals in the effluent solution also increased with the increase of TPH concentration, suggesting that the surfactant enhanced in-situ flushing be available to remove not only oil but heavy metals from contaminated sites. The removal efficiency of surfactant enhanced in-situ flushing was investigated at the real contaminated site in Korea. Results suggest that in-situ soil flushing could be a successful process to remediate contaminated sites distributed in Korea.

Effect of Dietary Mogchotan Supplementation on Fattening Performance, Fatty acid Composition and Meat Quality in Pigs (사료내 목초탄 첨가가 비육돈의 비육능력, 지방산 조성 및 육질에 미치는 영향)

  • This study was conducted to examine the effects of dietary Mogchotan(the mixture of charcoal and pyroligneous acid, 80:20, w/w) supplementation on fattening performance, fatty acid composition and the physico-chemical characteristics of meat in pigs. The present study was also stressed to investigate the possibility of industrial utilization of charcoal and pyroligneous acid as a livestock feed additive. Weight gain and feed conversion in pigs fed the Mogchotan supplemented diet were higher than those of the control group. In fatty acids composition, palmitic acid(C16:0) contents of Mogchotan treatment groups were lower than that of control group. However, Mogchotan supplementation increased C16:1, C18:0, C18:1, C18:2 and C18:3 contents when compared with control group pigs. Also, Mogchotan supplementation groups decreased saturated fatty acids level than control group. On the other hand, Mogchotan supplementation showed higher unsaturated fatty acids value, especially polyunsaturated fatty acids value compared to control group. The carcass pH of pigs fed the Mogchotan tended to be higher than control, but was not significantly different. The water holding capacity was significantly higher in pigs fed the 3.0% Mogchotan-supplemented diet than those of other treatment groups(p<0.05). Altogether, it has been suggested that dietary $1{\sim}3%$ of Mogchotan supplementation improved the fattening performance and meat quality in pigs.

The Effect of Alcohol Fermented Feedstuff Made of Byproducts on In vitro Fermentation Characteristics and NDF Disappearance in the Rumen (조사료 대용 산업부산물을 이용한 알코올 발효사료가 In vitro 반추위내 발효특성 및 NDF 분해율에 미치는 영향)

  • An in vitro study was conducted to examine the influence of alcohol-fermented feedstuff formulated with byproducts on the fermentation characteristics and NDF disappearance in the rumen. Dietary treatments were either a soybean curd-based alcohol-fermented feedstuff (AFS) or brewery grain-based alcohol-fermented feedstuff (AFB). The AFS and AFB are composed of 50% commercial beef cattle feed, 50% soybean curd dreg, 5% molasses and 0.5% yeast and 25% commercial beef cattle feed, 25% brewery grain, 25% soybean curd dreg, 25% corn grit, 5% molasses and 0.5% yeast, respectively. The change of ammonia, pH alcohol, volatile fatty acids, and NDF disappearance were measured at 0, 2, 4, 8 and 12 hr after in vitro incubation in the rumen. After 2 hr incubation, higher ammonia concentrations were resulted in AFS (12.47 mg/dl) and AFB (12.85 mg/dl) compared to control (11.84 mg/dl) (p<0.05). Ruminal pH of AFS and AFB were significantly higher than control during 1 to 6 hr fermentation, but the pH of AFS and AFB were decreased after 6 hr. At 12 hr fermentation, the alcohol concentration of AFS and AFB was significantly increased by 43.9% and 48.0%, respectively. The acetate concentration was rapidly decreased in control, while the concentration was slowly decreased in AFS and AFB with increasing the fermentation time. Lower concentrations propionate and butyrate were observed in AFS and AFB during every fermentation time (p<0.05). The NDF disappearance was significantly lower in AFS and AFB after 4 hr fermentation. These results suggest that alcohol fermented feedstuff can control the fermentation pattern in the rumen.

  • A barley(Hordeum vulgare L.) cultivar 'Youjin' with hooded spike type having good silage quality was developed at National Institute of Crop Science, RDA in 2016. 'Youjin' showed both high yielding and cold resistance through the preliminary and advanced yield trials(PYT, AYT) from 2012 to 2013. We conducted regional yield trials(RYT) of 'Youjin' in six locations around Korea for three years from 2014 to 2016. It had erect plant type, growth habit of II, the green leaf and hooded awn type. In the paddy field its heading date was April 24 and the maturing date was May 25. Plant height was 99 cm and the number of spikes per ㎡ was 696. It has high rate of leaf blades, resistance to BaYMV(Barley Yellow Mosiac Virus) and good winter hardiness. The average dry matter of Youjin was about 17.2 MT ha-1 in the field. And feed quality of 'Youjin' was 10.6% of crude protein content, 24.8% of ADF(Acid Detergent Fiber), 43.5 % of NDF(Neutral Detergent Fiber), 69.1% of TDN(Total Digestible Nutrients). And also 'Youjin' had grade I of silage quality.

Antioxidant Activities of Stachys sieboldii MIQ Roots (초석잠(Stachys sieboldii MIQ) 뿌리 추출물의 항산화 활성 고찰)

  • The root of Stnchys Sieboldif MIQ was extracted three times with methanol and extract was found to contain 3.02% of polyphenols and 1.97% of flavonoids. DPPH radical scavenging method, ferric thiocyanate method, and nitrite scavenging ability method were employed to investigate the constituents of the extract and to measure their activity on antioxidation. The fraction extracted by ethylacetate showed higher anti oxidation value than that of $\alpha-tocopherol$, butylated hydroxyanisole (BHA), and butylated hydroxytoluene (BHT) at the same concentration. UV-VIS spectral data of the extract by ethylacetate that was isolated on a silica gel column proved adsorption maxima in the range of 280∼330 nm. The fraction ES-RS that has $\lambda_{max}(nm)$ of band 1, 325nm and band II, 289nm exhibitd the strongest activity on antioxidation. ES-R5 fraction showed similar pattern to flavones by the analysis of UV-VIS spectral data.

An Evaluation of Vitek MS System for Rapid Identification of Bacterial Species in Positive Blood Culture (혈액배양 양성검체에서 패혈증 원인균 신속동정을 위한 Vitek MS 시스템의 유용성 평가)

  • The aim of this study was to shorten the time required for subculture and bacterial identification and obtain a simple and rapid identification method for new test methods for bloodstream infections. The following results were obtained using a mass spectrometer. In Vitek 2, 208 (81.8%) cases were well-identified and 45 isolates were not identified in blood cultures. Among 208 cases, 146 (57.5%) were Gram positive bacteria and 108 (42.5%) were Gram negative bacteria. In total, 233 were identified to the species level and 21 were identified to the genus level. The identification error was found to be Propionibacterium acnes as Clostridium bifermentans. The accuracy of Enterobacteriaceae, glucose non-fermentative bacilli (GNFB), and staphylococci were 81/83 (97.6%), 12/15 (80.0%), and 72/85 (84.7%), respectively. The concordance rate of Vitek 2 and Vitek MS by the direct method was 81.8% and 45 isolates were not identified. Most of the unidentified bacteria were Gram positive bacteria (N=37). The Gram positive bacteria were streptococci (14), coagulase-negative staphylococci (CNS) (11), enterococci (3), Staphylococcus aureus (2), Micrococcus spp. (2), Bacillus spp. (2) and Actinomyces odontolyticus, Finegoldia magna, and Peptostreptococcus spp. The results reporting time was reduced to 24~72 hours compared to the conventional method. The rate of identification of the aerobic and anaerobic cultures was similar, but the use of an anaerobic culture did not require a dissolution process, which could shorten the sample preparation time. These results suggest that the method of direct identification in blood cultures is very useful for the treatment of patients. In further studies, it might be necessary to further improve the method for identifying streptococci and CNS, which were lacking in accuracy in this study.

Improvement of the Fishing Gear and Fishing Method of the East-Sea Trawl Fishery (동해구 트롤 어구어법의 개량)

  • A serious of studies on the fishing gear and system of the East Sea trawl fishery was carried out to improve the fishing efficiency and the working conditions. As the first step of these studies, the fishing gear and system of the traditional East Sea trawl were checked in order to solve the some problems, such as the poor sheering efficiency of net mouth, the inconvenient fishing system of the side trawl and etc. And then the fishing system was reorganized from the side trawl into the stern trawl by setting up the net drum system on the stern deck, and introduction of two types of new designed nets, one for mainly the midwater trawl and the other for the bottom trawl. The results of the field experiment on the modified system and nets can be summarized as follows : 1. the modified system was well worked and could save the man-labour by about 80%. 2. The sheering efficiency of the improved net, A type was improved to 20 m height and 30 m width in the net mouth, and that of B type net, to 10 m height and 33 m width, compared with 1.5 m height and 15 m width in the traditional net. 3. Catch efficiency of pink shrimp in A or B type net was better about 3 or 5 times than that of traditional net, and in B net, for herring and other bottom fishes is better about 2 times than that of the traditional net.
