Effect of plant growth regulators on plant regeneration from the Belamcanda chinensis (범부채 (Belamcanda chinensis)의 식물체 재분화에 미치는 식물생장조절제의 영향)

  • To establish the optimum conditions of in vitro plant regeneration, the leaf, rhizome, and root explants of Belamcanda chinensis were cultured on the MS medium supplemented with different concentration of 2,4-D and BA. The callus induction was more effective in the root explants than the leaf and rhizome explants, and was the best on MS medium containing 3.0 mg/L 2,4-D or 1.0 mg/L 2,4-D and 3.0 mg/L BA. The highest numbers of shoots were regenerated when callus were cultured on MS medium containing 3.0 mg/L 2,4-D for 4 weeks. However, the multiple shoots were induced on MS medium supplemented with the combination of 2,4-D and BA. The normal root formation from shoot was effective on the MS medium lacking any plant growth regulators. For acclimatization, the regenerated plantlets were cultured on MS medium without sucrose and plant growth regulators for 2 weeks, and then transferred to the pot.

  • Effect of BA concentrations and culture methods on in vitro plant multiplication from shoot-tip cultures of Wasabia japonica was studied. Shoot-tips with leaf primordia and apical meristem were cultured on MS basal medium for all the experiments. Liquid medium for 2 weeks followed by semi-solid medium for 4 weeks containing 1.0 mg/L BA was the best to number of shoots (22.8) and shoot length (3.5 cm). Shoots proliferated could be divided into ca. 5 to 11 of cultures for the multiplication of plantlets. Divided plantlets showed root formation (90%) well onto MS basal medium without growth regulators like IBA and NAA. After rooting, all the plantlets transferred into the pots containing composed soil (bio-media Co., peatmoss $8{\sim}10%$, coir dust $66{\sim}70%$, zeolite $13{\sim}17%$, vermiculite $3{\sim}7%$, perlite $2{\sim}4%$) and grown well into whole plants with multiple shoots.

  • A protocol for the in vitro propagation of Chamaecyparis obtusa was established in the present study. Multi-shoots were initiated from apical shoot explants from germinants after 10 weeks of culture on Litvay medium (LM) supplemented with different concentrations of cytokinin. The effects of pre-treatment with high concentrations of cytokinin and varying concentrations (0.2 to 5.0 mg/L) of zeatin on in vitro shoot elongation and shoot multiplication were investigated. Optimal shoot growth was achieved on LM medium, with over 10-mm shoots after 10 weeks of culture. In the anti-browning tests, ethanesulfonic acid triggered the least browning in the shoot tips. The highest multi-shoot induction was observed in the 0.5-mg/L zeatin treatments, which yielded 80% induction of shoots after 10 weeks of culture, and maximum shoot elongation was observed in the LM basal medium without the hormone. The highest rooting rates were 65% under 0.2 mg/L indole-3-butyric acid.

Plant Regeneration from Leaf derived Callus of Hybrid Kiwi (Actinidia deliciosa × A. arguta) (잡종키위 (양다래×다래)의 엽조직 캘러스로부터 식물체 재분화)

  • Whole plants were regenerated from callus induced from leaf explants in hybrid kiwi (Actinidia deliciosa${\times}$A. arguta). Callus was induced from leaf explants which cultured on MS solid medium supplemented with combination of auxin (2,4-D, NAA: 0.1~0.5 mg/l) and cytokinin (BA: 0.1~0.2 mg/l). them, the highest callus formation (96.2%) was obtained from the treatment of 0.5 mg/1 2,4-D+0.1 mg/l NAA+0.05 mg/l BA. In the experiment of adventitious shoots induction from primary shoots, only a few shoots were produced in the treatment of 1.0 mg/l BA+0.05 mg/l IBA or 2.0 mg/l BA+0.05 mg/l lBA. As the callus were transferred to the secondary shoot-inducing medium, multiple shoots were obtained from the medium supplemented with 1.0, 2.0 or 5.0 mg/l zeatin in addition to the mixed treatments of BA, thidiazuron (TDZ) or zeatin. However, no multiple shoots were induced on the BA-contained medium of concentrations. Therefore it turned out that addition of BA to medium was less effective for induction of multiple shoots from callus in Actinidia deliciosa${\times}$A. arguta. For producing adventitious roots from shoots, the best frequency of rooting (83.3%) were recorded on the treatment of in vitro rooting (Standardi (St)+1.0 mg/l IBA). On the other side, the lowest result (40.0%) were shown in the treatment of 500 mg/l IBA, 1 hr. Whole plants with shoots and roots were recovered and acclimatized successfully.

Factors Influencing Somatic Embryo Induction and Plant Regeneration in Aralia elata Seem. (두릅(Aralia elata)의 체세포배 유도, 발아 및 식물체 재분화에 미치는 요인)

  • In order to find optimum conditions for somatic embryogenesis from different individual (2-year-old) in Aralia elata were cultured on MS medium supplemented with 1.0 mg/L 2,4-D, 3% sucrose, and 0.3% gelrite. We also investigated the effect of MS medium salt concentration, BA and ABA on the embryo germination and plant regeneration. While noticeable difference was observed on somatic embryo induction among different individual tree, no apparent difference was seen in both germination and regeneration frequencies. Compared with nonembryogenic calli, embryogenic calli tended to look yellow and/or pale brown in color, slowly growing and soft in their texture. Regardless of BA or ABA treatment, half-strength MS salt medium proved to be better than full strength MS medium in both embryo germination and plant regeneration. Both hypocotyl and cotyledon developments were slightly promoted by adding 0.1 mg/L BA. However, its effect on germination and regeneration seemed inferior to control. ABA treatment on somatic embryos at their torpedo and early cotyledonary stages resulted in poor response in germination and regeneration. Although most regenerated plantlets varied greatly in cotyledon number and shape, they could be developed into normal plants after 4 weeks in culture. More than 95% plantlets were acclimatized in an artificial soil mixture, successfully transplanted to nursery bed and grew normally without any phenotypic abnormalty.

Micropropagation of Juvenile and Mature Tree of Corylopsis coreana by Axillary Bud Culture (액아배양에 의한 유묘 및 성숙 히어리나무의 기내번식)

  • We have developed an in vitro micropropagation system via shoot formation from axillary buds using nodal segments of Corylopsis coreana. Explants from both juvenile tree (one-year-old greenhouse stock seedlings) and mature tree (ten-years-old tree in nursery) were compared with regard to propagation efficiency. Combined treatment of both BA and zeatin were effective on shoot proliferation since the best result was obtained on MS medium supplemented with 0.5∼3.0 mg/L zeatin and 0.2 mg/L BA. Generally, juvenile explants were better in both shoot proliferation and growth than mature explants. However, as the duration of in vitro culture was proceed to 6 months, explants from mature tree also produced three shoots per explant. Distinctive differences in rooting and adaptability to soil of shoots obtained from mother trees. Whereas shoots originated from juvenile explants rooted as high as 97%, those from adult explants showed 62% rooting. Similar result was also observed in soil acclimatization. The plantlets derived from juvenile plants survived 67%, while only 48% of those from adult trees survived. The results showed a possibility of the micropropagation of Corylopsis coreana through shoot formation from axillary buds. In addition, the advance of the research still remain to enhance the frequency of acclimatization of plantlets from mature trees for practical application.

Plant Regeneration and Effect of Auxin and Cytokinin on Adventitious Shoot Formation from Seedling Explant of Taraxacum platycarpum (민들레 [Taraxacum platycarpum]유식물 절편으로부터 부정아 발생에 미치는 auxin과 cytokinin의 영향 및 식물체 재생)

  • Taraxacum platycarpum has been used as a medicinal plant. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on medium with various kinds of growth regulators. Adventitious shoot formation was achieved when cytokinin was used alone. Shoot formation was higher on MS medium containing 2 mg/L BAP compared to that with 2 mg/L kinetin and 2 mg/L 2-ip. Among root, hypocotyl and cotyledon, roots were the best explant for the adventitious shoot induction. Adventitious shoot formation from roots declined markedly by the combination of both 0.1 mg/L NAA and 2 mg/L BAP, while shoot formation from cotyledons was stimulated by the same combination. Root formation from the regenerated shoots was achieved on 1/3MS medium containing 0.2 mg/L NAA. Regenerated plantlets was acclimatized and transplanted to the soil, showing 100% survival.

Optimization of apical tip culture condition for In Vitro propagation of 'Gisela 5' dwarf cherry rootstock (양앵두 왜성대목 'Gisela 5'의 기내번식을 위한 정단배양조건의 최적화)

  • Based on the results in this study, here we propose a systematic micropropagation process for 'Gisela 5' that is one of the important dwarfing cherry rootstocks. When the apical tips detached from newly developed shoot in spring season were cultured on the half strength MS media with 0.5 mg/L IBA and 0.5 ~ 1.0 mg/L BA, the cultures scored the highest acquisition rate at 90% for normal shoot with vigorous growth and without hyperhydricity. As next step, the young shoots maintained in vitro well multiplied on the full strength MS medium supplemented with 0.5 mg/L IBA and 0.5 mg/L BA, in which multiplication rate was approximately nine-fold. Given the half strength MS medium containing 2.0 mg/L IBA, each transplanted shoot further developed robust roots. Finally, the plantlets were easily acclimatized in the compost consisted of vermiculite, perlite, and peatmoss in the proportion of 1:1:1. We expect that the results are useful for cherry cultivation and its rootstock production.

Cucumber green mottle mosaic virus Moved into the Non-Host Figleaf Gourd Passing through Cucumber in Grafting System (오이/흑종호박 접목에서 오이녹반모자이크바이러스의 비기주 대목인 흑종호박으로 이동)

  • Cucumber green mottle mosaic virus (CGMMV) was not infected in figleaf gourd by sap inoculation. However CGMMV was detected by RT-PCR from the figleaf gourd collected from a field growing cucumber grafted onto figleaf gourd in Cheonan, Chungcheongnam Province in 2008. Which field showed 100% infection rate of the virus disease. In the experiment grafted with cucumber onto figleaf gourd, transportation of CGMMV through cucumber to figleaf gourd was confirmed by RT-PCR when the virus was mechanically inoculated on the leaves of the cucumber. The amplified DNA concentration of the virus on electrophoresis gel was much higher in the cucumber than in the figleaf gourd. However, the virus particles from the figleaf gourds were not observed under electron microscopy, also sap of the figleaf gourds was not transmittable to Nicotiana benthamiana. To identify the existence of CGMMV particle, the virus was purified from figleaf gourd and cucumber growing together in the graft system. CGMMV solution extracted from the cucumber represented a typical absorption spectrum of the virus but that from the figleaf gourd did not. Only a few CGMMV particles were observed in the purified preparation from the figleaf gourd. These results confirmed that CGMMV only passed through figleaf gourd in the grafting system. This study indicated that figleaf gourd is not a host of CGMMY.

Effect of Thidiazuron on the Formation of Micro-tubers and Plantlet Regeneration of Pinellia ternata T. (Thidiazuron이 반하(半夏)의 자구(子球) 및 식물체 재생에 미치는 영향)

  • This experiments were conducted to determine the effect of thidiazuron on forming tuberlets and plant regeneration of Pinellia ternata T. by tissue culture. The addition of $5\;{\mu}M$ TDZ to the medium had better regeneration than that of any other treatments of NAA and TDZ. At the combination treatments of NAA and TDZ, as the level of thidiazuron increased, the rate of shoot regeneration was incresed while the increment of NAA concentration inhibited the rate of shoot regeneration. The supplement of $5\;{\mu}M$ thidiazuron produced the best number of micro-tubers per explant and the number of micro-tuber formed was 25 in MS medium and 29 in MG medium on 30 day culture, respectively. Microtuber formation was the best on MG medium with 1.0 mg/l NAA and $5\;{\mu}M$ thidiazuron. MG medium was superior to MS and B5 medium for the growth of tuberlets. Half strength of MS medium with NAA 2 mg/l was the most effective for root formation. Rooting ability on nursery soil of plantlets produced in in vjtro was good as a 80% after 3 weeks.
