As seed production program is growing prosperously in various fishes in southern Korea, disease problems in larval and juvenile stages have emerged as a new research object. The following results were obtained from investigation about environmental factors related to mass mortalities of young Kinki red seabream, Pagrus major in process of artificial seedling production. Total length of red seabream larvae hatched was 2.93mm, and became 18.83~20.12mm at day 40. The first noticeable mortality of red seabream larvae (7.98~9.37mm) occurred in 25~30 day-old fish with the survival rates of 59.8~60.3%. Thereafter the mortality of larvae decreased, survival rate was 20.5~25.45% in day 40. After 20~30 days, the quality of pond water was bellow II class. During the experimental period COD, $PO_4$-P, $NO_2$-N, $NO_3$-N and $NH_$-N increased up to 3, 7, 34, 6 and 8 times, respectively, compared to initial ones. The number of viable bacteria in pond water and seabream larvae were $6.3{\times}10^6$~$2.3{\times}10^7$ cfu/ml, 4.3~$7.4{\times}10^6$ cfu/g in day 25, respectively. Among the isolated bacteria from the diseased red seabream in day 25, Vibrio spp. was considered to be the causative organism. External symptoms of this disease were floated, spined near the surface and inflated abdomen. When the isolated strain of the Vibrio was bathed to seabream larvae, $LD_50$ of seabream larvae was over $10^6$ cfu/ml of Vibrio spp.
During the storage of these two species in a large conservation tank, fertilized eggs were collected and the offspring were raised. During culturing of the offspring, individuals with mixed characteristics of these two species were observed, and 96 individuals were randomly tested using microsatellite markers applicable to both species. Among the 96 individuals, 15 individuals with mixed morphological characteristics were confirmed to be hybrids showing both of genotypes red seabream and blackhead seabream. Additionally, based on sequence analysis of mitochondrial cytochrome oxidase subunit 1 (mtDNA CO1), 81 showed 99% nucleotide sequence identity to that of black sea bream, and the remaining 15 individuals showed over 99% sequence identity to that of red seabream. So, hybrids were produced by female red seabream and male blackhead seabream. These results suggest that hybrids may form in nature between these two species if their habitats overlap due to the influence of humans or global climate change.
Olive flounder (Paralichthys olivaceus) is one of the most popular farmed fish in Korea. Genetic variability of the fish was investigated by means of microsatellite DNA markers. All of the 8 microsatellite loci were analyzed in this study. For the confirmation of genetic variation during a shift in generation, microsatellite variability was compared within the same hatchery strains but produced in different spawning years. When genetic variability of farmed flounders produced in 2006 and 2007 was compared with that of 2003, a marked reduction of genetic variability was observed in the 2006 and 2007 populations. Mean number of alleles per locus and expected mean heterozygosity decreased from 9.75 and 0.796 (in 2003 population) to 7.78 and 0.785 (in 2006 population), respectively. Moreover, we have observed the distortion of allele frequency. These results show that reduced genetic variability of farmed olive flounder in processed generation has lower numbers of alleles and genetic variability than these of wild fish. Our results suggest that to have a sustainable aquaculture of this species, there is need for scientific broodstock management based on genetic variation and more intensive breeding practices to improve genetic diversity and to avoid detrimental inbreeding effects.
The hatchability of the artificially induced hybrid between two groupers (family Serranidae), red spotted grouper (Epinephelus akaara) and brown-marbled grouper (E. fuscoguttatus) that lives in different habit environment was investigated. There was no difference in the required time of each developmental stages after fertilization between hybrid (red spotted grouper ♀ ${\times}$ brown-marbled grouper ♂) and purebred (red spotted grouper ♀${\times}$♂) and required 25.6 hours to hatch at incubated in $25^{\circ}C$, but a noticeable unequal cleavage in cell size was observed in hybrid eggs unlikely to purebred. The hatching rate of fertilized eggs of hybrid was generally low across the four incubate temperatures (22, 25, 28, $31^{\circ}C$) with highest 9.8% in $25^{\circ}C$. This study demonstrated the possibility of artificial hybridization between two groupers, red spotted grouper and brown-marbled grouper, thus preparing the groundwork on developmental characteristics, deformities of hatched larvae and early survival ability for further studies on aquaculture.
Nucleotide metabolism in endothelium is variable between different species. Recent studies demonstrated that this variability could contribute coagulation dysfunction, even though organs of the alpha 1,3-galactosyltransferase gene knockout pig were transplanted into the primate. CD73 (ecto-5'-nucelotidase) is an enzyme at cell surface catalyzing the hydrolysis of adenosine triphosphate to adenosine, which plays role on a substance for anti-inflammatory and anti-coagulant. Thus, overexpression of CD73 in endothelial cells of the pig is considered as an approach to reduce coagulopathy. In this study, we constructed a human CD73 expression vector under control of porcine Icam2 promoter (pIcam2-hCD73), which is expressed specifically at endothelial cells, and of CMV promoter as a control (CMV-CD73). First, we transfected the CMV-CD73 vector into HEK293 cells, and then confirmed CD73 expression at cell surface by flow cytometry analysis. Next, we transfected the pIcma2-CD73 and CMV-CD73 vectors into primary porcine fibroblasts and endothelial cells. Consequence was that the pIcma2-CD73 vector was expressed only at the porcine endothelial cells, meaning that the pIcam2 promoter lead to endothelial cell-specific expression of CD73 in vitro. Finally, we nucleofected the pIcam2-hCD73 vector into passage 3 fibroblasts, and enforced hygromycin selection of 400mg/ml. We were able to obtain forty three colonies harboring pIcam2-CD73 to provide donor cells for transgenic cloned porcine production.
Glucocorticoid is activating mammary gland cells for lactating animals, resulting in increasing abilities of the milk synthesis. Expression of the prolactin receptor(PRL-R) in mammary gland cells was closely associated with milk production. To increase lactation ability for the Korean Native Goats at mid-lactation period. 0.05, 0.1. and 0.2 g of hydrocortisone was administrated with 5 $m\ell$ of saline. and injected into vein. For the control, 5 $m\ell$ of saline was administrated in to vein. After 24 H, the mammary gland tissue was collected, and mRNA expression rates were investigated for the alpha-casein and PRL-R using competitive PCR(polymerase chain reaction). There was no significant differences between treatment and control groups for the mRNA expression rate of PRL-R in mammary gland cells after 24 h of administration of hydrocortisone. The rate of mRNA expression for the alpha-casein was increased 37%, 630%, and 380% at 0.05, 0.1, and 0.2 g of hydrocortisone administration groups, respectively, comparing with control group. The results suggested that PR L-R mRNA expression of mammary gland cell by administration of hydrocortison was not significant, but increase of the alpha-casein mRNA expression my be differences of expression of functional proteins in the cell and expression patterns of protein secretion time to out of the cell. This study showed increase of alpha-casein mRNA expression by administration of hydrocortisone at mid-lactation period of Korean native goat.

Culture method of somatic cells is one of the important factors in the production of transgenic pigs by somatic cell nuclear transfer. In this study, we established an efficient culture method of porcine fetal fibroblasts. Porcine fetal fibroblasts were isolated from 33-day-old fetuses. The proliferation of porcine fetal fibroblasts was analyzed by different serum types and culture media. The cultures in medium supplied 15% ES screened FBS showed faster increase in cell number than 15% FBS. Also, fetal fibroblasts have been propagated continuously for $7{\sim}8$ passages in ES modified DMEM and DMEM medium. We transfected $PGK-neo^r$ vector (pKJ2) into porcine fetal fibroblasts to estimate colony formation in this culture condition. The formation of colonies was confirmed in the medium containing $300\;{\mu}g/ml$ G418 at 12 day. These data show that this culture system can be used screening of porcine somatic cells transfected transgene.
The objective of this study was to investigate the comparison of transferable embryos and pregnancy rate between Hanwoo and Chickso. The results obtained were as follows: No differences were observed in the efficiency of superovulation rates on Hanwoo 78%, and Chickso 85%, respectively. The mean number of total embryos are each $14.76{\pm}2.16$ and $6.23{\pm}1.07$. So the mean number of transferable embryos are each $10.94{\pm}1.91$ and $4.58{\pm}1.05$. In addition, the mean number of total Hanwoo embryo from <10 and $10{\leq}$ of corpora luteum was $0.50{\pm}0.50$, $11.56{\pm}1.92$, respectively. In case of Chickso, The mean number of transferable embryo from <10 and $10{\leq}$ of CL was $2.75{\pm}1.39$, $6.00{\pm}1.00$, respectively. The pregnancy rates were Hanwoo 40%, and Chickso 37% following transfer of fresh embryos produced in vivo. Also, the pregnancy rates of Chickso 60% were significantly greater (p<0.05) than the Hanwoo 42.48% following transfer of following transfer of frozen embryos, respectively. In conclusion, these results suggest that Chickso may be effectively used for transferable embryos production in Hanwoo. Although the transferable embryos number was not enough, it seems the Chickso greatly affect pregnancy rate. The results indicated that the possibility of transferable embryos from Chickso for embryo transfer could be confirmed in this study. Based on the present findings, it was suggested that it is very important to evaluate in vivo embryo production and pregnancy rate of embryo transfer following superovulation for effective Hanwoo and Chickso production.
This study was conducted to examine the efficiency of enucleation and blastomere isolation from recipient oocytes and donor embryos, respectively and to determine the effect of oocyte age and electric voltage on the fusion rate and in vitro development of the fused oocytes in rabbit nuclear transplantation. Immature oocytes collected from ovarian follicles were matured in vivo for 12 h in TCM-199 containing FCS and hormones and in vivo matured oocytes were collected 17 to 18 h post-HCG. The fresh and frozen donor embryos of 8- to 16-cell stage were collected from the oviduct of superovulated does. The proportion of successfully enucleated oocytes was greatly lower in in vitro matured oocytes (42.3%) than that (62.7%) in in vivo matured oocytes The level of cytochalasin B for in vivo matured oocytes did not affect the efficiency of enuleation, but 7.5 $\mu$g /mL cytochalasin B for in vitro matured oocytes showed a high enucleation rate significantly. The isolation efficiency of a single blastomere nucleus did not differ between 8- and 16-cell stage embryos. The percentage of single blastomeres isolated from 16-cell stage fresh embryos after 0.5% pronase treatment was greatly higher at 16-min treatment (94.4%) than at 8-min(78. 1%) and the blastomeres(61.5%) isolated from frozen-thawed embryos after 16-min pronase were significantly fewer than those of fresh embryos. The age of recipient oocytes affected nuclear fusion rate. The reconstituted oocytes fused at 24-h age showed slightly higher fusion rate (77.8%) than those (65.0%)fused at 18-h age. The fusion rate of in vitro and in vivo matured oocytes inserted with fresh blastomere did not differ among electric voltages, but the cleavage rate and development to morula-blastocysts of in vitro matured oocytes was more higher under 0.6 kV/cm than under 0.8 to 1.2 kV/cm, while the cleavage rate and development of in vivo matured oocytes was higher under 0.8 to 1.0 kV/cm than under 1.2 kV/cm. The fusion and cleavage rate fol1owing insertion with frozen-thawed blastomere was not different between the in vitro and in vivo matured oocytes and was similar to those from fresh blastomere insertion.

Characterization of quantitative trait loci (QTL) was investigated in the experimental cross population between Berkshire and Yorkshire breed. A total of 512 F$_2$ offspring from 65 matting of F$_1$ parents were phenotyped the carcass traits included average daily gain (ADG), average backfat thickness (ABF), tenth rip backfat thickness (TRF), loin eye area (LEA), and last rip backfat thickness (LRF). All animals were genotyped for 125 markers across the genome. Marker linkage maps were derived and used in QTL analysis based on line cross least squares regression interval mapping. A decision tree to identify QTL with imprinting effects was developed based on tests against the Mendelian mode of QTL expression. To set the evidence of QTL presence, empirical significance thresholds were derived at chromosome-wise and genome-wise levels using specialized permutation strategies. Significance thresholds derived by the permutation test were validated in the data set based on simulation of a pedigree and data structure similar to the Berkshire-Yorkshire population. Genome scan revealed significant evidences for 13 imprinted QTLs affecting growth and body compositions of which nine were identified to be QTL with paternally expressed inheritance mode. Four of QTLs in the loin eye area (LEA), and tenth rip backfat thickness (TRF), a maternally expressed QTL were found on chromosome 10 and 12. These results support the useful statistical models to analyse the imprinting far the QTLs related carcass trait.
