Antimicrobial resistance and resistance transfer of Vibrio parahaemolyticus and Morganella morganii from commercial fisheries products (시판 수산물에서 분리한 Vibrio parahaemolyticus와 Morganella morganii의 항균제 내성과 내성 전이)

  • The purpose of this study was to investigate the antimicrobial resistance and resistance transfer of Vibrio parahaemolyticus and Morganella morganii isolated from fish products purchased from fish markets in Yeosu April - December 2017. These bacteria were identified by biochemical test and PCR results, and the transfer of antimicrobial resistance was confirmed by the broth mating method. To isolate the transconjugants formed during conjugation, TSA medium containing 50 ㎍/ml of ampicillin (AMP), and 150 ㎍/ml of streptomycin (SM) or 30 ㎍/ml of oxytetracycline (OT) was used. M. morganii isolates showed low susceptibility to AMP, amoxicillin (AML), and colistin (CT), erythromycin, OT, and tetracycline, compared to V. parahaemolyticus resistance to AMP, AML, and CT. The conjugation of V. parahaemolyticus or M. morganii with Escherichia coli resulted in the separation of V. parahaemolyticus and M. morganii showing SM resistance as transconjugants. Meanwhile, Edwardsiella tarda transconjugants showing AMP and AML resistance were obtained from the broth mating of V. parahaemolyticus and E. tarda. But the transfer of the VPA0477 which is a β-lactamase gene of V. parahaemolyticus was not confirmed. These results suggest that resistance transfer between pathogenic bacteria is bidirectional and progresses in a wide variety of patterns.

Effect of Squid and Octopus on the Quality Characteristics of Kimchi during Fermentation (문어와 오징어의 첨가량에 따른 배추김치의 숙성 중 품질특성에 미치는 영향)

  • This study was carried out to investigate the chemical and microbiological properties of Kimchi containing different amounts of squid and octopus during fermentation at $4^{\circ}C$ for 56 days. Kimchi were assigned to one the of the following 7 treatment groups: CK (control Kimchi; no addition), SK-5 (added 5% of squid), SK-10 (added 10% of squid), SK-20 (added 20% of squid), OK-5 (added 5% of octopus), OK-10 (added 10% of octopus), and OK-20 (added 20% of octopus). The quality characteristics of CK and other treatments were determined by measuring the pH, titratable acidity, reducing sugar, volatile basic nitrogen (VBN), and microbial analysis. During fermentation, Kimchi containing squid and octopus had higher pH and lower acidity values than the CK group (P<0.05). The titratable acidity was increased in all treatment groups, whereas the reducing sugar content decreased gradually during fermentation. The reducing sugar contents were highest in the CK group. The VBN value increased in all treatment groups with increasing storage period, and the CK group had lower VBN value than the other treatment groups containing squid and octopus. The numbers of total bacteria and lactic acid bacteria rapidly increased up to the second week, and this slowly increased thereafter.

Development of Analytical Method for Colistin in Fish and Shrimp using Liquid Chromatography Mass Spectrometry (LC-MS/MS를 이용한 수산물 중 콜리스틴 분석법 개발)

  • Colistin is a last resort antimicrobial agent against multi-drug resistant Gram-negative bacteria. This study was conducted to develop an analytical method to determine colistin in fish and shrimp. The analytes were confirmed and quantified via liquid chromatography-tandem mass spectrometry (LC-MS/MS) in the positive ion mode using multiple reaction monitoring (MRM). The sample was extracted with acidified 5% methanol (containing 0.5% formic acid). Then, solid phase extraction (SPE) was used for cleanup. Matrix-matched calibration curves were linear over the calibration ranges (0.05-1.2 mg/kg) for all the analytes into blank sample with $r^2$ > 0.99. All the values fulfilled the criteria requested by the Codex guidelines. Average recoveries ranged from 85.9% to 107.9%. The repeatability of measurements, expressed as the coefficient of variation (CV, %), was less than 15%. The limit of detection (LOD) was 0.02 mg/kg, and the limit of quantitation (LOQ) was 0.05 mg/kg. This improved method showed higher accuracy and acceptable sensitivity to meet the CAC guideline requirements and is applicable for the analysis of residual colistin (A+B) in fish and shrimp.

Emergy-Based Value of the Contributions of the Youngsan River Estuary Ecosystem to the Korean Economy (에머지 방법론을 이용한 영산강 하구생태계의 기여 가치 평가)

  • An emergy concept was used to evaluate the contributions of the Youngsan River Estuary to the Korean economy from systems and ecosystem service perspectives. Total emergy input to the estuary was $9.42{\times}10^{20}$ sej/yr with the chemical potential of river inflow accounting for 73% of the total and tidal energy for the remaining 27%, reflecting the unique environmental characteristics of estuaries where rivers meet sea. From the systems perspective, the estuary ecosystem contributed 179.9 billion em\/yr (14.91 million em\/ha/yr or 13,526 em$/ha/yr). Four ecosystem services of the Youngsan River Estuary were selected for the emergy evaluation: fishery production, pollutant removal, aesthetic function, and scientific research information. Aesthetic function contributed the most to the Korean economy with 179.9 billion em\/yr, followed by fishery production (101.1 billion em\/yr), pollutant removal (32.6 billion em\/yr), and scientific research information (934 million em\/yr). Emergy-based values of the selected ecosystem services of the estuary were higher than those calculated by economic methodologies.

Changes in Cholesterol Contents of Some Marine Products during Processing (수산물의 콜레스테롤함량과 가공중의 변화)

  • This experiment was carried out to investigate the changes of cholesterol contents during the processing of various marine products. For the above purpose, squid, eel, mackerel, baby clam and sea mustard were sun-dried, roasted, salted-dried, boiled-dried and salted, respectively. In raw samples, the highest content of cholesterol was 761 mg/100g in squid, following by eel, baby clam, mackerel and sea mustard in order, and there was no significant relationship between cholesterol and lipid contents. The cholesterol contents of the used samples were decreased during the processing except baby clam. The decreasing ratios of the cholesterol contents during the processing were $66.4\%$ in squid, $43.6\%$ in eel, $41\%$ in mackerel and $58\%$ in sea mustard. While the cholesterol content of baby clam was increased by 1.6 times compared with that of raw sample. Considered from the results, it seems that the changes of cholesterol content during processing depended upon the changes of lipid content in the sample.

A Causality Analysis of the Tangerine Market by Distribution Channel (감귤시장의 유통단계별 가격 인과성 분석)

  • The purpose of this study is to investigate price transmissions between wholesale and retail markets regarding Jeju tangerines by employing co-integration analysis and vector error correction model. The results of this study are summarized as follows: First, the long-run equilibrium relationship was found among wholesale and retail markets in time series for level by distribution channel. Second, a short-run causality relationship was observed between wholesale and retail markets. Third, the long-run causality relationship between wholesale market and retail markets was found bidirectional and feedback effect. These results imply that the wholesale price performs a central role in establishing price in the tangerine market, and the wholesale market influences tangerine price. In conclusion, for the development of a competitive tangerine industry, it is necessary to aggressively promote the policy of supply and demand control of tangerine production through organizing producers.

Contents of Arsenic in Some Fisheries Caught in Western Coast (서해안 지역 수산물의 비소함량에 관한 연구)

  • In Korea, a small proportion of fishery products are contaminated with a considerable amount of potentially hazardous contaminants. So, there is generally a high risks to consumers. Inorganic contaminants with the greatest potential for toxicity are Sb, As, Cd, Pb, Hg, Se and the Sulfides. As appears to be much more variable than the others. This study was performed to assess the levels of the As in fisheries caught from the waters of the western coast of Korea. The samples included 26 kinds of fishes, 18 kinds of Mollusca, 3 kinds of Crustacea and 7 kinds of salted fishes. The Moisture content of molluscans was significantly higher than that of others (p<0.05). And the As contents of crustaceans were significantly higher than those of fishes, molluscans and salted fishes (p<0.05). The As content of Turban shell was 51.62 ppm, which was the highest among the samples. Also, the As contents of eel, cuttle fish and blue crab were found to be higher than in other fisheries. This variability offers a solution for the reduction of exposure to As through restricting the harvest of aquatic organism from the western coast of Korea.

Evaluation of Thermal Processes for Canned Marine Products (1) Canned Boiled Sea-mussel in Brine and Canned Smoked Sea-mussel in Oil (수산물통조림의 살균조건에 관한 연구(1) 홍합 보일드 통조림 및 홍합 훈제 기름담금 통조림)

  • The present studies were conducted to evaluate the sterilizing efficiency of the thermal processes for the canned sea-mussel products, such as canned boiled sea-mussel in brine (packed into No. 1 flat can) and canned smoked sea-mussel in oil (packed into No. 3B square can), with the purpose of deciding the adequacy of the processes. Heat penetration was tested three times with three cans at a time for each canned product. The tip of the applicator was fixed on the position a little below the geometrical center of the can. The test cans were placed in the middle layer of the basket in which the same canned products were loaded with, and the test cans were arranged to the front, the middle and the rear in the retort. The heat penetration curve of the canned boiled sea-mussel in brine showed a broken logarithmic heating curve while that of the canned smoked sea-mussel in oil showed a simple logarithmic heating curve. The calculated $F_0$ value for the canned boiled sea-mussel in brine was 25.33 and the canned smoked sea-mussel in oil was 13.84. Additionally, the nomographs represents the relationship between $F_0$ values and B values(process time including $42\%$ of come-up time) for the canned sea-mussel products were constructed.

Application of a Peptide Nucleic Acid-Based Asymmetric Real-Time PCR Method for Rapid Detection of Vibrio cholerae (비브리오 콜레라 신속 검출을 위한 펩티드 핵산 기반 비대칭 real-time PCR 방법의 적용)

  • Vibrio cholerae is a very important pathogenic bacterium that has to be monitored in seafood and ships' ballast water. Various methods have been developed to identify this bacterium, yet these methods are time-consuming and have limitations for their sensitivity to detect contamination. The purpose of the present study was to develop a robust and reliable method for identifying V. cholerae. Peptide nucleic acid (PNA) probes were developed to use for PNA-based asymmetrical real-time PCR techniques. The toxigenic Cholera enterotoxin subunit B (ctxB) gene was selected as a target for detecting V. cholerae and the gene was synthesized as a positive template for conventional and real-time PCR. Real-time PCR primers and PNA probes were designed and standard curves were produced for the quantitative analysis. The selected PNA probes reacted specifically to V. cholerae without any ambiguity, even among closely related species, and the detection limit was 0.1 cfu/100 mL. Taken together, the PNA probes and asymmetrical qPCR methods developed in this present study could contribute to the rapid, accurate monitoring of V. cholerae in marine environments, and as well as in seafood and ships' ballast waters.

Distribution and exposure assessment of indicator PCBs in Food (식품 중 indicator PCBs의 분포와 노출평가)

  • Seven indicator PCBs (IUPAC nos 28, 52, 101, 118, 138, 153, 180 congeners) concentrations were measured in food samples including cereal (polished rice), meats, eggs, milk and dairy products, and fisheries and products as representation for the general Korean populations during 2006-2007, and was analyzed using isotopic dilution method. Fishes had the highest average level as 39.8 ng/g, 1.4 ng/g for milk and dairy products, and 0.9 ng/g for meats. The hairtail out of fishes was contaminated at the level of 15.4 ng/g, 5.4 ng/g for pacific mackerel and spanish mackerel, and 4.5 ng/g for yellow croaker. The ratio for indicator PCBs in overall food was contributed as follows; 35.8% for PCB-153, 16.2% for PCB-138, 16.1% for PCB-101, 13.4% for PCB-118, 8.8% for PCB-180, 6.9% for PCB-52, and 2.9% for PCB-28. The hexa-CBs including PCB-153 and 138 were more predominated, and the next was penta-CBs including PCB-101 and 118 in food. For estimated daily intake (EDI) in average and $95^{th}$ percentile, fishes and products out of overall food were taken to represent over 50%. However, it was estimated that there was no adverse health effect for Korean.
