Preparation and Characterization of Silk Fibroin/Gelatin Hybrid Scaffolds (실크 피브로인/젤라틴 하이브리드 지지체의 제조 및 특성분석)

  • Silk fibroin is a biocompatible and slowly biodegradable natural polymer. This natural polymer has excellent mechanical properties, non-toxicity, and non-immunogenic properties and has been demonstrated to support tissue regeneration. Also, gelatin is a natural material derived from collagen by hydrolysis and has an almost identical composition as that of collagen. Silk fibroin/gelatin scaffolds have been fabricated by using the freeze-drying method. To establish the scaffold manufacturing condition for silk fibroin and gelatin, we made scaffolds with various compositions of gelatin, glutaldehyde and silk fibroin. The silk fibroin/gelatin scaffolds were characterized using SEM, DSC, and water absorption ability tests. The cellular proliferation was evaluated by WST assay. These results suggested that a scaffold containing 8% of gelatin, 1% of glutaldehyde and 0.3 g of silk fibroin provided suitable characterstics for cell adhesion and proliferation. In conclusion, the silk fibroin/gelatin scaffold may serve as a potential cell delivery vehicle and a structural basis for tissue engineering.

Effects of the Actindia chinensis on Loperamide-induced Constipation in Rat (제주산 참다래가 Loperamide로 유도된 변비에 미치는 영향)

  • Loperamide-induced constipation reduced gastric emptying, small-intestinal and colonic motility, and these effects were prevented by Actindia chinensis(Gold Kiwi Fruit, GKF). In this study, the effects of Actindia chinensis on constipated male Sprague-Dawley rats induced by loperamide(2 mg/kg, s.c.,5 days) were investigated. Rats were randomly assigned to the normal control rats(regular diet), constipated rats(regular diet plus loperamide), constipated rats treated with 2.5% GKF(regular diet supplemented with 2.5% GKF plus loperamide), constipated rats treated with 5% GKF (regular diet supplemented with 5% GKF plus loperamide). There was less fecal excretion and lower fecal water content in loperamide-treated rats than in control rats. Oral administration of GKF blocked the decrease of fecal excretion and fecal water content in the loperamide-treated rats. Mucus production of crypt cell and mucus contents at fecal and mucosa surface were reduced by loperamide-treated rat. But colonic crypt cell contained increased mucin in the GKF treated group and mucus layer stained with alcian blue was significantly thicker in GKF treated rats compared with in loperamide-treated rats. In isolated rat ileum, loperamide produced inhibition of ileal motility. Pretreatment with methanolic extracts of GKF in isolated rat ileum prevented inhibition by loperamide. These findings indicated that the GKF was effective for alleviation of inhibition of colonic peristalsis by loperamide and that GKF might be of value in the prevention of constipation.

효과적인 일함수 조절을 위한 그래핀-고분자의 적층 구조

  • 그래핀은 뛰어난 기계적, 화학적, 광학적, 전기적 특성을 가지고 있는 2차원 물질로, 대면적 합성법과 전사 공정을 통해 다양한 기판에서의 사용이 가능해지면서 차세대 전자 소자로 활용하기위한 활발한 연구가 이루어지고 있다. 디스플레이, 태양전지의 전극과 전계 효과 트랜지스터의 채널로 적용한 연구에서 우수한 결과들을 보이고 있다. 특히, 금속/금속 산화물 전극은 염료 감응형 태양전지와 유기 발광 다이오드 구조에서 화학적으로 불안정할 뿐 아니라 일함수가 고정되어 쇼트키 접촉이 형성되면 저항을 낮추기 어렵지만, 그래핀은 금속/금속 산화물 전극보다 화학적으로 안정하고 일함수의 조절이 가능해 옴 접촉 형성에 용이하다. 그래핀의 일함수를 조절하는 연구는 크게 공유결합과 비공유 결합을 이용한 방법이 시도된다. 공유 결합을 이용한 방법은 합성과정에서 그래핀의 구조에 내재된 결함 혹은 새로운 결함을 형성하여 다른 원소를 첨가하는 방법이다. 이러한 방법은 그래핀의 결함 영역에서 작용하기 때문에 그래핀 전자 구조의 높은 수준 조절을 위해선 그래핀 구조의 파괴가 동반된다. 반면, 비공유 결합을 이용한 방법은 전하 이동 도핑 효과를 이용해 그래핀의 전자 구조를 제어하는 방법으로, 금속/금속산화물/기능기와 그래핀의 적층으로 복합 구조를 형성하는 방법이다. 금속/금속 산화물과의 복합구조는 안정적인 p-형 도핑이 보고되었지만, n-형 도핑은 대기중의 수분, 산소 그리고 기판과의 상호작용에 의해 대기중에서 불안정해 추가적인 피막공정이 요구된다. 기능기를 이용한 적층 구조는 그래핀과 기판사이의 상호작용 혹은 그래핀 전자 구조를 다양한 기능기를 이용해 제어하는 것으로, 이극성을 가진 자기정렬 단일층(self-assembled monolayers)이 대표적인 방법이다. 공간기(spacer)의 길이나 말단기(end group)의 종류로 p-형과 n-형의 도핑 수준을 제어할 수 있지만, 흡착기(chemisorbing groups)의 반응성이 기판의 화학적, 물리적 표면상태에 의존하기때문에 기판 선택이 제약되며 전처리 공정이 요구될 수 있는 한계가 있다. 본 연구에서는 다양한 기판에 적용가능한 용액 공정을 이용해 그래핀과 고분자를 적층하였고, 안정적이고 효과적으로 일함수를 낮추는 구조를 확인하였다.

Effects of Temperature and Relative Humidity on Water Soluble Vitamin Contents in Commercial Vitamin Tablet (저장 온도 및 상대습도가 비타민 정제 중 수용성 비타민 함량의 변화에 미치는 영향)

  • Effects of temperature and relative humidity on contents of water-soluble vitamins (vitamins $B_1,\;B_2,\;B_3,\;B_6$, and C) of two commercial tablets ("Multivitamin Dandelion" and "Chewable vitamin C") were investigated. When stored at various temperatures (25, 35, and $45^{\circ}C$) with cap, all measured vitamins were stable and degraded very slowly during 24 weeks of storage; low relative humidity (11% RH) without cap also provided stability during storage period. At higher relative humidities (75 and 100% RH), contents of all water-soluble vitamins, except vitamins $B_2\;and\;B_3$, decreased significantly at early storage period. These results showed that stability of water-soluble vitamins is highly dependent on relative humidity rather than storage temperature.

  • The mass balances of calcium salts during the manufacturing processes of Tofu were established by conductometric method and chemical analysis method. During the manufacturing processes of soy milk and Tofu, 66% of solid and 63% of calcium was transfered from soy-bean to the soy milk, and 47.8% of total solid from soybean was transfered to the Tofu, respectively. When the $CaCl_2$ was used as coagulant, calcium contents in Tofu $(Y_{Tofu},\;mg{\cdot}Ca/g{\cdot}Tofu,\;wet\;basis)$ and drained solution $(Y_{drained\;soln},\;mg{\cdot}Ca/ml{\cdot}drained\;soln.)$ were linearly increased with the amounts of $CaCl_2(C,g{\cdot}CaCl_2/ml{\cdot}soy\;milk)$ added in soy milk, and correlative equations between them were obtained as $Y_{Tofu}=0.3369\;C+1.2689$ for Tofu$(moisture\;content:\;81.5{\pm}0.5%)$ with r=0.9898, and $Y_{drained\;soln}= 0.2899C+0.0399$ for drained solution with r= 0.9991. It was proved that conductometric method was reasonably applicable to the measurement of calcium contents of the products from every processes of Tofu manufacture except soy-bean. However the conductometric method was not recomendable in the case of $CaSO_4$ as coagulant due to its low solubility ana uneven distribution in soy milk and Tofu tissue.

분리 대두 단백질과 옥수수 전분을 혼합하여 제조한 필름의 특성

  • 단백질, 탄수화물, 유지 등과 같은 생고분자를 소재로하여 제조하는 가식성 또는 생분해성 필름은 식품표면을 코팅하거나, 식품의 내부에 사용하여 식품을 외부의 충격으로부터 보호하고 식품의 저장수명을 증가시키는 동시에 수분, 기체 및 용질의 이동을 방지하는 기능을 한다. 이러한 가식성 필름의 소재 중 하나인 전분은 비교적 저가의 재료이고 기체투과를 방지하는 성질은 우수하지만, 탄력성이 적은 단점을 가지고 있다. 생고분자필름의 물리적 강도를 증진시키고 수증기의 투과성을 낮추는 방법으로 단백질과 탄수화물을 복합적으로 사용한다 본 실험에서는 분리 대두 단백질(SPI)과 옥수수 전분을 혼합하여 가식성의 생분해성 필름을 제조하고 이들의 이화학적 특성에 대해 연구하였다. SPI 필름은 SPI 5g에 증류수 100$m\ell$을 가해 glycerol 2g을 첨가해 제조하였으며, 옥수수 전분필름은 옥수수 전분 6g에 증류수 100$m\ell$을 가해 glycerol 1.2g을 첨가해 제조하였다. SPI와 옥수수 전분의 혼합필름은 SPI와 옥수수 전분의 양을 1:1 비율로 혼합하여 제조하였다. SPI 필름, 옥수수 전분 및 SPI와 옥수수 전분 용액의 혼합필름의 각각의 수증기 투과도는 7.49$\times$$10^{-11}$, 2.69$\times$$10^{-11}$, 4.24$\times$$10^{-11}$ g.m/m$^2$.s.Pa이었다. 본 실험으로 SPI의 단일재료로만 필름을 제조하였을 때보다 SPI 용액에 옥수수 전분용액을 혼합해 필름을 제조하면 탄력성을 유지하면서 수증기 투과도를 낮출 수 있다는 결과를 얻을 수 있었다.현미밥의 유리아미노산 분석 결과 필수아미노산을 포함한 다량의 아미노산이 존재하였으며 혈압강하 기능성 성분인 GABA(${\gamma}$ -amino butyric acid) 함량은 현미가 4.7 mg/100 g, 발아현미 20.8 mg/100 g이었으며, 동결건조 후 분석한 발아현미밥의 GABA 함량은 5.7 mg/100 g이었다. 발아현미밥의 관능검사 결과는 색, 윤기, 맛, 부착성, 응집성, 탄력성이 A가 유의적으로 높았으며, 외관의 경우도 A가 더 높은 평가를 받았다. 또한 전반적으로 A가 B에 비해 유의적인 차이를 나타내진 않았지만 더 나은 선호도를 나타내었다.좋게 평가되었다.*값은 63-68사이로, a*값은 0.13에서 -0.89사이를 b*값은 2-5값 사이에서 변화하여 제조한 죽의 색이 옅은 황색임을 알 수 있었다. 고형분 함량, 퍼짐성과 pH에 대한 $R^2$은 각각 0.4280, 0.5433과 0.2406임을 볼 때 버섯, 당근, 대파의 비율에 따라 제조한 쇠고기 야채 쌀죽의 고형분 함량, 퍼짐성과 pH는 설정된 범위내에서 그 유의성이 인정되지 않아 큰 영향을 미치지 않음을 알 수 있었다. 관능검사 결과, 색과 향에 대한 반응표면 회귀식의 $R^2$은 각각0.6000과 0.7825이고 P-value는 각각 0.4290과 0.0942로서 5% 수준에서 유의한 상관성이 없음을 확인할 수 있었다. 맛과 점성에 대한 $R^2$은 0.8717과 0.8068이고 P-value는 각각 0.0195 (p <0.05)와 0.0612로서 야채의 배합비에 따라 맛에 있어서 유의확률 5%수준에서 그 유의성이 인정되었으며, 전체적인 기호도에 대한 유의성은 $R^2$이 0.8463이고 P-value는 0.034

Quality Characteristics of Dumpling Shell Containing Pleurotus eryngii Powder (새송이버섯 분말을 첨가한 만두피의 품질 특성)

  • This study was conducted to investigate the quality characteristics of dumpling shells with added Pleurotus eryngii powder (PP) (2%, 4%, 6%). The weight after cooking and volume of the dumpling shells increased with the addition of PP. The water absorption of the dumpling shells was not significantly different by the addition of PP. Turbidity after cooking increased and hardness decreased by adding PP. The L value after cooking decreased with the addition of PP, and the b value was highest when the content of PP was 6%. Adhesiveness increased by adding PP and springiness decreased when PP content was above 4%. Chewiness and gumminess decreased by adding the powder. There were no differences among the control, 2%, and 4% samples in cohesiveness values, but cohesiveness significantly decreased with the addition of 6% of powder. Color characteristics showed low values when PP was added at more than 6% according to sensory characteristics tests. No significant differences were found in flavor, taste, smoothness, moisture, or adhesiveness of the dumpling shells after cooking according to additions of PP. Chewiness was higher by adding PP compared to the control, but no significant differences were found by additions of PP.

A Study for Polyol-in-Oil Type Lip Makeup Cosmetics with Natural Pigments (천연색소를 함유하는 유중폴리올(Polyol-in-Oil) 립메이크업 제품에 관한 연구)

  • Lip make-up products can be easily taken into body with food. For this reason, those products are requires to meet lots of qualifications compared with other cosmetic products. In addition, concerns about safety on synthesized tar pigments is constantly issued. Thus, demand of natural pigments is gradually increased and many kinds of natural pigments have been developed. However, there are some problems when natural pigments are applied to cometic products instead of synthetic ones. There is a reason that most of natural pigments consist of hydrophilic materials of sort of anthocyanin, but the existing lip make-up products is anhydrous oil dispersion type without water consisting oil and wax. Therefore, when watersoluble natural pigments are applied to anhydrous lip make-up products, color expression is lower and phase separation occurs due to the instability of the product. In addition, natural pigments have disadvantages that they can easily change by pH, heat and sunlight. There are troubles of stability because it is not easy to adjust for these factors in case of anhydrous forms. Aim of study is to develop lip make-up products which have not only safe to human but being high in expression of color by using natural pigments and securing stability of colorant as natural pigments are offered to polyol in oil emulsion. Then, lip make-up products which have heavy moisture while having not dryness that is created when the moisture evaporates are developed.

Preparation and Characteristics of High Performance Cathode for Anode-Supported Solid Oxide Fuel Cell (연료극 지지체식 고체산화물 연료전지용 고성능 공기극 제조 및 특성 연구)

  • Anode-supported solid oxide fuel cell (SOFC) was investigated to increase the cell power density at intermediate temperature through control of the cathode structure. The anode-supported SOFC cell were fabricated by wet process, in which the electrolyte of $8mol\%\;Y_2O_3-stabilized\;ZrO_2 (YSZ)$ was coated on the surface of anode support of Ni/YSA and then the cathode was coated. The cathode has two- or three- layered structure composed of $(La_{0.85}Sr_{0.15})_{0.9}MnO_{3-x}(LSM),\;LSM/YS$ composite (LY), and $La_{0.6}Sr_{0.4}Co_{0.2}Fe_{0.8}O_3{LSCF)$ with different thickness. Their single cells with different cathode structures were characterized by measuring the cell performance and ac impedance in the temperature range of 600 to $800^{\circ}C$ in humidified hydrogen with $3\%$ water and air. The cell with $LY\;9{\mu}m/LSM\;9{\mu}m/LSCF\;17{\mu}m$ showed best performance of $590mW/cm^2$, which was attributed to low polarization resistance due to LY and to low interfacial resistance due to LSCF.

Effect of Calcium Chloride Concentration on Roadside Ground Cover Plant Growth (염화칼슘 처리농도가 가로변 지피식물의 생장에 미치는 영향)

  • The purpose of this study was to provide information on management and apply it to a roadside ground cover plant understanding the capacity of calcium chloride in the plant. The experimental group was composed of the ratio control group of calcium chloride, 0.5%, 1.0%, and 3.0% in 500g of soil. Plant materials were selected and measured according to their ecological characteristics such as ground cover plant, Pachysandra terminalis, Hosta plantaginea, Trachelospermum asiaticum, Vitex rotundifolia, Euonymus japonica and Callicarpa japonica. The acidity of the amended soil was increased gradually depending on the treatment and conductivity was continually decreased. The EX-Ca increased after the treatment, but decreased in the middle of the experiment. Pachysandra terminalis, Trachelospermum asiaticum and Euonymus japonica were able to grow and survive at the ratio of 0.5%. Hosta plantaginea and Vitex rotundifolia were able to survive at the ratio of 1.0%. Hosta plantaginea, the possible state can absorb salts due to moisture and, can be applied to ground cover plants in the roadside. The growth and development of Callicarpa japonica was poor and the leaves were open to grow for calcium chloride treatment except the control group. It was concluded that Callicarpa japonica was very sensitive to calcium chloride.
