The Effects of Ethyl Pyruvate on Lipopolysaccharide-induced Acute Lung Injury (리포다당질에 의한 급성폐손상에서 Ethyl Pyruvate의 효과)

  • Background: Ethyl pyruvate (EP) is a derivative of pyruvate that has recently been identified by both various in vitro and in vivo studies to have antioxidant and anti-inflammatory effects. The aim of this study was to determine the effect of EP on lipopolysaccharide (LPS)-induced acute lung injury (ALI). Methods: 5 weeks old, male BALB/c mice were used. ALI was induced by an intratracheal instillation of LPS 0.5mg/Kg/$50{\mu}L$ of saline. The mice were divided into the control, LPS, EP+LPS, and LPS+EP groups. In the control group, balanced salt solution was injected intraperitoneally 30 minutes before or 9 hours after the intratracheal instillation of saline. In the LPS group, a balanced salt solution was also injected intraperitoneally 30 minutes before or 9 hours after instillation the LPS. In the EP+LPS group, 40mg/Kg of EP was injected 30 minutes before LPS instillation. In the LPS+EP group, 40mg/Kg of EP was injected 9 hours after LPS instillation. The TNF-$\alpha$ and IL-6 concentrations in the bronchoalveolar lavage fluid (BALF), and that of NF-$\kappa$B in the lung tissue were measured in the control, LPS and EP+LPS groups at 6 hours after instillation of saline or LPS, and the ALI score and myeloperoxidase (MPO) activity were measured in all four groups 24 and 48 hours after LPS instillation, respectively. Results: The TNF-$\alpha$ and IL-6 concentrations were significantly lower in the EP+LPS group than in the LPS group (p<0.05). The changes in the concentration of these inflammatory cytokines were strongly correlated with that of NF-$\kappa$B (p<0.01). The ALI scores were significantly lower in the EP+LPS and LPS+EP groups compared with the LPS group (p<0.05). In the EP+LPS group, the MPO activity was significantly lower than the LPS group (p=0.019). Conclusion: EP, either administered before or after LPS instillation, has protective effects against the pathogenesis of LPS-induced ALI. EP has potential theurapeutic effects on LPS-induced ALI.

Analysis of Fatigue Truck Model for LRFD Code (LRFD 설계기준(안) 피로설계트럭 모형 분석)

  • A reliability analysis of the fatigue failure of highway steel bridges was performed by applying the Miner's fatigue damage rule for the fatigue design truck proposed for the LRFD code and for the current DB 24 truck. The limit state function for fatigue failure is expressed as a function of various random variables that affect fatigue damage. Among these variables, the statistical parameters for the equivalent moment, the impact factor, and the loadometer were obtained by analyzing recently measured domestic traffic data, and the parameters for the fatigue strength, the girder distribution factor, and the headway factor were obtained from the measured data reported in literature. Based on the reliability analysis, the fatigue truck model for the LRFD code was proposed. After applying the proposed fatigue truck to the LRFD code, 16 composite plate and box girder bridges were designed based on the LRFD method, and the LRFD design results for the fatigue limit state were compared with those by the current KHBDC.

  • Nuclear fuel requires high reliability and safety and therefore contains debris filtering devices to prevent failure-inducing debris from entering it. The debris filtering performance of nuclear fuel is one of the most important factors for fuel integrity. Therefore, the performance must be evaluated and the measurement must be reasonable. In this study, a calculation method of the comprehensive filtering efficiency using the weighted mean was proposed to establish a standard filtering efficiency index. To confirm the suitability of the proposed method, representative debris specimens were selected and the filtering efficiency with the weighted mean was compared with the efficiency of the arithmetic mean. The weighting factor of the weighted mean was introduced to produce a fair evaluation. In addition, the analysis of the debris filtering mechanism was performed according to the size of debris specimens, and the main dimensions of the filtering feature for commercial nuclear fuel.

Radioprotective Effect of Quercetin Post-Treatment against γ-Irradiation-Induced Hepatocellular and Hematopoiectic System Damage in Mice (감마선 조사로 유도된 간세포와 조혈계 손상 마우스에서 퀘르세틴 투여 후의 방사선방호 효과)

  • This study was designed to evaluate the therapeutic effect of quercetin against radiation-induced hepatocellular and hematopoiectic damage in BALB/c mice. Mice were exposed to 6 Gy of ${\gamma}$-radiation and orally administered quercetin (25, 50 mg/kg b.w.) for 7 consecutive days. ${\gamma}$-Irradiation caused marked elevation of serum aspartate aminotransferase and alanine aminotransferase, levels as well as reduction of spleen index, thymus index, and the number of white blood cells. In addition, ${\gamma}$-irradiation induced significant elevation of lipid peroxidation as well as reduction of antioxidant enzyme activities, including superoxide dismutase, catalase, and glutathione peroxidase. However, post-treatment with quercetin resulted in a significant recovery of all of these parameters. These results suggest that quercetin acts as a potent radioprotector against irradiation-induced cellular damage in mice.

Retrofit Measures Based on Seismic Retrofit Priority of Existing Bridges (교량의 내진보강 우선순위를 이용한 합리적인 보강방안 선정기법)

  • The retrofit priority of existing and retrofitted bridges is examined and compared to determine effectively the seismic retrofit method of bridges. For the retrofit prioritization of bridges a quantitative procedure is proposed firstly based on seismic damage probabilities and total failure cost due to the damage of seismic vulnerable components. Using the proposed procedure, the retrofit priority of four typical girder-type bridges is determined. In addition, the ranking indices of bridges retrofitted by steel jackets and cable restrainers are revaluated for comparing with the results of existing bridges. Application of retrofitting method can considerably decreases damage possibilities of retrofitted components but may increases those of adjacent vulnerable components. Therefore, the seismic retrofitting effects based on the global motions of existing and retrofitted bridges should be examined to determine efficiently the retrofitting method. For evaluating the retrofitting effects the ranking indices obtained from the proposed procedure is found to be utilized effectively.

Effects of Different Milling Methodes on Physico-chemical Properties & Products (제분방법이 쌀가루 및 제품의 특성에 미치는 영향)

  • Rice flour obtained by four different milling methods, Pin mill. Colloid mill, Micro mill, and Jet mill, were used to investigate physico-chemical properties and product. Crude fat, ash, and protein contents of rice flours between different milling methods were similar. Blue value, amylose content, and damaged starch which related to properties of rice flour were reduced in the order that of Jet mill, Micro mill, Colloid mill, and Pin mill. Water absorption index, water solubility index, and water retention capacity increased as damaged starch increased. Hardness of gel(15%) is the highest value for Pin mill. The finer granules (Jet mill) had lower gelatinization onset(To) and peak(Tp) than any other rice flours. Those result are simillar with amylogram properties. Enthalpy of gelatinization increased as damaged starch increased. Jet mill had the highest score (p<0.05) of overall test in sensory evaluation and good paste properties.

  • Diffusion tensor images were obtained to investigate the damage of brain white matter in non-smokers and an adequate drinking group (less than 10 points) selected by scores of the Korean versions of alcohol use disorders identification test questionnaire. Factional anisotropy (FA) values, according to the variables of age and body mass index (BMI), were not statistically significant in all regions of the brain white matter after measurement of factional anisotropy (FA) values by the tract-based spatial statics (TBSS) method. In other words, age and body mass index (BMI) do not significantly affect the microstructural changes of the brain white matter.

노스리지 지진하의 강 뼈대 구조물에 대한 사례 연구

  • 노스리지 지진하의 모멘트 저항 강 뼈대 구조물에 대한 사례연구결과를 요약하면 다음과 같다. 1) 탄성과 비탄성 동적해석 방법의 신뢰도가 구조물의 동적거동의 모형화에 크게 영향을 받는다는 것을 알 수 있었고 구조물의 동적거동의 모형화를 위한 보다 개선된 지침과 기준의 필요성이 부각되었다. 2) 비탄성 횡방향 정적해석은 6층높이의 건물까지는 비탄성동적해석과 유사한 결과를 보였으나 10층 이상의 건물에서는 고차 모드 효과를 고려하지 못하여 상당히 상이한 결과를 나타냈다. 3) 응답스펙트럼 해석은 노스리지 지진하에서 탄성 시간이력 해석에 비하여 100%까지 상이한 결과를 보였다. 특정지진에 대한 구조물거동의 상세조사시 응답 스펙트럼 해석 대신 시간이력해석을 수행하는 것이 바람직하다. 4) 탄성 부재력의 저항능력 비와 소성 회전각 등의 거동지수 등은 현존하는 건물의 연결부 손상을 검사하기 위한 지침을 마련하는데 도움을 줄 수 있지만 특정 연결부를 검사에서 배제시키는 유일한 근거로 사용되어서는 안될 것으로 판단된다.

  • 계수적 구간 불확실 플랜트를 안정화하는 강인 제어기의 설계의 방법으로서 고정차수 극 배치(FOPA : Fixed Order Pole Assighnment) 알고리즘을 이용할 수 있다. FOPA 알고리즘에 의해서 강인제어기 계수의 집합은 선형제약조건으로 표현되고, 이 조건을 만족하는 임의의 한점은 주어진 불확실 시스템을 안정화한다. 본 논문에서는 선형제약조건으로 주어진 제어기 계수의 집합에서 외란의 에너지를 최소화하는 제어기를 구하는 알고리즘을 제안한다. 일반적으로 전역 최적해을 구하는 문제는 BMI(bilinear matrix inequalities)로 표현되지만 제어기의 계수를 고정했을 때는 LMI(lineal matrix inequalities)로 간략화되기 때문에 제어기계수에 대한 최소화와 성능지수에 대한 최소화를 반복함으로써 국부 최적해를 구할 수 있다. 제안한 알고리즘의 효용성을 보이기 위해 제어기 설계 예를 보이고, 그 성능을 비교 분석한다.

  • Objects: $^{99m}-lactosylated$ human serum albumin (LSA) is a newly synthesized radiopharmaceutical that binds to asialoglycoprotein receptors, which are specifically presented on the hepatocyte membrane. Hepatic uptake and blood clearance of LSA were evaluated in rat with acute hepatic injury induced by dimethylnitrosamine (DMN) and results were compared with corresponding findings of liver enzyme profile and these of histologic changes. Materials and Methods: DMN (27 mg/kg) was injected intraperitoneally in Sprague-Dawley rat to induce acute hepatic injury. At 3(DMN-3), 8(DMN-8), and 21 (DMN-21) days after injection of DMN, LSA injected intravenously, and dynamic images of the liver and heart were recorded for 30 minutes. Time-activity curves of the heart and liver were generated from regions of interest drawn over liver and heart area. Degree of hepatic uptake and blood clearance of LSA were evaluated with visual interpretation and semiquantitative analysis using parameters (receptor index : LHL3 and index of blood clearance : HH3), analysis of time-activity curve was also performed with curve fitting using Prism program. Results: Visual assessment of LSA images revealed decreased hepatic uptake in DMN treated rat, compared to control group. In semiquantitative analysis, LHL3 was significantly lower in DMN treated rat group than control rat group (DMN-3: 0.842, DMN-8: 0.898, DMN-21: 0.91, Control: 0.96, p<0.05), whereas HH3 was significantly higher than control rat group (DMN-3: 0.731,.DMN-8: 0.654, DMN-21: 0.604, Control: 0.473, p<0.05). AST and ALT were significantly higher in DMN-3 group than those of control group. Centrilobular necrosis and infiltration of inflammatory cells were most prominent in DMN-3 group, and were decreased over time. Conclusion: The degree of hepatic uptake of LSA was inversely correlated with liver transaminase and degree of histologic liver injury in rat with acute hepatic injury.
