Cosmetical Properties of Polysccharides from the Root Bark of Ulmus davidiana var. Japonica (유근피로부터 추출한 다당류의 화장료적 특성)

  • The fruit or root of Ulmus davidiana var. Japonica and its extract has traditionally been used as anti inflammatation medicines as well as a remedy of the digestive ulcer for a long time. Ulmus davidiana var. Japonica has also been used to cure palliative and diuresis, because it contains lot of mucus and tannin. In this study, we examined the moisturizing effect and anti-inflammation effect of polysaccharides extracted from Ulmus davidiana var. Japonica to investigated whether it can be used as a cosmetic ingredient or not. The polysaccharides were extracted from Ulmus davidiana var. Japonica. Then the hydrolyzed was obtained through the hydrolysis. It has been ascertained that the polysaccharides we mainly composed of rhamnose, galactose, and glucose through HPLC experiments, and it has 20,000 of the molecular weight and 89.3 dL/g of intrinsic viscosity In a moisturizing test of measuring water loss in a desiccator and moisture content by Comeometer CM820, Ulmus davidiana root extract showed a similar moisturizing effect as hyaluronic acid. And its anti-inflammation effect on carrageenin-induced edema evaluated by measuring the thickness of swelling over paw induced by carrageenin (25 uL of $2\%$ carrageenin saline injection), the $\%$ swelling over paw treated with Ulmus davidiana root extract was much less than that of paw treated with carrageenin only. The anti-inflammatory effect of Ulmus davidiana root extract was almost similar to that of ketoprofen. These results can say that Ulmus davidiana root extract can be effectively used as a cosmetic ingredient.

Property and Bio-adhesiveness in Hydrogel Material with Content of Ketorolac and Gardeniae Fructus Hydrolysis Products (케토롤락과 치자엑스 가수분해물이 함유된 하이드로겔제의 물성 및 생체 부착성)

  • This study manufactured hydrogel, which was contained NSAIDs(non-steroidal anti-inflammatory drugs) Ketorolac tromethamine and hydrolyzed products of gardeniae fructus extract, and experimented viscosity, surface tension, tensile strength and bio-adhesiveness by using hairless mouse. Thus, it was performed in expectation for being probably able to develop as effective auxiliary agent of periodontal disease after non-surgical or surgical periodontal treatment. As a result, the following conclusions were obtained. 1. Out of KGE and KGH gel materials, the content of ketorolac tromethamine was 1.02~0.97%. The content of geniposide was 0.34% in KGE gel A and C. However, it got lower to 0.11% in KGH gel B and D. The content of genipin wasn't shown in KGE gel A and C, but was shown with 0.13% in KGH gel B and D. 2. As for viscosity according to temperature in gel material, the gel, which used independently Carbopol 940 as gel inoculant, maintained the higher viscosity than the gel, which added Poloxamer 407. The surface tension in each material showed 34.77~40.58 dyne/cm at 37. As for tensile strength in material, KGH gel B was shown the higher tensile strength in about 3.5 times compared to the control group. 3. As for bio-adhesiveness, the back-skin upper part(epidermis) and abdomen skin were shown to be 50.62 N in KGH gel B, thereby having indicated higher value in about 5 times compared to control group. The back-skin lower part(dermis) and abdomen skin were shown to be 35.93 N in KGH gel B, thereby having indicated higher value in about 3.5 times compared to control group.

Skin Permeation and Periodontits Alleviation Effect of Hydrogel Containing Ketorolac Tromethamine and Hydrolysis Products of Gardeniae Fructus (케토롤락트로메타민과 치자가수분해물이 함유된 Hydrogel 제의 피부투과 및 치주염완화효과)

  • After producing 4 kinds of hydrogel materials by mixing the composite preparation in Gardeniae Fructus extracts with ketorolac tromethamine, which is NSAIDs (nonsteroidal anti-inflammatory drugs), through the experiment of measurement in the tensile strength, in the skin permeation, and in the periodontal-pocket reduction rate, the following conclusions were obtained. 1. The tensile strength of a drug indicated the tensile strength 3.5-fold higher compared to the control group, in KGH gel B. 2. The skin-permeation volume in ketorolac tromethamine was highest with $105.62{\mu}m/cm^2$ in KGH gel B for 8 hours, and the permeation volume in geniposide was relatively high with $73.8{\mu}m/cm^2$ in KGE gel A, but the permeation volume in genipin, which is the hydrolysis, represented the highest permeation amount with $50.17{\mu}m/cm^2$ in KGH gel B. 3. In terms of the periodontal-pocket reduction rate, after 4 weeks, KGE gel A showed the falling rate of 23.85% compared to the control group, but did not indicate the significant difference, and KGH gel B represented the reduction rate of 29.46% compared to the control group, thus it indicated the significantly treatment effect.

Reparability and Surgical Indications of Rotator Cuff Tears (회전근 개 파열의 봉합 가능성 및 수술 적응증)

  • Purpose: The reparability of rotator cuff tears is very important to establish the indication of the surgical treatment of rotator cuff tears. We reviewed the factors that effect to reparability of rotator cuff tears, and considered about the surgical indication of rotator cuff tears. Materials and Methods: The reparability was considered by the surgical reparability and the postoperative healing potentials of rotator cuff. The natural history of rotator cuff tear gives us the information to make decision about the surgical treatment or the conservative treatment. Results: There are 3 general categories of factors that help predict the healing potential of a rotator cuff tear. These include surgical technique, biology, and environmental issues. Surgeon can control the surgical technique by himself. The control of biology comes from careful operative indications by age, acute trauma history, chronicity, and tear size. And the control of environmental factor can give better outcomes by cessation of smoking and nonsteroidal anti-inflammatory drug (NSAID). Conclusion: A better understanding of the natural history and the reparability of rotator cuff tear will help us to estabilish the indication of surgical treatement of rotoator cuff tears, and lead to satisfactory clinical outcomes.

Conservative Treatment of Impingement Syndrome and Rotator Cuff Tear (충돌 증후군 및 회전근 개 파열의 보존적 치료)

  • The conservative treatment for impingement and rotator cuff tear includes rest, nonsteroidal anti-inflammatory drugs (NSAIDs), local steroid injection and physiotherapy depending on the purpose to relieve the pain and inflammation, in addition, stretching exercise to recover flexibility and strengthening exercise to recover the function could be used. When these conservative treatments are divided into multiple steps, the first one contains pain relief, modification of daily activity and stretching exercise. Second step includes strengthening exercise of the anterior/posterior cuff and peri-scapular muscles and eventually. The third step includes training program to return to job, housework and hobby activities and maintain. Thus, the key of these step wise approach for the treatment of impingement and rotator cuff tear is exercise program. Understanding of various exercise program and apply to the patients properly is most important for the conservative treatment of impingement and cuff tear.

In vivo Antinociceptive Antiinflamatory and Antioxidative Effects of the Leaf and Stem Bark of Kalopanax pictus in Rats (음나무 잎 및 수피의 진통소염효과 및 아주반트로 유발된 산화적 스트레스에 대한 효과)

  • The leaves (KPL) of Kalopanax pictus (KP) are used as a vegetable or a functional food in Korean society. The stem bark (Kalopanacis Cortex, KPS) has been traditionally used to treat neurotic pain, rheumatoid arthritis and diabetic disease. This research was undertaken to demonstrate that the leaf extract of KP (KPL) has also the antinociceptive and antiinflammatory effects like the extract (KPS) of Kalopanacis Cortex and to compare the activity levels of several extracts obtained from KP. Antinociceptive and antiinflammatory effects were measured against the extracts described as followings; KPL-1 (the MeOH extract obtained from the leaf shoot of KP collected on May), KPL-2 (the MeOH extract from KP collected on June), KPL-3 (the MeOH ectract from KP with no thorns), KPS-1 (MeOH extract from KPS of a Korean habitat), KPS-2 (MeOH extract from KPS of a Chinese habitat). The antimociceptive test undertaken by acetic acid-induced writhing, hot plate-, and tail-flick methods using mice. The anti-inflammatory test was also undertaken by measuring the edema in the carrageenan-induced test. The order of activity potency in the antinociceptive and antiinflammatory assays was commonly shown as followings: KPL-3>KPS>1>KPS-2>KPL-1>KPL-2. This order was also observed in acetic acid-induced vascular permeability test. The antiinflammatory activity in carrageenan-induced assay was also observed as the following order: KPL-3>KPS- 1>PS-2>KPL-1>KPL-2. In addition, adjuvant-induced rats were used for a model to assess the oxidative stress. Treatment of the rat with the extracts reduced serum thiobarbituric acid-reactive substances (TBARS), hydroxy radical(OH) and superoxide dismutase(SOD) activity caused by FCA together together with the inhibition of hepatic TBARS level and lipofuscin content. The above finding suggests that the leaf extract has the antinociceptive and antinflammatory activity. It is also suggested that KPL-3 with more potent activity than other tested extracts could be developed for a new available biomaterial.

Effects of Kalopanax Pictus Extracts and Their Related Origin on Gastric Lesions (해동피 및 유사생약 추출물의 위 손상에 대한 효과)

  • Kalopanax pictus has pharmacologically anti-inflammatory and analgesic effect and is known to respond to treatment of backache, knee pain and etc. In this study, we investigated the effects on gastric lesions of Kalopanax pictus both from Korea (KPK) and China (KPC) compared with their related origin, Znthoxylum ailanthoide both from Korea (ZAK) and China (ZAC), and Korean Bombax malabaricum (BMK). In preliminary screening, KPK and KPC shown effective inhibition of HCI EtOH-induced gastritis in rats. To elucidate their protective effects on gastric lesions, we assessed inhibition of H. pylori colonization, 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activities, reducing power test, and inhibition of lipid peroxidation. KPK was the most effective from antioxidant assays. KPK also shown the inhibition of indomethacin-induced gastric ulcer in rats. Gastric secretion in rats, KPK reduced the secretion of gastric juice and total acidity and raised pH. Therefore, it is possible that KPK can be developed as health functional food and natural medicine. In addition, it can contribute to the standardization with objectivity and reliability for KPK through the criteria establishment of the precise origin of medicine, the prevention of indiscriminate distribution of imitation, and the rising rate of dependence on imports of medicinal herbs, and mixing prevention of low-quality goods.

Study on the Projectile Velocity Measurement Using Eddy Current Probe (와전류 탐촉자를 이용한 총구 탄속 측정에 관한 연구)

  • Nowadays the weapon systems are employed air bursting munition (ABM) as smart programmable 40 mm shells which have been developed in order to hit the target with programmed munition that can be air burst after a set distance in the battlefield. In order to improve the accuracy of such a bursting time, by measuring the speed of the munition from the barrel, weapon systems calculate the exact time of flight to the target and then the time information must be inputted to the munition. In this study, we introduce a device capable of detecting a shot at K4 40 mm automatic grenade. The shot is composed of a rotating copper band to convert linear motion into rotary motion when it passes through the barrel, the steel section is exert the effect of fragment and aluminum section to give fuze information. The aluminum section was used to detect munition using eddy current method. To measure muzzle velocity by means of non-contact method, two eddy current probes separated 10 cm was employed. Time interval between two eddy current probe detection times was used as muzzle velocity. The eddy current probe was fabricated U-shape Mn-Zn ferrite core with enamelled copper wire, and 200 kHz alternating current was used to detect inductance change. Measured muzzle velocity using the developed sensor was compared to the Doppler radar system. The difference was smaller than 1%.

Development of a Recombinant Streptomyces griseus with sprA and sprB Genes for Proteolytic Enzyme Production (Streptomyces griseus IFO13350 유래 sprA 및 sprB 유전자를 이용한 Pretense 생산균주 개발)

  • Pronase, a protease produced for commercial purpose by Streptomyces griseus, was composed of serine protease, alkaline protease, aminopeptidase and carboxypeptidase complex, and it has been widely used as anti-inflammatory drugs for human therapy. In this study, we developed a new integration vector, pHJ101 derived from pSET152, containing strong promoter, ermE, to overexpress a certain protease gene. Specific PCR primers for cloning of sprA (a gene for S. griseus protease A) and sprB (a gene for S. griseus protease B) genes were designed from the basis of nucleotide sequence in databases and amplified by PCR. Plasmid pHJ201 and pHJ202 were constructed by inserting of amplified each gene in a vector pHJ101. S. griseus HA and S. griseus HB were respectively obtained by conjugal process of a parent strain, S. griseus IFO 13350 with the recombinant Escherichia coli harboring plasmid pHJ201 or pHJ202. When protease activity was measured in flask cultivation, produced protease levels of S. griseus HA and S. griseus HB increased about 5.3 times and 5 times, respectively, more than that of parent strain. And, the constructed integrating plasmid pHJ101 was applicable for overexpression of a certain gene in Streptomyces sp.

Effect of Saururi Herba Seu Rhizoma on anti-inflammatory properties in RAW264.7 cell line and murine models of inflammation (삼백초(三白草)의 소염작용(消炎作用)에 대(對)한 실험적(實驗的) 연구(硏究))

  • Purpose : The purpose of this research was to investigate the effects of Saurui Herba Seu Rhizoma(SHSR) on Anti-inflammatory properties in Raw264.7 cell line and murine models of inflammation. Methods : To investigate the effects of Saurui Herba Seu Rhizoma(SHSR) on anti-inflammation, we study cytotoxicity effects of SHSR on Mouse Lung Fibroblast Cells and Peritoneal Macrophages, Inhibitory effects of SHSR on the nitric oxide (NO) release, the ROS production, and the interleukin-6 production. Results : The cytotoxicity of SHSR on mouse lung fibroblast Cells and Raw264.7 cell line was not observed. SHSR in RAW264.7 cell line inhibited $IL-1{\beta}$, IL-6 mRNA gene expression depending upon the concentrations of extract and inhibited IL-18 mRNA gene expression at 100 ${\mu}g/ml$ of extract. SHSR in RAW264.7 cell line inhibit COX-2 mRNA gene expression at 100, 10 ${\mu}g/ml$ of extract. SHSR in RAW264.7 cell line inhibited NOS-II mRNA gene expression depending upon the concentrations of extract. SHSR in RAW264.7 cell line didn't inhibit $TNF-{\alpha}$ mRNA gene expression. SHSR in RAW264.7 cell line decreased IL-6 production depending upon the concentrations of extract. SHSR in RAW264.7 cell line decreased $ITNF-{\alpha}$ production according to the concentrations of extract. SHSR in RAW264.7 cell line inhibited NO release specially SHSR 100, 10 ${\mu}g/ml$ concentrations of extract. SHSR inhibit ROS production depending upon the concentrations of extract. Conclusion : These results suggest that SHSR can be used treating a lot of women disease caused by inflammation.
