Scaffolds of tissue engineering should be biocompatible and biodegradable for cell attachment, proliferation and differentiation. In the various scaffold fabrication, 3D printing technique can make the three dimensional scaffold with interconnected pores for cell ingrowth. Polycaprolactone (PCL) is biodegradable polyester with a low melting temperature and has been approved by the Food and Drug Administration (FDA). In this study, PCL scaffold was fabricated by 3D bioprinting system and surface modification of PCL scaffold was controlled by NaOH treatment. Morphological change and wetability of NaOH-treated scaffold were observed by SEM and contact angle measurement system. The remnant of PCL treated with NaOH was measured by ATR-FTIR. In vitro study of scaffolds was evaluated with WST-1 and ALP activity assay. NaOH treatment of PCL scaffolds increased surface roughness, hydrophilicity, cell proliferation and osteogenic differentiation. These results indicate that NaOH-treated PCL scaffold made by 3D bioprinting has tissue engineered potential for the development of biocompatible material.
This study presents a fabrication method of cell-chip using aqueous solution based surface modification. The applications of cell-chip have potential for fundamental study of genetics, cell biology as well as cancer diagnostics and treatment. Conventional methods for fabrication of cell-chip have been limited in economic loss and environmental pollution because of the use of harsh organic solvent, complex process of silicon technology, and expensive equipment. In order to fabricate cell chip, we have proposed simple and eco-friendly process combined polyelectrolyte multilayer coating with microcontact printing. For the proof of concept, the cell chip can be applied to analyze the different expression of cell surface glycans and derivatives between cancer and normal cells. Our proposed method is useful technique for the application of novel cancer diagnostics and basic medical engineering.

최근 사회적 이슈로 동물실험에 대한 규제가 강화되고 있어 동물실험을 대체할 새로운 방안의 중요성이 부각되고 있다. 이에 따라 현재 동물실험의 대체 방안의 하나로 3D 프린팅 기술을 활용한 3차원으로 배양된 인공장기에 대한 연구가 활발히 진행 중이다. 하지만 실시간으로 세포의 변화를 모니터링 할 수 있는 기술에 대한 연구는 많이 이루어지지 않고 있다. 본 연구에서는 3차원으로 배양된 세포에서 약물반응에 따른 세포변화를 실시간으로 분석할 수 있는 고감도 온도 및 임피던스 측정 바이오센서를 제작하였다. 센서 제작에 앞서 바이오센서로 사용하기 위해서는 세포를 안정적으로 성장시킬 수 있는 물질을 사용해야하며, 반도체공정으로 박막증착이 쉽고 물질변화가 크지 않도록 높은 work function(백금의 work function : 5.12~5.93 eV)을 가져야한다. 또한 온도 및 임피던스 측정을 위해 지표로 사용할 수 있는 TCR(Temperature Coefficient of Resistance)값이 온도에 따라 선형적으로 증가하는 특성을 가져야 한다. 위 조건들을 고려하여 센서물질로 백금을 선정하였다. 박막공정 및 열처리를 통하여 추출된 백금의 TCR은 $2045.9ppm/^{\circ}C$의 값을 가졌고, 추출된 백금의 TCR과 관계된 온도센서의 오차범위는 $0.01^{\circ}C$내에 있다. 이는 실시간으로 세포 변화를 분석할 수 있는 지표로써 활용되며, 고감도의 온도센서로써의 역할을 하기에 충분한 값이다.

Cardiovascular disease is one of the leading causes of death across the world, and gold-standard treatments such as percutaneous coronary intervention and artery bypass grafting have various limitations including myocardial damage and subsequent maladaptive cardiac remodeling. To overcome this, stem-cell therapies are emerging as a promising strategy for cardiovascular regeneration. Endothelial progenitor cells (EPCs) have high potential to proliferate and differentiate into endothelial cells for vascularization and tissue regeneration, and several clinical trials have explored EPC function in tissue repair in relation to clinical safety and improving cardiac function. Consequently, EPC has been suggested as a feasible stem-cell therapy. However, autologous EPC transplantation in cardiovascular disease patients is restricted by risk factors such as age, smoking status, and hypertension that lead to reduced bioactivity in the EPCs. New approaches for improving EPC function and stem-cell efficacy have therefore been suggested, including cell priming, organoid culture systems, and enhancing transplantation efficiency through 3D bioprinting methods. In this review, we provide a comprehensive understanding of EPC characteristics, therapeutic approaches, and the current state of clinical research into EPCs as stem-cell therapy for cardiovascular disease.

The ability of 3D/4D printing technology to create arbitrary 3D structures provides a greater degree of freedom in the design of printed structures. This capability has influenced the field of electronics and biomedical applications by enabling the trends of device miniaturization, customization, and personalization. Here, the current state-of-the-art knowledge of 3D printed electronics and biomedical applications with the unique and unusual properties enabled by 3D/4D printing is reviewed. Specifically, the review encompasses emerging areas involving recyclable and degradable electronics, metamaterial-based pressure sensor, fully printed portable photodetector, biocompatible and high-strength teeth, bioinspired microneedle, and transformable tube array for 3D cell culture and histology.

Patterning bacteria and cells on substrates has potential applications in molecular biology, antimicrobial drug screening, environmental monitoring and tissue engineering. We developed a technique to deposit two-dimensional array of bacterial cells onto an agar plate by modifying commercially available thermal inkjet printers. The concentration of the bacterial solution in the cartridge was carefully determined to ensure a single cell suspension in a droplet ejected from a nozzle. We measured quantitatively the effects of the bacterial concentration and the agar concentration on patterning performance. Bacterial patterning by inkjet printer is a low-cost and versatile technique which may replace the existing sophisticated methods.

The achievement of tissue engineering can be highly depending on the capability to generate complicated, cell seeded three dimensional (3D) micro/nano-structures. So, various fabrication techniques that can be used to precisely design the architecture and topography of scaffolding materials will signify a key aspect of multi-functional tissue engineering. Previous methods for obtaining scaffolds based on top-down are often not satisfactory to produce complex micro/nano-structures due to the lack of control on scaffold architecture, porosity, and cellular interactions. However, a bioprinting method can be used to design sophisticated 3D tissue scaffolds that can be engineered to mimic the tissue architecture using computer aided approach. Also, in recent, the method has been modified and optimized to fabricate scaffolds using various natural biopolymers (collagen, alginate, and chitosan etc.). Variation of the topological structure and polymer concentration allowed tailoring the physical and biological properties of the scaffolds. In this presentation, the 3D bioprinting supplemented with a newly designed plasma treatment for attaining highly bioactive and functional scaffolds for tissue engineering applications will be introduced. Moreover, various in vivo and in vitro results will show that the fabricated scaffolds can carry out their structural and biological functionality.

In this study, we used the ESD method to prepare the protein microarrays for observation the stem cell responses to pattern size, space and shapes. The ESD method allows a reduction in spot size, high efficiency of substance transfer, and high rate in fabrication as a result of ability to simultaneously deposit thousands of identical spots. Typical electro spraying conditions for the deposition of proteins were a voltage of $3{\sim}5keV$ and the humidity under 30%. The patterns of masks have a variety of shapes, spaces, and hole sizes from 10 um to $300{\mu}m$. Three kinds of proteins(collagen, fibronectin, and vitronectin dissolved in PBS) are deposited in a dry state, preserving the functional activity of proteins. Stem cells were cultured on each protein patterned sample at $37^{\circ}C$ for 1day.

Lips have a defect in maintenance of moisture due to their thin layer. As aging progresses, lips lose volume and redness, and become wrinkled. Fat grafting and filler surgery have been used to achieve attractive lips, but little research has been reported to develop better materials to replace the present methods. Recently, a study suggests that the increase of adipocyte number can be enhancing the expansion endogenous fat. In previous study, we identified that the efficacy of Magnolia officinalis bark extract (MOBE) was effective on the induction of adipogenic differentiation. In this study, we confirmed that MOBE enhanced the differentiation of human adipose-derived stem cells on the fat mimic 3D structure built by 3D bioprinting method From further experiments in human, we established a method to quantify the severity of lip wrinkle by measurement of standard deviation of gray value using Image J software. Finally, we found that topical treatment with 1% MOBE formulated lip balm significantly improved the lip wrinkle after using for 12 weeks. In conclusion, these findings suggest that MOBE has great potential, as a cosmetic ingredient, to reduce the lip wrinkle through the effect of promoting adipogenic differentiation.
