Immunohistochemical Study on the Cerebral Ganglion of African giant Snail, Achatina fulica (아프리카 왕달팽이(Achatina fulica) 뇌신경절의 면역조직화학적 구조)

  • An immunohisochemical study on the cerebral ganglion of the African giant snail, Achatina fulica. was conducted by applying the AB/AY staining and the avidin-bovine-peroxidase complex staining methods. The followings are the results obtained throughout the study. The cerebral ganglion of Achatina fulica is an ellipsoidal body of 2 x 1 mm in size, which is connected by the cerebral commissure of 1 mm in diameter. The cross-section through the cerebral ganglion, shaped like a butterfly, is divided into the medio-dorsal parts, the latero-dorsal parts, the caudo-dorsal parts, and the lateral lobes. In the medio-dorsal and latero-dorsal parts, the LG cells and the DG cells are found mixed, although the LG cells are dominant. In lateral lobe, however, the Y cells are quite dominant, while the LG cells and the DG cells are seldom found. The LG cells are 20-70 $\mu\textrm{m}$ in sizes and circular or ellipsoidal in shapes. They are stained light green with the AB/AY. 1 - 3 nucleoli are found in karyolymph, where granular chromantins are evenly distributed. In cytoplasm, it is found that the secretory granules are evenly developed.

Selective Accumulation of Rhodamine B in Müller cells in Rabbit Retina (Rhodamine B 염료의 토끼 망막 뮬러세포에 대한 선택적 염색)

  • Purpose: In this study, we investigated the dye to staining for selective accumulation in rabbit retina. Methods: Rhodamine B was injected into the vitreous body in rabbit. After 24 h, the isolated retina was checked condition of cell staining on the microscope. We used conventional immunocytochemical techniques for recognizing cell type. Results: Well-labeled nuclei were seen in the middle of the inner nuclear layer of the rabbit retina. The number and distrbution of the accumulating cells were similar to those of the m$\ddot{u}$ller glia. To identify m$\ddot{u}$ller cell, we used antibody directed against vimentin. Rhodamine B-immunoreactive nuclei also were labeled with antivimentin antibody. We found that Rhodamine B was accumulated selectively in retinal m$\ddot{u}$ller cell. Conclusions: Specific accumulation in rabbit retinal m$\ddot{u}$ller cell occurred when Rhodamine B was applied to living retina.

태아 십이지장 내분인포군집의 형태 및 면역조직화학적 특성

  • 임신 26주, 27주의 생명력이 없는 한국인 태아 2예를 대상으로 십이지장 점막에서의 장내분비세포군집(일부 일본인 학자들이 명명한 "Segi's cap")의 출현, 형태 및 구조를 관찰하고. 이들 세포군집에서 내분비세포들의 존재 및 면역조직 화학적 특징들을 관찰하고자, 파라핀조직절편과 냉동박절 후 hematoxvlin과 eosin염색. Azan염색, Comori법, 그리고 면역조직화학염색 표본을 제작, 관찰하여 다음과 같은 결과들을 얻었다 세포들의 군집 형태는 장 내강을 향해서는 오목한 형태였고, 기저막을 향해서는 볼록한 형태였다. 군집을 이룬 세포들은 중층을 이루었고. 술잔세포도 장 내캉목에서 관찰되었다 또한. 내강쪽에 위치한 세포들의 경우 미세융모들의 줄무의가장자리는 관찰되지 않았다 Leuenkephalin, somatostatin, substance p, vasoactive intestinal polypeptide 및 5-HT 항혈청을 이용하여 면역조직염색한 결과 somatostatin 양성반응세포와 5-HT 양성반응세포만이 관찰되었다. 이들 somatostatin 양성반응 세포와 5-HT 양성반응세포들은 대개가 원추형의 세포들로서 개구형을 이루고 있음을 관찰한 수 있었다.관찰한 수 있었다.

Application of Periodic Acid Thiocarbohydrazide Silver Proteinate Physical Development ( PA-TCH-SP-PD) Stain to Observation of Sertoli Cell (세르톨리세포 관찰을 위한 PA-TCH-SP-PD 염색의 적용)

  • The purpose of this study was to investigate the applicability of periodic acid thiocarbohy-drazide silver proteinate, physical development (PA-TCH-SP, -PD) stain to the seminiferous tubules for the ultrastructural studies of Sertoli cell column and Sertoli cell processes. In the Sertoli cell cloumn and Sertoli cell processes, high concentration of the reactive granules were observed under transmission electronmicroscope (TEM) after PA-TCH-SP-PD stain. Also some reactive granules were seen in the spermatogonium cytoplasm, clearly. These reactive granules specifically stained with PA-TCH-SP, -PD make the Sertoli cell column, Sertoli cell processes and spermatogonium cytoplasm easy to distinguish from nucleus of the germ cells, spermatocyte, spermatid and residual body which did not contain the reactive granules. This result indicates that the PA-TCH-SP, -PD stain is superior to other traditional electronic double stain methods for the ultrastructural studies of Sertoli cell.

  • 새로운 노화방지 물질로 개발한 3-APPA가 노화에 의해 야기되는 여러 변화들, 특히 세포 증식, 유전자 수준 및 단백질 수준에서의 collagen의 생합성 변화, 면역조직화학염색을 이용한 collagen 생합성의 변화등을 세포배양 및 동물실험을 통하여 측정하였다. MTT assay를 이용한 인체 피부 섬유아세포의 증식 실험에서 3-APPA는 무처치군에 비교해서 최고 2배의 섬유아세포 증식 효능을 나타내었으며, $^3$[H]-proline incorporation 방법을 이용한 단층세포 배양 및 3차원 dermal equivalent 섬유아세포 배양에서 무처치군 및 vitamin C 처리군에 비해 최고 1.5배의 collagen 생합성 증가를 나타내었다. 그러나 type I alpha-procollagen mRNA expression에는 영향을 미치지 않는 것으로 나타났다. H&E 염색을 이용한 hairless mice의 피부에 대한 형태학적 변화 및 type I pM procollagen antibody를 이용한 면역조직화학염색에서, 3-APPA는 collagen 생합성을 증가시키는 것으로 나타났다. 이상의 결과에서 3-APPA는 섬유아세포 배양 및 hairless mouse를 이용한 실험에서 피부 섬유아세포 증식을 촉진시키며 collagen 생합성을 증가시켜 피부노화를 억제 할 수 있는 물질임을 밝혔다.

Morphological Study on the TUNEL Reaction of Mouse Ovary by X-ray Irradiation (방사선 조사선량에 따른 생쥐 난소의 TUNEL 염색반응에 관한 형태학적 연구)

  • This research investigated morphological changes of ovarian follicle according to dose of irradiation when adult mice were exposed to X-rays from 6 MeV LINAC. At day 3 after irradiation of 200 cGy, 400 cGy and 600 cGy X-ray to the hole body of mice, the ovaries collected and stained with TUNEL. The normal follicles and atretic follicles were identified to apoptosis by the staining with TUNEL. In the atretic follicles of the normal ovary, the apoptotic bodies were well appeared and stained brown color. Almost of the follicles following irradiation are stained with TUNEL, but the sensitivity of reaction is weaker than that in irradiation of 400 cGy and 600 cGy X-ray. The granulosa cells of the radiated normal follicle by 400 cGV are shown brown color. In this stage, the nucleus of granulosa cells in the atrectic follicles are condensed and picknotic feature. The size of the radiated follicle by 600 cGy are decreased than the normal follicles. The atropic follicles are filled with apoptotic bodies which change of granulosa cells and theca cells by influence of X-ray. All of cell in the follicles are strongly positive stained with TUNEL by irradiation of 600 cGy.

마우스 골수세포와 말초혈액을 이용한 Giemsa와 Acridine orange를 이용한 염색법 비교

  • Acridine orange를 이용한 염색법은 OECD guideline 474에 채택되어 널리 사용되고 있는 염색법으로 Giemsa를 이용하였을 때 보다 artifact를 구별하기 쉬우며 숙련되지 않는 관찰자도 쉽게 소핵을 구별할 수 있다는 장점을 가지고 있다. 본 시험에서는 신약의 초기 screening단계에서 유전독성여부를 알아보기 위한 방법으로 마우스 골수세포와 말초혈액을 이용하여 Geimsa와 Acridine orange염색법을 비교하였다. (중략)

THE BONE CHANGES OF THE MIDPALATAL SUTURE AFTER RAPID PALATAL EXPANSION IN YOUNG ADULT DOGS (유성견에서 상악골 급속확장 후 정중구개봉합부에서의 골조직 변화)

  • Rapid palatal expansion(RPE) is a method of inducing the new bone formation in the palate by separation of the midpalatal suture, which can be done conveniently by placing heavy force across the maxillary dental arch. This experiment was undertaken to examine the histologic changes after RPE and during retention period. Four young adult dogs(a control dog, three experimental dogs) aged 4 to 6 months old were used for this experiment. Expansion screw($Hyrax^{\circledR}$, Dentarum Inc.) was delevered to the palate and fumed 180 degrees every morning and evening for 8 days, giving a total expansion of 7.2mm. A control dog was sacrified at the starting point of this study without any treatment and three experimental dogs were sacrified after RPE, 14-day retention, and 28-day retention in each. Thereafter, those samples were observed with hematoxylin-eosin(H-E) stain, ground section(Villanueva stain), alkaline phosphatase(ALP) stain, tartrate-resistant acid phosphatase(TRA) stain. The results were as followings: 1. After RPE, collagen fiber bundles were stretched along the midpalatal suture and few osteoblasts were flattened-inactive state and also, a little osteoid tissues was observed. Few multinucleated osteoclasts which had TRAP-positive activity in their cytoplasm were seen in horizontal section, whereas a few osteoclasts were seen in frontal section, especially in the nasal floor side of palatal bone. 2. After 14-day retention, collagen fiber bundles were stretched along the midpalatal suture and few osteoblasts which had ALP-positive activity in their cytoplasm were seen. Few multinucleated osteoclasts which had TRAP-positive activity in their cytoplasm were seen in horizontal section, whereas a few osteoclasts were seen in frontal section, especially in the nasal floor side of palatal bone. 3. After 28-day retention, collagen fiber bundles were arranged like those of control dog and osteoblasts which showed a lot of immature bone formation were cuboidal shape and exhibited ALP-positive activity in their cytoplasm. Few multinucleated osteoclasts which had TRAP-positive activity in their cytoplasm were seen in horizontal section, whereas a few osteoclasts were seen in frontal section, especially in the nasal floor side of palatal bone. According to the above results, the new bone formation after rapid palatal expansion was examined after 14-day retention and significantly increased after 28-day retention.

Immunohistochemical Expression of Galectin-3, Cytokeratin 19 and HBME-1 in Papillary Microcarcinoma of the Thyroid Gland (갑상선 미세 유두암종에서 Galectin-3, Cytokeratin 19와 HBME-1의 발현)

  • With the wide use of ultrasonography and fine needle aspiration of the thyroid gland, the incidence of papillary microcarcinoma of the thyroid gland is rapidly increasing nowadays. To improve the diagnostic accuracy of histopathologic findings of papillary thyroid carcinoma, various molecular markers have been used recently. We analysed the expression of galectin-3, cytokeratin 19 and HBME-1, using immunohistochemical technique in 37 cases of papillary microcarcinoma of the thyroid gland to evaluate the diagnostic value of these molecular markers. Immunohistochemically, galectin-3 expression was found in 37 cases of papillary microcarcinoma. Its localization was mostly cytoplasmic. Cytokeratin 19 expression was found in 36 cases. It was mostly localized to the cytoplasm and membrane. HBME-1 expression was found in all cases. Its localization was plasma membrane. The expression of these three molecular markers was negative in the adjacent normal thyroid tissue and accompanying benign lesions, although there are scattered foci of incomplete positive staining in cases of Hashimoto's thyroiditis. Our findings suggest that the immunohistochemical staining using antibodies for galectin-3, cytokeratin 19 and HBME-1 is an useful adjunctive method for the histopathological diagnosis of a papillary microcarcinoma of the thyroid gland.

Morphological Study on the Mast Cell of Proventriculus in Pheasant (Phasianus colchicus) (꿩 전위의 비만세포에 관한 형태학적 연구)

  • Mast cells have been studied extensively in various animals including rats and mice, whereas little is known the morphological data about pheasant mast cells. Here, morphological features of Korean pheasant mast cells are described in this study using light and electron microscopes. For light microscopy, mast cells had many metachromatic granules stained with toluidine blue in the cytoplasm. The fixation with $10\%$ neutral buffered formalin blocked staining of most mast cells but a modified Karnovsky solution proved to be a good fixative. In Korean pheasants, toluidine blue stained more mast cells than did alcian blue. For electron microscopy, the mast cells of the Korean pheasant were round, oval, spindle-like and irregular form and occasionally had a few short cytoplasmic processes. These cells had membrane-bounded granules and poorly developed organells. Some granules in the cytoplasm of the mast cells had bilayer membrane. Most granules were round shape and the membrane of several granules was concave or convex. The granules were composed of three parts, homogenous, particulate and reticular pattern.
