Anti-diabetic mechanism of melania snail (Semisulcospira libertina) protamex hydrolysates (다슬기 protamex 가수분해물(MPH)의 항당뇨 기작 연구)

  • Melania snail (Semisulcospira libertina) was traditionally used as the healthy food in Korea. It was generally known to improve liver function and heal a diabetes. The aim of this study was to elucidate the anti-diabetic mechanism of melanian snail hydrolysates treated with protamex (MPH) by investigating the inhibitory action on protein tyrosine phosphatase 1B (PTP1B), the improving effect on the insulin resistance in C2C12 myoblast and the protective effect for pancreatic beta-cell (INS-1) under the glucose toxicity. The melania snail hydrolysates treated with protamex (MPH), which showed the highest degree of hydrolysis (43%), and inhibited effectively PTP1B activity ($IC_{50}=15.42{\pm}1.1{\mu}g/mL$), of which inhibitory effect was higher than usolic acid, positive control ($IC_{50}=16.65{\mu}g/mL$). MPH increased the glucose uptake in C2C12 myoblast treated with palmitic acid. In addition, MPH increased insulin mRNA expression level by over 160% with enhanced cell viability in INS-1 cell under the high glucose concentration (30 mM). These results suggest that MHP may improve the diabetic symptom by the inhibiting the PTP1B activity, increasing the glucose uptake in muscle cell and protecting the pancreatic beta-cell from glucose toxicity.

Antioxidative Activity and Cytotoxicity of Fermented Allium victorialis L. Extract (산마늘 발효추출물의 항산화활성과 세포독성)

  • This study was conducted to investigate the antioxidative activity and cytotoxicity of fermented Allium victorialis extract. The results were as follows; The total polyphenol content of A. victorialis extract was 2.63 mg/g, and that of fermented A. victorialis extract was 1.65 mg/g which decreased a little by fermentation. The total flavonoid content of A. victorialis extract was 57.77 mg/g, and that of fermented A. victorialis extract was 62.27 mg/g, and this could increase a little from fermentation. Electron donating ability of A. victorialis extract was lower than vitamin C(97.71%), but before fermentation it was 82.29% and after fermentation it became 82.40%. Nitrite scavenging ability of A. victorialis extract before and after fermentation showed lower numerical value than that of butylated hydroxytoluene(BHT) at pH 2.5 but that of A. victorialis extract expressed higher than that of BHT. Superoxide dismutase-like activity showed relatively low level, 15%. Nitrite production increased by A. victorialis extract but was inhibited after fermentation. Methyl diamphetamine (MDA) content was inhibited with increased concentration of A. victorialis extract compared with $H_2O_2$ treatment but there was not any difference before and after fermentation. Therefore, production of lipid peroxide(LPO) was inhibited by A. victorialis extract. Cell viability of fibroblast cell was tend to slightly decrease with increased concentration of A. victorialis extract, but not different with control.

Change of Gene Expression Pattern of Mycobacterium tuberculosis H37Rv Against Host Immune Response in Infected Mouse Lung (결핵균 H37Rv에 감염된 마우스의 폐에서 면역 반응에 대항하는 Mtb 유전자의 발현 변화)

  • Mycobacterium tuberculosis (Mtb) is one of the most successful pathogens to infect one third of world population. Th1-mediated immunity against Mtb infection is known as critical to express mycobacteriostatic function but it is not sufficient to resolve the infection. In this study, to verify the possibility Mtb itself change the gene expression to survive against host immune response, expression pattern of selected H37Rv genes, 16S rRNA, acr, fbpA, aceA, and ahpC, during the course of infection was measured with absolute quantitation method using real-time RT-PCR. The total number of transcripts of 16S rRNA increased during the course of infection, which was coincide with the increasing CFU. The total number of fbpA transcripts per CFU, which encode typical secreted Mtb antigen, Ag85A, increased for 10 days of infection before decreasing. The number of transcripts of acr per CFU, which encode heat shock protein, ${\alpha}$-crystallin, increased during the infection, and ahpC and aceA, they both are enzymes produced in oxidative stressful condition, increased for 20 days and then slightly decreased on day 30. These findings are one of survival strategy of pathogen evading host immune response lead to persistent infection inside host cells.

Enhancement of Antioxidant and Whitening Effect of Fermented Extracts of Scutellariae baicalensis (황금 발효물의 항산화 및 미백 효과 증진)

  • Scutellariae baicalensis (S. baicalensis) has been traditionally used for anti-inflammatory effect. This study was designed to compare the antioxidant and whitening effects of S. baicalensis extract and its fermented extract by Leuconostoc mesenteroides (L. mesenteroides). Fermented extract of S. baicalenins was prepared by inoculation of L. mesenteroides after the extraction procedure with 70% ethanol. S. baicalensis extract and its fermented extract was investigated via high-performance liquid chromatography (HPLC). Simultaneous qualitative analysis of two bioactive components; baicalin and baicalein was achieved by comparing their retention times ($t_R$) and UV spectra with those of the standard components. Cell viability test results indicated that both S. baicalensis extract and its fermented extract were non-toxicity. In DPPH radical scavenging ability, $SC_{50}$ values of the fermented extract was $34.43{\mu}g/mL$ as a result of more effective than S. baicalensis extract. In nontoxic concentration rage, fermented extract of S. baicalensis showed strong melanin production inhibitory effect in ${\alpha}$-melanocyte stimulating hormone (MSH)-stimulated B16F10 cell ($IC_{50}=68.17{\mu}g/mL$). These results suggested that fermented extracts of S. baicalensis has considerable potential as a cosmetics ingredient with an antioxidant and anti-wrinkle and whitening effects.

Characteristics and Biocompatibility of Electrospun Nanofibers with Poly(L-lactide-co-ε-caprolactone)/Marine Collagen (전기 방사법을 통해 제조된 Poly(L-lactide-co-ε-caprolactone)/Marine Collagen 나노파이버의 특성 및 세포친화력 평가)

  • The uniform nanofibers of poly(L-lactide-$co$-${\varepsilon}$-caprolactone) (PLCL) with different contents of marine collagen (MC) were successfully prepared by electrospinning method. The effects of the major parameters in electrospinning process such as tip to target distance (TTD), voltage, nozzle size and flow rate on the average diameter of the electrospun nanofiber were investigated in generating composite nanofiber. The diameter and morphology of the nanofibers were confirmed by a scanning electron microscopy (SEM). Also, we measured a water contact angle to determine the surface wettability of the nanofibers. The average diameter of the nanofibers decreased as the value of TTD, MC contents, and voltages increased in comparison with that of pristine PLCL nanofiber. In contrast, the diameter of the nanofibers increased as the flow rate and inner diameter of nozzle increased in comparison with that of pristine PLCL. In addition, the hydrophilicity of the nanofiber and attachment of MG-63 cells on the sheets increased as incorporated collagen contents increased. Therefore, the marine collagen would be a potential material to enhance cellular interactivity of synthetic materials by mimicking the natural tissue.

Induction of Apoptosis by Bee Venom in A549 Human Lung Epithelial Cancer Cells through Modulation of Bcl-2 and IAP Family and Activation of Caspases (Bcl-2 및 IAP family의 발현 변화와 caspase 활성을 통한 봉독의 인체폐암세포 apoptosis 유도)

  • Bee venom is used to treat inflammatory diseases in Korean traditional medicine and has been known to inhibit proliferation and induce apoptosis in cancer cells. However, the molecular mechanisms involved in bee venom-induced apoptosis are still uncharacterized in human lung cancer cells. In the present study, we investigated the effects of bee venom on the apoptosis of A549 human lung epithelial cancer cells. Treatment of bee venom inhibited the cell viability and induced apoptosis in a concentration-dependent manner as measured by hemocytometer counts, fluorescence microscopy and flow cytometry analysis. Bee venom-induced apoptosis in A549 cells was associated with a marked inhibition of anti-apoptotic Bcl-2 expression without significant changes in the levels of Bax and Bcl-xL. Bee venom treatment also inhibited the levels of IAP family members such as cIAP-1 and cIAP-2 and induced the proteolytic activation of caspase-3 and caspase-9. Although further studies are needed, the present results suggest that apoptotic signals evoked by bee vemon in A549 cancer cells may converge caspases activation through a down-regulation of Bcl-2 rather than an up-regulation of Bax. These findings provide important insights into the possible molecular mechanisms of the anti-cancer activity of bee vemon in human cancer cells.

Mesenchymal Stem Cells Ameliorate Adriamycin Induced Proteinuric Nephropathy (Adriamycin 유발 신병증에서 중간엽 줄기세포의 완화 효과)

  • Purpose : Glomerulonephropathy (GN) often manifests as proteinuria and progresses to chronic renal failure without specific therapy. Mesenchymal stem cell (MSC) has been tried as a therapeutic agent in experimental GN, and previous studies showed that administration of MSC concomitantly to the insult inducing GN or via intra-renal administration ameliorated proteinuria. The purpose of this study was to test the therapeutic potential of MSC administered via intravenous route at the time of clinically evident proteinuria. Methods : MSCs were administered intravenously via tail vain into the mice with adriamycin (ADR) induced nephropathy (ADR-GN), two weeks after ADR injection when massive proteinuria was evident. To test the capacity of MSC modulate the cytokine production in the inflammatory milieu, the concentrations of IFN-$\gamma$ and IL-10 were measured in the supernatant of in vitro mixed lymphocyte culture (MLC) with or without additional MSC. Results : MSCs administered intravenously into the proteinuric mice with ADR-GN accelerated the recovery of this experimental GN with disappearance of proteinuria in two weeks when the saline treated (control) mice still showed significant proteinuria. The mice treated with MSC also had a tendency of better survival. Addition of MSC decreased IFN-$\gamma$ and increased IL-10 in the supernatant of MLC. Conclusion : This study showed that MSC had a therapeutic potential even when administered in a more clinically relevant setting into a proteinuric glomerulonephropathy model. Further study to verify the mechanism and long-term safety of this phenomenon is required.

Up-Regulation of NAG-1 and p21 Genes by Sulforaphane (브로콜리 유래 sulforaphane에 의한 NAG-1과 p21 유전자의 발현 조절)

  • We investigated the anti-proliferative activity of sulforaphane and expression changes of NAG-1 and p21 genes in response to sulforaphane treatment in human colorectal HCT116 cells. The results showed that sulforaphane decreased cell viabilities in a dose-dependent manner and induced expression of NAG-1 and p21 proteins in a dose-dependent and time-dependent manner. In addition, we found that NAG-1 expression by sulforaphane was not dependent on the presence of p53, whereas p21 expression was dependent on p53 presence. The results indicated that up-regulation of NAG-1 was not related with the activity of a dietary histone deacetylase inhibitor of sulforaphane. ATF3 induction was detected from 2 hr after sulforaphane treatment, indicating that ATF3 could be a transcription factor to up-regulate NAG-1 expression. The results of this study may help to increase our understanding of the molecular mechanism of anti-cancer activity mediated by sulforaphane in human colorectal cancer cells.

Acute toxicity effets of formaline to flounder, Paralichthys olivaceus (넙치 (Paralichthys olivaceus)에 대한 포르말린의 급성독성 효과)

  • Twenty-four hours, acute toxicity and the histopathological effect of formaline to flounder, Paralichthys olivaceus larvae were examined. The $LC_{50}$ values obtained to farmaline were 2,520 ppm in 1 hour treatment. 1.610 ppm in 2 hours treatment, 868 ppm in 4 hours treatment and 141 ppm in 24 hours treatment. Many pathological features such as hypertrophy of mucous and epithelial cells in secondary gill lamella, hyaline droplet degeneration of tubular epithelial cells in the proximal convoluted segment of renal tubules, focal or massive necrosis in liver cells and pycnotic nucleus in heart cells were recognized. The above results were discussed in relation to the application of formaline as therapeutic agent in flounder disease.

Effects of medicinal herb water extracts on expression of hepatic glucokinase, pyruvate dehydrogenase and acetyl-CoA carboxylase mRNA (한약재 물 추출물이 간세포 Glucokinase, Pyruvate Dehydrogenase, Acetyl-CoA Carboxylase mRNA 발현에 미치는 영향)

  • We studied the anti-diabetic effects of medicinal herb water extracts on expression of hepatic glucokinase (GCK), pyruvate dehydrogenase (PDH), and acetyl-CoA carboxylase (ACC) mRNA. The medicinal herbs used for experiments were Cornus officinalis (CO), Paeonia suffruticosa Andrews (PSA), Discorea japonica Thunb. (DJ), Rehmannia glutinosa (RG), Lycium chinense (LC), and Pyrus pyrifolia (PP). For GCK mRNA expression, CO, RG, and LC water extracts exhibited a more effective activity than other extracts. Cells treated with RG and LC water extracts showed an increase in expression of PDH mRNA to 191% and 124%, respectively, compared to control. Expression of ACC mRNA was significantly higher in LC water extract. These data indicate that CO, RG, and LC water extracts stimulates expression of hepatic GCK, PDH, and ACC mRNA.
