  • An entomopathogenic bacterium, Photorhabdus temperata ssp. temperata (Ptt), suppresses insect immune responses and facilitates its symbiotic nematode development in target insects. The immunosuppressive activity of Ptt enhances pathogenicity of various microbial pesticides including Bacillus thuringiensis (Bt). This study was performed to select a cheap and efficient bacterial culture medium for large scale culturing of the bacteria. Relatively cheap industrial bacterial culture media (MY and M2) were compared to two research media, Luria-Bertani (LB) and tryptic soy broth (TSB). In all tested media, a constant initial population of Ptt multiplied and reached a stationary phase at 48 h. However, more bacterial colony densities were detected in LB and TSB at the stationary phase compared to two industrial media. All bacterial culture broth gave significant synergism to Bt pathogenicity against third instars of the diamondback moth, Plutella xylostella. Production of bacterial metabolites extracted by either hexane or ethyl acetate did not show any significant difference in total mass among four culture media. Reverse phase HPLC separated the four bacterial metabolites, which were not much different in quantities among four bacterial culture broths. This study suggests that two industrial bacterial culture media can be used to economically culture Ptt in a large scale.

  • To isolate a novel gene for antibacterial peptide, an inducible clone(BmInc8) was selected by differential screening strategy from Bombyx mori cDNA library prepared from lavae injected with Escherichia coli. This clone contained a cDNA insert of 564 nucleotides and encoded 59 amino acids with an apparent molecular mass of 6.3 kDa. The cDNA sequence of BmInc8 had 61.2% identity compared to that of the bactericidin from Manduca sexta and also the deduced amino acids sequences from this insert had 65% identity compared to that of the cecropin D peptide Hyalophora cecropia. The transient expression assay of this insert using prokaryotic expression vector system revealed that the expressed peptide displayed the antibacterial activity. The cDNA sequence was deposited in GenBank under the accession number U30289.

  • Selection procedures for breeding genic male sterile lines for resistance to both Phytophthora blight caused by Phytophthora capsici and bacterial spot caused by Xanthomonas euvesicatoria were executed to $F_3$ and $F_4$ generations derived from a cross between a Phytophthora resistant genic male sterile (GMS) breeding line and a bacterial spot and Phytophthora resistant breeding line. Resistance to P. capsici was originally introduced from KC294(CM334) and KC263(AC2258), the well-known sources of resistance to P. capsici. Resistance to bacterial spot was introduced from KC47(PI244670). GMS lines with high resistance to P. capsici were obtained and the selected lines are expected to be quantitatively resistant also to bacterial spot.

Bacterial Meningitis in Children in One Tertiary Hospital (소아의 세균성 수막염)

  • Purpose : We performed a retrospective study on bacterial meningitis in children, pertaining to the causative organisms, bacterial resistance to antibiotics and the effect of recent introduction of Haemophilus influenzae type b(Hib) vaccine. Methods : We analysed the forty-three cases of bacterial meningitis which had been treated at the Inha University Hospital from June 1996 to June 2003. Results : Nineteen cases(44.2%) of them were infants younger than 2 months of age, and 29 cases(67.5%) younger than 1 year of age. The common causative organisms under 2 months of age were group B streptococcus(GBS)(47.4%), E. coli(21.1%), and Klebsiella pneumoniae(21.1%). In the age group beyond 2 months of age, S. pneumoniae were seen in 50 %, H. influenzae in 16.7% and N. meningitidis in 16.7%. All of the five cases of Hib meningitis had not been vaccinated for Hib. There has been no Hib meningitis cases since 2001. Overall fatality rate was 4.5%, and complication occurred in 39%. Complications was significantly less frequent in patients resumed to be treated within 48 hours after onset compared to after 48 hours after onset. Penicillin resistance of S. pneumoniae and GBS isolated among bacterial meningitis cases was high. Conclusion : Timing of treatment after the onset of the disease appeared the most important factor for prognosis of bacterial meningitis. The cases of H. influenzae meningitis have decreased probably due to Hib vaccination.

Microbial Changes of Salted and Fermented Shrimp by High Hydrostatic Pressure Treatment (초고압처리에 의한 새우젓의 미생물변화)

  • This study was conducted to enhance the storage stability of fermented shrimp with different salt contents using a high hydrostatic pressure. The effects of the magnitude of pressure and treatment time on the microorganisms of the fermented shrimp were investigated. The highest microbial counts with respect to the salt levels were observed at 18% salt, showing $3.4{\times}10^5\;CFU/g$ for general bacteria, $6.4{\times}10^4\;CFU/g$ for halophilic bacteria, $4.2{\times}10^5\;CFU/g$ for yeast and $3.0{\times}10^4\;CFU/g$ for halophilic yeast. The degree of sterilization increased with the magnitude of pressure and treatment time, and the sterilization could be analyzed by the first order reaction kinetics. The sterilization rate constants $(k_p)$ of the halophilic bacteria was lower than that of general bacteria. The $log(k_p)$ increased linearly with pressure and the slope of the regression line of the halophilic bacteria was greater than that of general bacteria, indicating that the sterilization of the halophilic bacteria was more dependent on the pressure. High hydrostatic pressure treatment was an effective non-thermal sterilization method for the salted and fermented shrimp, and the optimum treatment condition was for 10 min at 6,500 atm.

Antagonistic Effect of Lactobacillus sp. Strain KLF01 Against Plant Pathogenic Bacteria Ralstonia solanacearum (세균성 시들음병에 대한 식물성 유산균(Lactobacillus sp.)의 저해효과)

  • An antagonistic bacterial strain KLF01 was isolated from rhizosphere of tomato and identified to be Lactobacillus sp. by biochemical and genetic analysis. This strain showed antagonism against the used plant pathogenic bacteria like Ralstonia solanacearum, (bacterial wilt), Xanthomonas axonopodis pv. citri, (Citrus canker), Xanthomonas campestris pv. vesicatoria (Bacterial spot), Eriwinia pyrifoliae (Shoot-blight) and Eriwinia carotovora subsp. carotovora group (Potato scab) through agar well diffusion method. In planta test done by drench application of strain KLF01 $(4{\times}10^8 cfu/ml)$ into the experimental plot containing tomato (Solanum lycopersicum L.) cultivar 'Lokkusanmaru' and red pepper (Capsicum annuum L.) cultivar 'Buja' plants, in pot test post-inoculated with the plant pathogenic bacteria, R. solanacearum significantly reduced the disease severity, compared to the non-treated plants.

The Effect Of Seawater Concentration Of The Survival Of Fecal Pollution Bacteria (분변성 오염세균류의 담수 및 해수에 대한 저항성)

  • The effect of fresh water and various concentrations of aged seawater on the survival of fecal pollution bacteria, Escherichia coli, type I, Aerobacter aerogenes type I, and Streptococcus faecalis type were determined. Survivals of bacteria were measured by the membrane silter technique. Three species of bacteria indicate more tolerance in fresh water than in seawater. After 14-day incubation in fresh water, survival rates of bacteria were 90% withe E.coli, 20% with A.aerogenes, and 0.6% with Str.faecalis. However, the survival rate of fecal pollution bacteria decreases as the concetnration of seawater is increased. Generally, the death rate of E.coli is least affected by concentration of seawater. A.aerogenes is eliminated more rapidly with higher concentration of seawater, while Str.faecalis marks rather slight variation of elimination in various concentrations of seawater. In 100- percent seawater (Cl 18.1 ), the days required for 99.9% elimination of bacteria were 4.5 days with A.aerogenes, and 6.5 days with E.coli and Str.faecalis.

Current Status of Photobiological Hydrogen Production Technology Using Unicellular Marine Cyanobacterial Strains (단세포성 해양남세균 종주를 이용한 광생물학적 수소생산 기술)

  • Among various microscopic organisms producing photobiological hydrogen, cyanobacteria have long been recognized as the promising biological agents for hydrogen economy in 21 century. For photobiological production of hydrogen energy, marine unicellular $N_2$-fixing cyanobacteria have been evaluated as an ideal subgroup of Cyanophyceae. To develope the hydrogen production technology using unicellular $N_2$-fixing cyanobacteria, 3 important factors are pre-requisite: 1) isolation of the best strain from marine natural environment, 2) exploration on the strain-specific optimal conditions for the photobiological hydrogen production, and finally 3) application of the molecular genetic tools to improve the natural ability of the strain to produce hydrogen. Here we reviewed the recent research & development to commercialize photobiological hydrogen production technology, and suggest that intensive R&D during next 10-15 years should be imperative for the future Korean initiatives in the field of the photobiological hydrogen production technology using photosynthetic marine unicellular cyanobacterial strains.

Inhibitory Activity of Marine Bacterium on the Growth of Vibrio anguillarum (Vibrio anguillarum에 대한 항균성 세균의 분리(Vibriostatic activity of marine bacteria))

  • The antibacterial activity of marine bacterium(Pseudomonas aeruginosa JYMB1-3) was assayed against Vibrio anguillarum with the aim of evaluating the possible use for biocontrolling fish disease as probiotic strain. Inhibition test on the solid medium showed that vibrios were especially sensitive to the JYMB1-3. Edwardsiella tarda, Streptococcus sp. and Staphylococcus epidermidis were also sensitive to that strain, however the antibacterial abilities were varied to the pathogens. The vibriostatic activity of antibacterial substance produced from $10^7$ cells of the strain for 24 hours was equivalent to $2.5{\mu}g$ of chloramphenicol.

Occurrence of Off-Odor and Distribution of Thermophilic Bacteria from Rice and Cooked Rice Stored at Electric Rice Cooker (쌀과 취반백미의 고온성 세균 분포 및 이상취 발생)

  • The distribution of mesophilic and thermophilic bacteria in milled rice was not different according to growing land and variety of rice. However, The number of these bacteria were abundant in milled rice of lower milling degree or longer storage period. The growth of thermophilic bacteria in cooked rice of electric rice cooker was rapidly increased during storage of lower temperature below 75 $^{\circ}C$. Thermophilic bacteria were not appeared just after cooking. After cooked rice was stored far 18∼24 hon thermophilic bacterial growth was rapidly increased by changing spore to vegetative cell. The positive relation between cell number of thermophilic bacteria and occurrence of off-odor was slightly observed. The number of thermophilic bacteria in upper cooked rice of oven and was higher than those in inner and lower cooked rice. Major volatile compound of milled rice cooked and stored in electric rice cooker was hexanal oriented from rice. After long storage, it was confirmed that furan was major volatile compound as off-odor.
