Reaction Mode of Transglucosidase from Aspergillus niger for Production of Isomaltooligosaccharides (Aspergillus niger 유래의 Transglucosidase의 이소말토올리고당 생성반응 특성)

  • The research was undertaken to characterize the reaction mode of transglucosidase (TG) from Aspergillus niger for the production of isomaltooligosaccharides such as isomaltose, panose and isomaltotriose. TG hydrolyzed maltose to glucose units and produced panose and glucose by transglucosylation. TG hydrolyzed panose to maltose and glucose when panose was used as an initial substrate. The reaction patterns of products when isomaltose, isomaltotriose or isomaltotetraose were used as substrates were different from the case when maltose was used as a substrate. Maltotriose and maltose showed the same formation pattern of products. TG also produced isomaltooligosaccharides from maltooligosaccharides. The production of panote by TG from maltose was mathematically described by Michaelis-Menten kinetics. The kinetic constants, $V_{max}$ (the maximum velocity) and $K_m$ (Michaelis constant), were estimated by Lineweaver-Burk plot to be 400 M/min and 21.4 mM, respectively.

  • Characteristics of cell growth and heavymetal adsorption by recombinant Saccharomyus cerevisiae strains harboring multiple copies of the CUP1 gene encoding metallothione (MT) protein were studied in batch cultures. Recombinant S. cerevisiae strains harboring multiple copies of the CUP1 gene were superior to the host and wild-type yeast strains in terms of cell growth and heavy metal removal, indicating that the copy number of the CUP1 gene for MT expression played an important role in the adsorption of heavy metals. It was suggested that the CUP1 promoter for the MT expression is induced by manganese and zinc as well as copper An optimum copper concentration for MT expression and concomitant adsorption of heavy metals by recombinant S. cerevisiae was found to be 0.31 mM. A nonionic surfactant Triton X-100 enhanced cell growth by 17.7% and removal of zinc by 6.1% compared with the control case.

Effects of Fermentation Conditions on Production of Erythritol by Candida magnoliae (Candida magnoliae의 발효 조건이 erythritol의 생산에 미치는 영향)

  • This study was carried out to examine the effects of fermentation conditions on the production of erythritol by osmophilic yeast Candida magnoliae. It was found that sucrose was superior to glucose as carbon source and 109 g/L erythritol was produced from 400 g/L sucrose. When yeast extract was used as nitrogen source, maximum values of yield and productivity for erythritol were obtained at 15 and 20 g/L of yeast extract, respectively. A mixture of 15 g/L yeast extract and 3 g/L ammonium phosphate allowed more efficient utilization of sucrose and hence resulted in 149 g/L of erythritol, 0.37 g erythritol/g sucrose of erythritol yield and $0.78\;g/L{\cdot}hr$ of erythritol productivity. A batch fermentation supplemented with 40 g/L KCl resulted in an erythritol concentration of 167 g/L and an erythritol yield of 0.42 g erythritol/g sucrose.

  • P. stipitis CBS5776 was cultivated to examine the characteristics of ethanol fermentation for hexoses (mannose, g1ucose, and galactose) and pentoses(xylose and arabinose). Glucose was the best carbon source among the sugars used in terms of ethanol yield. Glucose was used to produce ethanol with an yield coefficient 0.376g ethanol/g glucose, whereas mannose was converted to produce ethanol with an yield coefficient 0.326g ethanol/g mannose. P. stipitis CBS5776 was also grown in a mixture of sugars to study the pattern of carbon utilization. The yeast utilized glucose and mannose firsts and then galastose and xylose as carbon sources. Arabinose was partially used for biomass when it was present as a sole carbon source, but it was not metabolized at all in a mixture of carbon sources. P. stipitis produced $12.2g/\ell$ ethanol with a yield coefficient 0.332 g ethano1/g sugar in a mixture of sugars.

  • In a typical web environment, it is difficult to determine the update and re-computation status of WebView content or the transition of WebView processing included in web page. If an update to one of data is performed before a read operation to that, we could get a wrong result due to the incorrect operation and increase a complexity of the problem to process. To solve this problem, lots of researchers have studied and most of these problems at the single user environment is not problems. However, the problems at a distributed environment might be occurred. For this reason, in this paper, we proposed the extended state transition graph and algorithms for each status of WebView for explaining WebView state in the distributed environment and analyze the performance of using the materialized WebView and not. Additionally, also analyze the timing issues in network and effectiveness which follows in size of WebView contents.

Characterization of Enzymatic Properties of Streptomyces albus Amylase Expressed in Recombinant Bacillus subtilis (재조합 Bacillus subtilis 에서 발현된 Streptomyces albus 유래 amylase 의 효소공학적 특성)

  • The research was undertaken to characterize enzymatic properties of Streptomyces albus amylase expressed in recombinant Bacillus subtilis. Molecular weight and pI of the purified enzyme were estimated to be 50 kD by SDS-PAGE and 4.3 by isoelectric focusing. The optimum temperature and optimum pH were $45^{\circ}C$ and 6.0, respectively. D-and Z-value were estimated to measure thermostability of the purified enzyme. The Z-value was estimated $17.7^{\circ}C$, which is lower than typical amylase. Maltotetraose was produced as a major component from soluble starch in the early state of reaction but gradually degraded to maltose. Thin layer chromatography was also performed to analyze the reaction products. The parameters involved in Michaelis-Menten enzyme kinetics were found to be the maximum velocity of 0.37 mM/min and the Michaelis constant of 0.13%, respectively.

  • We introduce how to fit random effects models via a SRC-Stat statistical package. This package has been developed to fit double hierarchical generalized linear models where mean and dispersion parameters for the variance of random effects and residual variance (overdispersion) can be modeled as random-effect models. The estimates of fixed effects, random effects and variances are calculated by a hierarchical likelihood method. We illustrate the use of our package with practical data-sets.

Effectiveness of 32-element Surface Coil Array for Accelerated Volume-Targeted Breath-Hold Coronary MRA (체적 지향형 호흡정지 자기공명 조영술의 가속화에 대한 32채널 코일 어레이의 효용성)

  • Purpose : To compare 12 and 32-element surface coil arrays for highly accelerated coronary magnetic resonance angiography (MRA) using parallel imaging. Materials and Methods : Steady state free precession coronary MRA was performed in 5 healthy volunteers at 1.5 T whole body MR scanner using both 12 and 32-element surface coil arrays. Left anterior descending and right coronary artery data sets were acquired for each volunteer. Data sets were sub-sampled for parallel imaging using reduction factors from 1 to 6. Mean geometry factor (g-factor), maximum g-factor, and artifact level were calculated for each of the two coil arrays. Results : Over all reduction factors, the mean and maximum g-factors and artifact level were significantly reduced using the 32-element array compared to the 12element array (P << 0.1). The mean g-factor was sensitive to the imaging orientations of coronary arteries while the maximum g-factor and artifact level were independent of orientation. Conclusion : The 32-element surface coil array significantly improves artifact and noise suppression for highly accelerated coronary MRA using parallel imaging. The increased acceleration factors made feasible with the 32-element array offer the potential to enhance spatial resolution or increase volumetric coverage for 3D coronary MRA.

Antioxidant and Anti-fibrotic Properties of Root Extract of Lythrumsalicaria L. in $CCL_4$-Induced Liver Fibrosis Rat Model (사염화탄소로 섬유화가 유도된 흰쥐 간에서 털부처꽃 뿌리 추출물의 항산화 및 섬유화저해 활성)

  • Fifty percent ethanol extract of Lythrum salicaria Linne root (LSR) was tested in vitro on antioxidant activity, and furthermore was investigated on antioxidative and fibrosis protecting activities in $CCL_4$-induced liver fibrosis rat model. Ratio of hepatic GSH/GSSG (reduced glutathione/oxidized glutathione) as bio-parameter of antioxidant level in $CCL_4$ plus LSR-treated rats for 6 weeks significantly increased from 2.8- to 5.7-fold than that of $CCL_4$-treated rats at p < 0.05. Thiobarbituric acid reactive substances (TBARS) contents in $CCL_4$ plus LSR-treated rats ranged from 1.57- to 2.19-fold of normal rats and were lower than those in $CCL_4$ plus silymarin-treated rats ($1.78{\sim}2.46$-fold of normal rats) (p < 0.05). Amounts of hydroxyproline of liver tissue showing the content of total collagen, a parameter of fibrosis, in $CCL_4$ plus LSR-administrated rat livers were $4.9{\sim}8.8{\mu}g$/mg ($-47{\sim}-71%$, compared with that in $CCL_4$-treated rat livers ($16.6{\mu}g$/mg tissue), which were significantly lower than those in $CCL_4$ plus silymarin-administrated rats being $8.4{\sim}11.7{\mu}g$/mg ($-30{\sim}-50%$). This collagen reducing effect of liver tissue in $CCL_4$ plus LSR-treated rats was supported by histological observation using microscopy assay. From the results, we conclude that the root of L. salicaria have efficient antioxidant potential and effective antifibrotic activities.

Candida magnoliae에 의한 erythritol 생산을 위한 유가식 공정의 개발

  • Two-stage fed-batch culture was peformed to improve the volumetric productivity of erythritol. In the growth phase dissolved oxygen was maintained to 20% and the feed medium was automatically supplied to the fermenter by pH-stat mode. The cell yield was 0.76 g-cell/g-glucose. In two-stage fed-batch culture, 41% of total erythritol conversion yield with 187 g/L of erythritol concentration and 2.79 g/L-h of maximum erythritol Productivity were obtained when 400 g/L of glucose was directly added in the form of non-sterile powder at production phase. The erythritol productivity increased in parallel with cell mass. The metabolic shift in the biosynthetic pathway of erythritol was caused by dissolved oxygen concentration. The production of gluconic acid was observed when the dissolved oxygen in the medium was maintained over 40% during the production phase, whereas the dissolved oxygen concentration lower than 40% caused the production of citric acid. But the butyric acid was produced independently with dissolved oxygen concentration in the medium. The production of organic acids such as gluconic acid, citric acid, and butyric acid was decreased by addition of mineral salts.
