  • 우리나라는 국토의 약 68%가 산악지형으로 포장의 경사와 토양중의 자갈이 밭작물의 재배와 기계화에 장애가 되고 있다. 포장중의 자갈은 토양의 보습력과 통기성을 향상시키고 경사지에서의 토양침식을 방지하는 긍정적 측면이 있으나, 자갈이 토양 표면에 산재할 경우 작물의 발아와 입모를 억제하고, 작물생육을 위한 토양환경을 조성하는 경운작업 재배관리 작업을 어렵게 하기 때문에, 농민들은 포장내의 자갈을 제거하기 위해 많은 노력을 기울이고 있다. 특히 관행 로타리 작업 시에는 자갈이 대부분 토양 표면에 노출됨으로 생육환경 및 기계작업 환경을 더욱 악화시키게 된다. 2cm이상의 자갈이 토양 표면에 있을 경우 기계 파종이나 이식이 어렵고 발아이후의 생육도 좋지 않게 된다. 그러나 지중에 존재하는 자갈은 토양의 투수성을 향상하여 경사지에서 토양침식을 막아주고, 수분과 온도의 보지력을 크게하고 토양 통기성을 향상시키므로 뿌리의 발육을 촉진해 준다.   • 표고 시설재배에서의 고품질 표고(동고, 화고)의 생산환경 분석을 위하여 차광재배와 시설재배사의 환경을 분석하여 각 시설내의 환경조건을 분석하였다. 차광재배사와 시설재배사의 기상환경 분석 결과, 시설재배의 습도는 차광재배에 비하여 20-40%높게 나타났으며, 70-80% 이상인 기간이 차광재배에 비하여 약2배 가량 높게 나타났다. 생육기간중 시설재배에서의 최고온도가 저온성 표고의 생육 온도보다 2-4$^{\circ}C$ 높아 생육에 악 영향 준 것으로 판단된다. 골목 하부로 갈수록 시설재배에서 4-5$^{\circ}C$ 높게 나타났다. 시설재배의 $CO_2$의 농도는 차광재배에 비하여 100-200ppm 정도 높게 나타났다. 이상의 결과로부터 시설내의 환기촉진과 과습방지가 시설내의 고품질 표고 생산을 위한 환경개선 방법이라고 판단된다.

Bioactivity of the Extract of Coptis chinensis: In-vitro Antifungal Activity against Phytophthora capsici and Growth-promotion Effect in Red-pepper (황련 추출물의 고추역병에 대한 In-vitro 항진균 활성 및 고추 생육촉진 효과)

  • To investigate anti-phytopathogenic fungal activity of Coptis chinensis, the methanol extract and its organic solvent fractions were prepared. Using the extract and the fractions, in-vitro spore-germination inhibition and mycelial-growth inhibition activities were evaluated against Colletotrichum gloeosporioides, Phytohpthora capsici, Pyricularia grisea, Rhizoctonia solani, Botryosphaeri dothidea, Glomerella cingulata, respectively. Treatment of the methanol extract (500 mg/mL) into the spore of phytopathogenic fungi completely inhibited germinations for 5 days, except B. dothidea, and showed strong antifungal activities against P. grisea and B. cinerea, and antioomycetes activity against P. capsici. The minimal growth inhibition concentrations of the methanol extract against P. grisea, B. cinerea and P. capsici were 300, 300, and 500 mg/mL, respectively. For practical application of C. chinensis in red-pepper field, the hot-water extract (1,000 mg/mL) was prepared in commercial facility, after evaluation of heat stability and solvent-extraction yields of antifungal substances. The 3-times leaf-spray of the extract from June to August, 2008 did not show any deleterious effect to red-pepper. In fact, the leaf-spray promoted plant growth including leaf, root and fruit. The average weight and rind of each fruit were increased to 119% and 117% comparison to those of without treatments. Our results suggest that C. chinensis is a useful source for control of red-pepper diseases and plant growth.

  • A promoting effect of ginseng saponin on the growth recovery rate of Saccharomyces rouxii which was treated by heat was confirmed in previous report(22). In order to deduce the promoting effect on the growth recovery of the heat stressed yeast, the effect of ginseng saponin on the activity and the heat stability of the amylase produced by Sacchirromyces rouxii were observed. The amylase showed the highst activity at 0.01% of saponin. At this concentration, the activity increased about 23% compared to the control. Furthermore, the ginseng saponin showed a protective effect against thermal inactivation of the amylase produced by Saccharomyces rouxii.

Development of Microbial Inoculant Using By-product of Oriental Herbal Medicine (한약재박을 이용한 미생물제제의 개발)

  • The development of microbial inoculant was conducted using a by-product of oriental herbal medicine. The constituent of the by-product, which was high in organic matter, was 11.3% of crude protein, 5.1% of crude lipid, 49.7% of NDF (neutral detergent fiber), and 33.8% of ADF (acid detergent fiber). Microorganisms isolated from the by-product of oriental herbal medicine were 35 species. Among them, 6 bacterial species, 4 fungal species, 2 actnomycetes species, and 1 yeast species were effective in the utilization of the by-products. The 13 strains screened were tested for the plant growth-promoting effect in soybean seedling. BL-333 strain was found to increase the soybean yield by about 23% as compared with control. The strain BL-333 was identified as Paenibacillus marcerans. P. marcerans BL-333 showed high anti-fungal activities against virulent fungi, especially Fusarium sp. and Collectotrichum sp. Yields of plants which were inoculated with microbial inoculant prepared with P. marcerans BL-333 and by-product of oriental herbal medicine were found to be higher than control by $3{\sim}24%$. The yield was especially promoted in lettuce, radish, chinese cabbage and cucumber plants.

  • Healthy seedling generation is the major concern in overcoming adverse effects of biotic and abiotic stresses during tender stage of development in vegetables and horticultural crops. Because of this, priority is given to research leading to the generation of healthy seedlings in crops subjected to transplanting and bedding. In this study, growth pouch experiments were conducted to determine the effect of inoculation of six different strains of Methylobacterium sp. namely, M. oryzae CBMB20, M. phyllosphaerae CBMB27, M. suomiense CBMB120, and Methylobacterium strains CBMB12, CBMB15 and CBMB17 on the seedling development of the vegetable crops cabbage, Chinese cabbage and cucumber; and horticultural crops tomato and red pepper. Crops treated with the test strains generally showed higher seedling dry matter accumulation compared to the control. Significantly higher accumulation was exhibited by CBMB12, CBMB17, and CBMB20 in cabbage, as well as for CBMB27 and CBMB120 on tomato and Chinese cabbage, respectively. Furthermore, all the strains promoted root elongation in cucumber and tomato seedlings while in Chinese cabbage and red pepper, root elongation was observed with CBMB120 and CBMB12 inoculation, respectively. Large scale nursery study is needed to develop a thorough protocol for healthy seedling development with the use of these strains.

Biological Control of Plant Pathogens by Bacillus sp. AB02. (Bacillus sp. AB02를 이용한 식물 병원균에 대한 생물검정)

  • In the greenhouse fields for fruits and vegetables during the winter in Korea, there are serious damages by the sclerotium diseases due to the low temperature and humidity. This study was carried out to select an antagonic agent for the biological control of the sclerotium diseases. The 55 antagonic agents were selected from the rhizosphere in soil where the fruits and vegetables were cultivated in the green house fields, and strain AB02 among the tested isolates was estimated to be the strongest antagonist against the sclerotium disease. Using strain AB02, the antifungal spectrum was tested against 5 different plant pathogens. According to the results of the test, strain AB02 . showed the high antagonistic effect against Botrytis cinerea and Sclerotinia sclerotiorum. For the experiment of biological control against the sclerotium disease, it was estimated the suppression effect and the control effect by the strain in the pot experiment using the green perilla. According to the result of the pot experiments, the suppression effect was 40% and the control effect was 62%, respectively. For the stimulation effect of the tested plant growth by strain AB02 compared to the control, it was improved as 120% for the total length, 141% for the liveweight, 121% for the total number of leaves, 185% for the leaf area, and 327% for the liveweight of the root, respectively. Strain AB02 showing the antagonistic effect against the sclerotium disease and the stimulation effect for the plant growth was identified as Bacillus sp.

Effects of Rooting Promoters and Light Intensity on Rooting and Root Growth of Rose Cuttings (장미의 삽목발근에 미치는 발근촉진제 및 광도의 영향)

  • This study was conducted to determine the effect of rooting promoters and light intensity on rooting and root growth of rose cuttings. The cuttings, both 'Noblesse' and 'Red Velvet', were rooted up to 100% with Rootone treatment, while less than 100% with other growth promoter treatments. In 'Red Velvet', percentage of rooting increased with increasing IAA, NAA, and IBA concentrations. In 'Noblesse', however, the rate increased up to $500mg{\cdot}L^{-1}$ in NAA and IBA, and $1000mg{\cdot}L^{-1}$ in IAA, and then decreased above the concentrations. In 'Noblesse', root number, length, and weight increased by Rootone treatment. In 'Red Velvet', root number, and length increased by IBA treatment, while root weight was highest in Rootone treatment. In both cultivars, rooting was accelerated and the rate increased under high light intensity and root growth increased also. Between both cultivars, propagation and root growth of 'Red Velvet' decreased dramatically with decreased light intensity.

  • Plug seedlings of bell pepper(Capsicum annuum L.) were grown for 50 days in controlled environment chambers under 12 hrs per day photoperiodic condition with sixteen different day and night temperature regimes to investigate the possibility of height control. The seedlings were then transplanted to greenhouse to investigate the growth, flowering, and yield afterward. Plant height and stem length of seedlings were mainly affected by day temperature rather than night temperature. Internode elongation was suppressed by a negative DIF and was enhanced by a positive DIF even with the same average daily temperature (ADT). Leaf unfolding rate was influenced more by ADT than by DIF. Fresh and dry weights increased as ADT increased. Leaf area and stem diameter increased until temperature increased up to 24$^{\circ}C$ day and night temperature and decreased above 24$^{\circ}C$, The position at which the first flower was initiated was lowered as ADT increased. The first flower degeneration was not obvious up to 24$^{\circ}C$ ADT but increased rapidly above 24$^{\circ}C$ ADT. Seedling compactness(Dry weight per plant height :mg.mm$^{-1}$ ) was greater under -DIF than +DIF condition. In conclusion, DIF treatment was an applicable technique to control stem elongation and growth rate such as leaf unfolding rate and position at which first flower was initiated could be controlled by ADT.

  • The growth extent of Leuconostoc mesenteroides and Lactobacillus plantarum in the medium which contain sterilized extract of each kimchi minor ingredient (green onion, garlic, ginger, raw red pepper, and red pepper powder) was examined. All minor ingredients decreased the growth of Lac. plantarum, and this effect of garlic is the most distinctive, ginger had the positive effect on the growth of Leu. mesenteroides, and garlic had the negative effect on the growth of Leu. mesenteroides. When the growth extent of two bacteria in the medium which contain sterilized successive extracts of each of garlic, ginger and red pepper powder was examined, the butanol fraction of garlic was reprsented the negative effect on the growth of Leu mesenteroides and Lac. plantarum, and the water fraction of ginger and red pepper powder were represented the positive effect on the growth of Leu. mesenteroides.
