  • 1. 서론 : 생물기술이 급속히 발전함에 따라 다양한 생물제품들(예, 단백질, 효소, 항생제 등)이 생산되고 있으나, 그 생산량은 극단적으로는 기질용액 1g당 10$^{-8}$g 정도로까지 낮아, 이를 고순도로 정제하기 위해서는 다단계의 공정이 필요하여 분리정제의 비용이 크다. 따라서 생물제품의 산업적 생산이 경제성을 갖기 위해서는 생물제품을 보다 경제적으로 분리정제할 수 있는 효율적인 공정개발이 필수적이다. (생략)

  • Effect of immobilization on the production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) by Nicotiana tabacum cells was investigated using polyurethane foam as immobilization matrices. The cell activity and the hGM-CSF production were maintained for 16 days in spite of 3 times of media exchange. Under the same conditions of temperature and agitation rate, maximum concentrations of hGM-CSF in a 500-mL spinner flask and 100-mL Erleuneyer flasks were 17.3 ${\mu}g/L$ and 9.8 ${\mu}g/L$, respectively. Consequently high hGM-CSF production could be possible in spinner flask when the rate and amount of media exchange were optimized.

  • The objective of this study was to estimate dynamics of litter production and soil organic carbon of Pinus densiflora forest, Quercus mongolica forest, and Rhobina pseudo-acasia forest at Mt. Nam as a part of Korea National Long-Term Ecological Research (KNLTER) from 2008 to 2009. Litter production of P. densiflora forest was the highest in October 2008, 2009 and the lowest in January 2008 and December 2009. Litter production of Q. mongolica forest was the highest in November and the lowest in February in 2008 and 2009. Litter production of R. pseudo-acacia forest was the highest in November in 2008 and October in 2009 and the lowest in January in 2008 and December in 2009. It means that leaves of P. densiflora forest shed earlier than deciduous oak forests in Korean central region. An average of litter production for 2 years was 7.07, 6.36, $4.66ton\;ha^{-1}$ in P. densiflora forest, Q. mongolica forest, R. psuedo-acacia forest, respectively. An average of soil organic carbon matter for 2 years was 88.3, 76.5, $84.2ton\;ha^{-1}$ in P. densiflora forest, Q. mongolica forest, R. psuedo-acacia forest, respectively.

Diversity, Production and Nutrients Concentration of Mushrooms in a Pitch Pine Forest (리기다소나무림의 버섯의 다양성, 생산량 및 영양염류 함량)

  • Diversity, production and nutrients concentrations of mushroom in a pitch pine forest (Pinus rigida) were studied from March through November 2000 in Kongju, Chungnam Province. Fifteen $2{\times}2m$ permanent quadrats were established randomly in the study area. 32 species of mushrooms were identified in the permanent quadrats during the study period. Although mushrooms occurred from June to October, the heighest diversity and biomass production occurred in August and September. Seasonal dominant species were Collybia confluens in July, Cantharellus lutescens in August and September, respectively. Annual production of mushrooms were 12.13kg $Dwha^{-1}yr^{-1}$. Seasonal production was 0.71kg $Dwha^{-1}$ for July, 3.95kg $Dwha^{-1}$ for August and 6.92 kg $Dwha^{-1}$ for September, respectively. Average concentrations of N, P, K, Ca and Mg in mushrooms was $38.81{\pm}4.82, \;0.90{\pm}0.44,\;28.47{\pm}7.81,\;0.24{\pm}0.16\;and\;0.76{\pm}0.26mg\;g^{-1}$, respectively, which were much higher than those in leaf litter and soil of the study site. Amount of N, P, K, Ca and Mg accumulated in mushrooms in 2000 was 463.41, 10.26, 345.21, 3.14 and $8.99g\;ha^{-1}yr^{-1}$, respectively.

  • 국내에서 바지락 양식은 경기도 연안의 간석지에서 1912년에 시작되었다고 보고되어 있으며, 1980년부터 본격적인 양식이 시작되어 1990년에 최고 생산량을 기록하였다. 그러나 최근에는 매립에 의한 서식장 감소, 양식장의 밀식에 따른 자가 오염 및 환경변화 등에 의한 바지락 폐사에 따라 생산량 감소가 야기되어 지속적인 바지락 양식장 유지관리의 중요성이 대두되고 있다. 곰소만 조간대 및 주변 해역에서의 기존연구는 지질학적 특성뿐 만 아니라, 생물학적 특성, 퇴적물의 물리적 특성, 그리고 해역의 수리역학적 특성을 포함한 포괄적인 연구에 관해서는 미흡한 실정이다.   • Erinacine which is produced from Hericium erinaceus mycelia is known as stimulating activity of nerve growth factor(NGF) synthesis. Thus. this work was concentrated on the maximum production of Hericium erinaceus mycelia. Media compositions were studied as the chemical conditions that affected cell growth. The modified media was ditermined from the results, including glucose 20g/L and yeast extract 10g/L.

Production Condition and Utilities of Extracellular Biopolymer from Bacillus licheniformis (Bacillus licheniformis가 생산하는 Extracellular Biopolymer의 생산조건 및 이용특성)

  • A bacterium that produce biopolymer was isolated from Gochujang, one of Korean traditional fermented foods, and identified as Bacillus licheniformis. The production of biopolymer was highest and 34mg/250ml, when the baterium was cultivated in condition of sucrose 6.0%, Yeast extract 0.1%, peptone 0.l%, NaCl 3.0%, and pH 6.0. The 1% solution of this biopolymer was able to form a translucent and glossy film. And the solution of biopolymer was found to precipitate Kaolin solution and also stabilize the suspension of lactic acid bacteria within the test range of 0.25-1.00%.   • Rhodospirillum rubrum KCTC 1372 produced hydrogen from glucose for first 48hrs culture under the anaerobic photosynthetic conditions, and after 48hrs culture the hydrogen production was decreased by the accumulation of producing organic acids in broth. Only 41% of glucose was consumed and 143mL/day/L hydrogen were produced after 96hrs culture. However the hydrogen production and glucose consumption were substantially increased when the pH of the culture broth were controlled to 6.8-7.2. After 96hrs culture, 450mL/day/L hydrogen were produced, and about 80% glucose was consumed. Specific hydrogen production rate was 48.33mL/hr/g cells under pH not controlled, but 45.42mL/hr/g cells under pH controlled.

  • The purpose of this work was to investigate the characteristics of a biosurfactant-producing bacterium isolated from crude-oil contaminated soils. During the incubation of strain HK-3 with 1% crude-oil, bacterial growth pattern, the amount of biosurfactant production, and pH changes were monitored. In order to examine the effect of supplemented carbons on the production of biosurfactant, cultivation of HK-3 cells in BH media with different carbons (e.g. glucose, dextrose, mannitol, citrate, or acetate) revealed that the production of biosurfactant reached the maximal level at the 72 h incubation with mannitol, which the area of clear zone was measured to approximately 7.64 $cm^2$. Identification test using the BIOLOG system, morphology study based on scanning electron microscopy and the 16S rRNA sequence-based phylogenetic analysis assigned strain HK-3 to a Pseudoalteromonas species, designated as Pseudoalteromonas sp. HK-3 which was registered in GenBank as [FJ477041].
