Column experiment for contaminated coastal sediment bioremediation using biostimulating agent (연안오염저질의 생물정화를 위한 생물활성촉진제의 효능 평가 : 현장 컬럼실험)

  • This study evaluated the effectiveness of biostimulating agent in contaminated coastal sediment. The study was conducted via column tests in coastal sea wherein two separate columns were employed for two different polymers used and another column for a blank. The biostimulating agent was made by mixing sea sediment with biostimulants viz acetate, nitrate, sulfate. The biostimulating agent was then rolled into balls, dried and coated with either Cellulose Acetate (CA) or Polysulfone (PS) to control the release of the biostimulants. The pH was around 7.6~8 for 4 months while COD, TP and TN were significantly lower in the column containing biostimulating agents. Heavy metal(Fe, Zn, Cd, Cr, Pb, Cu) was converted to stable forms and PS coated biostimulating agent had a high efficiency of heavy metals distribution.

Effect of the Applied Biostimulant Depth on the Bioremediation of Contaminated Coastal Sediment (연안오염퇴적물에 주입한 생물활성촉진제의 깊이가 생물정화효율에 미치는 영향)

  • This study investigated the optimum depth for the application of bioremediation in contaminated coastal sediment using a lab scale column experiment. The biostimulants were placed in the top surface of the sediment facing seawater, 3cm, 6cm and 10cm of the depth from the surface, respectibely. During the experiment, the changes of organic pollutants and heavy metal fractions in the sediment were monitored in 1 month and 3month time intervals. The organic pollutants found during various analysis such as chemical oxygen demand, total solids and volatile solids, significantly reduced when the depth of the biostimulant was 3cm or less. In contrast, at a depth of over 6cm, the reduction of organic pollutants decreased, and the results were similar to the control. Heavy metals fractions in the sediment also changed with the depth of the biostimulants. The exchangeable fraction of the metals was quite reduced at the sediment surface in the column, but the organic bound and residual fractions considerably increased at a depth of 3cm. Based on this study, the optimum biostimulants depth for in-situ bioremediation of contaminant coastal sediment is 3cm from the sediment surface.

Study on the elution of biostimulant for in-situ bioremediation of contaminated coastal sediment (오염된 연안저질의 현장생물정화를 위한 미생물활성촉진제의 용출특성 연구)

  • A study on the elution characteristics of biostimulating agents (sulfate and nitrate) from biostimulants which are used for in-situ bioremediation for the coastal sediment contaminated with organic matter was performed. The biostimulating agents were mixed with the coastal sediment, and then massed the mixture into ball. Two kinds of ball type biostimulant were prepared by coating the ball surface with two different polymers, cellulose acetate and polysulfone. A granular type biostimulant (GTB) was also prepared by impregnating a granular activated carbon in the biostimulating agent solution. The image of scanning electron microscopy for the biostimulant coated with cellulose acetate (CAB) showed that the inner side of the coating layer consisted of irregular and bigger size of pores, and the surface layer had tight structure like beehive. For the biostimulant coated with polyfulfone (PSB), the whole coating layer had a fine structure without pore. The elution rate of the biostimulating agents for the CAB was higher than that for the PSB, and the elution rate for the GTB was considerably higher than that for the PSB in distilled water as well as in sea water. The elution rate of the biostimulating agents in turbulent water flow was about 3 times higher than that in standing water, and the elution rate of nitrate was higher than that of sulfate from the stimulating agents.

Polymer coating for controlled release of biostimulants from Biostimulant balls (생물활성촉진제의 용출율 제어를 위한 폴리머 코팅)

  • We prepared biostimulant balls using sea sediment mixed with biostimulants viz acetate, nitrate and sulfate. The Biostimulant balls were coated with Cellulose Acetate (CA) and Polysulfone (PS) to control the release of the biostimulants. SEM images showed that CA coating was porous and irregular in the inside and very uniform and tight like beehive while PS coating was the same in the inside and outside and not porous. Biostimulants release was found to be high in sea water compared to distilled water. The release of nitrate was higher compared to sulfate. In turbulent environment the release of bionutrients was 50% higher than static environment.

Effect of surfactants and Environmental Factors on the Bioremediation of Phenanthrene (토양내 Phenanthrene의 생물학적분해에 미치는 계면활성제와 환경변수의 영향)

  • The biodegradation of ($C^{14}$)phenanthrene was studied in water and soil-water systems with nonionic surfactants and biosurfactant : polyoxyethylene alkyl ester($C_{17}$$H_33$COO($C_2$$H_4$O)nH) and sophorolipid. The extents of so1ubilization and biodegradation were monitored by radiotracer technique. Experimental results showed that surfactant concentrations above the critical micelle concentration were toxic to the phenanthrene-degrading bacteria in soil or active sludge and the presence of surfactant micelles inhibited mineralization of PAHs. Solubility and bioavailibility of phenanthrene in water and soil-water system were enhanced by mixed surfactants system. The optimum water content and hydrogen concentration were 30% (w/v) , pH 7, respectively.

A Pilot Scale Biostimulation Study for In-situ Remediation of Organic Pollutants and PAHs contained in Coastal Sediment (연안 해역퇴적물에 함유된 유기오염물과 PAHs의 현장정화를 위한 파일럿 규모의 생물활성촉진연구)

  • A pilot scale biostimulation experiment was performed under field conditions to degrade organic pollutants and PAHs (Polycyclic Aromatic Compounds) in contaminated coastal sediment. During the experiment, the seawater temperature around the sediment varied from $16.5^{\circ}C$ to $21^{\circ}C$, while the sediment pH was stable at 8.4-8.5. The experiment was conducted over a one year period with a control group and a sample group with a applied biostimulant composed of acetate, sulfate, and nitrate. Chemical oxygen demand decreased 39% in the control group and 79% in the sample group. Volatile solids were reduced from 15 to 7 g/kg in the control group and 2.5 g/kg in the sample group. Out of the 2-, 3- ,4- ,5-, and 6-rings of the 16 vital PAHs, the compound including naphthalene (2-ring), took 2 months to degrade completely, while the degradation efficiency was 55.6% in the control group during the same period. In the case of the 3-ring and 4-ring PAHs, complete degradation in the sample and 46% - 100% degradation in control was observed after one year of the experiment. The 5-ring and 6-ring PAHs were degraded to about 77%-100% in the sample group and 26%-87% in the control group during the one year of the experiment. The study results show that biostimulation is a very effective method to improve the degradation rate of organic pollutants and PAHs (Polycyclic Aromatic Compounds) in contaminated coastal sediment.

  • LHRH분비와 환denylate cyclase활성에 미치는 PGE2, Opioid, 그리고 Ca2+의 영향을 흰쥐의 시상하부 조직을 사용하여 조사하였다. K+(30mM)에 의한 LHR광분비촉진은 Ca2+의존적인데 반하여, PGE2에 의해 촉진되는 LHR노 분비는 세포의 Ca2+농도에 의존하지 않았다. PGE2에 의한 LHR기 분비와 CAMP합성은 PGE2농도(1$\times$10-7M-1$\times$10-4M)에 비례하여 촉진되었으며, $\beta$-endorphin (1x10-3M)은 PGEa에 의한 LHRH 분비촉진과 CAMP합성을 공히 억제하였다. 오피오이드 수용체의 길항제인 Naloxone(Ix10-"M)은 $\beta$-endorphin에 의한 저해효과를 극복시켜서, LHR광 분비와 CAMP합성은 각각 회복되었으나, CAMP합성은 부분적인 회복을 보인 반면에, LHRH분비는 PGE2에 의한 촉진효과보다도 더 활성화되었다. 결론적으로 LHRH분비에 미치는 오퍼오이드의 억제작용은 PGE2-cAMP의 세포내 전달과정을 저해함으로써 유발되는것으로 추정 된다.정 된다.

  • Cytotoxicity and antibacterial activity of preservatives were examined. Fibroblast cell L929 was used for cytotoxicity experiment and Pseudomonas aeruginosa A TCC27853, Staphylococcus aureus ATCC25923. Trichoderma reesei ATCC6967 were used for antibacteria and antifungi. Benzalkoniumchloride(BAK) as synthetic preservative and chitosan as natural preservative were used. Minimum inhibitary concentration (MIC) of BAK was 0.1 % for P. aeruginsa and 0.001% for S. aureus and 0.1 % for T reesei MIC of chitosan was 2% for P. aeruginosa and 1 % for S. aureus.

Use of Activated Soil to Bioaugment Degradation of Atrazine in Soils (토양 내의 Atrazine의 생물학적 분해 촉진을 위한 활성토의 이용)

  • Effectiveness of activated soil containing directly enriched atrazine-degrading soil microorganisms as an inoculant to bioaugment degradation of atrazine in soils was investigated. A Wooster silt loam (Typic Fragiudalf) was spiked with atrazine at a rate of 4 mg/kg soil three successive times to create activated soil. Atrazine degradation was significantly enhanced (p < 0.05) after the first treatment. After the second treatment, there was an increase in the number, based on MPN, of microorganisms utilizing atrazine as a C- and N-source by 3 logs and 1 log of magnitude, respectively. Inoculation of typical agricultural soils collected from Ohio with activated soil at a rate as low as 0.5% reduced the extractable atrazine remaining in soils to the level below 2% of that initially recovered (initially added at a rate of 4 mg/kg soil) after 4 days. Inoculation at a higher rate was required to achieve the same result in soils with non-typical properties (pH of 4.5 or organic matter of 43% w/w). Activated soil was stable, in terms of atrazine degradation activity, at least up to 6 months when it was kept at low temperature (< $10^{\circ}C$) and moistened (water content above 15%). The results of this study indicate that microorganisms capable of degrading atrazine are relatively easily enriched in soil to create activated soil. Use of activated soil can be a practical option for bioremediation of contaminated soils.

Characterization of Chinese Cabbage Phospholipase D by a Multistirring Batch System Bioreactor (다중 교반형 생물반응기에 의한 배추 Phospholipase D의 특성연구)

  • Phospholipase D catalyzes the phosphatidohydrolysis and transphosphatidylation of phospholipid in the biological systems. In this study we were partially purified phospholipase D from Chinese cabbage and the characterization of the enzyme was carried out in a multistirring batch system bioreactor. The enzyme showed optimum activity at pH ,5.6, highest activity at 37$^{\circ}C$ and Ca2+ is important for the enzyme activity. Optimum concentrations of Ca2+ for phosphatidohydrolysis was 20 mM and for transphosphatidylation was 40 mM, respectively. Some organic solvents such as diethylether, isopropylether and butylacetate were activated the enzyme activity. On the other hand, EDTA, Ba2+, Mn2+ and Zn2+ showed inhibitory effect on the enzyme activity. The base acceptors in transphosphatidylation by the Chinese cabbage phospholipase D were tested. Various poly-and monohydroxy alcohols were found to be active.
