Characteristics of Culture Conditions for the Production of Biosurfactant by Bacillus pumilus IJ-1 (Bacillus pumilus IJ-1의 생물계면활성제 생산을 위한 배양 특성)

  • To characterize the culture medium for the biosurfactant production by Bacillus pumilus IJ-1, the influences of various carbon, nitrogen and mineral sources were assessed. As a result, the highest biosurfactant production was observed after 96 h cultivation containing 0.5% (w/v) tryptone. The strain was able to grow and produce biosurfactant at 0-10% (w/v) NaCl, in the pH range of 5-10, and at $20-45^{\circ}C$. Optimal culture conditions for the biosurfactant production were at $20^{\circ}C$ and pH 9.0 after 72 h incubation and the surface tension of biosurfactant was 27.0 dyne/cm.

  • Optimal culture conditions were characterized for production of crude biosurfactant of Bacillus subtilis JK-1. During incubation of B. subtilis JK-1, the bacterial growth pattern, changes of the surface tension at variable temperatures, pH and NaCl concentrations in bacterial culture medium were studied. The strain was able to grow and produce biosurfactant at $15-45^{\circ}C$, in the pH range of 6-10, and at 0-10% (w/v) NaCl. In case, culture broth pH was gradually changed to neutral or weak alkaline. Optimal culture conditions for crude biosurfactant production were at $35^{\circ}C$ and pH 7.0 after 48 h incubation and the surface tension of biosurfactant was 24.0 mN/m. Besides, as the concentration of NaCl was increased from 0 to 10% (w/v), the growth was decreased, pH of the culture broth was converted from weak alkaline to acidic, and the surface tension rised.

Removal Characteristics of Tetracycline, Oxytetracycline, Trimethoprime and Caffeine in Biological Activated Carbon Process (생물활성탄 공정에서 Tetracycline, Oxytetracycline, Trimethoprime 및 Caffeine 제거특성)

  • In this study, The effects of three different activated carbon materials (each coal, coconut and wood based activated carbons), empty bed contact time (EBCT) and water temperature on the removal of pharmaceutical 4 species (oxytetracycline, tetracycline, trimethoprime and caffeine) in BAC filters were investigated. Experiments were conducted at three water temperature (5, 15 and $25^{\circ}C$) and four EBCTs (5, 10, 15 and 20 min). The results indicated that coal based BAC retained more attached bacterial biomass on the surface of the activated carbon than the other BAC, increasing EBCT or increasing water temperature increased the pharmaceutical 4 species removal in BAC columns. In the coal-based BAC columns, removal efficiencies of oxytetracycline and tetracycline were 87~100% and removal efficiencies of trimethoprime and caffeine were 72~99% for EBCT 5~20 min at $25^{\circ}C$. The kinetic analysis suggested a firstorder reaction model for pharmaceutical 4 species removal at various water temperatures (5~$25^{\circ}C$). The pseudo-first-order reaction rate constants and half-lives were also calculated for pharmaceutical 4 species removal at 5~$25^{\circ}C$. The reaction rate and half-lives of pharmaceutical 4 species ranging from 0.0360~0.3954 $min^{-1}$ and 1.75 to 19.25 min various water temperatures and EBCTs, could be used to assist water utilities in designing and operating BAC filters.

Antioxidant and Antimicrobial Activities of The Extracts from Native Camellia japonica in Korea (국내 자생 동백나무(Camellia japonica L.) 추출물의 항산화 및 항미생물 활성)

  • This research has been undertaken to increase availiability of native Camellia japonica leaf and flower in Korea as a edible-medicinal resource. Chemical compositions, antioxidant and antimicrobial activities in different parts of camellia were investigated. Crude protein contained the highest in young leaves(14.22%) but less than 10% in different parts. The contents of crude fat and crude ash were 60.48% in seeds and 5.16% in mature leaves, respectively. Methanol extract of young leaf, flower, and flower bud in camellia showed strong antioxidant activity compared with different parts. Also, antioxidant activity of these was higher than that of BHT, but weaker than that of VtC. Antioxidant activity of various parts in camellia were in order of young leaf > flower bud > flower > mature leaf > stem > bark. Extracts of mature leaf and bark had a remarkable antibacterial activity(0 CFU/$m\ell$) on Bacillus subtilis. The young leaf extract displayed effective growth inhibition against B. subtilis, Candida albicans, and Trichosporon beigelii.

Synthesis of Carboxylate-Based Anionic surfactant from Coconut Oil Source and Characterization of Interfacial Properties (코코넛 오일로부터 유래된 카르복실레이트계 음이온 계면활성제의 합성 및 계면 특성에 관한 연구)

  • In this study, a carboxylate-based anionic surfactant SLEC-3 was prepared from coconut oil and the structure was elucidated by using FT-IR, 1H-NMR and 13C-NMR analysis. Measurements of interfacial properties such as critical micelle concentration, static and dynamic surface tensions, emulsification index, and foam stability have shown that SLEC-3 is better in terms of interfacial activity and more effective in lowering interfacial free energy than those of SLES, which has been widely used as a conventional anionic surfactant in the detergent industry. Biodegradability, acute oral toxicity and dermal irritation tests also revealed that SLEC-3 surfactant possesses excellent mildness and low toxicity, indicating the potential applicability in detergents and cleaner products formulation.

  • 생물학적 처리공정내 미생물은 물리화학적, 생태학적으로 처리공정의 운전조건과 유입하·폐수 특성에 따라 존재량 및 반응조내 존재하는 우점종이 달라진다. 특히,폐수의 생물학적 처리시 유입수내 존재하는 독성 및 저해물질은 생물반응조내 존재하는 미생물의 양을 감소시키거나 미생물의 종류를 단순화시키거나 환경조건 및 폐수 특성변화에 능동적으로 대처할 수 있는 능력을 감소시킨다. 따라서 이러한 문제점을 극복하기 위한 방안으로 생물능력향상(bio-augmentation)이 필요하며, 여러 가지 방법으로 생물능력향상을 이룰 수 있다. 본 연구에서는 생물능력향상을 위하여 기존의 효소활성이 강한 미생물군을 투입함으로써 처리효율향상에 미치는 영향 및 제한성을 고찰하였다. 처리효율 향상을 위해서는 생존성, 폐수에 대한 순응기간 및 미생물의 체류가 매우 중요하였으며, 특히 누입된 미생물군이 처리효율을 향상시키기 위해서는 일정기간 이상의 순응기간이 필요하였다. 한편, 토착미생물군인 UB-1의 경우 기존 생물능력향상 미생물군과 거의 동일한 처리효율을 나타내고 있으며, 사전에 순양기간을 거친 UB-2의 경우도 기존 생물능력향상 미생물군과 비슷한 처리효율을 보였다. 따라서 본 결과로 볼 때 생물능력향상 미생물군 투입에 의한 처리효율의 향상은 크지 않으며, 하·폐수의 생물학적 처리공정에 적요하는데는 다소 제한성이 있은 것으로 판단되었다.

Flocculating Properties of Bioflocculant Biopol32 from Pseudomonas sp. GP32 (Pseudomonas sp. GP32가 생산하는 생물고분자응집제 Biopol32의 응집특성)

  • The flocculating properties of bioflocculant Biopol32 produced by Pseudomonas sp. GP32 were investigated for application in industrial wastewater treatment. The major flocculating substance of bioflocculant Biopol32 was identified as polysaccharide. Many anionic flocculants need a counter ion to induce higher flocculating activity. The flocculating activity of bioflocculant Biopol32 was markedly increased by the addition of cationic ions ($Ca^{2+}$, $Al^{3+}$). The flocculating activity of bioflocculant Biopol32 was the most effective when 7.0 mM $CaCl_2{\cdot}2H_2O$ as coflocculant was added. The flocculating activity on the effect of pH and the temperature of the bioflocculant Biopol32 was compared with anionic commercial flocculant (polyacrylamide) and bioflocculant (zooglan from Zoogloea ramigera). In kaolin suspension, the highest flocculating activity was obtained at the bioflocculant Biopol32 concentration of 1.5 mg/l. A high flocculating activity was observed in the pH range of 5.0 to 8.0. The flocculating activity of bioflocculant Biopol32 was sustained up to $60^{\circ}C$, but decreased rapidly at over $70^{\circ}C$. In the batch culture, the charge density of bioflocculant Biopol32 was compared with flocculating activity. The larger the anionic charge density and apparent viscosity of bioflocculant Biopol32, the higher the flocculating activity. Therefore, we confirmed that the flocculating activity and apparent viscosity of bioflocculant Biopol32 was closely related to the charge density of bioflocculant Biopol32.

Antioxidant Properties of 7 Domestic Essential Oils and Identification of Physiologically Active Components of Essential Oils against Candida albicans (식물정유 7종의 항산화능 분석 및 Candida albicans 생장 억제 정유의 생리활성 성분 구명)

  • In this study, we selected two essential oils, Citrus unshiu and Cinnamomum cassia with superior antioxidant effects from the essential oils of 7 wild plants in South Korea and examined their antimicrobial activity against Candida albicans, which causes dermatitis to identify the antimicrobial components in the essential oils. As a result of measuring DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, SC50 value of the Citrus unshiu essential oil was 0.010 mg/mL, while for the Cinnamomum cassia essential oil, SC50 value was 0.09 mg/mL. In addition, when ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity was measured, SC50 value of the Citrus unshiu essential oil was 0.09 mg/mL, while for the Cinnamomum cassia essential oil, it was 0.06 mg/mL, exhibiting high antioxidant activity. For the minimum inhibitory concentration (MIC), the essential oil of Cinnamomum cassia was 1.25 mg/mL and that of Citrus unshiu was 5 mg/mL, demonstrating a high antimicrobial activity of the Cinnamomum cassia essential oil. Through the thin layer chromatography (TLC) bioassay, we assessed the antimicrobial activity against C. albicans according to the fraction components of the two essential oils. Also, by using preparative TLC (prep. TLC), we obtained the active fractions, and by performing GC/MS analysis of the components with the same Rf value, we identified the antimicrobial-active components. As a result, the main components having antioxidant and antimicrobial activities were cinnamyl acetate, eucalyptol, linalool, and citral of the Cinnamomum cassia essential oil and linalool from the Citrus unshiu essential oil. Also, based on the analysis of the fractional components that showed antioxidant and antimicrobial activities in both of the two essential oils, it was found that linalool has antioxidant activity, while cinnamyl acetate, eucalyptol, citral, and geranyl acetate have antioxidant and antimicrobial activities.

  • For the screening of anti-inflammation and antioxidative activities, ethanolic extract of 40 species of traditional herbal medicines were examined in vitro their hyaluronidase inhibitory effect and radical scavenging activity. Astragali Radix, Eucommia Cortex, Schizandrae Fructus, Scutellaria Radix and Moutan Radicis Coritex showed more than 50% hyaluronidase inhibitory effects. Moutan Radicis Cortex, Paeoniae Radix Alba, Plantaginis Semen and Sorbus commixta Hedl. showed more than 90% in electron donating activity. The scavenging effects of ethanolic extracts on hydroxyl radical showed significantly activity.

오미자(Schizandra Chinesis Baillon)추출물의 분획별 항상화활성 효과

  • To search for antioxidant compounds from plant resources, ethanol extracts of 10 plants species were investigated using DPPH method and SOD activity by NBT method. The highest activity was shown in the ethanol extracts of Schizandra Chinesis Baillon. And than, antioxidant effects and total polyphenol concentration were investigated in solvent fraction of ethanol extracts from Schizandra Chinesis Baillon. The DPPH inhibitory activity and SOD activity of ethyl acetate fraction (DPPH 51.6%, SOD 66.2%) and butanol fraction(DPPH 60.7%, SOD 67.4%) showed strong biological activities.
