A Study on Energy Saving Effect from Automatic Control of Air Flowrate and Estimation of Optimal DO Concentration in Oxic Reactor of Wastewater Treatment Plant (하수처리장의 포기조 최적 DO 농도 산정 및 공기송풍량 자동제어를 통한 에너지 절감 효과 도출)

  • It is important to keep stable effluent water quality and minimize operation cost in biological wastewater treatment plant. However, the optimal operation is difficult because of the change of influent flow rate and concentrations, the nonlinear dynamics of microbiology growth rate and other environmental factors. Therefore, many wastewater treatment plants are operated for much more redundant oxygen or chemical dosing than the necessary. In this study, the optimal control scheme for dissolved oxygen (DO) is suggested to prevent over-aeration and the reduction of the electric cost in plant operation while maintaining the dissolved oxygen (DO) concentration for the metabolism of microorganisms in oxic reactor. For optimal control, The oxygen uptake rate (OUR) is realtime measured for the identification of influent characterization and the identification of microorganisms' oxygen requirement in oxic reactor. Optimal DO seT-Point needed for the microorganism is suggested based on real time measurement of oxygen uptake of microorganism and the control of air blower. Therefore, both stable effluent quality and minimization of electric cost are satisfied with a suggested optimal setpoint decision system by providing the necessary oxygen supply requirement to the microorganisms coping with the variations of influent loading.

Microbial Activity Analysis for the Selectively Sterilizing of Government-controlled Bulk Public Archives (대량 공공 기록물의 선별적 소독을 위한 미생물 활성도 분석 연구)

  • An archive is a collection of documents or records. Currently, most archived documents are made of paper. Paper is susceptible to biological damage and deterioration due to its material properties. To control the biological damage, treatment with chemical disinfectants and control of the storage environment are often used. In government-controlled bulk public archives, all documents are chemically sterilized before storage. However, an extremely large quantity of public records have been produced, and storage space and conservation management are gradually reaching their limits. In this study, 60 species of microbes were identified using a genetic method. We successfully applied the adenosine triphosphate (ATP) bioluminescence method to detect microbial contamination on paper documents. A calibration curve of the ATP bioluminescence as a function of the microbe quantity was obtained, and the microbial activity on non-sterilized paper archives from 1951 was analyzed using an ATP luminometer. It was found that the microbial activity was suppressed or reduced in climate-controlled storage environments at $22^{\circ}C$ and 55% relative humidity. We anticipate that these results will be used to establish selective sterilization systems for government-controlled bulk public archives.

  • Droplet based microfluidic systems have been developed for the application of biological and chemical research field. A picoliter droplet in microfluidic device provides a compartmentalized and well-defined reactor in miniaturized system. The microfluidic system with small droplets can reduce reagent cost and enhance efficiency through automated high-throughput screening system. In this review, we summarize the functionality of droplet based microfluidic system including droplet generation, precise droplet control, and various applications. In addition, this article reviews current applications in chemistry and biology, and discuss advantages of droplet based microfluidics compared with conventional manner.

  • 육제품 제조용 starter culture에 대한 연구는 기초과학을 바탕으로 관련 미생물의 생리대사 특성은 물론 이들의 환경적응성이나 상업적인 생산공정개발에 이르기까지 상당한 발전을 거듭해 왔다. 특히, 최근에 와서 유전공학의 응용으로 starter균주의 대사능력을 조절할 수 있고 생물학적 안전성을 쉽게 검증할수 있을 뿐만 아니라 원하는 활력을 안전하게 보존할 수 있도록 하는 연구도 진행되고 있다. 미생물의 생육환경인 육의 발효과정에 대한 연구를 통하여 육제품 starter culture개발의 근간이 되는 지식을 얻게 되었으며 어떤 요인이 발효과정중 starter 균주의 대사능력과 성장에 영향을 주는지 또한 발효과정중 starter culture간의 상호작용 및 starter와 윈래 혼합육에 존재하는 균들사이에서 일어나는 상호작용에 관한연구, 나아가서는 Bacteriocine과 Bacteriophage가 육제품제조에서 어떤 의미를 갖고 있는지 등에 관한 연구도 활발히 이루어지고 있다. 특히 이런 연구결과들을 통하여 이들 starter의 실제 상업적인 이용에 있어 원료육이나 부재료, 향신료의 선정은 물론 제조 공정을 제어하여 제품의 안전성을 기대할 수 있을 뿐만아니라 하나의 starter culture로 여러 종류의 발효sausage를 제조하여도 제품마다 최상의 능력을 발휘할 수 있는 새로운 균주의 개발에도 기여할 수 있다. 스타타컬쳐에 대한 연구상황은 균주별로 다르나 유산균에 대하여 가장 많이 연구되어 있고 Micrococci에 대한 연구도 상당히 진전되어 있다. 특히할만한 점은 아시아의 발효생선이 육제품 발효에 이용되는 Staphylococcus carnosus종의 habitat로 밝혀진 것이다. 뿐만 아니라 곰팡이 균주도 실제 praxis에 적합하게 개발시킬수 있다. 따라서 앞으로 발효육제품제조에 있어 starter culture가 갖는 의미는 매우 중요하며 특히 짧은 숙성기간을 거치는 발효소시지의 제조에 있어서는 필수불가결한 공정의 한 분야로 자리잡게 될 것이다.

Sensory Motor Coordination System for Robotic Grasping (로봇 손의 힘 조절을 위한 생물학적 감각-운동 협응)

  • In this paper, human motor behaving model based sensory motor coordination(SMC) algorithm is implemented on robotic grasping task. Compare to conventional SMC models which connect sensor to motor directly, the proposed method used biologically inspired human behaving system in conjunction with SMC algorithm for fast grasping force control of robot arm. To characterize various grasping objects, pressure sensors on hand gripper were used. Measured sensory data are simultaneously transferred to perceptual mechanism(PM) and long term memory(LTM), and then the sensory information is forwarded to the fastest channel among several information-processing flows in human motor system. In this model, two motor learning routes are proposed. One of the route uses PM and the other uses short term memory(STM) and LTM structure. Through motor learning procedure, successful information is transferred from STM to LTM. Also, LTM data are used for next moor plan as reference information. STM is designed to single layered perception neural network to generate fast motor plan and receive required data which comes from LTM. Experimental results showed that proposed method can control of the grasping force adaptable to various shapes and types of greasing objects, and also it showed quicker grasping-behavior lumining time compare to simple feedback system.

  • The interaction between artificial fish and aquatic surroundings and the fish's realistic locomotion are very important elements to construct virtual aquariums. In general, the artificial fish in virtual aquariums used to be created by 3D modeling tools, and was repeatedly showing the simple and constant form of swimming. This paper will analyze the sorts of biological forms of fish-swimming and the propelling and turning characteristics. Then, we propose a method of the basic swimming and turning of artificial fish to generate various and natural-looking locomotion. It is possible to make a explorable virtual aquarium more immersive by using interactive interfaces together.

  • A prototype of expert system which can support the operation for the municipal waste water treatment plant located at Kyoung-Ki Do, Kwang-ju Kun was developed and tested. This system provides (i) tracking the cause of the problem, (ii) analysis, and (iii) solution Knowledge-base consists of about 100 production-rules for the biological wastewater process, such as bio-reactor and final clarifier. Rules were obtained from the analysis of the problems such as sludge bulking. The system provides stable process control and management and effectively helps inexperienced operators with advanced and standard technologies. Future works will focus to develop a statistical process control model and associate with expert system. The control model can process the operation data statistically; analyze the relationship between affecting factors and control variables; and provide optimum operation parameters.

Isolation of Bacterial Strains Inhibiting the Growth of Microcystis aeruginosa and Cyanobacterium Growth Inhibition Assay (녹조 원인 남세균 Microcystis aeruginosa의 생장을 억제하는 세균균주의 분리 및 남세균 생장 억제능 검정)

  • The objective of this study was to isolate algae growth inhibiting microorganism to biologically control Microcystis aeruginosa, which is a harmful cyanobacterium. Various bacterial strains were isolated in this study, and four bacterial strains of M1~M4 exhibited remarkable growth inhibiting activity against M. aeruginosa. Based on the 16S rRNA analysis, the isolated M1~M4 strains were identified, and isolated four strains were rod-type and gram-negative. In particular, as well as respective single strain, co-culture of the isolated M1~M4 strains showed obvious algicidal activity against M. aeruginosa. When mixed four strains were inoculated, about 50% of the chlorophyll a was reduced after two days, about 70% after four days, and about 80% after seven days. From these results mentioned above, the four bacterial strains may contribute to the control of harmful M. aeruginosa.

Preparation of Monodisperse PEGDA Microparticles Using a Dispensing Needle Based Microfluidic Device (주사기 바늘 기반의 미세유체 장치를 이용한 단분산성 PEGDA 입자의 제조)

  • This study presents a novel method for preparing monodisperse polyethylene glycol diacrylate (PEGDA) microparticles in a dispensing needle based microfluidic device. The microfluidic devices are manufactured by manually assembling various off-the-shelf products without using additional equipment. In this microfluidic device, the volumetric flow rates of the dispersed phase of PEGDA solution and the continuous phase of oil are controlled to generate monodisperse PEGDA droplets. The PEGDA droplet contains photo-initiator thus it is crosslinked to microparticle by photopolymerization at the ends of the device. The particle size is easily controlled by adjusting the volume flow rate and the size of the microfluidic device. The monodispersity of the particles is calculated by a coefficient of variation of 2.57%. To demonstrate the biological applications of PEGDA particles, cells are encapsulated and observed for proliferation and viability.

Grazing Effects of Freshwater Bivalve Unio douglasiae of the North Han River on the Cyanobacterial Bloom Waters (북한강 수계에 분포하는 말조개의 남조류 섭식특성)

  • A freshwater bivalve (Unio douglasiae) was examined to assess the filtering rate (FR) on the cyanobacterial assemblage in a hypertrophic lake. Animal U. douglasiae used in the present study was collected using a hand-operated dredge from the North Han River (Gapyeong, Korea). The FR was measured at different feeding conditions such as feeding interval (1, 4, 7, and 24 h), mussel size (4.2$\sim$8.1 cm, n=23), prey concentration (506.7, 409.8, 327.5, 199.7 and 88.6 ${\mu}g\;L^{-1}$), and mussel density (0.5, 1.0 and 1.5 indiv. $L^{-1}$). On the applied feeding interval, the maximum FR (0.21 L $g^{-1}h^{-1}$) and minimum feces production (FP, 0.12 mg $g^{-1}h^{-1}$) were observed at 1 and 24 hr, respectively. Both weight-based FR and FP were not correlated with the mussel size, and the values lied in a limited range with some degree of variation. Likewise, no significant relations between FR and FP were observed in the mussel size. The FR values were negatively correlated with food concentration, but positively with FP. For the food concentrations, the maximum FR (0.41 L $g^{-1}h^{-1}$) and FP (0.16 mg $g^{-1}h^{-1}$) were 88.6 ${\mu}gL^{-1}$ and 327.5 ${\mu}gL^{-1}$, respectively. These results indicate that U. douglasiae collected from the North Han River, although the filtering rate were slightly less than Keum River mussel, may be applied as a strategic bio-filter to mitigate cyanobacterial bloom in eutrophic lake.
