RAPD Analysis and Cordycepin Concentration of Hybrided Cordyceps militaris Strains by Mating (교잡된 Cordyceps militalis 균주의 RAPD 분석 및 생리활성물질인 cordycepin 함량 측정)

  • The C. militaris belongs to entomopathogenic fungi, which have their specific sequences in internal transcribed spacer regions (ITS1 and ITS2) depending on species. In this study, to identify the phylogenetic relationship of the strain hybrided by mating of C. militaris, we compared DNA sequences of ITS regions and 5.8S ribosomal DNA (rDNA) repeat unit of hybrid strain and its parental strains. The result revealed that hybrid strains are C. militaris species. In addition, cordycepins produced by hybrid strains and other strains of C. militaris were analyzed by HPLC with 20mM $KH_2PO_4$ of mobile phase and C-18 columns. The result indicated that the strain hybrided by mating produce higher concentration of phytochemical cordycepin than other C. militaris strains.

Analysis of High School Science Textual Descriptions of Scientifically Debatable Compounds According to the Experimental Results by MBL: A Case Study of Carbonic Acid in Water and Aqueous Solution of Carbon Dioxide (MBL 실험 결과를 토대로 한 과학적으로 논의 되고 있는 화합물의 고등학교 과학 교과서 기술 분석: 이산화탄소 수용액과 탄산 수용액의 경우)

  • The purposes of this study are to investigate the description of scientifically debatable carbonic acid in the Korean high school textbooks, characterize the physical properties of 'carbonic acid solutions' by using an MBL set-up and compare the properties with textual ones. Four different aqueous solutions of carbon dioxide have been prepared and analyzed: naturally aerated aqueous solution, dry ice-dissolving solution, $CO_2$-bubbling solution and commercial carbonic acid water. Experimental findings showed that pH and conductivity of these 4 solutions ranged from 3.85 to 5.66 and from 0.21 ${\mu}S$/cm to 272.1 ${\mu}S$/cm, respectively. Out of these solutions, the dissociation constant($K_{a1}$) of the bubbling solution at room temperature could be calculated to $5.7{\times}10^{-7}$ which value is comparable to the textual $4.3{\times}10^{-7}$ within experimental errors, which means that textual compound is not pure carbonic acid but the equilibrated mixture of carbonic acid and the aqueous solution of carbon dioxide. On the other hand, textual analysis showed that most of high school textbooks used carbonic acid as an example of weak acid and buffer solution of the blood but none of them distinguished the carbonic acid from the aqueous solution of carbon dioxide. Only one textbook, however, tiered two species in the chemical equation.

Development of Microsatellite Markers for Discriminating Native Korean and Imported Cattle Breeds (한국 재래품종과 외래품종의 구별을 위한 초위성체 마커의 개발)

  • Three Korean native cattle (KNC) and seven exotic breeds (Chikso, Hanwoo, Jeju black, Holstein, Japanese black, Charolais, Angus, Hereford, Simmental, and Cross breed) were characterized by using five microsatellite (MS) markers (INRA30, TGLA325, UMN0803, UMN0905, and UMN0929) from the sex chromosome. Genetic diversity was evaluated across the 10 breeds by using the number of alleles per locus, allele frequency, heterozygosity, and polymorphism information content (PIC) to search for locus and/or breed specific alleles, allowing a rapid and cost-effective identification of cattle samples, avoiding mislabeling of commercial beef. It was divided into two main groups from STRUCTURE analysis, one corresponding to KNC and the other to exotic cattle breeds. These results also showed specific genetic differences between KNC and exotic breeds. Nei's standard genetic distance was calculated and used in the construction of a neighbor-joining tree. Results evidenced a correspondence between genetic distance, breeds' history, and their geographic origin, and a clear separation between KNC and exotic breeds. Overall, this study evidenced that DNA markers can discriminate between domestic and imported beef, contributing to the knowledge on cattle breeds' genetic diversity and relationships by using MS markers of the sex chromosome. These markers would be useful for inhibitory effect about false sales and for building an effective tracking system.

밀배아 분말첨가가 식빵의 품질특성에 미치는 영향

  • 식생활의 서구화로 제빵 산업이 발달하고, 빵을 주식으로 하는 인구가 늘고 있다 또한 기존의 재료보다는 기능성이 첨가된 부재료를 활용한 건강 지향적인 식품의 수요가 증가하고 있는 추세이다. 최근 밀배아의 용도 다양화를 위하여 팔은 연구가 진행되고 있으며, 상업적으로 이용되고 있다. 소맥의 제분과정에서 부산물로 얻어지는 밀배에는 식물 중에서 가장 많은 천연 비타민 E 복합체가 이상적인 상태로 함유되어 있다. 밀배아에 들어 있는 비타민 E는 천연 비타민 E 복합체와 합성 비타민 E로 구분되어진다. 천연비타민E는 8가지 이성체를 가진 자연 비타민 E를 말하며, 4종류의($\alpha$$\beta$${\gamma}$$\delta$)의 토코페롤과 4종의 토코페롤의 복합체이다. 비타민 E는 사람을 포함한 고등동물에 필수적인 영양소로 식물성기름, 곡류, 배아 등에 많이 들어 있으며, 여러 이성질체중 $\alpha$-토코페롤이 체내에서 가장 생물학적 활성이 큰 것으로 알려졌으며, 생체막조직에서 인지질의 천연항산화제 역할을 하여 체내대사를 정상으로 유지하는데 큰 기능을 하는 것으로 보고되고 있다. 우리나라에서 식용으로 소비되는 밀은 거의가 수입에 의존하고 있으며, 밀배아는 거의 전량이 가축의 사료용으로 사용되고 있고 근래에 와서는 배아유, 배아떡, 복합 조미료, 쿠키, 배아를 첨가한 국수 등에 이용되고 있다. 따라서 본 실험에서는 식빵제조시 밀배아 분말을 0, 2, 4, 6, 8, 10%씩 첨가하여 기능성 식빵을 제조하였을 때의 반죽의 물성, 호화특성을 살펴보고 빵으로 제조하였을 때의 식빵의 품질 특성으로서 1차 발효 손실률, 굽기 손실률, 부피, 질감, 노화도, 관능적 특성 및 mould-free shelf life를 측정하였다.te, ferric citrate가 유제품에 사용하기에 적합한 것으로 나타났다. 생이용성을 평가하기 위하여 우유에서 low molecular weight components(ILC)를 분리하고 철분과 복합체를 형성시킨 다음, 철분 결핍된 쥐의 소장에서 loop을 형성시켜 ILC-철분 복합체를 injection하여 철분 흡수도를 조사하였다. Ferrous lactate 100ppm에서 약 25.6%흡수되었고 ferric citrate 100ppm은 24.7%, ferrous sulfate는 19.7%흡수되었다. ILC를 첨가하지 않은 100ppm 철분염 용액은 ferrous sulfate를 제외하고는 흡수도가 감소되었다. 철분 결핍된 쥐에게 gavage 방법에 의하여 철분강화우유를 투여하였을 때 철분 25ppm 시료에서는 ferrous sulfate가 12.5%로 가장 높았고 ferrous lactate는 8.1%, ferric citrate는 6.5% 흡수되었다. 철분 100ppm수준에서는 흡수율이 낮아져 ferrous sulfate는 25ppm 시료보다 절반이하 수준이었다. Ferric citrate는 차이가 거의 없었으며 ferrous lactate는 70%수준이었다. 이상의 결과에서 철분강화우유에 사용하기 적합한 철분염은 ferrous lactate, ferric citrate였는데 특히 ferrous lactate는 제품의 이화학적 품질, 생이용성 측면 모두에서 가장 좋은 것으로 나타났다.다 높았으며, 1회당 평균 8.1$\pm$5.1개의 난포란을 회수하였다. investigation can be separated into sampling and analytical uncertainties, it can be used as a criterion where the resources for the inves

  • Printmaking is a field of fine arts and is placed on a vague boundary that is perceived as a commercial product with a popular character due to the speciality of editions. Advances of modern science and technology has developed a new technique of printmaking, and the fusion of printmaking and computer has shown the possibility of reproduction art. Reproduction printmaking has been heavily influenced by photography and extended areas, and the various forms of printing have brought about many changes and attempts by stimulating the possibilities of indirect art at various angles. As the history of printmaking and technology closely relate, the development of computer makes widespread expansion of plural artistry, technological and artistic change. A new conceptual shape can be created on the copied image simply by placing the material of the print on the smoothly flat surface expressed in digital form. The process and the result of such work show the area of unique work which is different from the value of the $\grave{a}$ la carte art or the characteristics of the material given by the print. The deprecated perception of reproducibility evaluated the value of the work in a direct sense of printmaking. It is undeniable that it is devalued by a bundle of works regardless of the value of each edition. However, the physical properties of the prints on the paper are brought up with hand drawings drawn on the canvas by hand. And it becomes an opportunity to show new aspect and change through the process of combining digital print information on paper. The diversity of media is sometimes a controversy of identity between art and technology. In the future, it should be discussed how the limit of the media which can be enjoyed in the field of art can be set as a standard.

Validation of diacylglycerol O-acyltransferase1 gene effect on milk yield using Bayesian regression (베이지안 회귀를 이용한 국내 홀스타인 젖소의 유량형질 관련 DGAT1유전자 효과 검증)

  • DGAT1(diacylglycerol O-acyltransferase1) gene is well known as a major gene of milk production in dairy cattle. This study was conducted to investigate how the DGAT1 gene effect on milk yield was appeared from the genome wide association (GWA) using high density whole genome SNP chip. The data set used in this study consisted of 353 Korean Holstein sires with 50k SNP genotypes and deregressed estimated breeding values of milk yield. After quality control 41,051 SNPs were selected and locations on chromosome were mapped using UMD 3.1. Bayesian regression of BayesB method (pi=0.99) was used to estimate the SNP effects and genomic breeding values. Percentages of variance explained by 1 Mb non-overlapping windows were calculated to detect the QTL region. As the result of this study, top 1 and 3 of 2,516 windows were seen around DGAT1 gene region and 0.51% and 0.48% of genetic variance were explained by these two windows. Although SNPs on the DGAT1 gene region are excluded in commercial 50k SNP chip, the effect of DGAT1 gene seem to be reflected on GWA by the SNPs which are in linkage disequilibrium with DGAT1 gene.

ACE Inhibitory and Antioxidative Activities of Silkworm Larvae (Bombyx mori) Hydrolysate (번데기 가수분해물의 ACE 저해활성과 항산화활성)

  • In order to utilize the silkworm larvae (Bombyx mori) protein, defatted silkworm protein was hydrolysed by four enzymes (pepsin, trypsin, neutrase and alcalase) at various hydrolysis times (6, 12, 18, 24 and 30 hr) and suspension concentrations (2, 5, 10, 15 and 20%). Protein solubility index, ACE (angiotensin converting enzyme) inhibitory activity and antioxidative activity of silkworm protein hydrolysates were investigated. The optimum condition of hydrolysis was 10% suspension concentration and 18 hr. Protein solubility index of trypsin treatment was higher than other enzyme treatments. ACE inhibitory activity and $IC_{50}$ value of antioxidative activity of neutrase treatment were 86.16% at $100\;{\mu}g/mL$ and $352.75\;{\mu}g/mL$, respectively; also, these values were higher than other enzyme treatments.

Applicable Properties of Electrolyzed Acid-Water as Cleaning Water (세정수로서의 전해산화수 적용 특성)

  • To enlarge application field of electrolyzed acid-water(EAW) on food industry, the changes of EAW properties by storage conditions and heating were investigatet. It was showed that storing EAW in closed container is mon effective to keep up the oxidation-reduction potentials(ORP), hyperchloride content and pH than stored in opened ones. ORP of EAW stored in closed container could be kept mon than 1 month as 1,150 mV levels. Ruing heating from 2$0^{\circ}C$ to 95$^{\circ}C$, ORP was increased to 1,150 mV levels at 95$^{\circ}C$ after gradual decrease to 5$0^{\circ}C$. Tyrosinase activity was decreased approximately to 26%~35% in EAW having a 950 mV~1,140 mV ORP. Also it was confirmed that EAW has anti-browning effect as sliced apple and potato, and their juices treated with EAW had conspicous difference in their $\Delta$E value. 12 kinds of pesticides such as aldrine, capful diazinon, diedrin, $\alpha$-endosulfan $\beta$-endosulfan, endosulfan sulfate, endrin, $\alpha$-BHC, o,p'-DDT, procymidone, PCNB added in EAW were recovered from ND~73.6% comparing to ones added in distilled water. The recovered amounts of pesticides, procymidone and diazinon in lettuce after soaking in EAW were 1.12 ppm and ND, compared with those of amounts soaked in distilled water were 3.67 ppm and 3.05 ppm respectively. So, it seems that EAW has potentials to promote the degradation of pesticides.

Comparison of characteristics in commercial fermented vinegars made with different ingredients (시판 발효식초의 원료에 따른 특성 비교)

  • The quality characteristics of commercial fermented vinegars made with different ingredients were compared. The pH levels of the persimmon, fig, and brewing and rice vinegars were 3.60, 3.37, and 2.62, respectively. The total acid contents of the brewing, apple, and plum vinegars ranged from 6.33 to 6.57%. The free amino acid contents were detected in the following order: brewing vinegar (521.05 mg/100 g) > fig vinegar (358.89 mg/100 g) > persimmon vinegar (353.02 mg/100 g) > rice vinegar (122.31 mg/100 g) > plum vinegar (103.52 mg/100 g). The free amino acid contents of the commercial fermented vinegars were 56.85~358.89 mg/100 g, and their gamma-aminobutyric acid (GABA) contents, 0.21~27.22 mg/100 g. In particular, the GABA content of the fig vinegar was 1.3- to 100-fold higher than those of the other vinegars. The total polyphenol compound and total flavonoid contents were detected in the following order: persimmon vinegar > fig vinegar > brewing vinegar > rice vinegar. Hence, the results of this study can provide a new alternative for making functional vinegars containing organic acid and GABA.

  • Infestations of two spotted spider mite (TSSM), Tetranychus urticae Koch, on glasshouse rose (Rosa sp.) grown by an arching method, were determined by counts of the number of TSSM per leaflet in Buyeo, Chungnam Province, for a 2-yr period. Binomial sampling plans were developed based on the relationship between mean density per leaflet (m), and proportion of leaflets infested with ( T mites (PT), according to the empirical model In (m) = a+p In (-ln (1 -PT)). T was defined as tally threshold, and set to 1, 3, 5, 7, and 9 mites per leaflet. Increasing sample size had little effects on the precision of the binomial sampling plan, regardless of tally threshold. However, the precision increased with higher tally thresholds. There was a negligible improvement in precision with T ) 7 mites per leaflet. T= 7 was chosen as the best tally threshold for estimating densities of TSSM based on the precision of the model. Independent data set was used to evaluate the model. The binomial model with T= 7 provided reliable predictions of mean densities of TSSM observed on the commercial glasshouse roses.
