Anti-oxidative properties of ginseng (인삼의 항산화 작용)

  • Clinical and animal studies have shown that free radical overload is an important cause for a variety of diseases. Although ginseng has been recognized as antioxidant, how it modulates anti-oxidative process at the molecular level remains unknown. Free radical production is induced by tumor necrosis factor-$\alpha$ (TNF-$\alpha$) under the stress condition, and (TNF-$\alpha$) release is activated by TNF-$\alpha$-converting enzyme (TACE). Since TACE inhibitor is also well known for anti-inflammatory agent, ginseng seems to show anti-oxidative activity by repressing TACE pathway. Further studies on signal transduction would be warranted to elucidate molecular action mechanisms of ginseng on anti-oxidation and anti-inflammation.

The Role of Increased Oxidative Stress in the Development of Diabetic Nephropathy (당뇨병성 신증의 발생에 있어서 산화성 스트레스의 역할)

  • The pathogenesis of diabetic nephropathy is still not completely understood while renal disease is one of the most common disabling complications of diabetes. We, in the present study, investigated the possible involvement of oxidative stress in the development of diabetic nephropathy. To hasten the development of diabetic nephropathy, streptozotocin was injected to unilaterally nephrectomized rats (NEPH-STZ). Eight weeks later, NEPH-STZ rats developed severe hyperglycemia, proteinuria, and hypertension. The kidneys of these rats showed compensatory hypertrophy and mesangial expansion. In contrast, the rats with streptozotocin injection alone (STZ) did not increase urinary protein excretion. Nephrectomized non-diabetic rats (NEPH) developed increased urine protein excretion, but without prominent renal morphological changes. However, oxidation of renal cortical tissue protein significantly increased in all 3 groups of NEPH, STZ and NEPH-STZ in comparison to control rats (CONT). The result indicates the non-specificity of the oxidative tissue damage and suggests that the oxidative damage is hardly a sole mechanism leading to the development of the diabetic nephropathy. However, it would still be a contributing factor considering that the oxidative stress is a common final pathway mediating tissue damages in chronic diabetic complications and other serious illness.

Inhibitory Effects of Panax ginseng C. A. Mayer Treated with High Temperature and High Pressure on Oxidative Stress (산화적 스트레스에 대한 고온고압처리 인삼의 억제 효과)

  • Reactive oxygen species (ROS) are produced by oxidative stresses which cause various chronic diseases such as diabetes and obesity. Ginseng (Panax ginseng C.A. Mayer) has been reported to contain various biological activities such as anti-cancer, anti-diabetic, neuroprotective, radioprotective, anti-amnestic and anti-aging effects. In this study, we investigated the effects of Panax ginseng, treated with high temperatures and high pressures, on oxidative stress in C2C12 myoblasts and 3T3-L1 adipocytes. Oxidative stress was induced in the C2C12 cells through the introduction of $H_2O_2$ (1 mM), and cells were then treated with various ginseng preparations: dried white ginseng (DG), steamed ginseng (SG) and high temperature and high pressure treated ginseng (HG). In addition, 3T3-L1 preadipocytes were treated with various ginsengs for up to 8 days following standard induction of differentiation. Our results show that HG treatment significantly protected oxidative stress in both cell lines and enhanced gene expression of antioxidant enzymes. Therefore, in this study, we investigated the protective effects of ginseng on the oxidative stress of adipocytes and muscle cells.

Physiological Roles of Bacillus subtilis thiol peroxidase gene in response to oxidative stress (산화적 스트레스에 대한 Bacillus subtilis의 thiol peroxidase 유전자의 생리적인 기능)

  • In Order to investigate the physiological role of thiol peroxidase in Bacillus subtilis, a thiol peroxidase (btpx) knock-out mutant was generated by homologous recombination. The growth of btpx knock-out mutant in aerobic condition showed a similar pattern with that of wild type of Bacillus subtilis 168/ But btpx knock-out mutant showed a retarded growht in response to oxidative stress such as $H_2O_2$, cumene hydroperoxide (CHP) treatments.

Effects of hot water extracts of roasted radish against renal oxidative stress induced by high-fat diet (고지방식사로 유도된 신장 산화스트레스를 개선하는 가압볶음 무말랭이 열수추출물 효과)

  • The antioxidant and anti-inflammatory effects of roasted dried radish (RDR) against renal oxidative stress were examined in high-fat diet (HFD)-fed mice. The HFD was prepared by adding lard to chow diet to provide 50% of the calories from fat. Hot water extracts of dried radish (DR) or RDR were administered orally to mice at 237 mg/kg bw/day, whereas distilled water was administered as a vehicle for 12 weeks. Compared to the control group, renal reactive oxygen species, peroxynitrite, and thiobarbituric acid reactive substance level in the DR or RDR group were significantly decreased, whereas the glutathione level was increased (p<0.05). Protein expressions of antioxidant factors such as nuclear factor erythroid 2-related factor-2, heme oxygenase-1, glutathione S-transferase, superoxide dismutase, catalase, and glutathione peroxidase were significantly increased in the DR and RDR groups; however, nuclear factor-kappa B expression was suppressed (p<0.05). These antioxidant and anti-inflammatory effects of RDR were found to be significantly greater than those of DR.

Enhanced drought and oxidative stress tolerance in transgenic sweetpotato expressing a codA gene (CodA 고발현 형질전환 고구마의 산화 및 건조 스트레스 내성 증가)

  • Glycine betaine (GB) is one of the compatible solutes that accumulate in the chloroplasts of certain halotolerant plants under salt or cold stress. The codA gene for choline oxidase, the enzyme that converts choline into GB, has been cloned from a soil bacterium Arthrobacter globiformis. We generated transgenic sweetpotato plants [Ipomoea batatas (L.) Lam] expressing codA gene in chloroplasts under the control of the SWPA2 promoter (referred to as SC plants) and evaluated SC plants under oxidative and drought stresses. SC plants showed enhanced tolerance to methyl viologen (MV)-mediated oxidative stress and drought stress due to induced expression of codA. At $5{\mu}M$ of MV treatment, all SC plants showed enhanced tolerance to MV-mediated oxidative stress through maintaining low ion leakage and increased GB levels compared to wild type plants. When plants were subjected to drought conditions, SC plants showed enhanced tolerance to drought stress through maintaining high relative water contents and increased codA expression compared to wild type plants. These results suggest that the SC plants generated in this study will be useful for enhanced biomass production on global marginal lands.

Effects of Open versus Laparoscopic Cholecystectomy on Oxidative Stress in Dogs (개에서 산화 스트레스 상태에 대한 일반 개복술 대 복강경 수술을 통한 담낭절제술의 영향)

  • In the present study, we investigated and compared the oxidant-antioxidant status of dogs undergoing open and laparoscopic cholecystectomy. Ten male beagle dogs, 4-6 years old, weighing 7-12 kg were used. The animals were randomly assigned to one of two groups according to the type of surgical procedure; open (group 1, n = 5) or laparoscopic cholecystectomy (group 2, n = 5). Heart rate, mean arterial pressure, end-tidal carbon dioxide, peripheral oxygen saturation, and respiratory rates were measured. Plasma total oxidant status (TOS) and total antioxidant status (TAS) levels for the evaluation of oxidative stress were measured. The ratio of the TOS to the TAS gave oxidative stress index (OSI), which is an indicator of the degree of oxidative stress. TOS, OSI and TAS levels were significantly changed after surgery in comparison to levels of before anesthesia. There were no significant different between groups. In the present study, we showed that oxidative stress parameters were found similar in dogs underwent laparoscopic or open cholecystectomy.
