Comparison of Yield and Forage Quality of Silage Corns at Different Planting Dates (파종시기에 따른 국내 육성 사일리지용 옥수수의 수량 및 사료가치의 품종간 비교)

  • This study was conducted to evaluate yield and forage quality of domestic silage corn hybrids at different planting dates. Days to silking of Suwon169 and P3394 were 62 days, respectively, the shortest among corn hybrids and Gangdaok was the longest. Cheonganok was more resistant to lodging than other hybrids. Kwangpyeongok and Gangdaok were more excellent to stay-green than other hybrids. Ear ratios to total dry matter of other domestic hybrids except Gangdaok were similar with those of DK697 and P3394. Ear ratios to total dry matter were similar in planting dates, May 3 and May 21 with 46% and 45% but June 11, low with 41%. Fresh yield of Gangdaok was the highest of all hybrids and other hybrids except Gangdaok were similar. Dry matter (DM) yields of domestic hybrids except Cheonganok were similar to those of DK697 and P3394. Total digestible nutrients (TDN) yields of domestic hybrids were no significant difference compared with those of DK697 and P3394. Fresh yield, DM yield and TDN yield of all hybrids were similar in planting dates, May 3 and May 21 but those of all hybrids were reduced in planting date, June 11 compared with May 3 and May 21. All hybrids were no significant difference in acid detergent fiber (ADF) which had values ranging from $44.2{\sim}46.2%$. All hybrids were no significant difference in neutral detergent fiber (NDF) which had values ranging from $58.1{\sim}59.9%$. There were no significant differences in ADF and NDF according to planting dates. Relative feed value (RFV) of P3394 was the lowest of all hybrids. Other hybrids except P3394 had values ranging from $84.1{\sim}85.0$ and was no significant difference in RFV. It is concluded that domestic hybrids tested in this study have high forage quality as well as high production similar to those of imported corn hybrids.

Effects of Glucose and Acrylic acid Addition on the Biosynthesis of Medium-Chain-Length Polyhydroxyalkanoates by Pseudomonas chlororaphis HS21 from Plant Oils (Pseudomonas chlororaphis HS21에 의한 식물유로부터 Medium-Chain-Length Polyhydroxyalkanoates 생합성이 미치는 포도당 및 아크릴산의 첨가 효과)

  • The characteristics of cell growth and medium-chain-length polyhydroxyalkanoate (MCL-PHA) biosynthesis of Pseudomonas chlororaphis HS21 were investigated using plant oils as the carbon substrate. The organism was efficiently capable of utilizing plant oils, such as palm oil, corn oil, and sunflower oil, as the sole carbon source for growth and MCL-PHA production. When palm oil (5 g/L) was used as the carbon source, the cell growth and MCL-PHA accumulation of this organism occurred simultaneously, and a high dry cell weight (2.4 g/L) and MCL-PHA ($40.2\;mol{\%}$ of dry cell weight) was achieved after 30 hr of batch-fermentation. The repeating unit in the MCL-PHA produced from palm oil composed of 3-hydroxyhexanoate ($7.0\;mol{\%}$), 3-hydroxyoctanoate ($45.3\;mol{\%}$), 3-hydroxydecanoate ($39.0\;mol{\%}$), 3-hydroxydodecanoate ($6.8\;mol{\%}$), and 3-hydroxytetradecanoate ($1.9\;mol{\%}$), as determined by GC/MS. Even though glucose was a carbon substrate that support cell growth but not PHA production, the conversion rate of palm oil to PHA was significantly increased when glucose was fed as a cosubstrate, suggesting that bioconversion of some functionalized carbon substrates to related polymers in P chlororaphis HS21 could be enhanced by the co-feed of good carbon substrates for cell growth. In addition, the change of compositions of repeating units in MCL-PHAs synthesized from the plant oils was markedly affected by the supplementation of acrylic acid, an inhibitor of fatty acid ${\beta}-oxidation$. The addition of acrylic acid resulted in the increase of longer chain-length repeating units, such as 3-hydroxydodecanoate and 3-hydroxytetradecanoate, in the MCL-PHAs produced. Particularly, MCI-PHAs containing high amounts of unsaturated repeating units could be produced when sunflower oil and corn oil were used as the carbon substrate. These results suggested that the alteration of PHA synthesis pathway by acrylic acid addition can offer the opportunity to design new functional MCL-PHAs and other unusual polyesters that have unique physico-chemical properties.

Geological Structure of Okcheon Metamorphic Zone in the Miwon-Boeun area, Korea (미원-보은지역에서 옥천변성대의 지질구조)

  • The Miwon-Boeun area in the central and northern part of Okcheon metamorphic zone, Korea, is composed of Okcheon Supergroup and Mesozoic Cheongju and Boeun granitoids which intruded it. The Okcheon Supergroup consists mainly of quartzite (Midongsan Formation), meta-calcareous rocks (Daehyangsan Formation, Hwajeonri Formation), meta-psammitic rocks (Unkyori Formation), meta-politic rocks (Munjuri Formation), meta-conglomeratic rocks (Hwanggangni Formation) in the study area, showing a zonal distribution of NE trend. Its' general trend is locally changed into NS to EW trend in and around high-angle fault of NS or NW trend. This study focused on deformation history of the Okcheon Supergroup, suggesting that the geological structure was formed at least by four phases of deformation. (1) The first phase of deformation occurred under ductile shear deformation of top-to-the southeast movement, forming sheath fold or A-type fold, asymmetric isoclinal fold, NW-SE trending stretching lineation. (2) The second phase of deformation took place under compression of NW-SE direction, forming subhorizontal, tight upright fold of M trend in the earlier phase, and formed semi-brittle thrust fault (Guryongsan Thrust Fault) of top-to-the southeast movement and associated snake-head fold in the later phase. (3) The third phase of deformation formed subhorizontal, open recumbent fold through gravitational or extensional collapses which might be generated from crustal thickening and gravitational instability. (4) The fourth phase of deformation formed moderately plunging, steeply inclined kink fold related to high-angle faulting, being closely connected with the local change of NE-trending regional foliation into NS to EW direction of strike in the vicinity of the high-angle fault.

Analysis of Genetic Variation in Pre-Harvest Sprouting at Different Cumulative Temperatures after Heading of Rice (벼 출수 후 적산온도에 따른 수발아 발생의 품종간 차이 및 변이 분석)

  • Pre-harvest sprouting (PHS) refers to seed germination during ripening, due to loss of dormancy before harvest. As PHS in rice causes decrease in grain yield and quality, tolerance to PHS is an important trait of Japonica cultivars in Korea. It is important to investigate the related genes and environmental factors, because PHS is a quantitative trait. In this study, we examined PHS rates at three different times according to the cumulative daily mean temperature after heading (CTAH) for 5 rice cultivars released in Korea for 5 years from 2013 to 2017 to determine the effect of environmental factors on PHS. ABA content in ripening spikelets was analyzed to understand how it was related to PHS tolerance. PHS rate increased as CTAH increased from $800^{\circ}C$ to $1200^{\circ}C$. PHS rate was significantly different (p < 0.001) among the cultivars, showing Dasanbyeo, Jounbyeo, and Nampyeongbyeo to be PHS-tolerant, and Jopyeongbyeo and Gopumbyeo to be susceptible at all the CTAH of 800, 1000, and $1200^{\circ}C$. In 2015 and 2016, PHS rates were relatively higher, because of high temperature and frequent rainfall during the ripening period. In each cultivar, ABA content decreased as CTAH increased from $800^{\circ}C$ to $1200^{\circ}C$. However, there was no significant correlation between ABA content and PHS tolerance among the cultivars.

Comparison of Forage Production and Feed Value of Winter Forage Crops in Jeju (제주지역에서 동계사료작물의 사초생산성 및 사료가치 비교)

  • This experiment was conducted to compare forage production and feed value of winter forage crops at the experimental field of Livestock Division, National Institute of Subtropical Agriculture from 2007 to 2008. The experiment was arranged in a randomized block design with three replications. The five winter forage crops used in this experiment were barely ('Youngyang' and 'Yuyeon'), oats ('Samhan' and 'Swan'), wheat ('Kumkang' and 'Woori'), triticale ('Shinyoung') and italian ryegrass ('Hwasan 101' and 'Florida-80'). The species having the fastest heading date was wheat (5th April) while the heading date of oats ('Samhan') was the latest as 28th April. The dry matter (DM) content of wheat was the highest among the five winter forage crops. Italian ryegrass ('Hwasan 101') showed the highest CP content by 11.2% and oats (Swan) showed the lowest content by 9.8%. Average acid detergent fiber (ADF) and neutral detergent fiber (NDF) content were 28.6 and 50.8% respectively. The five winter forage crops have narrow range of total nutrient digestibility (TDN) from 64.0% to 69.1%. Italian ryegrass ('Hwasan 101') showed the highest DM yield by 25,951 kg/ha, but the DM yield of barely and wheat low by 13,892 kg/ha ('Yuyeon') and 14,815 kg/ha ('Woori'), respectively. Italian ryegrass and oats have higher forage yield and feed value in Jeju. Also, oats has high grain and forage yield. The results demonstrated oats can cultivate as alternative winter forage crop for whole crop silage barely in Jeju.

Chemical and Spectroscopic Characterization of Soil Humic and Fulvic Acids and Sorption Coefficient of Phenanthrene: A Correlation Study (토양 휴믹물질의 화학적.분광학적 특성에 따른 페난트린 흡착상수와의 상관성 규명에 대한 연구)

  • In this study, the organic carbon normalized-sorption coefficients (Koc) for the binding affinity of phenanthrene (PHE) to 16 different soil humic and fulvic acids of various origins were determined by fluorescence quenching. The humic and fulvic acids used in this study were isolated from 6 different domestic soils including Mt. Hanla soil, IHSS standard soil and peat as well as Aldrich humic acid and characterized by elemental composition, ultraviolet absorption at 254 nm, composition of main structural fragments determined by CPMAS $^{13}$C NMR. The Koc values($\times$10$^4$, L/kg C) for each of HA and FA samples were in the range of 1.48$\sim$8.65 and higher in HA compared to that of FA(3.13$\sim$8.65 vs 1.48$\sim$2.48) in the experimental condition([PHE]/[HS] = 0.02$\sim$0.2(mg/L)/(mg-OC/L), pH 6). The correlation study between the structural descriptors of humic and fulvic acids and log Koc values of phenanthrene, show that the magnitude of Koc values positively correlated with the UV$_{254}$ absorptivity([ABS]$_{254}$) and two $^{13}$C NMR descriptors (C$_{Ar-H,C}$, $\sum$C$_{Ar}$/$\sum$C$_{Alk}$), while negatively correlated with the independent descriptors of the(N+O)/C atomic ratios and $^{13}$C NMR descriptors (I$_{C-O}$/I$_{C-H,C}$). These results confirmed that the binding affinity for the hydrophobic organic compound, phenanthrene are significantly influenced by the polarity and aromaticity of soil humc and fulvic acids.

Prediction of Dietary Protein-Energy Balance by Milk Urea Nitrogen and Protein Contents in Dairy Cow (젖소의 우유 중 단백질과 요소태질소 측정에 의한 사료의 에너지와 단백질 균형 상태 예측)

  • Milk urea nitrogen (MUN) and Milk protein (MP) are being used as indicators of the protein-energy balance and for actual farm feeding practices. The purpose of this study was to investigate the MUN and MP concentrations of individual cows and bulk tank milk to evaluate the dietary protein-energy balance from lactating Holstein cows. Mean MUN and MP concentrations in the milk samples obtained from 132,636 cows of 4,856 herd during Jan. 1999 to Dec. 2001 were 16.2 5.2mg/dl and 3.30 0.35%, respectively. The highest values were found during summer and lowest valued during winter in MUN. But, the average contents of MP were the highest during winter and the lowest during summer. In order to evaluate protein-energy balance for feeding, we set the level of recommended MP range of 2.90${\sim}$3.29% in early lactation considering a negative energy balance. The recommended level of MP in mid-lactation and late lactation were set as 3.10${\sim}$3.49%, and 3.30${\sim}$3.69%, respectively. Recommended MUN range of 12${\sim}$18 mg/dl was determined through the whole lactation period. Individual cows milk were analyzed by the 9 types based on this levels of MP and MUN in this study. Among the total cows investigated, 26.8%, 25.8%, and 22.2% have shown the recommended criteria of MP and MUN values, respectively. Also, of total herds surveyed, 11.6% had MUN values lower than 12.0 mg/dl and 32.9% had values higher than 18.0 mg/dl and 44.5% of total herd have not met with the recommended criteria of MP values in bulk tank milk. In case of MP, out of the total herd surveyed, 26.0% had MP values lower than 3.10% and 24.0% had values higher than 3.30% and 50.0% had MP values outside the recommended interval (3.1${\sim}$3.3%). This study has indicates that many dairy farms are under improper feeding management practice of the dietary protein-energy balance.

AICAR (5-aminoimidazole-4-carboxamide-1-β-D-ribonucleoside) Decreases Protein Synthesis in C2C12 Myotubes Cultured in High Glucose Media (근육세포 내 Glucose 농도와 AICAR에 의한 단백질 합성 저해)

  • AMP-activated protein kinase (AMPK) maintains energy homeostasis in skeletal muscle. Nonetheless, its functional role on protein synthesis with different nutrient availability has not been elucidated. Therefore, the purpose of this study is to examine the effect of AMPK activity on protein content in C2C12 myotubes incubated with low (5 mM; LG) or high (25 mM; HG) glucose media. LG stimulated (p<0.05) AMPK and acetyl CoA carboxylase (ACC) activity compare to those in HG group. Total protein content was higher in myotubes cultured with HG than in those cultured with LG and further increased by AICAR (5-amino-1-${\beta}$-D-ribofuranosyl-imidazole-4-carboxamide). Myotubes cultured with HG showed 7.5% lower UbFL (Ubiquitin Firefly Luciferase)-to-SV40 (Simian virus40) ratio compared to those in LG. Glucose level did not change the phosphorylation level of mammalian target of rapamycin (mTOR). Interestingly, administration of AICAR significantly increased phosphorylation level of mTOR in myotubes cultured with LG but not in those with HG. Overall, this data indicate that AMPK activity and protein turnover are finely regulated in response to different glucose concentration.

Processing of anchovy based powder for instant soup packed in tea bag and the taste compound of its extractives (Tea bag 포장한 멸치 복합 분말의 가공 및 그 추출물의 정미성분)

  • This study was carried out to prepare anchovy based powder for instant soup packed in tea bag which can be used handily as a extractives, and to determine the taste compounds of extractives of anchovy based powder for instant soup. The anchovy based powder for instant soup was made by adding of 72% in the redried anchovy, 14% in the sea tangle, 7% in the mushroom and 7% in the katsuobushi to the total mixtures. And the anchovy based powder for instant soup was packed in tea bag. The desirable extraction time are 5 min in package in tea bag with air permeability $100\;m^3/m^2/min$ and over 20 min in package in tea bag with air permeability $65\;m^3/m^2/min$, respectively. Judging from the result of extraction rate of total nitrogen, color and sensory evaluation in extractives of anchovy based powder for instant soup extracted by optimal extraction time, the quality in extractives of instant soup packed in tea bag with air permeability $100\;m^3/m^2/min$ was superior to those of instant soup unpacked in tea bag and of instant soup packed in tea bag of air permeability $65\;m^3/m^2/min$. The principal taste compounds of extractives of anchovy based powder for instant soup packed in tea bag with air permeability $100\;m^3/m^2/min$ were IMP (9.26 mg/100ml in 5% solution) and free amino acids (24.30 mg/100 ml in 5% solution) such as histidine, proline, lysine and taurine. Total creatinine, betaine and TMAO were seemed to act an auxiliary role in taste of extracives of anchovy based powder for instant soup packed in tea bag with air permeability $100\;m^3/m^2/min$.

Formation of biogenic amines and bioactivities of Makgeolli under different fermentation conditions (발효조건에 따른 막걸리의 Biogenic Amines(BAs)의 생성과 생리활성)

  • Generation of biogenic amines (BAs) and physiological activities of Makgeolli, Korean rice wine, were investigated during fermentation under different temperatures (20, 25, and $30^{\circ}C$) and time (3, 5, and 7 days). The pH was 3.96~4.36 during fermentation and the acidity increased proportionally with temperature and time. Alcohol and total phenolic contents peaked on day 5 at 25 by 9.0~9.8% and $3.01{\pm}0.07mg/g$, respectively. The organic acid mostly produced was lactic acid, which increased gradually with temperature and time. Formation of biogenic amines comprised of tryptamine and putrescine was observed during fermentation: 6.91, 11.70, and 15.63 mg/mL of tyramine on day 7 at 20, 25, and $30^{\circ}C$, respectively. Antioxidant activities from DPPH and FRAP assay were high in the order of $25^{\circ}C$>$20^{\circ}C$>$30^{\circ}C$ while that from ABTS assay was high regardless of temperature. Processed at 1 mg/mL and $30^{\circ}C$, the inhibitory effect on ${\alpha}$-amylase was $67.99{\pm}0.11$, $73.64{\pm}0.43$, and $75.51{\pm}0.26%$ on days 3, 5, and 7, respectively, which increased proportionally with temperature and time. Inhibitory activity on lipid accumulation in 3T3-L1 adipocytes was the highest in Makgeolli fermented on day 5 at $25^{\circ}C$. This study shows that fermentation at $25^{\circ}C$ for 5 days yields Makgeolli of the best quality with high bioactivities but no biogenic amines.
