Production System of Virus-free Apple Plants Using Heat Treatment and Shoot Tip Culture (열처리와 경정배양을 이용한 바이러스 무병 사과 생산 시스템)

  • In worldwide, viral diseases of apple plants has caused the serious problems like reduced production and malformation of fruits. Also, the damages of apple plants by virus and/or viroid infection (Apple chlorotic leaf spot virus, Apple stem grooving virus, Apple mosaic virus, and Apple scar skin viroid) were reported in Korea. However there is few report about the protection approach against the infection by apple viruses. Therefore, this paper introduced the experimental protocol for the development of virus-free apple cultivars (Danhong, Hongan, Saenara, Summerdream). Apple plants were treated at $37^{\circ}C$ for 4 weeks and shoot tips were cultured in vitro. After heat treatment, the detection of apple viruses was performed by RT-PCR using virusspecific detection primers in new apple cultivars. With the heat treatments followed by in vitro shoot tip culture, the proportion of virus-free stocks of 'Danhong', 'Hongan', 'Saenara', and 'Summerdream' was 28%, 16%, 12%, and 12%, respectively. Taken together, this approach can be a good tool for production of virus-free apple stocks.

Development of New Hybrid Cultivar 'Semil' in Zoysiagrass (한국잔디 신품종 '세밀(Semil)' 개발)

  • This study was carried out to develope new zoysiagrass cultivar Semil' (Registration no. 1749). Artificial crossing between the collected lines of typical Z. matrella, NM1 (collected in South Korea) and the different ecotype of Z. matrella, ASm (collected in South Korea) was made to develop a F1 hybrid plant (NM1 ${\times}$ ASm). Among the 30 progenies, (NM1 ${\times}$ ASm)-1 (Semil) showed superior performance in density rating. 'Semil' showed genetically light green color, with narrow leaf width ($2.7{\pm}0.12mm$), and low plant height ($10.3{\pm}3.7cm$), Length to lowest leaf blade of this cultivar was $1.8{\pm}0.15cm$, which may allow low mowing height upto 12 mm.

  • The National Forest Seed and Variety Center aims to write a manual for cultivation and characteristic inspection of new seed varieties in Korea, serving dual purposes. The first is the enhancement of the test guidelines, particularly the distinctness, uniformity and stability (DUS) test, for new seed variety, where research and examination are conducted based on the test guidelines. However, if the crop-specific test guidelines are initially reorganized, the details are unclear. Therefore, it seeks to maintain a written record to prevent confusion when the person in charge is changed. The second is to encourage the development of new varieties. By incorporating general characteristics and cultivation techniques in the manual, it seeks to encourage breeders to develop new varieties. Additionally, it serves as an important chronological record of the creation process, thereby helping future manual authors. This article introduces the current status of new plant variety protection and outlines the significance of creating a manual for cultivation and characterization of Actinidia arguta for screening new varieties.

New Columnar Apple Variety 'Tinkerbell' for Pollenizer (사과 무측지성 수분수용 신품종 '팅커벨')

  • This research was performed to breed new columnar-typed apple variety for pollinator in apple orchard. We cross-fertilized 'Hongro' that was an ovary parent and 'Maypole' a pollen parent in 2003. We gathered seeds in 2003 and sowed after breaking of dormancy in 2004. Seedlings was been growth and development till flowering. After fruit setting, We investigated characteristics of growth and fruit including mating compatibility from 2010 to 2012, then finally selected '03-2-95' named 'Tinkerbell' in 2012, registered as new variety on Korea Seed & Variety Service in 2015. The full bloom stage of 'Tinkerbell' was later 2 days then of 'Maypole'. Tree habit of 'Tinkerbell' was alike to of 'Maypole', for example weak tree vigor, columnar type. But there were many different characteristics between 'Tinkerbell' and 'Maypole'. Fruit shape of 'Tinkerbell' was ovoid whereas of 'Maypole' was ellipsoid. Fruit of 'Tinkerbell' was bigger, smoother skin, no waxy bloom than of 'Maypole'. Fruit of 'Tinkerbell' was more delicious than of 'Maypole' because of having higher soluble sugar content and lower titratable acidity. 'Tinkerbell' was excellent pollinator in apple orchard. Because of having high fruit setting rate with major apple varieties and a narrow tree width that was advantageous as pollinator in apple high density system.

'Hongan', a New Mid-Season Apple Cultivar (중생종 사과 '홍안(紅顔)' 육성)

  • A new cultivar 'Hongan' was originated from an artificial cross between 'Fuji' and 'Jonathan' carried out at National Institute of Horticultural & Herbal Science in 1993. The cultivar was preliminarily selected among the elite siblings for its high fruit quality in 2002. After regional adaptability test at five districts for subsequent four years as 'Wonkyo Ga-32', it was finally selected in 2006. 'Hongan' is characterized by the following. Optimum harvest time is late September. Shape of the mature fruit at 309 g on mean weight is globosity with a light red skin on a greenish yellow ground and yellowish white flesh. The fruit contains favorable total soluble solids at $13.7^{\circ}Brix$ and an acidity at 0.28%. Safe storage period for the fruit is 3 weeks at room temperature. It is resistant to Bitter rot. 'Hongan' also reveals a physiological cross compatibility with leading cultivars such as 'Gamhong' and 'Tsugaru' in addition to its maternal parent 'Fuji'. Tree topology is semi-spreading with vigorous growth habit.

Ecology of Marssonina Blotch Caused by Diplocarpon mali on Apple Tree in Kyungpook, Korea (사과나무 갈색무늬병의 발생생태)

  • Apple Marssonina blotch, caused by Diplocarpon mali, which has been increasing on apple trees and become one of the most serious diseases on apple trees in Korea since the begining of 1990's. In this study, ecology of Marssonina blotch including disease incidence and spore dispersals was surveyed from 1992 to 1995 in Kyungpook, and factors influencing the incidence of the disease were analyzed. Marssonina blotch began to occur on apple leaves in June and was observed commonly in most of apple orchards after August, and increased rapidly in September. The incidence of this disease was high at the year of low temperature and a lot of precipitation. The conidia discharge began to occur in May and continued to October, and the peak period of spore release was in August and usually more than 70% of total spore release of the year released from August to September. The incidence of the disease was high in the northern and mountain are as such as Yeongjoo, Chungsong, Andong, and relatively low in the southern areas such as Kunwi, Yongchon. Jonathan cultivar was the most susceptible to Marssonina blotch, and Jonagold, Sekaiichi was secondly susceptible and the next Fuji was more susceptible than Tsugaru. The incidence of the disease was relatively high in orchards which cultivation management of irrigation, drainage, air circulation, fertilization, and fungicide spraying were poor.

Breeding of a New Mid-season Apple Cultivar 'Yeohong' (중생종 사과 '여홍(麗紅)' 품종 육성)

The cultivar was preliminarily selected among the elite siblings for its high fruit quality in 2002. After regional adaptability test at five districts for subsequent four years as 'Wonkyo Ga-31', it was finally selected in 2007. 'Yeohong' is characterized by the following. Optimum harvest time is late September. Shape of mature fruit at 278 g on mean weight is oblate a light red skin on a greenish yellow ground and yellowish white flesh. The fruit contains favorable total soluble solids at $14.3^{\circ}Brix$ and titratable acidity at 0.4%, which results in a gustatory harmony between sugars and acids. Safe storage period for the fruit is 3 weeks at room temperature. It is not resistant to Bitter rot. 'Yeohong' also reveals a physiological cross compatibility with leading cultivars such as 'Hongro' and 'Tsugaru', but a physiological cross incompatibility with 'Fuji'. It has weak growth habit.

Photo-aging regulation effects of newly bred Green ball apple (신품종 그린볼 사과의 광노화인자 조절효과)

  • In this study, extracts from the Green ball apple peel (GBE) and the newly bred green ball apple from Korea showed inhibition effects on photo-aging factor regulation associated with skin aging. To investigate the inhibition effect on photo-aging factor regulation in skin, GBE was treated with UVB to induce photo-aging related factors in CCD986sk fibroblast cells. Photo-aging factor regulation effects showed that GBE inhibited UVB-stimulated matrix metalloproteinase (MMP)-1 and MMP-9 protein synthesis in collagen type I alpha 2 chain (COL1A2), MMP-1, MMP-9, and tissue inhibitors of metalloproteinase (TIMP)-1 protein expression. The expression of COL1A2 and TIMP-1 protein was significantly increased. The mRNA expression levels of COL1A2, MMP-1, MMP-9, hyaluronan synthase (HAS)2, transforming growth factor (TGF)-β, and TIMP-1 were decreased by GBE. The expression of TIMP-1 and TGF-β, which are regulators involved in matrix metalloproteinase and type I procollagen expression, was found to increase with increasing expression of COL1A2. The expression of HAS2, which is involved in the production of hyaluronic acid, one of the structural proteins constituting the skin, was also confirmed. Therefore, GBE showed excellent efficacy against photo-aging factor regulation and could be used as functional material to prevent and treat skin aging.

Effect of the Exposed Length of Dwarf Rootstock M.9 on Growth and Yield of 'Seohong', 'Summer Dream' and 'Honggeum' Apples (사과 '서홍', '썸머드림', '홍금'의 생장 및 수량에 미치는 M.9 대목 노출길이의 영향)

  • The effect of different exposed length of M.9 rootstock on growth and yield was tested in the new apple cultivars. The 'Seohong', 'Summer Dream', and 'Hongguem' grafted on M.9 rootstock were planted in March 2010. The trees were planted in such a way that the exposed length of the rootstock to be 10, 20 and 30 cm. The result showed that as decreasing exposure length, TCA of 'Seohong', 'Summer Dream', and 'Hongguem' was increased between $3^{rd}$ and $6^{th}$ year after planting. The TCA of 20 cm and 30 cm exposure length showed respectively 70% and 60% in 'Seohong', 88% and 66% in 'Summer Dream', and 55% and 41% in 'Hongguem' of the TCA with 10 cm exposure length on 6-year-old trees. Tree height, canopy width, shoot length and terminal shoot length were also increased according to decreased exposure length in 6-year-old trees. The cumulative yield of three cultivars was higher in the tree with a lower exposure length between $4^{th}$ and $6^{th}$ year after planting. The cumulative yield of 20 cm and 30 cm exposure length showed respectively 77% and 63% in 'Seohong', 85% and 76% in 'Summer Dream', and 73% and 58% in 'Hongguem' of the cumulative yield with 10 cm exposure length in 6-year-old trees. Considering tree growth and yield, the optimum exposure length of M.9 was 10 cm for 'Seohong', 'Summer Dream' and 'Hongguem'.

The cultivar was preliminarily selected among the elite siblings for its high fruit quality in 2003. After regional adaptability tests in five districts for four subsequent years as 'Wonkyo Ga-34', it was ultimately selected in 2008. Optimum harvest time is late September. Mature fruit has mean weight of 233 g and is conic with light red skin on a greenish yellow ground and yellowish white flesh. The fruit contains a favorable total soluble solids content at $13.8^{\circ}Brix$ and titratable acidity at 0.43%, which results in gustatory harmony between sugars and acids. It is not resistant to bitter rot or Marssonina blotch. 'Picnic' exhibits a physiological cross compatibility with leading cultivars such as 'Fuji', 'Hongro' and 'Tsugaru'. Tree topology is semi-spreading with a weak growth habit.
