Isolation of Symbiotic Frankia Strain from the Root Nodule of Alnus hirsuta (물오리나무의 뿌리혹으로부터 Frankia 공생균주의 분이)

  • An endophyte was isolated from the root nodule of alnus hirsuta. The isolated endophyte was identified as a Frankia sp. through morphological characteristics. Their infectivity and effectivity were confirmed by nitrogen-fixing root nodules induced on inoculated Alnus seedlings. Reisolated endophyte from the induced nodule showed identical morphological characteristics as the first isolate, showing the nodule was induced by the first isolate. Consequently, the first isolate was confirmed as a true symbiont of Almus hirsuta root nodule. The isolate was designated as a Frankia SNU 014201 strain.

Root Nodule Specific Proteins of Alnus hirsuta (물오리나무(Alnus hirsuta)의 뿌리혹 특이 단백질)

  • Root nodule specific proteins of Alnus hirsuta were examined. SDS-PAGE pattern of the Alnus root nodule was simpler than that of soybean, showing five nodule specific proteins whose molecular weights were 48, 40, 36, 26 and 19 kD, respectively. Among them, 48 kD protein existed most abundantly and were composed of two subunits whose pI value were 4.0 and 4.3, respectively. The 48 kD protein seemed to be a heme containing protein based on reaction with diaminobenzidine. Although 19 kD protein was present in small amount, it was most similar to leghemoglobin in terms of its molecular weight.

Resistance of Commercial Pepper Cultivars to Root-knot Nematodes (시판 고추품종에 대한 뿌리혹선충 저항성 검정)

  • Ninety two pepper (Capsicum annuum L.) cultivars were screened for resistance to two common species of root-knot nematodes, Meloidogyne incognita and M. arenaria. All 92 pepper cultivars were resistant to M. arenaria (0-3 eggmass/plant) but were susceptible to M. incognita (76-678 eggmass/plant). Susceptibility to M. incognita were slightly differed; 'Geomok' and 'Shintaepung' had relatively less eggmass (<100 eggmass/plant) when compared to those 29 very susceptible culivars such as 'Bulggotcheoreum' (>300 eggmass/plant). Therefore, pepper is highly recommended as a high-valued rotation crop to only those greenhouses infested with M. arenaria, but should restrict for M. incognita.

Occurrence of plant parasitic nematodes in Codonopsis lanceojata field and its damage by Meloidogyne hapia (더덕포장 선충발생 상황 및 당근뿌리혹선충에 의한 피해)

  • This study was conducted to survey plant-parasitic nematodes and their damages on Codonopsis lanceolata Trautv at 116 fields of 15 major cultivation areas in Korea. Among the 9 plant-parasitic nematode genera detected in the fields, Ditylenchus citri, Meloidogyne hapla, M. incognita, Pratylenchus neglectus, Tylenchorynchus clayton were identified. M. hapla Chitwood showing the highest field infection rate of 61.9% followed by Tylenchus spp. 16.1%, Pratylenchus neglectus 7.8%, Ditylenchus citri 5.1%, and Helicotylenchus sp. 5.1%. Average field infection rate of root-knot nematodes in 2-year-old C. lanceolata was 67.0%, and the average plant infection rate was 60.2%. The average yield decrease rate was 10%. The damage rate by the root-hot nematode increased as the cultivation year extended in Jeonbuk province, and the damage was also higher in flat land than in mountainous areas.

  • Two species of root-knot nematode, Meloidogyne incognita and M. hapla, were used in tests of soybean cultivars for resistance. Sixteen soybean cultivars were planted in the 20-cm clay pot and inoculated with each of M. incognita and M. hapla. Four soybean cultivars, Eundaedu, Baegcheon, Dongbugtae and Danyeobkong were resistant; Kwangdu, Hwanggeumkong, and Buseog were moderate, and the rest nine cultivars were susceptible to M. incognita. Jangyeobkong only was resistant, Danyeobkong, Hill, Dongbutae, Hamandaerip and Chungbugbaeg were moderate, and the rest ten cultivars were susceptible to M. hapla. There were no cultivars resistant to both nematodes although Dongbugtae and Danyeobkong were resistant to M. incognita and moderately resistant to M. hapla.

Crown Gall of Chrosanthemum Caused by Agrobacterium rubi and A. tumefaciens (Agrobacterium rubi와 A. tumefaciens에 의한 국화 뿌리혹병)

  • Crown gall on lower stem and root of chrysanthemum(Dendranthema grandiflorum Kitamura) was observed at Hwasung and Gumi in 2001 and 2004, respectively. Tumors were semi-round with rough surface texture of dark brown color. Nine isolates inducing gall formation on lower stem of chrysanthemum among twenty isolates from the tumor tissues were characterized. Their colonies were convex, glistening, circular with an entire edge and whitish or tannish cream in color on potato dextrose agar supplemented with 0.5% $CaCO_3$. The virulent isolates were rod-shaped with peritrichous flagellae, gram negative, aerobic and growing on D1M agar. Among nine virulent isolates, one isolate was identified as Agrobacterium rubi and eight isolates were A. tumefaciens based on biochemical and physiological characteristics, fatty acid profiles and substrate utilization patterns. A. tumefaciens had strong pathogenicity and broad host range compared with A. rubi. This is the first report on crown gall of chrysanthemum in Korea. To our knowledge, crown gall of chrysanthemum caused by A. rubi is first report in this study worldwide.

Effects of Root-knot Nematodes, Meloidogyne hapla, M. incognita, M. arenaria and M. javanica on Growth and Yield of Tomato (토마토에 대한 뿌리혹선충의 주요 종별 피해정도)

  • The effects of infection severity of susceptible tomato varieties, Rutgers and Boksu 2, by root-knot nematode, meloidogyne hapla, M. arenaria or M. javanica on plant growths and tomato yields were investigated. The inoculum levels of each nematode species were 0, 1,000 and 10,000 nematodes per 42cm diam. pot. Tomato yield was reduced by the nematode infection. Severity of infection was on the decreasing order of M. javanica, M. incognita, M. arenaria, M. hapla, ranging from $80\%$ by M. javanica to only $7\%$ by M. hapla. Yield reduction by infection of M. hapla was more prominent in Boksu 2 than in Rutgers. However the results were vice versa for the other nematode species, The top fresh-weight of Rutgers inoculated with 10,000 nematodes was greater than root weight, regardless of the nematode species, whereas plant height and top fresh weight decreased with increasing root weight when inoculated with inoculum density of 1,000/pot.
