Detection of Overwintering Sites Inhabited by Cherry Witches' Broom Pathogen Taphrina wiesneri with Species-specific PCR in Korea (PCR을 이용한 벚나무 빗자루병균(Taphrina wiesneri)의 월동부위 검출)

  • Taphrina wiesneri, a pathogen of cherry witches' broom, is highly pathogenic to Prunus yedoensis Matsumura which are widely planted in parks and streets in South Korea. In order to control the disease, it is crucial to know the life cycle of the fungus. We attempted to detect the fungus tentatively overwintering in shoots and branches of cherry trees both having witches' broom and healthy before flowering and leafing in spring using PCR with species-specific primer set (TwITSF and TwITSR). Genomic DNAs were extracted from the symptomatic and the asymptomatic shoots or branches. Results indicated that T. wiesneri is present in leaf buds and inner bark not only in symptomatic branches but also in the asymptomatic branches in diseased trees. However, the fungus was not detected in flower buds of the symptomatic trees and any samples of healthy trees.

Surveys on Disease Occurrence in Major Horticultural Crops in Kangwon Alpine Areas (강원도 고랭지 주요 원예작물의 병해 발생 상황)

  • These surveys were conducted to check the occurrence of disease in various horticultural crops in alpine areas, especially Daekwallyong areas, Pyongchang-Gun, and Hyeongseong-Gun in Kangwon province. TuMV on Chinese cabbage was one of the most serious diseases, especially in 1994 and 1997. The incidence of soft rot and clubroot has been increased gradually. Brittle root rot on Chinese cabbage was significantly decreased. Soft rot, gray mold, downy mildew, powdery mildew, bottom rot and Alternaria leaf spot were the common diseases on most vegetable crops. Gray mold (Botrytis cinerea) on celery, cercospora leaf spot (Cercospora sp.) on melon, powdery mildew (Erysiphe cichoracearum) on lettuce, and clubroot (Plasmodiophora brassicae) on parsley are newly found in Korea. The most common and predominant diseases were viruses, especially CMV, TMV, TuMV, BBMV, and gray mold, wilts, and cercospora leaf spot on many flowers in alpine areas. Fusarium oxysporum f. sp. eustomae causing wilting on lisianthus (Eustoma grandiflorum), Turnip mosaic virus causing mosaic and color breaking on stock, Cercospora spp. causing cercospora leaf spot on various wild lily, Cladosporum echinulatum causing leaf spot on carnation, and phytoplasma causing witches' broom on statice (Limonium sinuatum) and blazing star (Liatris spp.) were newly found during these surveys in Korea.

Detection method of Genetic Variation of Mulberry Dwarf Phytoplasma by PCR-SSCP Analysis (PCR-SSCP 분석법에 의한 뽕나무 오갈병 파이토플라스마의 유전변이 검출기법)

  • Single-strand conformation polymorphism (SSCP) analysis of MD and JWB phytopalsma isolates which amplified PCR products using the R16F2n/R2 phytoplamsa universal primer pair were compared for variations of their nucleotide sequence. The MD and JWB phytoplasmas were clearly distinct each of the band patterns from about 1.2 kb PCR products. To clearly distinct of close SSCP band patterns, the MD and JWB phytoplasma PCR products were mixed and performed to detect their polymorphism. The SSCP band patterns show all of bands of MD and JWB on single lane and easily distinct their each band patterns. The PCR-SSCP analysis was possible to detect of 1.2 kb nucleotide sequence and near close band patterns were easily distinct by mixing two samples.

Occurrence of Sesame Phyllody Disease in Korea and Detection of Its Phytoplasma (참깨 엽화병의 발생과 파이토플라스마의 검출)

  • In August 1996, phyllody disease of sesame (Sesamum indicum L.) caused by phytoplasmas was observed at Boeun, Chungbuk Province, Korea. Symptoms included extreme proliferation of growing tips and numerous small leaves, giving the infected plant a witche's-broom effect. Parts or all of the floral parts were transformed into green leaf-like structures, and little or no seeds were produced. Transmission Electron microscopy revealed the presence of phytoplasmas in the phloem sieve elements of infected plant. Since the infected sesame plants were growing near by phytoplasma infected jujubes (Zizyphus jujubu), we tried a polymerase chain reaction (PCR) technique to identify these two causal phytoplasmas. The DNA extracted from the stems of infected sesame plant was PCR-amplified using a primer set specific to 16S rRNA gene of known phytoplasmas. The amplification generated a 1.4kb band in both sesame samples and phytoplasma-infected jujubes, which also suggests the sesame plants were infected with phytoplasmas. The restriction digestion of the amplified band by four different enzymes, AluI, HaeIII, HinfI or TaqI revealed that the phytoplasmas infecting jujubes and sesame plants were of different groups.

Control of Jujube Witches'-Broom by Post-Harvest Trunk Injection of Oxytetracycline (옥시테트라싸이클린의 수확후수간주입(收穫後樹幹注入)에 의(依)한 대추나무 빗자루병(病) 방제(防除))

  • Post-harvest trunk injection of oxytetracycline-HCl(OTC) was attempted for the control of jujube witches'-broom. Diseased jujube(Zizyphus jujuba) trees with 9 to 16cm trunk diameters were treated with OTC ranged from 2g to 6g according to the size of trunk diameter. OTC dissolved in 0.51 or 1.01 of water was transfused into diseased trees by gravity flow injection during September and October. All these OTC concentrations prevented symptom development for two growing seasons and restored previously severely diseased trees to normal conditions. There was no differences in control effect between 0.51 and 1.01 OTC solutions. With these high OTC concentrations, no phytotoxicity was observed in the new leaves of the following year. Mycoplasma -like organism (MLO)-specific fluorescence was absent in the phloem of recovered tissue when examined by fluorescence microscopy using DAPI(4'-6-diamidino-2-phenylindole.2HCl) staining, indicating the disappearance of MLO by the action of OTC.
