Effects of Cross-Pretreatment of Cadmium and Arsenic on Lethality of Cadmium or Arsenic to Mice (카드뮴과 비소의 생쥐 치사독성에 대한 카드뮴과 비소의 교차전처리효과)

  • Whether the pretreatment of sublethal arsenic or cadmium may prevent from lethality of arsenic or cadmium to mice, respectively, and also the protection against to lethality of arsenic or cadmium which might be induced by pretreatment of arsenic or cadmium may be related with their hepatic glutathione contents were investigated. When sodium arsenite or cadmium chloride was subcutaneously injected to mice (ICR strain) using lethal doses, all mice of both group were killed. The mortality of mice which were subsequently injected with lethal arsenic 24 hours after pretreatment of sublethal arsenic was decreased, and the same result was obtained in the case of cadmium. Sublethal pretreatment of arsenic or cadmium prior to lethal arsenic or cadmium treatment to mice, respectively, didn't decrease hepatic glutathione contents of the survived mice, while decreases of that contents in liver were observed in the mice just after they died. Cadmium pretreatment decreased mortality of mice which subsequently injected with lethal arsenic, while arsenic pretreatment didn't protect against cadmium lethality. These results indicate that protection against arsenic or cadmium lethality to mice induced by pretreatment of sublethal arsenic or cadmium may be directly related to other factors induced by sublethal camium pretreatment, not to hepatic glutathione contents.

Trends of Arsenic Maximum Levels on Agricultural Commodities and Processed Agricultural Products (농산물 및 농산가공품 중 비소 허용기준에 관한 국내외 동향)

  • Although concerns about Arsenic (As) contamination in agricultural foods have currently increased, there in on adequate international risk management standards for As particularly on agricultural commodities and processed agricultural products. This scenario holds true also in Korea. Australia, and New Zealand has determined the As maximum level (ML) but only on cereals grains which is based on total As contents. ln addition, Japan has regulated the ML based on trivalent As contents in agricultural commodities, which do not have legal restrictions. On the other hand, China has developed a systemic risk management to restrict the As contamination above MLs in agricultural commodities and processed agricultural products based on inorganic and total As contents. The establishment of an adequate analytical method for As specification in agricultural foods is essential to determine the acceptable level of As in agricultural food. Probabilistic approach may remove some uncertainties in calculating human risk assessment from As. It should be reviewed in terms of maximum levels to set the best scenario based on a realiability and availability to achieve effective As management on agricultural foods in Korea.

The As-removal Effects of Pyrite Including Arsenopyrite after Process for Use in Medicine. (유비철석을 함유하는 황철석 약광물의 수치 후 비소 제거효과)

  • As pyrite is commonly associated with arsenopyrite, the use of pyrite including arsenopyrite for medicine requires close attention on arsenic toxicity. The toxicity was reduced by traditional processing operations include heating and quenching in vinegar. To verify the scientific effects of this process, pyrite containing many crystals of arsenopyrite was processed at temperatures from 45$0^{\circ}C$ to 85$0^{\circ}C$ and through as many as 5 processing cycles. Arsenopyrite completely disappeared when processed only once at $650^{\circ}C$ while it remained even after 5 processing cycles at 45$0^{\circ}C$. Arsenic was most abundant in medicinal mineral samples processed at 45$0^{\circ}C$ and sharply decreased when processed at $650^{\circ}C$ or 85$0^{\circ}C$ And arsenic extraction test in water was carried out from the processed pyrite medicine on the assumption that pyrite medicines with the lowest As metal content would be most desirable. Arsenic were most abundant in water extracted from medicinal mineral samples processed at 45$0^{\circ}C$ and sharply decreased when processed at $650^{\circ}C$ or 85$0^{\circ}C$. But the extracted As concentrations in water exceeded drinking water standards even when processed at 85$0^{\circ}C$. Increasing temperature promoted elimination of arsenopyrite and reduction of As in medicinal minerals and the extraction solutions. But the effects of processing cycles at the same processing temperature were not clear. Heating temperature is more important than number of processing cycles for the removal of arsenic, and it is necessary to heat pyrite to over $650^{\circ}C$ to remove it.

  • INTRODUCTION : Arsenic is a ubiquitous element present in various compounds throughout the earth's crust. The use of arsenic compounds increased greatly during the 18th and 19th centuries, including its use in pigments and dyes, as a preservative of animal hides, in glass manufacture, agricultural pesticides, and various pharmaceutical substances. The causal association between human arsenic exposure, usually in the form of inorganic compounds containing trivalent arsenite (As$^{III}$) or pentavalent arsenate (As$^V$), and various forms of human cancer has been known for many years.

The availability of tropical copepod Nitocra sp. for marine ecotoxicological evaluation (해양생태독성평가를 위한 열대 요각류 Nitocra sp.의 이용 가능성)

  • Indigenous species are needed for more accurate toxicity assessments in tropical regions. Thus, we determined not only the optimum culture conditions for stable maintenance of Nitocra sp. isolated from tropical regions but also the availability of copepods for marine ecotoxicological evaluation. Experiments on temperature, salinity, and diet as factors for optimum culture conditions as well as acute and chronic toxicity tests for ecotoxicological assessment were carried out. Data on optimum culture conditions were analyzed for statistically significant observations using one-way analysis of variance (ANOVA). Optimum temperature and salinity for Nitocra sp. were $29^{\circ}C$ and 24~39‰, and Nitocra sp. fed Tetraselmis suecica had relatively faster development and higher survival than other microalga. Under optimum culture conditions, toxicity tests were carried out. The $LC_{50}$ level and NOEC (no observed effect concentration) levels of copper and arsenic were calculated in the acute toxicity test. In the chronic test of Cu and As, developmental time and survival traits were usable endpoints for toxicity assessments. As a result, tropical copepod Nitocra sp. seems to be a potential candidate organism for marine ecotoxicological evaluation.

Studies on the Properties of lntracelluar Arsenic Binding Substances in the Rat Liver (흰쥐 간조직의 세포내 비소결합물질의 특성에 관한 연구)

  • Sodium arsenite (As) aqueous solution containing 4 ppm wss given to male rats for 15 days as drinking water. Electrophoretic pattern of liver cytosol from As-fed rats appeared to be significantly different from that of the control rats. Although the normal protein content of the cytosol fraction of As-fed rat liver was decreased, 8 stress proteins were increased. In liver cytosol fractions of As-treated rat, one kind of arsenic-binding substance (AsBS) was observed. Molecular weight of AsBS was identified to be 500 D and composition of amino acid was glycine, glutamic acid and cysteine. Glutathione (GSH) appeared to bind to arsenic and GSH-As complex showed the same mobility as AsBS on gel filtration chromatography. GSH conjugated As prevented As from inhibiting respiration, conformational change and swelling-contration of mitochondria. According to the above results. it is concluded that in vfuo treated arsenic stimulated synthesis of stress protein, and arsenic-binding substance might be glutathione and have a protective role against arsenic toxicity.

Cytotoxicity of COX-2 Inhibitor (Nimesulide) in Non-small Cell Lung Cancer Cell Line (비소세포폐암 세포주에서 COX-2억제제(Nimesulide)의 세포독성)

  • In recent years, a combination of two demographic phenomena, an increased number of older people in the population and an increase in the incidence of lung cancer with age, has made it mandatory to develop therapeutic modalities with less toxicity for the treatment of inoperable elderly patients with lung cancer. Therefore, we investigated the correlation between COX-2 expression and cytotoxicity of Nimesulide, a specific COX-2 inhibitor. Material and Method: Immunohistochemical staining of COX-2 was performed. After exposure of Nimesulide, XTT analysis, FACS analysis and Hoechst staining were carried out. Result: COX-2 protein was expressed in non-treated A549 cells strongly, but not in H1299. Cytotoxicity of Nimesulide against A549 cell and H1299 cell were similar and $IC_{50}$ of Nimesulide in both cell lines were $70.9{\mu}M$ in A549 cell line and $56.5{\mu}M$ in H1299 cell line respectively. FACS analysis showed $G_0/G_1$ arrest in both cell lines and the S phase cell fraction was decreased. Morphologic assessment of apoptosis by Hoechst 33258 staining, many apoptotic cells were detected in both cell lines. Conclusion: Selective COX-2 inhibitor, Nimesulide, can inhibit the proliferation of non-small cell lung cancer cell lines in vitro. Inhibitory effect of Nimesulide are induction of apoptosis and $G_0/G_1$ arrest. There is no correlation between COX-2 expression and cytotoxicity of Nimesulide, a specific COX-2 inhibitor. Therefore, highly selective COX-2 inhibitors such as Nimesulide can be expected to lead to even greater efficacy of their use as adjuncts to various anticancer angents and radiation therapy for the treatment of high-risk patients.

Application of the Recombinant Bioluminescence Bacterium on the Toxicity Assessment of the Sole Chemicals and Soil Samples (유전자 재조합 생물 발광 균주를 이용한 순수 오염물과 토양시료의 독성도 평가)

  • Various factors affecting on the bioassay were investigated. Experiments with a low mixture ratio (cell to toxicant solution) of 0.5 : 9.5 (v/v) produced observable bioluminescence intensity for assay. Both sodium lactate and potassium nitrate stimulate bioluminescence activity; 2.6~4.0 times of control. Distilled water and MSM, which gave non significant effects on the bioluminescence activity, were determined as proper diluent or extract solutions. A wide range of toxic responses of metals and organics were observed. In general, organics were much less sensitive than metals. Samples collected from eleven sites showed the bioluminescence activity ranging from 29 to 111% of the control. Significant correlation between toxicity and total metal contents was not observed, but the toxicity of two groups, sorted based on the contaminated arsenic concentration in soils, was 44% and 20%, showing considerable differences.

Bioassessment and Comparison of Toxicity of Arsenics based on the Results of Various Bioassays (다양한 생물 검정법에 근거한 비소의 위해성 평가 비교)

  • The acute toxicity of arsenic compounds was assessed and compared using following four bioassays; bioluminescence activity of the recombinant strain RB1436, germination of four different seeds, ${\alpha}$-glucosidase activity produced by Bacillus lichemiformis, acute genetic revertant mutation using mutant strain Salmonella typhimurium. Different sensitivities were observed among tested bioassays, but generally the toxicity by arsenite was greater than that of arsenate. Among tested four seeds, sensitivities of Lactucus and Raphanus were greater than others, and these two seed types were appeared as proper type for bioassay. High revertant mutation ratio (5.1) was observed with 1 mg/L arsenite, indicating high mutagenicity. The sensitivity of ${\alpha}$-glucosidase activity on arsenic compounds was much lower than other methods. The evaluation of interactive toxic effects using various bioassays may comprise a useful tool for the bioassessment of environmental pollutants.
