Determination and Monitoring of Grayanotoxins in Honey Using LC-MS/MS (LC-MS/MS를 이용한 벌꿀 중 grayanotoxin 분석법 연구 및 실태조사)

  • This study was performed to establish analysis methods, and evaluated for grayanotoxin in domestic/foreign honey and wild honey. The molecular weight of grayanotoxins I, II and III, excluding grayanotoxin III that has been commercialized, were analyzed by LC-MS/MS. Then, the molecular structure of grayanotoxins I and II were analyzed by NMR. A total 111 samples (25 Korean honey, 21 Korean wild honey, 13 Korean honeycomb honey, 44 foreign honey, 8 foreign wild honey) were examined to determined whether or not each sample contained grayanotoxins I, II, and III. The honey samples were mixed with methanol and loaded into a tC18 cartridge, the filtrate was diluted with water, and the mixture was then analyzed by ESI triple-quadrupole LC-MS/MS. Grayanotoxins were only found in the foreign wild honey and were not detected in Korean honey, Korean honeycomb honey, or Korean wild honey. Three of the samples contained grayanotoxin I, II, and III, and one sample contained only grayanotoxins I and III. The lowest level for grayanotoxin I was 3.13 ${\pm}$ 0.00 mg/kg, and the highest level was 12.93 ${\pm}$ 0.01 mg/kg. The levels of grayanotoxin II were 0.84 ${\pm}$ 0.01 mg/kg, 0.92 ${\pm}$ 0.00 mg/kg and 1.08 ${\pm}$ 0.01 mg/kg, respectively. The lowest level of grayanotoxin III was 0.25 ${\pm}$ 0.01 mg/kg and the highest level was 3.29 ${\pm}$ 0.74 mg/kg. Through this study, safety management for foreign wild honey has been enabled.

  • A study was conducted to compare three analytical methods for determination of hexavalent chromium in the welding fume. Precision and accuracy arc documented for colorimetric, ion chromatographic, and inductively coupled plasma-optical emission spectroscopic method. Evidence is presented that welding fume can affect the oxidation of trivalent chromium. A simple sonication extraction method, proposed in this study, instead of hot alkaline extraction has the advantage of minimizing the potential for chromium oxidation.

Hemicellulose Recovery from Rice Straw using Dilute Sulfuric Acid (묽은 황산을 사용하여 볏짚으로부터 헤미셀룰로오스 회수)

  • Rice straw was pretreated using dilute sulfuric acid at reaction conditions covering two levels of reaction temperature (140, $150^{\circ}C$) and five levels of acid concentrations ($1.0{\sim}3.0%wt$). The production and decomposition rates of major components of rice straw indicating glucose, xylose, galactose and arabinose were investigated. The production rate of arabinose and the decomposition rate of xylose were greatest among them. The maximum attainable hemicellulose (xylose+galactose+arabinose) yield was about 80%. High acid concentration appears to favor the maximum yield but high temperature does not. The optimum condition was found to be $140^{\circ}C$, 2.5% and 20 minutes. The maximum glucose yields were almost same, around $16{\sim}18%$, regardless of reaction conditions.

  • The ability of sodium-exchanged clay particles as an adsorbent for the removal of commercial dyes, Methylene blue (MB) and Malachite green oxalate (MG) from aqueous solutions has been investigated under various experimental conditions. The effect of the experimental parameters, such as pH solution, agitation time, adsorbate concentration and adsorbent dose were examined. Maximum adsorption of dyes, i.e. >90% has been achieved in aqueous solutions using 0.03 g of clay at a pH of 7 and 298 K for both dyes. The adsorption process was a fast and the equilibrium was obtained within the first 5 min. For the adsorption of both MB and MG dyes, the pseudo-second-order reaction kinetics provides the best correlation of the experimental data. The adsorption equilibrium results follow Langmuir and Dubini-Radushkevich (D-R) isotherms with high regression coefficients $R^2$ > 0.98. The mean free energies $E_a$ of adsorption from D-R model were 3.779 and 2.564 kj/mol for MB and MG respectively, which corresponds to a physisorption process.

Influence of Oxygen-/Nitrogen-containing Functional Groups on the Performance of Electrical Double-Layer Capacitor (전기이중층 커패시터의 성능에 미치는 산소/질소 함유 관능기들의 영향)

  • In this study, activated carbons (ACs) were modified as electrode materials for an electric double layer capacitor (EDLC) by controlling oxygen- and nitrogen-containing functional groups. The morphological and chemical properties of ACs were analyzed through scanning electron microscopy (SEM), fourier transform infrared (FTIR) spectrometer, automatic elemental analyzer (EA) and Boehm titration. Also, charge/discharge tests were performed to investigate the EDLC performance. Oxygen- and nitrogen-containing functional groups were introduced on the surface of ACs through acid and urea treatments, respectively. ACs with nitrogen-containing functional groups showed 2 mA increase of gravimetric discharge capacity and quick achievement of maximum charge/discharge performance. However, ACs with oxygen-containing functional groups showed low discharge capacity and its gradual decrease during further cyclic test, since the functional groups interrupted adsorption/desorption of charges in the electrolyte on the surface of ACs.

  • As the studies on effective utilization of polyunsaturated lipids in sardine (Sardinops melanosticta), the conditions of deacidification, decoloring, and deodorization for processing of refined sardine oil were investigated In the process of refining, phosphoric acid treatment was not effective in removing phosphatides, and optimal condition to neutralize the crude sardine ill was treating for 30min at $40^{\circ}C$. with $0.5\%$ excess of 3M NaOH solution. Decoloring was optimized by adding $5\%$ bleaching earth and treating for 20min at $50^{\circ}C$ under vacuum, and deodorizing was done by steam destillation at $180^{\circ}C$ under 3 torr of vacuum. When deodorizing temperature exceeded $200^{\circ}C$, some changes occured in fatty acid composition and artifacts were appeared on GLC chromatogram.

  • The aim of this study was to investigate physiological chracteristics of Lentinus edodes in Korea. We tried to detect properties of the several wood-degradative enzymes and investigate patterns of the enzyme production. A specific carbon source was used in the enzyme induction media for each enzymes, and the crude extract was used for the enzyme solution. With these enzyme solution, we investigated optimum temperature and pH conditions of their reactions. Moreover we investigated transition patterns of the enzyme production of the several wood-degrad­ative enzmes from Complex and Saw dust media for the purpose of studying the mechanisms of the wood component degradation by this fungus. It was assumed that the order of the wood com­ponent degradation was cellulose, xylan, and then pectic substances, and that the synergistic effects of these substances also influenced the degradation of wood components.

  • Porcine small intestine submucosa (SIS) has been widely used as a biomaterial without immunorejection responses. Crosslinked SIS sponges were characterized for the possibility of the bio-interactive wound dressings and tissue engineered scaffolds. SIS powders were dissolved in 3% acetic acid aqueous solution at 48hrs followed by pouring into mold and then fabricated by freeze-drying method. SIS sponge was prepared by crosslinked with 1-ethyl-(3-3-dimethyl aminopropyl) carbodiimide hydrochloride (EDC) solution (deionized water: ethanol=5:95) with 1-100mM concentration for 24 hrs and Iyophilized. SIS sponges were characterized by scanning electron microscopy, differential scanning calorimeter, and Fourier transform infrared spectrometer and were tested their porosity and water absorption ability. It was observed that the concentration of EDC might be exceeded 50 mM to get good physical characteristics. In conclusion, it seems that SIS sponge could be very useful for the applications of wound healing and tissue construction.

Thermodynamics on the Mixed Micellar Formation of Dimethyldodecylamine Oxide in Water/n-Propanol (Dimethyldodecylamine Oxide 의 물/n-프로판올 용매에서 혼합미셸 형성에 관한 열역학적 연구)

  • The pseudophase separation model is used to describe the effects of pH and n-propanol on the mixed micellar formation of protonated and unprotonated dimethyldodecylamine oxides. Dimethyl-dodecylamine oxide surfactant molecules may exist in aqueous solution in either nonionic (unprotonated) or cationic (protonated) form, and they can be modeled thermodynamically as a binary mixture of cationic and nonionic surfactants. The composition of the binary mixture is varied by adjusting the solution pH. And activities, micellar compositions, and monomeric compositions of two surfactant species can be calculated directly from the experimental titration data by applying pseudophase separation model to the micellar system of DDAO in water/n-propanol. The critical micellar concentrations and the p$K_a$ values of the binary mixture systems are dependent on the micellar composition of the protonated cationic surfactant (X); especially they show the minimum phenomena when they are plotted against the micellar composition of the protonated cationic surfactant (X). The critical micellar concentration of the binary mixed DDAO system is generally decreased when n-propanol is added to the binary mixture system, and the degree of decrease is dependent on the concentration of n-propanol.

Preparation of Polysiloxane Composite Films with Microphase-Separated Silicone Oiol by Photocrosslinking (광가교 반응에 의한 미세 상 분리된 실리콘 오일을 함유하는 폴리실록산 복합체 필름의 제조)

  • Polysiloxanes with methacrylate groups at both terminals were synthesized by a hydrosilylation reaction between allyl methacrylate and hydride-terminated polysiloxanes. The polysiloxane methacrylates with high molecular weights could be prepared through the reaction of polysiloxane methacrylates and octamethylcyclotetrasiloxane with an acid catalyst. The structures of the prepared polysiloxane methacrylates were verified by $^1$H- and $^{29}Si-NMR.$ The polysiloxane methacrylates were freely miscible with silicone oils. Polysiloxane films with microphase-separated liquid silicone oil were prepared by photo-crosslinking the mixture of polysiloxane methacrylates and silicone oil. Scanning electron microscopy (SEM) of the films showed that the size of silicone oil droplets became smaller with a lower loading of silicone oil, lower molecular weight of polysiloxane methacrylate, and lower molecular weight of silicone oil.
