Genetic Analysis of the Ability of Callus Formation and Plant Regeneration in Seed Culture of Rice (벼의 종자배양에서 캘러스 형성과 식물체 재분화 능력의 유전)

  • This study was conducted to determine the inheritance of the ability of callus formation and plant regeneration in seed cultures of rice. The culturabilities of three Japonica rices,'Chucheongbyeo', 'Nagdongbyeo', and 'Daeribbyeo 1', were higher than those of Tongil type cultivars, 'Milyang 23' and 'Samgangbyeo 'The frequency for callus growth in F₂ populations of the three crosses, 'Milyang 23/Chucheongbyeo', 'Milyang 23/Daeribbyeo 1', and' Samgangbyeo/Nagdongbyeo', revealed a nearly normal distribution. The broad-sense heritability estimated from the ability of callus formation in the crosses were ranged from 83.8% to 90.1%. The frequency distribution of plant regenerability in F₂ population of 'Milyang 23/Daeribbyeo 1' showed a continuous variation. But the segregation mode of plant regenerability from seed-derived callus in the F₂ progenies of 'Milyang 23/Chucheongbyeo' and 'Samgangbyeo/Nagdongbyeo' appeared to fit the expected 3 : 1 ratio for the high and low regenerability. These results suggest that the high plant regenerability of 'Chucheongbyeo' and 'Nagdongbyeo' was regulated by a single dominant gene.

The Effects of Adult Attachment and Self-Differentiation on Clinical Competency of Nursing Students (성인애착과 자아분화가 간호대학생의 임상수행능력에 미치는 영향)

  • This study was conducted to understand the relationship between the clinical competency of nursing students and their adult attachment and self-differentiation of nursing students. Data collection was conducted from the third and fourth grade students who had experienced clinical practice in a nursing university at Daejeon, Chungcheong province in two nursing universities at Daejeon and Chungcheong province. Finally, 169 out of initial 200 questionnaires were used for analysis. The collected data were analyzed through descriptive statistics, Pearson's correlation effectiveness, and stepwise multiple regression using the SPSS Statistics 22 Windows program. Attachment avoidance in adult attachment was significantly correlated with self-differentiation (r=-240, p=.002) and clinical competency (r=-.237, p=.002). Clinical competency was significantly correlated with attachment avoidance (r=-.237, p=.002) and self-differentiation (r=.248, p=.001). The result of stepwise multiple regression indicates that self-differentiation and attachment avoidance (F=8.707, p<.001) predicts 8.4% in clinical competency. The most powerful predictor was self-differentiation (${\beta}=.203$, p=.008), followed by attachment avoidance (${\beta}=.188$, p=.0015). Therefore, in order to improve the clinical competency of nursing college students, it is deemed necessary to develop and apply educational and counseling programs capable of increasing their level of self-differentiation while decreasing their degree of attachment avoidance.

Comparison of Neural Cell Differentiation of Human Adipose Mesenchymal Stem Cells Derived from Young and Old Ages (연령별 지방 중간엽 유래 줄기세포의 신경세포로의 분화 능력 비교)

  • Recently, adipose mesenchymal stem cells (AdMSC) that are similar to bone marrow MSC and blood derived MSC are thought to be another source for stem cell therapy. However, the diseases that can be applied for stem cells therapy are age-dependent degenerative diseases. Accordingly, the present study investigated the growth and differentiation potential to neural cells of human AdMSC (hAdMSC) obtained from aged thirty, forty and fifty. The growth of cells and cell viability were measured by passage and neural differentiation of hAdMSC was induced in neural differentiation condition for 10 days. Our results demonstrated that cell number, viability and morphology were not different from hAdMSC by age and passage. Immunofluorescence analysis of neural cell marker (TuJ1, NSE, Sox2, GFAP or MAP2) demonstrated no significant differences in neural cell differentiation by age and passage. As the number of passage was increased, the mRNA level of MAP2 and Sox2 was decreased in hAdMSC from age of 50 compared to hAdMSC from age of 30. In conclusion, the present study demonstrated that ability of neural cell differentiation of hAdMSC was maintained with ages, suggesting that autologous stem cells from aged people can be applied for stem cell therapy with age-dependent neural disease with the same stem cell quality and ability as stem cell derived from young age.

Effects of Interpersonal Relationship, Self Differentiation on College Adjustment in Nursing Students (간호대학생의 대인관계능력, 자아분화가 대학생활적응에 미치는 영향)

  • The purpose of this study was to investigate the level of interpersonal relationship, self differentiation and adjustment to college life as well as to identify the factors influencing the adjustment to college life in nursing students. The study was conducted on 185 nursing students in D and S Province and from November 15th to December 5th 2016. The data was analyzed using t-test, one-way ANOVA, Pearson's correlation and Multiple regression. The mean score of the adjustment to college life was $3.30({\pm}0.49)$ and the interpersonal relationship was $3.73({\pm}0.41)$ and self differentiation was $3.07({\pm}0.32)$. There were positive correlations between adjustment to college life and interpersonal relationship(r=.614, p<.001) and self differentiation((r=.264, p<.001). Significant factors influencing adjustment to college life were interpersonal relationship, self differentiation which explained 39% of adjustment to college life. Based on these findings, programs that develop interpersonal relationship and self differentiation are necessary for college nursing students in order to promote college adjustment.

Varietal Difference and Inheritance of Plant Regenerability in Anther Culture of Rice (벼 악배양 효율의 품종간 차이와 식물체 재분화 능력의 유전)

  • This study was carried out to identify varietal difference and the inheritance of the ability of plant regeneration in anther culture of rice. The anthers of 33 Japonica, 11 Indica/Japonica, and 3 Indica rice cultivars were culture on $N_6-Y$_1$$ medium with 1 mg/L NAA and 2 mg/L kinetin. The remarkable variability in plant regeneration were observed among the genotypes, ranging from 0.0% to 19.4% Most of Japonica rices were resulted to have better culturability than that of Indica/Japonica and Indica type cultivars. Newly developed Japonica rices, 'Ilmibyeo' and 'Hangnambyeo' showed to have regenerability with the frequency of 19.4% and 18.1% respectively. The segregation mode for callus formation and plant regenerationin anther culture of F$_2$ population of 'Milyang 23/Chucheongbyeo' showed a continuous variation. The variation of plant regeneration frequency in anther culture of F$_2$ plants ranged from 0% to 33.3% with a mean of 6.3% The broad-sense heritability estimated from the ability of plant regeneration was considerably high (82.7%).

Factors Affecting Plant Regeneration from Seed-Derived Calli in Italian ryegrass (Lolium multiflorum Lam.) (이탈리안 라이그래스의 종자배양에 있어서 식물체 재분화에 미치는 몇 가지 요인)

  • In order to optimize the conditions of callus induction and plant regeneration in seed culture of Italian ryegrass, the effects of basic medium and carbon sources on seed culturability and genotypic difference of plant regenerability were investigated. MS medium was better than N6 and B5 medium in enhancing callus growth and plant regeneration. Sucrose was superior to maltose in plant regeneration as carbon source in the medium. The genotypic difference in plant regenerability was obvious among four cultivars of Italian ryegrass tested. 'Rio' and 'Jeanne' showed to have higher regenerability with the frequency of 38% and 56%, respectively.

Effect of Gallus gallus var. domesticus (Yeonsan ogolgye) Extracts on Osteoblast Differentiation and Osteoclast Formation (연산 오골계 물 추출물이 조골세포와 파골세포의 활성에 미치는 영향)

  • The effects of water extracts of Gallus gallus var. domesticus (Yeonsan ogolgye, GD) on the activities of osteoblast differentiation and the restraint of osteoclast formation were investigated. The water extract of GD in the human osteoblast "MG-63" cell, was examined in relation to alkaline phosphatase (ALP) activity and alizarin red stains. In order to observe the effects of osteoclasts formation, we analyzed RAW 264.7 cell tartrate-resistant acid phosphatase (TRAP) activity and TRAP stains. The ALP activity of the water extract of hen and cock flesh (3 years) were 133.8% and 129.6%, respectively. The ALP activity of flesh extracts was also higher than that of the skin extracts. Concerning the effects of age, the 3 years old flesh extracts had a higher activity than that of the one year old extracts. However the activity of the 3 years old skin extracts was lower than that of the one year old extracts. For gender conditions, the ALP activity of the hen extract was higher than that of the cock. The degree bone mineralization in the three years old hen flesh exhibited the highest rate, at 124.3%, amongst all the groups. The TRAP activity of the flesh extracts of the three years old cock revealed the lowest rate, at 31.8%, compared to the control. Our results demonstrate that the water extract of GD increases bone mineralization and osteoblast differentiation activity in MG-63 cells and enhances the inhibitory activity of bone-resorption in RAW 264.7 cells. In conclusion, the water extracts of GD seem to be effective in the prevention and treatment of bone related disorders.

Efficient Callus Induction and Plant Regeneration of Guineagrass (Panicum maximum Jacq.) (난지형 목초 기니아그라스의 효율적인 캘러스 유도 및 식물체 재분화)

  • Guineagrass (Panicum maximum Jacq.) is an important warm-season forage grass as well as biomass crop. It has both sexual and asexual mode of reproduction (apomictic) depending on cultivar. We developed efficient plant regeneration system for an apomictic (cv. Natsukaze) and a non-apomictic (Noh-PL1) guineagrass by optimizing the level of L-proline in the callus induction and that of $AgNO_3$ in plant regeneration medium. Among the L-proline concentrations tested, the best callus induction was achieved by using 2g/L L-proline in both the genotypes. Immature embryos proved to be the best explant source for tissue culture of guineagrass. The highest frequency of shoot regeneration was obtained on MS plant regeneration medium supplemented with 2 mg/L $AgNO_3$. These results provide a foundation for efficient tissue culture and genetic improvement of guineagrass.

Synergic Effect on Adipocytic Differentiation by a Combination of Thyroxine andGlucocorticoid in A549 Cells (A549 폐암세포주의 지방세포 분화에 미치는 티록신 및 당질 코르티코이드 호르몬의 상승 효과)

  • The present study examined the rate of cell growth and differentiation potential into adipocytes in A549 lung adenocarcinoma cells exposed to each adipogenic medium containing glucose metabolism hormones, such as thyroxine (T4) thyroid hormone and glucocorticoid (GC) adrenal steroid hormone, as well as pioglitazone (PGZ), a PPARγ agonist. Following each adipogenic treatment for 2 weeks, the rate of cell growth was significantly (p<0.05) inhibited, and the level of telomerase activity was significantly (p<0.05) decreased in the PGZ-based adipogenic medium containing both T4 and GC hormone compared with those containing each T4 or GC hormone. Moreover, the adiposome-like vesicles were highly reacted with Oil-Red O staining solution, and the levels of transcripts expressed in the differentiating adipocytes for adipogenesis, including adinopectin, leptin, and resistin, were significantly (p<0.05) increased in the PGZ-based adipogenic medium containing both T4 and GC hormone compared with those of the adipogenic medium containing each T4 or GC hormone, implying that adipocytic differentiation has fully occurred in the A549 cancer cells. Based on present observations, the PGZ-based adipogenic medium containing both T4 and GC efficiently induces inhibition of cell growth and cellular differentiation into adipocytes in A549 cancer cells rather than in the adipogenic medium containing only T4 or GC hormone. Adipogenic treatment could provide potential probability in cancer chemotherapy.

Effect of Copper on the Plant Regeneration from Seed Derived Callus of Orchardgrass (Dactylis glomerata L.) (오차드그래스의 종자유래의 캘러스로부터 식물체 재분화율에 미치는 Copper의 영향)

  • This study was conducted to investigate the effects of copper in the efficiencies of callus formation and plant regeneration of orchardgrass (Dactylis glomarata L.). Seeds were cultured on MS medium containing $2\;mg/{\ell}$ 2,4-D and different concentrations ($0.1-100\;{\mu}mol$) of copper sulfate. Plant regeneration was achieved on N6 medium containing $1\;mg/{\ell}$ NAA, $5\;mg/{\ell}$ kinetin and $0.1-100\;{\mu}mol$ of copper sulfate. Callus formation was not affected by copper incorporation into MS medium. However, the efficiency of plant regeneration was promoted by copper and the maximum efficiency was obtained when $70\;{\mu}mol$ copper was incorporated in the culture medium. The average number of regenerated plants from the seed-derived callus was also increased by copper.
