Genetic Analysis of the Ability of Callus Formation and Plant Regeneration in Seed Culture of Rice (벼의 종자배양에서 캘러스 형성과 식물체 재분화 능력의 유전)

  • This study was conducted to determine the inheritance of the ability of callus formation and plant regeneration in seed cultures of rice. The culturabilities of three Japonica rices,'Chucheongbyeo', 'Nagdongbyeo', and 'Daeribbyeo 1', were higher than those of Tongil type cultivars, 'Milyang 23' and 'Samgangbyeo 'The frequency for callus growth in F₂ populations of the three crosses, 'Milyang 23/Chucheongbyeo', 'Milyang 23/Daeribbyeo 1', and' Samgangbyeo/Nagdongbyeo', revealed a nearly normal distribution. The broad-sense heritability estimated from the ability of callus formation in the crosses were ranged from 83.8% to 90.1%. The frequency distribution of plant regenerability in F₂ population of 'Milyang 23/Daeribbyeo 1' showed a continuous variation. But the segregation mode of plant regenerability from seed-derived callus in the F₂ progenies of 'Milyang 23/Chucheongbyeo' and 'Samgangbyeo/Nagdongbyeo' appeared to fit the expected 3 : 1 ratio for the high and low regenerability. These results suggest that the high plant regenerability of 'Chucheongbyeo' and 'Nagdongbyeo' was regulated by a single dominant gene.

Expression of Organogenesis-related Genes and Analysis of Genetic Stability by ISSR Markers of Regenerants Derived from the Process of in vitro Organogenesis in Japanese Blood Grass (Imperata cylindrica 'Rubra') (기내배양 홍띠 단계별 재분화체의 기관분화 관련 유전자 발현과 ISSR에 기반한 유전적 안정성 분석)

  • The in vitro organogenesis is one of important issues in plant embryology, and somaclonal variations are existing in calli and/or regenerants induced from a process of the organogenesis with in vitro circumstances. In this study, expressions of organogenesis-related genes were evaluated and genetic stability of regenerants derived from the process of in vitro organogenesis were measured using ISSR markers in Imperata cylindrica 'Rubra', Poaceae. The expressions of organogenesis-related genes were detected all of regenerants at the process of the organogenesis. All ISSR markers produced with an average of 71 bands per in vitro-cultured regenerants, and the scorable bands were varied from two to eight with an average of 5.14 bands per a primer. The polymorphism rates of the in vitro regenerants were higher than that of mother plants (1.4%), showing 4.1% (pot-cultured regenerants), 4.3% (field-cultured regenerants), 4.2% (in vitro-cultured regenerants), 5.6% (calli with green shoots) and 1.4% (calli), respectively. The genetic similarity matrix (GSM) among all accessions ranged from 0.747 to 1.0 with a mean of 0.868. GSM of the regenerants showed differences (from 0.972 to 1.00) compared with that of mother plants (0.991). According to the clustering analysis, two independent groups were divided into; the one is mother plants and regenerants cultured at room and open field, the other is regenerants cultured in vitro. The results give a new insight for understanding the dynamics of organogenesis in monocot plant.

Plant Regeneration from Hypocotyl-Derived Protoplasts of Brassica oleracea var. capitata (양배추 배축 원형질체로부터 식물체 재분화)

  • Protoplasts were isolated from hypocotyl tissues of 5-day-old Brassica oleracea var capitata Green Challenger seedlings. Several media were used for protoplast culture and shoot regeneration. The shoot-regeneration rapacity of protoplast derived callus depended on the initial culture medium. Protoplasts were cultured in liquid medium (B5 medium supplemented with CaCl2, 2H2O 600mg/L, g1ucose 20g/L, D-mannito1 70g/L, NAA lmg/L, BA lmg/L, 2.4-D 0.25 mg/L)at 27$^{\circ}C$ under the dark After 5 to 10 days, cultlues were diluted with medium with a reduced osmotic stabilizer and then transferred to illuminated conditions. The culture medium was changed with the fresh medium at 7- to 10-day-intervals until the formation of microcallus. Hypocotyl protoplast-derived callus proliferated when transferred to MS medium supplemented with NAA lmg/L, BA 1mg/L and GA$_3$ 0.02mg/L. Upon transfer to MS basal medium without growth regulators, roots were produced. In an attempt to increase the regeneration frequency, 10g/L polyvinylpyrrolidone was added to the regeneration medium, but the shoot regeneration was mot improved. The regenerated whole plants were acclimated in a sterized soilless mixture(vermiculite 2;perlite 2;peat moss1) in a culture room.

Effect of Plant Growth Regulators and Medium Supplements on Plant Regeneration from Stolon of Zoysia matrella Merr (금잔디 포복경으로부터 식물체 재분화에 있어서 식물생장조절물질과 배지첨가물질의 영향)

  • To optimize tissue culture conditions for genetic transformation of Zoysia matrella (L.) Merr., we investigated the effects of different plant growth regulators and medium supplements on plant regeneration using stolon explants excised from mature plant grown in a green house. Plant regeneration frequency was 33.3% when stolon tissues with a node were cultured on the regeneration medium supplemented with 0.5 mg/L 2,4-D and 1 mg/L of kinetin. Comparing the basal media tested, MS medium showed higher plant regeneration performance than N6 or SH medium. Addition of 5 mg/L $AgNO_3$ with 10 mg/L cysteine improved frequency of plant regeneration up to 40%. Among different carbon sources, 3% sucrose was found to show the best for regeneration frequency. This rapid and efficient plant regeneration system would be useful for using genetic transformation experiments of manilagrass without intervening callus-mediated regeneration.

  • To establish the system of in uitro plant regeneration, the leaf segments of Sedum sarmentosum were cultured on MS media supplemented with different levels of 2,4-D, NAA and BA. The callus induction and growth showed a good response on MS medium supplemented with 3.0mg/L 2,4-D and 1.0mg/L BA, but a few callus induced on medium containing NAA and BA. In plant regeneration, combination of BA and NAA promoted shoot organogenesis from callus, and the highest frequency was obtained on MS medium supplemented with 0.2mg/L NAA and 3.0mg/L BA. When calli were transferred to the plant regeneration medium containing 0.2mg/L NAA and 3.0mg/L BA, healthy shoots without hyperhydricity were continuously induced (17.2 plantlets per callus) after 50 days of culture. When regenerated plantlets were transferred onto hormone-free MS medium, rooting was easily achieved from all of them.

  • The experiment was conducted to investigate optimal condition for callus induction and plant regeneration for transformation system of gerbera. Callus induction was more effective in 'white day' then other two cultivar 'Songsongee' and 'Love Song' The optimized plant growth regulators concentration on callus induction, was MS basal medium with NAA 0.1 mg/L+ TDZ 0.5 mg/L. The optimized plant growth regulators concentration on plant regeneration, which was used MS basal medium was IAA 1.0 mg/L + BA 1.0 mg/L + Zeatin 0.1 mg/L. The optimized petiole age for more effective plant regeneration was 32 days petiole after in vitro subculture and MS basal medium strength was 1/2 MS strength.

Efficient Fertile Plant Regeneration from Protoplasts of Javanica Rice and Their Ploidy Determination by Flow Cytometry (Javanica 벼 원형질체로 부터 효율적인 식물체 재분화와 flow cytometry에 의한 ploidy 검정)

  • The Southeast Asian javanica rice variety Tinawen was investigated for efficient protoplast culture and plant regeneration from cell suspension-derived protoplasts using a feeder cell culture method. Feeder cells of both Lolium multiflorum and Oryza ridleyi, either alone, or in combination, were employed and plants were regenerated from protoplast-derived colonies on several plant regeneration media. Dehydration of protoplast-derived colonies was also investigated as a means of enhancing plant regeneration. In the presence of L. multiflorum or O. ridleyi feeder cells, the protoplast plating efficiency ranged from 0.09% to 1.48%, depending on the feeder cell type and the age of the cell suspension. L. multiflorum feeder cells induced approximately 6-fold higher plating efficiency compared with those of O. ridleyi. The plant regeneration frequencies were 19.3-31.7% with L. multiflorum, 13.0-18.0% with O. ridleyi and 18.0-22.0% with a mixture of both in various plant regeneration media when protoplast-derived colonies were dehydrated, while for the non-dehydrated colonies, the values were 2.0-7.0%, 3.0-5.0% and 0-4.0%, respectively. Flow cytometric analysis of 34 protoplast-derived plants showed that the majority of plants were diploids and only 2 plants were tetraploids. The plants which were transferred to glasshouse were fertile.

Effects of Media and its Components on Callus Induction and Plant Differentiation in Rice Anther Culture (배 약배양에서 배지조성이 Callus 및 식물체분화에 미치는 영향)

  • Effects of media and its components on callus induction and plant regeneration were studied to increase the cultural efficiency in rice anther culture. The N$_{6}$ basic medium gave better results in callus induction than those of MS or Miller. The medium used for callus induction affects the plant regeneration. The frequency of plant regeneration from callus grown on Miller basic medium was lower than those of N$_{6}$ or MS. Most of calli derived from anthers, above 90%, were induced from 20 days to 40 days after anther inoculation. The cultural efficiency of modified N$_{6}$ basic medium which was composed of 31.5mM KNO$_3$ and 1.75mM(NH$_4$)$_2$SO$_4$ as nitrogen sources was higher than those of N$_{6}$ basic medium. Combination of NAA and Kinetin showed better results than that of 2, 4-D only in cultural efficiency. Effect of DL-alanine on callus induction in Indica variety, IR40, showed better response in the anthers pretreated for 6 days at 12$^{\circ}C$.
